Local Cervical Immunity in Women with Low-grade Squamous Intraepithelial Lesions and Immune Responses After Abrasion

  • Minor trauma to the uterine cervix is supposed to induce local immunity to prevent cervical lesions caused by human papillomavirus (HPV) infection. This study aimed to investigate the local cervical immunity in women with low grade squamous intraepithelial lesion (LSIL) and effects of abrasion after cryosurgery or Pap smear. One hundred women with LSIL and known results of HPV detection were recruited. HPV positive women were randomly divided according to abrasion into cryotherapy and Pap smear observation groups. Cervical tissues and cervico-vaginal lavage (CVL) were collected at 6 and 12 months after allocation. The levels of cytokines at first recruitment were compared with cytokine levels at 6 months after abrasions. The mRNA of IFN-${\gamma}$, TNF-${\alpha}$ and IL-10 in cervical tissues and these cytokines secreted in CVL were determined using real time PCR and ELISA, respectively. Anti-HPV16 IgG and IgA antibodies in CVL were assessed by western blotting. At first recruitment of women with LSIL (100 cases), IL-10 mRNA and cytokine in HPV positive group (60 cases) was significantly higher than negative group (40 cases). IFN-${\gamma}$ and TNF-${\alpha}$ mRNA level in both groups were comparable but their secretions in CVL were significantly increased in HPV negative group. After abrasion for 6 months in HPV-positive women, all mRNA and secreted cytokines were changed, but no significant difference was observed between cryotherapy and observation groups. When individuals were compared between first recruitment and after abrasion for 6 months, IFN-${\gamma}$ mRNA and anti-HPV16 L1 IgA antibodies were significantly increased in the cryotherapy group. The results suggest that modulation of local cervical immunities by abrasion might promote different effects in clearance of HPV-related cytological abnormalities.

제주도 서부 해역의 저염수층을 고려한 수중통신 성능 (Performance of Underwater Communication in Low Salinity Layer at the Western Sea of Jeju)

  • 해양에서의 음속은 수온, 염분, 압력에 의한 실험식으로 계산되며 해양에서의 평균 염분은 약 34 psu (practical salinity unit)로 수성이나 수평 거리에 따른 변화가 대부분 수 psu 이하이기 때문에 음속에 크게 영향을 마치지 못한다. 그러나 최근 여름철에 중국 양쯔강 범람에 의해서 24 psu 정도의 저염수가 제주 서부 해역으로 유입되는 사례가 발생하고 있으며 이 저염수는 음속에 영향을 미친다. 본 논문에서는 이러한 환경 변화가 수중통신에 미치는 영향을 분석하였다. 즉, 저염수로 인한 음속구조의 변화를 계산하였고, 저염수층 내에서 송수선 수심과 전달거리를 바꿔가며 음파 전달 경로를 모의하여 통신 채널을 추정하였으며, BPSK(Binary phase shift key) 변조방식을 이용하여 비트 오류율을 계산하였다. 동일한 실험 조건하에 저염수가 없는 경우의 성능을 비교하여, 저염수가 통신 성능에 어떠한 영향을 미치는가에 대해 분석하였다. 저염수는 수심 약 20m까지의 표층부에서 음속의 기울기를 양의 기울기로 변화시켜 음과 채널을 형성하였고, 표층부에서 대부분의 송수신 신호의 비트 오류율을 감소시키는 경향을 확인하였다. 본 논문의 저염수에 의한 수중 통신 성능에 미치는 영향을 분석한 결과는 정확한 해양 통신 및 탐지 성능분석을 하기 위해서는 해양환경의 변화를 고려하는 것이 매우 중요하다는 것을 시사한다.

