Blockade of Kv1.5 by paroxetine, an antidepressant drug

  • Paroxetine, a selective serotonin reuptake inhibitor (SSRI), has been reported to have an effect on several ion channels including human ether-a-go-go-related gene in a SSRI-independent manner. These results suggest that paroxetine may cause side effects on cardiac system. In this study, we investigated the effect of paroxetine on Kv1.5, which is one of cardiac ion channels. The action of paroxetine on the cloned neuronal rat Kv1.5 channels stably expressed in Chinese hamster ovary cells was investigated using the whole-cell patch-clamp technique. Paroxetine reduced Kv1.5 whole-cell currents in a reversible concentration-dependent manner, with an $IC_{50}$ value and a Hill coefficient of $4.11{\mu}M$ and 0.98, respectively. Paroxetine accelerated the decay rate of inactivation of Kv1.5 currents without modifying the kinetics of current activation. The inhibition increased steeply between -30 and 0 mV, which corresponded with the voltage range for channel opening. In the voltage range positive to 0 mV, inhibition displayed a weak voltage dependence, consistent with an electrical distance ${\delta}$ of 0.32. The binding ($k_{+1}$) and unbinding ($k_{-1}$) rate constants for paroxetine-induced block of Kv1.5 were $4.9{\mu}M^{-1}s^{-1}$ and $16.1s^{-1}$, respectively. The theoretical $K_D$ value derived by $k_{-1}/k_{+1}$ yielded $3.3{\mu}M$. Paroxetine slowed the deactivation time course, resulting in a tail crossover phenomenon when the tail currents, recorded in the presence and absence of paroxetine, were superimposed. Inhibition of Kv1.5 by paroxetine was use-dependent. The present results suggest that paroxetine acts on Kv1.5 currents as an open-channel blocker.

  • Amyotrophic lateral sclerosis is a devastating neurodegenerative disease with a complex genetic basis, presenting both in familial and sporadic forms. The hexanucleotide (G4C2) repeat expansion in the C9orf72 gene, which triggers distinct pathogenic mechanisms, has been identified as a major contributor to familial and sporadic Amyotrophic lateral sclerosis cases. Animal models have proven pivotal in understanding these mechanisms; however, discrepancies between models due to variable transgene sequence, expression levels, and toxicity profiles complicate the translation of findings. Herein, we provide a systematic comparison of 7 publicly available Drosophila transgenes modeling the G4C2 expansion under uniform conditions, evaluating variations in their toxicity profiles. Further, we tested 3 previously characterized disease-modifying drugs in selected lines to uncover discrepancies among the tested strains. Our study not only deepens our understanding of the C9orf72 G4C2 mutations but also presents a framework for comparing constructs with minute structural differences. This work may be used to inform experimental designs to better model disease mechanisms and help guide the development of targeted interventions for neurodegenerative diseases, thus bridging the gap between model-based research and therapeutic application.

벼 발아초기 종자를 이용한 고효율 단기형질전환 방법 (High-efficiency and Rapid Agrobacterium-mediated genetic transformation method using germinating rice seeds)

  • 벼의 염기서열 분석이 완료됨에 따라 유전자의 세포내 기능을 밝히기 위한 기능유전체 연구가 활발히 진행되고 있다. 이를 위해 효율적으로 아그로박테리움을 이용해 원하는 유전자를 식물체 내로 형질전환을 하기 위한 노력은 지금도 계속 진행되고 있다. 본 실험에서는 캘러스를 유기한 후 아그로박테리움을 이용해 접종하는 기존의 방법과 달리, 성숙 종자를 소독한 후 2,4-D가 포함된 액체배지에 24시간 침종하여 배 부분이 발아하기 시작하는 종자를 이용해 바로 아그로박테리움을 접종하여 체세포변이의 발생을 최소화하고 유전자를 포함하고 있는 아그로박테리움이 식물 조직내로 침투할 수 있는 효율을 증가시키며, 그 후 캘러스를 유기하여 재분화 시킴으로써 형질전환 식물체를 얻는 방법을 새롭게 수립하였다. 배양과정 중 공동배양 배지에 아그로박테리움 성장억제물질인 silver nitrate와 항산화 물질인 DTT를 첨가하여 공동 배양 기간을 7일 이상으로 늘림으로써 벼 형질전환효율을 증가시킬 수 있었고, PCR 분석을 통해 원하는 목표 유전자가 형질전환체에 안정적으로 도입이 되는 것도 확인할 수 있었다. 또한, 이 방법은 형질전환 효율이 낮은 일품벼와 같은 품종에도 적용할 수 있을 것으로 판단된다. 이러한 결과를 종합해 볼 때, 본 실험을 통해 얻어진 새로운 공동배양 방법은 우수한 농업적 형질을 가진 벼 육종 소재 및 품종 개발시 효율적으로 이용할 수 있을 것으로 생각된다.

