Chemical Composition of Cultured and Wild Codonopsis lanceolata Roots of Different Age Groups -I. Proximate Composition, Minerals and Protein Fractions- (더덕(沙蔘)의 년근별(年根別) 화학성분(化學成分)에 관(關)한 연구(硏究) -제1보(第1報) : 일반성분(一般成分), 무기질(無機質) 및 단백질(蛋白質) 분획(分劃)-)

  • Proximate compositions, minerals and protein fractions of the roots of cultured and wild Codonopsis lanceolata of different age groups were examined as the basic research for the study of their source of processed foods. The most abundant proximate composition of the roots of C. lanceolata was observed to be total sugars and next come crude protein, crude fiber, crude fat and ash in descending order irrespective of cultured and wild ones. The richest mineral contained in the roots was noticed to be K and followed by Mg and Ca. Generally increased tendency of crude protein, fat, ash, K, Mg, Ca, Mn, Zn, Cu and P contents were observed with older roots, however, decreased total sugars and Fe content. Lead and cadmium content was far bellow the authorized tolerance limits. The quantitative fractionation of the protein of the roots ranked albumin the highest content, followed by globuin, prolamin and glutelin. Decreased albumin content was observed with the older age roots, while increased globulin, prolamin and glutelin content. The minimum solubility of the soluble protein of the roots was found to be at pH 4.0 and maximum, at pH 10.0. Disc gel electrophoresis of the soluble protein of C. lanceolata roots showed almost similar patterns and numbers of bands. The molecular weight for main band protein was estimated to be about 90,000.

Effect of Dietary Fatty Acid and Vitamin E Supplementation in Antioxidant Systmes of the Second Generation Rat Brain Sections (식이지방산 조성 및 비타민 E의 보충이 제 2 세대 흰 쥐 뇌조직의 항산화 체계에 미치는 영향)

  • In this study, we examined the effects of dietary fatty acids and vitamin E supplementation on antioxidant systems in the rat brain regions. The Sprague Dawley rats were fed the experimental diets 3-4 wks prior to the conception. Experimental diet consisted of 10% fat(wt/wt) which were safflower oil(SO, poor in $\omega$3 fatty acids), mixed oil(MO, P/M/S ratio=1.03:1.45:1,$\omega$6/$\omega$3 ratio=6.3) and mixed oil supplemented with vitamin E(ME:MO+500mg vitamin E/kg diet). At 3 and 9 weeks of age of the newborn rats, frontal cortex(FC), corpus striatum(CS), hippocampus(H) cerebellum(CB) were dissected out from the whole brain. Activities of glutathione peroxidase(GSH-P(sub)x, superoxide dismutase(SOD) concentrations of malondialdehyde(MDA) were mesaured. Dietary fatty acids were not effective in antioxidative system for rat brain. However, when vitamin E was supplemented to the diet(ME), the activities of GSH-P(suh)x tended to increase in comparison to MO group. Therefore, the activites of GSH-P(suh)x of FC and H at the age of 3 weeks showed significant differences(p<0.05). The activities of Total-SOD tended to decrease in ME group compared to MO group. There were significant differences(p<0.05) in FC and CS at the age of 3 weeks. The activities of Mn-SOD tended to increase and Cu, Zn-SOD tended to decrease when vitamin E was supplemented. The activity levels of antioxidative enzymes at the age of 3 weeks and 9 weeks were similar. This suggested that the activity level of antioxidative enzymes reached to the adult level at the age of 3 weeks which is the end point of lactation period. The concentrations of MDA were not altered by experimental diets. When the activities of antioxidant enzymes were compared, the activities of antioxidant enzymes were the lowest in H and FC. In conclusion, the antioxidative system were not altered by dietary fatty acid at the age of 3 weeks and 9 weeks, but the supplementation of vitamin E altered the antioxidative systems. A Study of the Generation of Transgenic Chickens That Express Human SOD-3 Protein (사람의 SOD-3 단백질을 발현하는 형질전환 닭 생산 연구)