  • Taurine (2-ethaneaminosulfonic acid) is one of the major intracellular ${\beta}$ -amino acids in mammals and is required for a number of biological processes including membrane stabilization, osmoregulation, antioxidation, detoxification, modulation of calcium flux and neurornodulation. The taurine transporter (TAUT) which contains 12 hydrophobic membrane-spanning domains has been cloned from dog kidney, rat brain, mouse brain, human thyroid, placenta and retina. In this study, The TAUT cDNA from the human intestinal epithelial cell, HT-29 was cloned and sequenced. Reverse-transcription polymerase chain reaction (RT-PCR) was performed to amplify partial cDNA encoding human intestinal TAUT. The coding region of the PCR product was 732 bp long. The primers were designed to encode highly conserved amino acid sequences near the transmembrane domains III (IPYFIFLF) and Ⅵ (KYKYNSYR) both in human and mouse. The TAUT cDNA amplified was ligated into the pGEX 4T-1 expression vector. The resulting sequence of human intestinal TAUT cDNA (Accession number of NCBI Genebank is AF346763) was identical to the sequences of the TAUTs previously determined in the human placenta and retina except 3 base pairs from that of the reported human thyroid. TAUT specific antibodies were generated to use them as biological tools in the studies of the biological role of TAUT. Peptides of 149-162 amino acid residue (14 amino acids) of the TAUT were synthesized. The synthetic peptide used in this study was LFQSFQKELPWAHC. This region was chosen not only to avoid putative glycosylation sites but also to exclude regions of known homology with GABA transporters in the extracellular hydrophilic domains. The synthetic peptide, TAUT-1 was conjugated with carrier protein, kehole lympet hemocyanin (KLH) to use as an antigen. When used for immunization on a rabbit to produce polyclonal antiserum, the conjugates elicited high -titered specific anti-TAUT-1 antibodies, which reacted well with the ovalbumin (OVA) conjugated peptides in ELISA. The KLH-conjugated peptide was also used as immunizing antigen in BALB/c mice to produce TAUT specific monoclonal antibodies. From the culture supernatant of the hybridoma, the specificity of anti-TAUT-1 monoclonal antibodies was confirmed by ELISA. Further applications of more tools in TAUT expression analysis will be performed such as western blotting and flow cytometry.

Pd nanoparticles on poly(amidoamine) dendrimers modified single-walled carbon nanotubes as highly sensitive hydrogen gas sensors

  • In order to overcome the lack of reactivity with hydrogen gas ($H_2$) and utilize unique properties of Carbon Nano Tubes (CNTs) for the application to hydrogen sensors, there have been intensive works on the surface functionalization of CNTs with various types of nanoparticles including Pd. In the present work, we have investigated the effect of dendrimers and Pd nanoparticles to the hydrogen sensing properties of CNTs by comparing three types of samples: Pd/SWNTs (Sample I), Pd/dendrimer/SWNTs (Sample II) and heat-treated Pd/dendrimers/SWNTs (Sample III). As a result of IV measurement under the $H_2$ and air, sample I was found to have a high sensitivity (25%) to $H_2$, but to have a very slow response time (324 s) and recovery rate. On the other hand, Sample II was found to show much faster response time (3 s) and good recovery rate but lower sensitivity (8.6%) than Sample I which is due to induced dipole moments in the dendrimers. Interestingly, Sample III showed both fast response time (7 s) and high sensitivity (25%), indicating that the pyrolysis of the dendrimers during heat treatment which reduce the distance between the surface of the SWNTs and the functionalized Pd nanoparticles plays a key role in improving the sensitivity. The pyrolysis of the dendrimers in Pd nanoparticle-dendrimer-SWNTs was found to enable a significant electrical conductance modulation upon exposure to extremely low concentrations (10 ppm) of $H_2$ in air. Our results demonstrate that the Pd Nanoparticle-Grafted Single-Walled Carbon Nanotubes(SWNTs) with Dendrimers can be used to detect hydrogen, makingoutstanding properties such as fast response, and recovery time, high sensitivity, low detection limit at room temperature compared with other types of hydrogen sensors.

