Carbon Monoxide Ameliorates 6-Hydroxydopamine-Induced Cell Death in C6 Glioma Cells

  • Carbon monoxide (CO) is well-known as toxic gas and intrinsic signaling molecule such as neurotransmitter and blood vessel relaxant. Recently, it has been reported that low concentration of CO exerts therapeutic actions under various pathological conditions including liver failure, heart failure, gastric cancer, and cardiac arrest. However, little has been known about the effect of CO in neurodegenerative diseases like Parkinson's disease (PD). To test whether CO could exert a beneficial action during oxidative cell death in PD, we examined the effects of CO on 6-hydroxydopamine (6-OHDA)-induced cell death in C6 glioma cells. Treatment of CO-releasing molecule-2 (CORM-2) significantly attenuated 6-OHDA-induced apoptotic cell death in a dose-dependent manner. CORM-2 treatment decreased Bax/Bcl2 ratio and caspase-3 activity, which had been increased by 6-OHDA. CORM-2 increased phosphorylation of NF-E2-related factor 2 (Nrf2) which is a transcription factor regulating antioxidant proteins. Subsequently, CORM-2 also increased the expression of heme oxygenase-1 and superoxide dismutases (CuZnSOD and MnSOD), which were antioxidant enzymes regulated by Nrf2. These results suggest that CO released by CORM-2 treatment may have protective effects against oxidative cell death in PD through the potentiation of cellular adaptive survival responses via activation of Nrf2 and upregulation of heme oxygenase-1, leading to increasing antioxidant defense capacity.

A Study on Water Quality Changes of Geum River Subwatersheds: In Cases of Tributary (금강수계 소유역내 수질 변화 분석 -소하천을 대상으로-)

  • BACKGROUND: For effective subwatershed management, it is very important to select the tributaries for improving water quality and understand the characteristics of tributaries. Until now, however, the case study of main streams has been managed. 17 tributaries in Geum river subwatershed were monitored to regulate the source of water contaminations and identify their current situations in this study. METHODS AND RESULTS: As pollution indicators, such as biological oxygen demand($BOD_5$), chemical oxygen demand($COD_{Mn}$), suspended solid(SS), total nitrogen (T-N), total phosphate(T-P) and total organic carbon(TOC) in Geum river were examined from January to December in 2011. The results were as follows : The annual average concentration of nutrients in Yongdam reservoir upsteam was 0.7 mg/L for BOD, 3.0 mg/L for COD, 8.4 mg/L for SS, 2.905 mg/L for T-N, 0.035 mg/L for T-P and 1.6 mg/L for TOC. Water quality of Daechung reservoir upstream was mostly similar tendency in comparison to Yongdam reservoir upstream. Among the 22 tributaries, water quality in Daechung reservoir downstream was more polluted. T-N contents were significantly high in Miho B4 located Daechung reservoir downstream(annual average concentration: 13.53 mg/L). In cases of Miho A1, A2 and C1, pollution degree was worsened during rainy season expecially. CONCLUSION(S): For improving water quality of Geum river subwatershed, the tributaries in the Mihocheon area should be preferentially considered. Mihocheon tributary is the highest in pollution site, and thus a study on long-term effects should be research.

Amorphous Cr-Ti Texture-inducing Layer Underlying (002) Textured bcc-Cr alloy Seed Layer for FePt-C Based Heat-assisted Magnetic Recording Media

  • $Cr_{100-x}Ti_x$ amorphous texture-inducing layers (TIL) were investigated to realize highly (002) oriented $L1_0$ FePt-C granular films through hetero-epitaxial growth on the (002) textured bcc-$Cr_{80}Mn_{20}$ seed layer (bcc-SL). As-deposited TILs showed the amorphous phase in Ti content of $30{\leq}x(at%){\leq}75$. Particularly, films with $40{\leq}x{\leq}60$ kept the amorphous phase against the heat treatment over $600^{\circ}C$. It was found that preference of the crystallographic texture for bcc-SLs is directly affected by the structural phase of TILs. (002) crystallographic texture was realized in bcc-SLs deposited on the amorphous TILs ($40{\leq}x{\leq}70$), whereas (110) texture was formed in bcc-SLs overlying on crystalline TILs (x < 30 and x > 70). Correlation between the angular distribution of (002) crystal orientation of bcc-SL evaluated by full width at half maximum of (002) diffraction (FWHM) and a grain diameter of bcc-SL indicated that while the development of the lateral growth for bcc-SL grain reduces FWHM, crystallization of amorphous TILs hinders FWHM. $L1_0$ FePt-C granular films were fabricated under the substrate heating process over $600^{\circ}C$ with having different FWHM of bcc-SL. Hysteresis loops showed that squareness ($M_r/M_s$) of the films increased from 0.87 to 0.95 when FWHM of bcc-SL decreased from $13.7^{\circ}$ to $3.8^{\circ}$. It is suggested that the reduction of (002) FWHM affects to the overlying MgO film as well as FePt-C granular film by means of the hetero-epitaxial growth.