  • Lentiviral vector system is efficient vehicles for the delivery of exogenous genes, and it is generally used in the generation of transgenic chickens. In this study, we used recombinant lentiviral vectors to generate transgenic chicks that express the human superoxide dismutase-3 gene driven by the chicken ovalbumin promoter. It is well known that superoxide dismutases(SODs) are believed to play a crucial role in protecting cells against oxygen toxicity. There are three forms of SOD proteins: cytosolic Cu-Zn SOD, mitochondrial Mn SOD, and extracellular SOD(SOD-3). The recombinant lentivirus containing the human SOD-3 gene was injected into the subgerminal cavity of freshly laid eggs. Subsequently, the embryos were incubated to hatch using phases II and III of the surrogate shell ex vivo culture system. From 341 injected embryos, the 78 chicks hatched after 21 days incubation. The hatched chicks were screened for the human SOD-3 gene by using PCR. Two of 47 male chickens that survived to sexual maturity contained the human SOD-3 gene in their semen. These results showed that our transgenic chicken generation system was completely established.

Metal Concentrations in Lungs of Coal Workers' Pneumoconiosis Patients (탄광부 진폐증자의 폐조직내 금속농도)

  • The metal concentrations in lungs from 12 coal workers' pneumoconiosis(CWP) patients and 6 controls, who were not exposed occupationally to coal mine dust and metals during their life time, were analyzed by atomic absorption spectrophotometry. 1. Copper, lead, nickel, magnesium, manganese, zinc and iron concentrations in lungs of CWP patients were $1.10{\pm}0.088$, $1.12{\pm}0.068$, $0.22{\pm}0.020$, $113.7{\pm}1.31$, $0.19{\pm}0.012$, $10.2{\pm}1.54$, $426.7{\pm}2.63{\mu}g/g$ wet weight. 2. Copper, lead, nickel, magnesium, manganese, zinc and iron concentrations in lungs of controls were $1.10{\pm}0.013$, $0.85{\pm}0.007$, $0.10{\pm}0.008$, $87.6{\pm}1.29$, $0.18{\pm}0.005$, $10.6{\pm}1.44$, $164.9{\pm}3.29{\mu}g/g$ wet weight. 3. The ratios of concentrations for copper, lead, nickel, magnesium, manganese, zinc, and iron in lungs for CWP patients and controls were 1 : 1, 1.32 : 1, 2.20 : 1, 1.30 : 1, 1.06 : 1, 0.92 : 1, 2.58 : 1, respectively. There were significant differences in concentrations of lead, nickel, magnesium, iron by group(p<0.05). 4. There was no significant difference in metal concentrations of right upper lobe, right lower lobe, left upper lobe and left lower lobe for both CWP patients and controls (p>0.05, p>0.05). 5. In CWP patients lead was well correlated with nickel showing a rank correlation coefficient of 0.533, and zinc was correlated with copper showing a rank correlation coefficient of 0.476. 6. The concentrations of copper, nickel, maganese, and zinc in Korean CWP patients were lower than those in foreign CWP patients.

Effect of Chelating Agents on the Grwoth of Chinese cabbage and Availability of Nutrients in Plastic Film House Soils (시설재배지에서 킬레이트제 처리가 양분 유효도와 배추생육에 미치는 영향)

  • This study is conducted to evaluate the effects of chelating agents for improving plant growth and reusing accumulated nutrients in soils of plastic film house. Two experiments were carried out at follows: i) The incubation test was conducted using soils treated with 0, 300 mM of EDTA and DTPA to examine the availability of nutrients. ii) For the pot test, chinese cabbages were cultivated in soils with 0, 0.1, 0.5, 1, and 5 mM of EDTA and DTPA to examine the impacts of plant growth response. The application of chelating agents increased ther availability of soil nutrients in the following order: DTPA > EDTA > control. Inorganic concentration of chinese cabbages in DTPA treatments consderably increased in nitrogen, phosphate, iron and aluminium contents than that of the other treatments. The optimal concentration of DTPA for vigorous plant growth as 0.5 mM. Thus, DTPA was more effective than other chelating agents for healty growth of cabbages and the availability of nutrients accumulated in plastic film house.

