Estimation of Ruminal Degradation and Intestinal Availability of Crude Protein in the Animal-Origin Feedstuffs Using Mobile Nylon Bag Technique

  • Ruminal degradation characteristics and intestinal availability of crude protein (CP) in four animal-origin feeds (fish meal, meat meal, viscera meal, feather meal) were estimated by mobile nylon bag technique. Three ruminally and duodenally cannulated Holstein dairy cows (average body wt. 550kg) fed a diet containing 40% concentrate and 60% orchard grass hay on a dry matter (DM) basis. Assuming that the outflow rate of diet in rumen is 5% per hour (k =0.05), contents of quickly degradable CP (QDP), slowly degradable CP (SDP), and undegradable CP (UDP) in the rumen were 27.6%, 9.4%, 63.0% for fish meal, 34.3% 28.1%, 37,6% for meat meal, 43.9%, 12.5%, 43.6% for viscera meal, and 14.4%, 15.8%, 69.8% for feather meal, respectively. Intestinal CP degradability was 51.0% for fish meal, 27.2% for meat meal, 37.9% for viscera meal and 56.2% for feather meal. Available UDP in the intestinal tract was contained 288 g, 217 g, 246 g and 423 g per kilogram DM of diet in fish meal, meat meal, viscera meal and feather meal, respectively.

DETERMINATION OF THE APPARENT ILEAL DIGESTIBILITY OF PROTEIN AND AMINO ACIDS IN FEEDSTUFFS AND MIXED DIETS FOR GROWIG-FINSHING PIGS WITH THE MOBILE NYLON BAG TECHNIQUE

  • A series of experiments was conducted to determine the influence of various pepsin-HCL pretreatment factor, hereby the factors of duration of washing for the retrieved bags, inherent to the mobile nylon bag technique (MNBT), on apparent ileal digestibility of crude protein (AIDCP) and apparent ileal digestibility of dry matter (AIDDM). At last, the AIDCP and apparent ileal digestibility of amino acids (AIDAA) in maize, barley, wheat, rapeseed meal, cottonseed meal and three mixed diets were determined with the MNBT and ileo-rectal anastomis pigs (IRAT). For the MNBT techniques, bag measuring $25{\times}40$ MM and containing 0.75 g feedstuff samples, after pre-digestion in vitro, were introduced into the ileo-rectal anastomis pigs (IRAT) gastrointestinal tract through a duodenal cannula and recovered in the ileal digesta between 6 and 12 h. later. 1. The apparent ileal digestibility of dry matter (AIDDM) and crude protein (AIDCP) of the tested samples, with the exception of fish meal, determined by MNBT were not affected by the different pepsin-HCL pretreatment times in vitro between 2.5 h. and 4 h. 2. There was no significant (p > 0.05) difference of the AIDCP and AIDDM of maize determined by the MNBT among different pepsin concentration (0.03%, 0.07% and 0.1 %) treatment in vitro. 3. The AIDCP determined with the MNBT was affected by the washed and unwashed recovered bags from the ileal digesta. 4. The AIDCP and AID amino acids (AIDAA) of maize, barley, wheat, rapeseed meal, soya-bean meal, cottonseed meal and three mixed diets from the MNBT, with a solution of 0.01N HCL (PH 2) and 0.1% of pepsin concentration, a pepsin-HCL pretreatment time in vitro or 4h. and a washing time of the recovered bag from the ileal digesta compared well with those from the IRAT. The linear regression analysis showed a significant correlation (p < 0.01) of AIDCP and AIDDA between the IRAT and MNBT.

In situ 나일론백 그리고 모바일백 방법을 이용한 국내 부존사료자원의 반추가축용 사료 가치 평가 (Evaluation of non-conventional feeds for ruminants using in situ nylon bag and the mobile bag technique)

  • 본 연구는 반추동물 사료로서 한국에서 생산된 부존사료자원 15종의 화학적 조성, 소화율 그리고 에너지 가를 평가하기 위하여 실시하였다. 반추위 그리고 십이지장 캐뉼라가 장착된 거세한우 3두(평균체중 $520{\pm}20.20kg$)를 개별 계류식우사에 공시한 후 14일의 순치기간과 3일간의 시험기간을 두었다. 실험 방법으로는 화학조성 분석법, in situ 나일론백과 모바일백 방법을 이용하였다. 사료 내 화학적 조성을 분석한 결과, 엿밥, 깻묵, 장유박 및 두유박의 조단백질 함량은 30% 이상이었다. In situ 나일론백을 이용한 사료의 분해 특성을 분석한 결과, 비트펄프, 맥주박, 커피박, 엿밥, 수수겨, 임자박, 라면박 및 두유박의 반추위 미분해 단백질 함량은 50% 이상이었다. In situ 모바일백을 이용한 사료 내 가소화 영양소총량(TDN)의 분석은 비트펄프, 맥주박, 막걸리박, 수수겨, 깻묵, 라면박, 미강, 장유박, 비지, 두유박 및 밀기울이 50% 이상인 것으로 관찰되었다. 요약하자면, 상기 부존사료자원은 배합사료 또는 조사료를 대체할 수 있는 훌륭한 사료자원으로서의 높은 잠재적 가치를 가지고 있음을 시사하고 있다. 따라서 본 연구에서 얻은 부존사료자원의 화학적 조성, 소화율 및 에너지 가를 반추동물 섬유질배합사료 제조를 위한 기초 자료로 활용하고 이를 통해 사료효율 향상, 사료비용 절감 및 환경오염 감소에 기여하고자 한다.