Effect of 1,3-Benzopyrone on Chemotactic Activity of Peripheral Blood Leukocytes in the Dog (1,2-benzopyrone이 개 말초혈액 백혈구의 유주활성에 미치는 영향)

  • This study was undertaken to examine whether 1,2-benzopyrone affects on chemotactic activity of canine peripheral blood leukocytes. A modified Boyden chamber method was sed on chemotaxis evaluation. The direct treatments of 1,2-benzopyrone showed no ffects on the chemotaxis of peripheral blood mononuclear cells (PBMCs) and olymorphonuclear cells (PMNs). But chemotaxis of PMN was remarkably enhanced by ulture supernatant from PBMC but not PMN treated with 1,2-benzopyrone. Similarly, it as also increased by recombinant (r) interleukin (lL)­8. This chemotactic activity of MN was inhibited by addition of anti-rIL-8 polyclonal antibody. The chemotaxis of PBMC was not enhanced by culture supernatant from either PBMC or PMN treated with 1,2-benzopyrone. Therefore, these results suggested that the chemotactic activity of PMN ay be mainly mediated by IL-8-like factor(s) produced from PBMC treated with ,2-benzopyrone.

Purification and Characterization of an Acid Deoxyribonuclease from the Cultured Mycelia of Cordyceps sinensis

  • A new acid deoxyribonuclease (DNase) was purified from the cultured mycelia of Cordyceps sinensis, and designated CSDNase. CSDNase was purified by $(NH_4)_2SO_4$ precipitation, Sephacryl S-100 HR gel filtration, weak anion-exchange HPLC, and gel filtration HPLC. The protein was single-chained, with an apparent molecular mass of ca. 34 kDa, as revealed by SDS-PAGE, and an isoelectric point of 7.05, as estimated by isoelectric focusing. CSDNase acted on both double-stranded (ds) and single- stranded (ss) DNA, but preferentially on dsDNA. The optimum pH of CSDNase was pH 5.5 and its optimum temperature 55. The activity of CSDNase was not dependent on divalent cations, but its enzymic activity was inhibited by high concentration of the cation: $MgCl_2$ above 150 mM, $MnCl_2$ above 200 mM, $ZnCl_2$ above 150 mM, $CaCl_2$ above 200 mM, NaCl above 300 mM, and KCl above 300 mM. CSDNase was found to hydrolyze DNA, and to generate 3-phosphate and 5-OH termini. These results indicate that the nucleolytic properties of CSDNase are essentially the same as those of other well-characterized acid DNases, and that CSDNase is a member of the acid DNase family. To our knowledge, this is the first report of an acid DNase in a fungus.

Fluorene-Based Conjugated Copolymers Containing Hexyl-Thiophene Derivatives for Organic Thin Film Transistors

  • Two fluorene-based conjugated copolymers containing hexyl-thiophene derivatives, PF-1T and PF-4T, were synthesized via the palladium-catalyzed Suzuki coupling reaction. The number-average molecular weights (Mn) of PF-1T and PF-4T were found to be 19,100 and 13,200, respectively. These polymers were soluble in common organic solvents such as chloroform, chlorobenzene, toluene, etc. The UV-vis absorption maximum peaks of PF-1T and PF-4T in the film state were found to be 410 nm and 431 nm, respectively. Electrochemical characterization revealed that these polymers have low highest occupied molecular orbital (HOMO) levels, indicating good resistance against oxidative doping. Thin film transistor devices were fabricated using the top contact geometry. PF-1T showed much better thin-film transistor performance than PF-4T. A thin film of PF- 1T gave a saturation mobility of 0.001-0.003 cm2 V?1 s?1, an on/off ratio of 1.0 × 105, and a small threshold voltage of ?8.3 V. To support TFT performance, we carried out DSC, AFM, and XRD measurements.

A Study on the Dyeing Characteristics by Reproduction of Hwangsu Spring

  • To evaluate the Hwangsu spring as natural mordants effect on dyeing. The natural mordants effect of Hwangsu spring were analyzed by pH, temperature, trace elements, organic matter and color fastness. Hwangsu spring having pH 2.42 and $14.4^{\circ}C$ was harvested at the end of September, in Yeongcheon (Gyeongbuk Province, Korea) and left on a $0^{\circ}C$ and $25^{\circ}C$ before use. Hwangsu spring during storage at $25^{\circ}C$ turned brown by photochemical reaction. The Result of ICP analysis, it contained Fe(414.9 ppm), Al(88.9 ppm), Mn(4.9 ppm) and observed character by water analysis. Dyeing and post-mordant procedure; Cotton(KS K 0905) were dyed with clove powder for 30 min at $70^{\circ}C$. Hwangsu spring, Aluminium Sulfate $14-18H_20(Al_2(SO_4)_3{\cdot}14-18H_2O)$, Iron Sulfate Heptahydrate($FeSO_4{\cdot}7H_2O$) were used by post mordant at the same ratio. The dyed fabrics were treated with 20% each mordant solution at $25^{\circ}C$ for 10min. Comparison with a reproduced chemical mordant, the K/S values of cotton fabrics dyed with Hwangsu sping were increased.