액체배양을 이용한 단기 벼 형질전환 방법 (Rapid Agrobacterium-mediated genetic rice transformation method using liquid media)

  • 본 연구는 종자로부터 유도된 캘러스를 아그로박테리움을 이용해 감염하여 고체배지에서 배양하는 기존의 방법과 달리, 형질전환에 소모되는 노동력과 시간, 비용을 단축하고자 감염과 공동배양, 균제거와 캘러스 선발까지 액체배양을 시도하였다. 성숙 종자로부터 유도된 캘러스를 바로 아그로박테리움으로 감염함으로써 조직배양으로 인한 체세포 변이의 발생을 최소화하고 감염부터 그 후 캘러스선발까지 총 4단계의 시간과 비용을 최소화하였으며, 감염된 캘러스로부터 재분화 시킴으로써 형질전환 식물체를 얻는 방법을 새롭게 수립하였다. 배양과정 중 감염과 공동배양 기간을 3일로 단축시킴으로써 캘러스의 스트레스를 최소화 하였고, PCR 분석을 통해 원하는 목표 유전자가 형질전환체에 안정적으로 도입이 되는 것도 확인할 수 있었다. 이러한 결과를 종합해 볼 때, 본 실험을 통해 얻어진 새로운 액체배양 방법은 우수한 농업적 형질을 가진 벼 품종 개발시 효율적으로 이용할 수 있을 것으로 생각된다.

Allele and Genotype Frequencies of the Polymorphic Methylenetetrahydrofolate Reductase and Lung Cancer in ther Jordanian Population: a Case Control Study

  • Background: Methylenetetrahydrofolate reductase (MTHFR) is involved in amino acid synthesis and DNA function. Two common polymorphisms are reported, C677T and A1298C, that are implicated in a number of human diseases, including cancer. Objective: The association between MTHFR C677T and A1298C genotype and haplotype frequencies in risk for lung cancer (LC) was investigated in the Jordanian population. Materials and Methods: A total of 98 LC cases were studied for MTHFR C677T and A1298C polymorphisms, compared to 89 controls taken from the general population, employing the PCR-RFLP technique. Results: The frequency of the genotypes of MTHFR C677T among Jordanians was: CC, 59.6%, CT, 33%; and TT, 7.4% among LC cases and 49.4%, 40.2% and 10.3% among controls. No significant association was detected between genetic polymorphism at this site and LC. At MTHFR A12987C, the genotype distribution was AA, 29.5%; AC, 45.3%, and CC 25.3% among LC cases and 36.8%, 50.6% and 12.6% among controls. Carriers of the CC genotype were more likely to have LC (OR=2.5; 95%CI: 1.04-6; p=0.039) as compared to AA carriers. Smokers and males with the CC genotype were 9.9 and 6.7 times more likely to have LC, respectively ($OR_{smokers}=9.9$; 95%CI: 1.2-84.5, p=0.018; $OR_{men}=6.6$; 95%CI: 1.7-26.2, p=0.005). Haplotype analysis of MTHFR polymorphism at the two loci showed differential distribution of the CC haplotype (677C-1298C) between cases and controls. The CC haplotype was associated with an increased risk for lung cancer (OR=1.6; 95% CI: 1.03-2.4, p=0.037). Conclusions: The genetic polymorphism of MTHFR at 1298 and the CC haplotype (risk is apparently lower with the C allele at position 677) may modulate the risk for LC development among the Jordanian population. Risk associated with the 1298C allele is increased in smokers and in males. The results indicate that a critical gene involved in folate metabolism plays a modifying role in lung cancer risk, at least in the Jordanian population.

단삼에 의한 Candida albicans 바이오필름 발달의 억제 (Growth of Candida albicans Biofilm is Inhibited by Salvia miltiorrhiza)