Further Modifications to the Mobile Nylon Bag Technique to Determine Nutrient Digestibility for Swine

  • Previous studies conducted with swine have reported that the mobile nylon bag technique (MNBT) does not always accurately predict in vivo nutrient digestibilities. Therefore, in this study, the MNBT was modified so that nutrient digestibilities would more closely resemble those from conventional (Con) digestibility studies obtained using the indicator method. A total of 19 feeds were tested including five cereal grains, five legumes, three high protein sources and six mixed diets. The principle changes to the MNBT included the use of a fecal collection harness which minimized the number of bags lost. In addition, previous protocols involved pooling of bags within pig while in the present experiment all bags were analyzed separately to increase the precision of the test. Finally, chemical analyses were done using the entire nylon bag plus residue rather than opening.the bags and scraping out the contents. With the exception of the barley sample (p=0.01), dry matter digestibility (DMD) coefficients obtained with the MNBT were not significantly different from those obtained with the indicator method. The linear regression equation relating the MNBT to the indicator method was Con DMD=-O.77+1.02 MNBT DMD ($r^2=0.93$: p<0.0001). There was no significant (p>0.05) difference in gross energy digestibility (GED) coefficients determined using the MNBT or the indicator method for any of the 19 feeds. The regression line equation relating the MNBT to the indicator method was Con GED=-5.68+1.06 MNBT GED ($r^2=0.94$: p<0.0001). The MNBT was less effective in predicting in vivo crude protein digestibility (CPD) than it was in predicting dry matter and energy digestibility. Differences greater than five percentage units were observed for two of the legumes, Kabuli chickpeas (p=0.02) and the extruded pea-canola seed mixture (p=0.01) as well as for three of the mixed diets including the unheated hulled barley-based diet (p=0.01), the unheated hulless-barley based diet (p=0.08) and the barley-soybean meal based diet (p=0.008). The regression equation relating the MNBT to the indicator method was Con CPD=5.75 + 0.90 MNBT CPO ($r^2=0.76$; p<0.0001). This study indicates that the modified MNBT can be used for the rapid determination of dry matter and energy digestibility in a wide variety of ingredients. For the measurement of crude protein digestibility, the technique produces results similar to conventional digestibility studies for cereal grains and high protein feeds but tends to overestimate protein digestibility for legumes and mixed diets.

Effect of Mobile Bag and Sample Sizes on Intestinal Digestibility of Forage in Sheep

  • This study aimed to clarify the effect of mobile bag size and ratio of sample size to bag surface area on intestinal digestibility of forage in sheep. Four Suffolk ewes fitted with ruminal and proximal duodenal cannulae were fed second-cut Italian ryegrass (Lolium multiflorum Lam.) hay twice daily, and the same forage was used to measure intestinal digestibility. The forage samples were incubated in the rumen for 16 h and then in pepsin-HCl solution for 3 h before intestinal incubation. The incubated forage samples were placed in a nylon mobile bag. The bag sizes used were either 20 mm${\times}$20 mm (small bag size; SBS) or 30 mm${\times}$30 mm (large bag size; LBS) and the ratio of the sample size to the surface area of the bag was either 5.5 $mg/cm^{2}$ (low ratio; LR) or 11.0 $mg/cm^{2}$ (high ratio; HR) resulting in four different treatment conditions: SBS-LR, SBS-HR, LBS-LR and LBS-HR. Eight bags per animal were inserted through the duodenal cannulae at 15-min intervals and were subsequently collected from the feces of the animal. The mean intestinal bag transition time did not differ significantly between animals, but ranged from 23.2 to 27.0 h. The intestinal digestibility of dry matter (IDDM) ranged from 0.162${\pm}$0.019 g/g in the SBS-HR treatment group to 0.195${\pm}$0.018 g/g in the SBS-LR treatment. The intestinal digestibility of crude protein (IDCP) ranged from 0.610${\pm}$0.031 g/g in the LBS-LR treatment to 0.693${\pm}$0.018 g/g in the SBS-LR treatment. There was no difference in the IDDM and IDCP between different treatments. It was therefore concluded that the size of the mobile bag and the ratio of the sample size to the bag surface area did not influence the intestinal digestibility of forage. Future studies should use bags with high ratios of sample size to surface area in order to obtain sufficient residue for further analysis.