스마트 홈 헤이 홈 Air의 클라우드 아티팩트 원격 수집 방안 연구 (A Study on the remote acuisition of HejHome Air Cloud artifacts)

  • IoT(Internet of Things) 디바이스의 사용이 확대됨에 따라 경찰청의 디지털 포렌식 적용 범위가 스마트 홈 영역으로 확대되었다. 이에 따라 스마트 홈 플랫폼 데이터를 수집하기 위해 진행된 기존 연구들은 대부분 모바일 기기의 로컬 데이터 분석과 네트워크 관점의 분석 등의 연구가 주로 수행되었다. 하지만 증거 분석을 위해 유의미한 데이터는 스마트 홈 플랫폼의 클라우드 스토리지에 주로 저장되어있다. 따라서 본 논문에서는 사용자가 헤이 홈 앱 기반의 "헤이 홈 스퀘어" 서비스를 이용할 때 PC에 기록되는 Microsoft Edge, Google Chrome, Mozilia Firefox, Opera와 같은 웹 브라우저들의 쿠키 데이터베이스를 통해 사용자 계정의 accessToken을 획득하여 헤이 홈 Air 환경에서 클라우드에 저장된 데이터의 수집 방안을 연구했다. 데이터는 헤이 홈의 모회사가 제공하는 OpenAPI를 활용해 클라우드로 직접 접근하여 수집하였다. 본 논문에서는 스마트 온·습도 센서, 스마트 도어 센서, 스마트 모션 센서로 환경을 구성하여 실험를 수행했고 날짜 및 장소별 온·습도 데이터, 사용한 디바이스 리스트, 방 내 모션 감지 기록 등의 아티팩트를 수집할 수 있는 것을 확인하였다. 이와 같은 아티팩트 분석 결과를 통해 알 수 있는 사건 당시의 온·습도 등의 정보는 포렌식 수사 과정에서 단서로 활용될 수 있다. 또한 본 논문에서 제안한 OpenAPI를 활용한 클라우드 데이터 수집 방안은 데이터 수집 과정에서 발생할 수 있는 변조 가능성을 배제하고, API를 이용해 결과를 호출하기 때문에 디지털 포렌식의 원칙인 무결성의 원칙과 재현성의 원칙을 따른다.

분자핵의학 기법을 이용한 다약제내성 진단 (Detection of Multidrug Resistance Using Molecular Nuclear Technique)

  • 다약제내성이 발현된 암세포에서 세포내의 항암제를 세포외로 배출시키는 기전을 체내에서 비침습적인 방법으로 영상화 할 수 있는 SPECT와 PET는 악성종양의 진단과 평가에 중요한 역할을 할 것으로 판단되나, 아직까지도 Pgp와 MRP의 운반능을 적절하게 평가하는 핵의학적 영상방법을 정립하는데는 극복해야할 문제점들이 많다. 지금까지의 MDR영상에 관한 연구들은 대부분이 $^{99m}Tc$-표지 방사성의약품을 이용한 연구였으나, PET의 임상 응용이 증가함에 따라 보다 특이적이고 쉽게 응용될 수 있는 PET용 방사성 추적자의 개발도 이루어져야 할 것이다. $^{99m}Tc$-MIBI의 암 세포내 일방향(unidirectional) 섭취는 음성인 세포막 전하와 세포내 소립체 기질 전하에 의하여 결정되므로, MIBI의 섭취는 다른 지용성 양전하를 띤 막전위 추적자들과 유사하게 작용한다. $^{99m}Tc$-표지 방사성의약품은 암조직의 혈류 증가나 소립체 용적이나 활성도가 증가하면 섭취가 증가할 수 있어 보다 특이적인 MDR추적제의 개발이 필요한 것이다. 최근 Lorke 등은 약제감수성 및 내성 인체대장암세포인 $HT-29^{par}$ 세포와 $HT-29^{mdrl}$ 세포를 이용한 연구에서, 두세포 모두에서 $^{18}F$-FDG의 섭취가 있었고, MDR이 발현된 세포와 종양에서 $^{18}F$-FDG 섭취가 훨씬 낮았고, MIBI는 MDR이 없는 모세포에서도 매우 낮았음을 보고한 바 있다. 이 세포는 전자현미경검사에서 사립체가 풍부하지 않은 세포였다. 그러므로 이러한 결과로 보아 $^{99m}Tc$-MIBI 영상에서 종양이 보이지 않거나 섭취가 미약하다고 해도 MDR이 발현되었다고 단정할 수는 없게 된다. 즉 MDR의 발현유무를 정확하게 감별할 수 있기 위하여는 저항이 없는 세포에 MIBI가 충분하게 섭취되어야 한다는 것이 필수적인 요건이며, 종양세포 종류에 따라서는 FDG가 MDR의 marker가 될 수 있다는 것이다. Pgp 수송체는 ATP의존성 약제배출 펌프이므로 MDR세포는 에너지가 많이 필요하여, MDR 세포는 당분해율(rate of glycolysis)이 증가되어 있고 HT-29 mdrl 종양세포에서는 포도당 이동과정의 변화로 FDG 섭취가 감소되었다. 또한 Pgp가 점차 증가됨과 함께 plasma membrane transporter인 GLUT-1 level이 감소된다. 이러한 결과는 다약제내성의 영상화가 지금까지의 예상보다 보다 복합적이고 다양하므로 보다 많은 연구가 필요할 것이라는 점을 시사한다. 최근 시도되고 있는 생체광학 영상을 이용한 다약제내성 유전자 및 Pgp 발현 연구는 아직 시작단계이나, 분자 생물학적 영상법의 발전과 함께 MRI 기술등에도 이용될 수 있으므로 향후 많은 연구가 있을 것으로 기대된다.