  • Quartz veins from the Jukwangri area of Hwawon-myeon are epithermal quartz veins that are filling the NW or NE-trending faults within Precambrian metasedimentary rocks. Based on their prolongation and ore grades, No. 1 quartz vein can be traced for about 200 m and varies 0.1 to 3 m in thickness. Mineralization of No. 1 quartz vein can be divided into hypogene and supergene stages. Hypogene stage is associated with hydrothermal alteration minerals(phyllic and argillic zones) such as illite, sericite and sulfides such as pyrite, arsenopyrite, sphalerite. chalcopyrite, galena, argentian tetrahedrite. Supergene stage is composed of Fe-Mn oxide, Zn-Fe oxide and Pb oxide. Fluid inclusion data indicate that homogenization temperature and salinity of hypogene stage range from 187 to $306^{\circ}C$ and ken 0.0 to 6.2 wt.% eq. NaCl, respectively. They suggest that ore forming fluids were progressively cooled and diluted from mixing with meteoric water. Oxygen($-4.1{\sim}4.1%o$) and hydrogen($-107{\sim}-88%o$) isotope com-positions indicated that hydrothermal fluids were derived from meteoric and evolved by progressive mixing with meteoric water during mineralization.

Effect of Various Divalent Ions on the Calcium Current of Adrenal Medullary Chromaffin Cells in the Rat

  • It is well known that chromaffin cells of adrenal medulla secrete catecholamine in response to sympathetic nerve activation and the influx of $Ca^{2+}$ through the voltage dependent $Ca^{2+}$ channels (VDCC) in the cell membrane do a major role in this secretory process. In this study, we explored the effect of divalent cations on VDCC of rat chromaffin cells. Rat (Sprague-Dawley rat, 150-250 gm) chromaffin cells were isolated and cultured. Standard giga seal, whole cell recording techniques were employed to study $Ca^{2+}$ current with external and internal solutions that could effectively isolate VDCC currents $(NMG\;in\;external\;and\;TEA\;and\;Cs^{2+}\;in\;internal\;solution)$. The voltage dependence and the inactivation time course of VDCC in our cells were identical to those of bovine chromaffin cells. A persistent inward current was first activated by depolarizing step pulse from the holding potential (H.P.) of -80 mV to -40 mV, increased to maximum amplitude at around +10 mV, and became smaller with progressively higher depolarizing pulses to reverse at around +60 mV. The inactivation time constant $(\tau)$, fitted from the long duration test potential (2 sec) was $1295.2{\pm}126.8$ msec $(n=20,\;1\;day\;of\;culture,\;mean\;{\pm}S.E.M.)$ and the kinetic parameters were not altered along the culture duration. Nicardipine $(10\;{\mu}M)$ blocked the current almost completely. Among treated divalent cations such as $Cd^{2+},\;Co^{2+},\;Ni^{2+},\;Zn^{2+}\;and\;,Mn^{2+},\;Cd^{2+}$ was the most potent blocker on VDCC. When the depolarizing step pulse from -80 mV to 10 mV was applied, the equilibrium dissociation constant $(K_d)$ of $Cd^{2+}\;was\;39\;{\mu}M,\;K_d\;of\;Co^{2+}\;was\;100\;{\mu}M\;and\;K_d\;of\;Ni^{2+}];was];780{\mu}M.$ The principal findings of this study are as follows. First, the majority of $Ca^{2+}$ channels in rat chromaffin cells are well classified to L-type $Ca^{2+}$ channel in the view of kinetics and pharmacology. Second, all divalent cations tested could block the $Ca^{2+}$ current and the most potent blocker among the tested was $Cd^{2+}$.

A Study on Mineral and Alginic acid Contents by Different Parts of Sea Mustards(Undaria pinnatifida) (미역의 부위별 무기성분 및 알긴산 함량에 관한 연구)

  • This study was conducted to examine the proximate composition, alginic acid and mineral contents and the types of calcium and magnesium in the parts(frond, stipe, sporophyll) of sea mustards. Carbohydrates and the crude fat was the highest in sporophyll. The contents of alginic acid were $25.9{\sim}32.2%$. Total alginic acid(32.2%) and insoluble alginic acid in water(27.7%) was e highest in sporophyll. Calcium and magnesium was the highest in frond. Phosphorous, potassium and sulfur was the highest in sporophyll. Iron, zinc and mangane was the highest in frond. Ca/P ratio in frond and stipe was about $1.7{\sim}1.8:1$ levels. Calcium and magnesium soluble in sodium chloride was the highest in sea mustards. Calcium and magnesium soluble in water was the highest in frond. Calcium and magnesium soluble in hydrochloric acid was the highest in sporophyll.