THE INFLUENCE OF SELECTED CHEMICAL TREATMENTS ON THE RUMINAL DEGRADATION AND SUBSEQUENT INTESTINAL DIGESTION OF CEREAL STRAW

  • An experiment was conducted with three ruminally and intestinally cannulated non-lactating cows of Finnish Ayrshire breed, to assess the ruminal degradation characteristics of oat (Avena sativa), rye (Secale cereale) and rice (Oryza sativa) straw by the nylon bag technique, and the subsequent post-ruminal degradation of their rumen-undegraded residues by using the mobile bag technique, respectively. The straw samples were untreated or treated with aqueous $NH_3$ or with urea solution in cold or hot water. The untreated straw samples were milled or chopped, and the treated straw samples were chopped. The constant values a, b, and c were computed according to the exponential equation, where a = intercept of degradation curve at time 0, b = potentially degradable material, c = rate of degradation of band (a+b) = maximum potential degradability (asymptote). It was found that nitrogen contents of chemically treated straw were markedly increased by both $NH_3$ and urea treatments. Milling the samples attributed to a remarkable loss at 0 h incubation time as compared to chopping of the respective samples. However, chemical treatment markedly improved the b value and the subsequent (a+b) values for dry matter, organic matter, neutral-detergent fiber, and acid-detergent fiber of the samples. Furthermore, temperature of the water used in the urea solutions was considered essential, since urea in hot water rather than in cold water seemed to enhance the overall degradability. The disappearance of rumen-incubated straw residues from the mobile bags ranged from 4.5 to 9.6% for the parameters measured. On average, the OM disappearance from bags was clearly higher for the residues of urea treated straw compared to those of ammonia treated straw, but the disappearance of NDF tended, however, to be higher on the ammonia treatment.

ALKALINE HYDROGEN PEROXIDE TREATED ASPEN AS A RUMINANT FEED

  • Two experiments were conducted to evaluate the nutritive value of a diet containing 20% alkaline hydrogen peroxide(AHP) treated aspen sawdust for ruminants. In experiment 1, sheep fed treated aspen(treated) had higher (p < 0.05) average daily gain and improved feed/gain ratio compared to animals fed untreated aspen (untreated), Apparent NDF and ADF digestibilities of treated aspen were higher (p < 0.05) than for untreated material. A similar trend was observed for DM, organic matter and cellulose digestibilities coefficients. In experiment 2, the soluble and degradable DM and crude protein (CP) fractions tended to increase with AHP treatment. Treatment also increased (p<0.05) the degradation rate of the degradable fraction. Results of these experiment indicate that AHP treatment results in a substantial improvement in the extent of utilization of aspen sawdust by ruminants.

동물성 부산물 사료 세 종류에 대한 열처리가 조단백질의 반추위내 분해특성 및 하부장기내 이용성에 미치는 영향 (Effects of Heat Treatment of Three Animal by-products on Ruminal Degradation Characteristics and Intestinal Availability of Crude Protein)

  • 동물성 부산물 사료(우모분, 우지박, 내장분)단백질의 반추위내 분해특성과 하부장기내 이용성에 대한 열처리 효과를 구명하기 위하여 반추위와 십이지장에 누관이 장착된 Holstein 건유우 3두를 공시하였다. 시험사료에 대한 열처리는 149$^{\circ}C$가 유지되는 oven에서 4시간동안 처리한 후, 1 mm체를 통과시켰다. 시험사료의 반추위내 분해특성은 발효시간별 분해율에서 비선형회귀식을 유도하여 구하였고, 사료단백질의 하부장기내 이용성은 mobile nylon bag기법으로 추정되었다. 농후사료와 orchard grass를 60:40의 비율로 급여하였으며, 물과 mineral block은 자유섭취토록 하였다. 조단백질의 반추위내 유효분해도(k=0.05) 및 하부장기내 소실율에 있어서 우모분은 각각 30.2% 및 56.2%, 우지박은 75.0% 및 18.6% 그리고 내장분은 56.4% 및 37.9%였다. 시험사료에 대한 열처리효과에 있어서 조단백질의 반추위내 유효분해도는 우모분과 내장분은 증가하였으나 우지박은 감소되었고(P$<$0.05), 하부장기내 조단백질 소실율에서는 우지박은 증가된 반면, 우모분과 내장분은 감소되어(P$<$0.05) 상반되는 결과를 나타내었다. 반추위 미분해 사료단백질의 하부장기내 이용율은 우모분, 우지박 및 내장분에 대해서 각각 80.4%, 83.8% 및 86.9%였으며, 열처리를 함으로써 우모분과 우지박은 각각 94.0% 및 91.3%로 향상되었으나, 내장분은 76.5%로 낮아졌다(P$<$0.05).