구강 편평세포암종에서 $P16^{ink4}$ 유전자의 Methylation에 대한 연구 (($P16^{ink4}$ Methylation in Squamous Cell Carcinoma of the Oral Cavity.)

  • The p16 protein is a cyclin dependent kinase inhibitor that inhibits cell cycle progression from $G_1$ phase to S phase in cell cycle. Many p16 gene mutations have been noted in many cancer-cell lines and in some primary cancers, and alterations of p16 gene function by DNA methylation have been noticed in various kinds of cancer tissues and cell-lines. There have been a large body of literature has accumulated indicating that abnormal patterns of DNA methylation (both hypomethylation and hypermethylation) occur in a wide variety of human neoplasma and that these aberrations of DNA methylation may play an important epigenetic role in the development and progression of neoplasia. DNA methylation is a part of the inheritable epigenetic system that influences expression or silencing of genes necessary for normal differentiation and proliferation. Gene activity may be silenced by methylation of up steream regulatory regions. Reactivation is associated with demethylation. Although evidence or a high incidence of p16 alterations in a variety of cell lines and primary tumors has been reported, that has been contested by other investigators. The precise mechanisms by which abnormal methylation might contribute to carcinogenesis are still not fully elucidated, but conceivably could involve the modulation of oncogene and other important regulatory gene expression, in addition to creating areas of genetic instability, thus predisposing to mutational events causing neoplasia. There have been many variable results of studies of head and neck squamous cell carcinoma(HNSCC). This investigation was studied on 13 primary HNSCC for p16 gene status by protein expression in immunohistochemistry, and DNA genetic/epigenetic analyzed to determine the incidence, the mechanisms, and the potential biological significance of its Inactivation. As methylation detection method of p16 gene, the methylation specific PCR(MSP) is sensitive and specific for methylation of any block of CpG sites in a CpG islands using bisulfite-modified DNA. The genomic DNA is modified by treatment with sodium bisulfate, which converts all unmethylated cytosines to uracil(thymidine). The primers designed for MSP were chosen for regions containing frequent cytosines (to distinguish unmodified from modified DNA), and CpG pairs near the 5' end of the primers (to provide maximal discrimination in the PCR between methylated and unmethylated DNA). The two strands of DNA are no longer complementary after bisulfite treatment, primers can be designed for either modified strand. In this study, 13 paraffin embedded block tissues were used, so the fragment of DNA to be amplified was intentionally small, to allow the assessment of methylation pattern in a limited region and to facilitate the application of this technique to samlples. In this 13 primary HNSCC tissues, there was no methylation of p16 promoter gene (detected by MSP and automatic sequencing). The p16 protein-specific immunohistochemical staining was performed on 13 paraffin embedded primary HNSCC tissue samples. Twelve cases among the 13 showed altered expression of p16 proteins (negative expression). In this study, The author suggested that low expression of p16 protein may play an important role in human HNSCC, and this study suggested that many kinds of genetic mechanisms including DNA methylation may play the role in carcinogenesis.