Qualitative Analysis and Plasma Characteristics of Soil from a Desert Area using LIBS Technique

  • In this work, laser induced breakdown spectroscopy (LIBS) is used to investigate soil samples collected from different desert areas of Riyadh city in Saudi Arabia. Both qualitative analysis and plasma parameters are studied via the observed LIBS spectra. These experiments have been done using a Spectrolaser-7000 system with 50 mJ fundamental wavelength of Nd:YAG laser and detection delay time of 1 microsecond. Many spectral lines are highly resolved for many elements like Al, Fe, Mg, Si, Mn, Na, Ca and K. The electron temperatures Te and electron densities Ne, for the constituent of generated LIBS plasma, are determined for all the collected samples. It is found that both Te and Ne vary from one desert area to other. This variation is due to the change of the elemental concentration in different desert areas that affects the sample's matrices. Time dependent measurements have also been performed on the soil samples. While the signal-to-base ratio (SBR) reached its optimal value at 1 microsecond, the plasma parameters Ne and Te reach values of $4{\times}10^{17}cm^{-3}$ and 9235 K, respectively, at 2.5 microsecond. The later indicate that the plasma cooling processes are slow in comparison to the previously observed results for metallic samples. The observed results show also that in the future it is possible to enhance the exploitation of LIBS in the remote on-line environmental monitoring application, by following up only the values of Ne and Te for one element of the soil desert sample using an optical fiber probe.

Elemental Composition of the Soils using LIBS Laser Induced Breakdown Spectroscopy

  • Laser induced breakdown spectroscopy (LIBS) technique has been used for the elemental composition of the soils. In this technique, a high energy laser pulse is focused on a sample to produce plasma. From the spectroscopic analysis of such plasma plume, we have determined the different elements present in the soil. This technique is effective and rapid for the qualitative and quantitative analysis of all type of samples. In this work a Q-switched Nd: YAG laser operating with its fundamental mode (1064 nm laser wavelength), 5 nanosecond pulse width, and 10 Hz repetition rate was focused on soil samples using 10 cm quartz lens. The emission spectra of soil consist of Iron (Fe), Calcium (Ca), Titanium (Ti), Silicon (Si), Aluminum (Al), Magnesium (Mg), Manganese (Mn), Potassium (K), Nickel (Ni), Chromium (Cr), Copper (Cu), Mercury (Hg), Barium (Ba), Vanadium (V), Lead (Pb), Nitrogen (N), Scandium (Sc), Hydrogen (H), Strontium (Sr), and Lithium (Li) with different finger-prints of the transition lines. The maximum intensity of the transition lines was observed close to the surface of the sample and it was decreased along the axial direction of the plasma expansion due to the thermalization and the recombination process. We have also determined the plasma parameters such as electron temperature and the electron number density of the plasma using Boltzmann's plot method as well as the Stark broadening of the transition lines respectively. The electron temperature is estimated at 14611 °K, whereas the electron number density i.e. 4.1 × 1016 cm-3 lies close to the surface.

Shear bond strength of dentin bonding agents cured with a plasma arc curing light (전단접착강도와 관련된 Plasma Arc Curing Light의 중합효율평가)

  • The objective of this study was to compare dentin shear bond strength (DSBS) of dentin bonding agents (DBAs) cured with a plasma arc (PAC) light curing unit (LCU) and those cured with a light emitting diode (LED) LCU. Optical properties were also analyzed for Elipar freelight 2 (3M ESPE); LED LCU, Apollo 95E (DMT Systems); PAC LCU and VIP Junior (Bisco); Halogen LCU. The DBAs used for DSBS test were Scotchbond Multipurpose (3M ESPE), Singlebond 2 (3M ESPE) and Clearfil SE Bond (Kuraray). After DSBS testing, fractured specimens were analyzed for failure modes with SEM. The total irradiance and irradiance between 450 nm and 490 nm of the LCUs were different. LED LCU showed narrow spectral distribution around its peak at 462 nm whereas PAC and Halogen LCU showed a broad spectrum. There were no significant differences in mean shear bond strength among different LCUs (P > 0.05) but were significant differences among different DBAs (P < 0.001).