Determination of Optimal Conditions of Pressure Toasting on Legume Seeds for Dairy Deed Industry : I. Effects of Pressure Toasting on Nutritive Values of Lupinus albus in Lactating Dairy Cows

  • Whole lupinus albus seeds were pressure toasted at temperatures of 100, 118 and $136^{\circ}C$ for 3, 7, 15 and 30 min to study rumen degradation and post-rumen digestion and to determine optimal heating conditions for the Dutch dairy feed industry. In sacco nylon bag and mobile bag techniques were employed for rumen and intestine incubations to determine ruminal degradation characteristics and intestinal digestion of crude protein (CP) in 4 lactation rumen cannulated and 4 lactating intestinal cannulated Dutch dairy cows fed 47% hay and 53% concentrate according to Dutch dairy requirements. Measured rumen degradation characteristics were soluble fraction (S), undegradable fraction (U), potentially degradable fraction (D), lag time (T0) and rate of degradation (Kd) of insoluble but degradable fraction. Percentage bypass feed protein (BCP), ruminal microbial protein synthesized based on available nitrogen (N_MP) and that based on available energy (E_MP), true protein supplied to the small intestine (TPSI), truly absorbed BCP (ABCP), absorbed microbial protein (AVP) in the small intestine, endogenous protein losses in the digestion (ENDP), true digested protein in the small intestine (TAP or DVE in Dutch) and degraded protein balance (PDB or OEB in Dutch) were totally evaluated using the new Dutch DVE/OEB System. Pressure toasting decreased (p<0.001) rumen degradability of CP. It reduced S (p<0.05) and Kd (p=0.06), increased D (p<0.05) and U (p<0.01) but did not alter T0 (p>0.05), thus resulting in dramatically increased BCP (p<0.001) with increasing time and temperature from 73.7 (raw) up to 182.5 g/kg DM ($136^{\circ}C/15min$). Although rumen microbial protein synthesized based on available energy (E_MP) was reduced, true protein (microbial and bypass feed protein) supplied to the small intestine (TPSI) was increased (p<0.001) from 153.1 (raw) to 247.6 g/kg DM ($136^{\circ}C/15min$). Due to digestibility of BCP in the intestine not changing (p>0.05) average 87.8%, the absorbed BCP increased (p<0.001) from 62.3 (raw) to 153.7 g/kg DM ($136^{\circ}C/15min$). Therefore DVE value of true digested protein in the small intestine was significantly increased (p<0.001) from 118.9 (raw) to 197.0 g/kg DM ($136^{\circ}C/15min$) and OEB value of degraded protein balance was significantly reduced (p<0.001) from 147.2 (raw) to 63.1 g/kg DM ($136^{\circ}C/15min$). It was concluded that pressure toasting was effective in shifting degradation of CP of lupinus albus from the rumen to small intestine without changing intestinal digestion. Further studies are required on the degradation and digestion of individual amino acids and on the damaging effects of processing on amino acids, especially the first limiting amino acids.

In situ Ruminal and Intestinal Digestibility of Crude Protein and Amino Acids in By-product Feedstuffs

  • The objectives of this study was to evaluate the degradability and digestibility of crude protein (CP), rumen undegradable protein (RUP), and individual amino acids (AA) on six by-product feedstuffs (BPF) (rice bran, RB; wheat bran, WB; corn gluten feed, CGF; tofu residue, TR; spent mushroom substrate from Pleurotus ostreatus, SMSP; brewers grain, BG) as ruminants feed. Three Hanwoo steers (40 months old, $520{\pm}20.20kg$ of body weight) fitted with a permanent rumen cannula and T-shaped duodenal cannula were used to examine of the BPF using in situ nylon bag and mobile bag technique. The bran CGF (19.2%) and food-processing residue BG (19.7%) had the highest CP contents than other feeds. The RUP value of bran RB (39.7%) and food-processing residues SMSP (81.1%) were higher than other feeds. The intestinal digestion of CP was higher in bran RB (44.2%) and food-processing residues BG (40.5%) than other feeds. In addition, intestinal digestion of Met was higher in bran RB (55.7%) and food-processing residues BG (44.0%) than other feeds. Overall, these results suggest that RB and BG might be useful as main raw ingredients in feed for ruminants. Our results can be used as baseline data for ruminant ration formulation.