Aβ42로 유도된 알츠하이머 마우스 모델에서 이중 가공 인삼열매 추출물의 학습 및 기억 손실 개선 효과 (Double-processed ginseng berry extracts enhance learning and memory in an Aβ42-induced Alzheimer's mouse model)

  • AD는 뇌의 신경세포 사멸뿐만 아니라 학습 및 기억능 소실을 초래하는 퇴행성 뇌 질환이며, 기억과 관련된 주요 뇌 구역인 해마(hippocampus)는 콜린성 조절(cholinergic modulation)에 의해 영향을 받는다(Konishi 등, 2015). AD 병인에 대한 다양한 해석과 설명이 있으나 크게는 amyloid cascade hypothesis과 함께 cholinergic hypothesis가 주류를 이루고 있다. 몇몇의 연구에서 ChAT 합성, ACh 분비, nicotin 및 muscarinic 수용체 감소가 AD 뇌의 대뇌피질과 해마에서 관찰되어(Tata 등, 2014) 퇴행성 뇌질환에서의 중요성이 제시되었고, 이를 배경으로 한 acetylcholinesterase inhibitors (AChEI)가 AD 증상 완화의 목적으로 미국 FDA로부터 승인되어 시판되고 있다. 본 연구에서는 인삼의 활성 성분인 진세노사이드가 다량 함유된 PGBC가 $A{\beta}42$로 유도된 치매 모델에서 뇌세포 보호, ACh 분비 증가, 학습력/기억력 증가, ChAT 발현 증가를 확인하여 인삼열매 추출물의 치매 적용 여부를 확인하고자 하였다. 결과에서 언급한 바와 같이 PGBC는 익기 직전의 4년근 인삼 열매에 추출 및 발효 단계를 추가하여 확보된 물질로서, $A{\beta}42$ 섭취, 제거 및 ACh 분비 촉진 활성이 있는 Re, Rd, Rg3 함량이 증가되어(Kim 등, 2014; Jang 등, 2015), PGBC 자체로서 치매 인자에 대한 조절 효능이 예측되었다. 특히, 7증 7포 및 발효과정을 거친 이중가공 인삼 열매 추출물이 비 발효 증포 추출물에 비해 Rg3가 현저히 증가하는 사전 연구결과와 Rg3가 $A{\beta}42$ 제거 활성을 가지는 것으로 확인된 결과를 종합할 때 PGBC 투여가 AD 증상 완화의 조절자 역할을 할 것으로 생각된다(Kim 등, 2013; Jang 등 2015). 본 연구에 따르면, 마우스 치매 모델에 PGBC 처리 시 PAT 및 Morris water-maze test를 통해 대조군 대비 유의한 수준의 인지능력 개선, ACh 합성을 유도하는 ChAT 유전자 발현 증가, ACh 분비량 증가 등이 확인되어 전체적인 인지능 개선에 극적인 영향을 준 것으로 판단된다. 특히 $A{\beta}42$를 뇌 실로 주입(intracranial injection) 하여 나타나는 뇌세포 손상이 PGBC 투여를 통해 보호된 것으로 사료되었으며, 이는 주요 뇌세포 중 하나인 성상세포에서 관찰되는 GFAP 분석을 통해 확인되었다. 뇌 균질액을 이용한 AChE 활성 연구에서도 PGBC가 AChE를 현저하게 저해하는 것으로 확인되어, AD 환자에게 처방이 가능한 2대 의약품 군중 하나인 시냅스 내 신경전달물질 ACh bioavailability 증가 목적의 처방 보조요법 적용이 기대된다(${\check{C}}olovi{\acute{c}}$ 등, 2013). 결론적으로, 본 연구에 사용된 PGBC는 학습 및 기억력을 개선하는 활성물질을 포함하고 있어 1차적인 퇴행성 뇌질환 보조재로서 직간접적인 대증요법 역할과 2차적으로는 뇌 세포 보호를 통한 질병 악화 지연 소재로 개발이 기대되며, 보다 심도 있는 기전연구를 통해 천연물 신소재 개발도 가능할 것으로 사료된다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually.