Physicochemical Characteristics of Barley Powder Produced from Different Cultivars and with Different Degrees of Milling (품종 및 도정도별 보릿가루의 이화학적 특성)

  • This study investigated changes in physicochemical characteristics of barely powder produced from different cultivars (Dahan, Hinchalssal, Heugkwang, Heugnurl and Boseokchal) and with different degrees (15, 19, 23, and 27%) of milling. Crude protein content increased with decreasing milling degrees, with content ranges of 7.38-10.09, 8.01-10.58, 9.47-11.62, and 9.03-12.08%, respectively. Further crude lipid and ash content increased with decreasing milling degree. The crude lipid content was highest in Dahan-milled barley, while crude ash content was highest in Hinchalssal-milled barley. As the milling degree decreased, palmitic and stearic acid composition decreased and oleic and linoleic acid composition increased. Major minerals present in the barley powder were Ca, Fe, K, Mg, Mn, Na, and Zn. K and Mg content increased with decreasing milling degree. These results indicate that further studies are needed to optimize the cultivar usage and degree of milling for manufacture of various barley products.

Serum Mineral and Haematobiochemical Profile of Microfilariae Infected Cattle in India: Its Effects on Production and Therapy

  • A survey was under taken of six district of Northern India viz. Bareilly, Pilibhit, Udham Singh Nagar, Nainital, Almora and Rampur. The age, breed, sex and physiological status recorded. A total number of 854 cattle examined out of which lactating (274 cases), non lactating (302 cases) heifers (128 cases), calves (82 cases) and adult male (68 cases) were examined. An incidence of 4.92 percent (42) of microfilarisis was recorded. The highest prevalence was observed in Rudrapur District of Udham Singh Nagar (33.33%, 4/12), followed by Lalkaun in Nanital District (21.74%, 10/46), Rampur (12.50%, 2/16), Bareilly (8.16%, 8/98) and Pilibhit (1.22%, 1/82). No infection was observed in Almora region. Amongst 854 cattle of different group incidence was highest in adult male (12.20%, 10/82), followed by non lactating (3.82%, 12/314) and lactating (2.70%, 2/74), (7.64%, 12/157) was found in Heifers. For haemeto-biochemical, serum minerals estimations and therapeutic study 32 animals suffering from filariasis and 18 healthy animals were taken. 16 animals were treated with ivermectin $@200{\mu}g/kg $ body weight. Effect of this disease on production has also been estimated for which body weight and milk production was observed. The main clinical manifestations observed were anaemia, loss of appetite, debility, oedematous swelling especially in the abdominal region, increased heart rate, and respiration rate. Haematological changes indicated decrease in hemoglobin, total erythrocyte count, packed cell volume, erthrocyte fragility and neutrophil, whereas there was significant increase in erythrocytes sedimentation rate (ESR), total leukocyte count (TLC), lymphocyte and eosinophils. Biochemical changes showed significant reduction in the values of serum albumin, A : G ratio, where as there was significant increase in blood glucose, blood urea nitrogen (BUN), globulin, total lipid, total cholesterol, phospholipids, serum bilirubin. Serum mineral profile also altered markedly, which indicate a significant decrease in Ca, Cu, Fe, Zn, and Mn with increase value of Na and Cl. There was no significant change in P and K values. Enzyme pattern in micro filaria infected animal indicated increased level of AST, ALT, alkaline phosphatase, ornithine carbamyl transferase, sorbitol dehydrogenase, glutamate dehydrogenase, isocitric dehydrogenase and lactate dehydrogenase. In blood gas values and acid/base balance, there was an increase in $PVCo_2$ and $PVo_2$. It has been observed that microfilaria infected cattle showed decrease in body weight and milk production. Animal treated with ivermectin showed the return of these above values toward normalcy.