  • Candida albicans는 기회감염을 유발하는 주요한 병원성 진균 중의 하나이다. 캔디다증 치료과정에서 항진균제에 대한 내성이 흔히 발견되는데, 그 이유는 Candida가 바이오필름을 형성할 수 있기 때문이다. 이전의 연구에서 우리는 단삼(Salvia miltiorriza)의 에탄올추출물이 세포막의 투과성을 변화시키고 세포벽 합성을 저해하여 항캔디다 활성을 나타냄을 밝혔다. 본 연구에서는 10개 C. albicans 임상균주가 형성한 초기단계의 바이오필름을 대상으로 XTT 환원분석법으로 대사활성을 측정하니, $78{\mu}g/ml$ 단삼 에탄올추출물에 의해 바이오필름의 대사활성이 평균 51.3% 감소되었다. C. albicans 세포들이 폴리스티렌 표면에 부착하거나 germ tube를 형성하는 과정에서의 단삼 에탄올추출물의 영향을 현미경으로 분석하니, $39{\mu}g/ml$ 단삼 에탄올추출물에 의해 부착된 세포의 밀도는 현저하게 감소하였으나 germ tube 형성은 거의 억제하지 못했다. 단삼 에탄올추출물이 C. albicans SC5314 세포의 균사에 특이적인 유전자 발현에 미치는 영향을 qPCR로 분석한 결과, EAP1은 34.7% (p < 0.001), ALS1은 45.0% (p < 0.001), ALS3는 48.1% (p < 0.001), ECE1은 21.3% (p = 0.006) 억제하였다. 결론적으로 단삼의 에탄올추출물은 초기단계의 C. albicans 바이오필름 발달을 효율적으로 저해하며, 이는 EAP1, ALS1, ALS3 유전자의 발현억제에 따른 세포부착 억제와 관련이 있다. 더불어 단삼 에탄올추출물의 C. albicans 세포막 기능저해와 세포벽 합성억제에 의한 구조변화 또한 세포부착단계에서의 바이오필름 발달억제에 기여할 것으로 추정된다.

Lead Pollution and Lead Poisoning among Children in China

  • Lead is ubiquitous in the human environment as a result of industrialization. China's rapid industrialization and traffic growth have increased the potential for lead emissions. Lead poisoning in children is one of the most common public health problems today, and it is entirely preventable. Children are more vulnerable to lead pollution and lead in their bodies can affect their nervous, circulatory, and digestive systems. Children are exposed to lead from different sources (such as paint, gasoline, and solder) and through different pathways (such as air, food, water, dust, and soil). Although all children are exposed to some lead from food, air, dust, and soil, some children are exposed to high dose sources of lead. Significant sources of lead for China's children include industrial emissions (often close to housing and schools), leaded gasoline, and occupational exposure that occurs when parents wear lead-contaminated clothing home from work, burning of coal for home heat and cooking, contaminated food, and some traditional medicines. To assess the blood lead level in children in China, a large-scale study was conducted in 19 cities among 9 provinces during 1997 to 2000. There were 6502 children, aged 3-5 years, were recruited in the study The result indicates that the mean blood lead level was 8.83ug/dl 3-5 year old living in city area. The mean blood lead level of boys was higher than that of girls (9.1l ug/dl vs 8.73ug/dl). Almost 30 percent childrens blood lead level exceeded 10ug/dl. The average blood lead level was higher than that of in 1985 (8.83ug/dl vs 8.lug/dl). An epidemiological study was carried on the children living around the cottage industries recycling the lead from battery. Nine hundreds fifty nine children, aged 5-12 years, living in lead polluted villages where the lead smelters located near the residential area and 207 control children live in unpolluted area were recruited in the study. The lead levels in air, soil, drinking water and crops were measured. The blood lead and ZnPP level were tested for all subjects. The results show that the local environment was polluted. The lead levels both in the air and crops were much higher than that of in control area. In the polluted area, the average blood level was 49.6ug/dl (rang 19.5-89.3ug/dl). Whereas, in the unpolluted area, the average blood level was 12.4ug/dl (rang 4.6-24.8ug/dl). This study indicates that in some countryside area, some cottage industries induce seriously lead pollution and cause children health problem. For the introducing of unleaded gasoline in some large cities, such as Beijing and Shanghai, the blood lead level showed a declined trend since 1997. By 2000, the use of leaded gasoline in motor vehicles has been prohibited in China. The most recent data available show that levels of lead in blood among children in Shanghai decreased from 8.3ug/dl in 1997 to 7.6ug/dl in 1999. The prevalence rate of children lead poisoning (blood lead >10ug/dl) was also decreased from 37.8% to 24.8%. In children living in downtown area, the blood lead level reduced dramatically. To explore the relationship between gene polymorphisms and individual susceptibility of lead poisoning, a molecular epidemiological study was conducted among children living in lead polluted environment. The result showed that the subjects with ALAD2 allele has higher ZPP level, and the subjects with VDR B allele has larger head circumference than only with b allele. In the present study, we demonstrated that ALAD genotypes modify lead effects on heme metabolism and VDR gene variants influence the skull development in highly exposed children. The polymorphism of ALAD and VDR genes might be the molecular inherited factor modifying the susceptibility of lead poisoning. Recently, Chinese government pays more attention to lead pollution and lead poisoning in children problem. The leaded gasoline was prohibited used in motor vehicles since 2000. The government has decided to have a clampdown on the high-polluted lead smelters for recycling the lead from battery in countryside.
