Quality characteristics and antioxidant activities of cheonggukjang prepared with soybean and lotus seeds

  • Cheonggukjang (CGJ) is a famous traditional Korean food that is typically produced by fermenting steamed soybean seeds and has a unique flavor and taste. The objective of this study was to evaluate the effect of addition of lotus seeds on the quality and antioxidant activities of CGJ. Color value, 1,1-diphenly-2-picrylhydrazyl radical scavenging potential, and the amounts of total polyphenol, total flavonoid, mineral, and free amino acid were evaluated. The CGJ sample produced with lotus alone or a mixture of soybean and lotus produced in Korea showed relatively high antioxidant potential. The amount of essential and total free amino acids was also high in the sample prepared with lotus seeds grown in Korea. On the other hand, the total mineral content was low in the lotus-based samples. The results indicated that a mixture of an equal proportion of soybean and lotus seeds could be a good option to prepare nutritious CGJ.

  • $eta$-Sialon synthesis was investigated via the simulataneous reduction and nitriding of Wando pyrophyllite. When Wando pyrophyllite-graphite-$Si_3N_4$ seed mixture was heated at 135$0^{\circ}C$ for as long as 10 hours in 80% $N_2$-20%$H_2$ atomsphere $eta$-$Si_3N_4$ solid solution was mainly formed together with a small amount of $\alpha$-$Si_3N_4$ The value z of the forming $Si_{6-x}Al_2O_2N{8-z}$ was decreased with heating time.

In Vitro and Greenhouse Evaluation of Cucumber Growth Enhanced by Rhizosphere Microorganisms (실험실내와 비닐하우스에서 근권 미생물에 의한 오이 생육증진의 검정)

  • We developed an in vitro assay method for evaluating plant growth promotion and providing an evidence that the growth promotion is rendered by growth enhancing factors. The amendment of culture filtrates of Trichoderma harzianum T95 and Gliocladium virens G872 and G872B in Murashige and Skoog (MS) agar medium enhanced the cotyledon growth of cucumber in terms of fresh weight and primary leaf area of cucumber cotyledon cuttings, of which the treatment of G. virens G872B was the most effective. The mycelial culture filtrate of G872B was more effective in the growth promotion than its conidial germling filtrate. The addition of 1% sucrose in MS mineral medium with 0.1% culture filtrates of the antagonists (T95 and G872B) was optimum for enhancing the effect of the filtrates on the growth of cotyledon cuttings in vitro. When cucumber seeds treated with G872B, Pseudomonas putida Pf3 or the G872B-Pf3 mixture were planted in a greenhouse, the rate of seed germination, biomass of shoot and root, and yield of cucumber fruits were increased, especially by G872B or the G872B-Pf3 mixture. Correspondingly, cucumber fruit yields in early to middle-cycles of harvest were significantly greater in the plots of G872B than the control and Pf3-treated plots, and the final yield was highest in the plots of the G872B-Pf3 mixture applications.

Nutrient intake, digestibility and performance of Gaddi kids supplemented with tea seed or tea seed saponin extract

  • Objective: An experiment was conducted to determine the nutrient intake, digestibility, microbial protein synthesis, haemato-biochemical attributes, immune response and growth performance of Gaddi kids fed with oat fodder based basal diet supplemented with either tea seed or tea seed saponin (TSS) extract. Methods: Eighteen male kids, $7.03{\pm}0.16$ months of age and $19.72{\pm}0.64kg$ body weight, were distributed into three groups, $T_0$ (control), $T_1$, and $T_2$, consisting of 6 animals each in a completely randomized design. The kids were fed a basal diet consisting of concentrate mixture and oat fodder (50:50). Animals in group III ($T_2$) were supplemented with TSS at 0.4% of dry matter intake (DMI), and group II ($T_1$) were supplemented with tea seed at 2.6% of DMI to provide equivalent dose of TSS as in $T_2$. Two metabolism trials were conducted, 1st after 21 days and 2nd after 90 days of feeding to evaluate the short term and long term effects of supplementation. Results: The tea seed ($T_1$) or TSS ($T_2$) supplementation did not affect DMI as well as the digestibility of dry matter, organic matter, crude protein, neutral detergent fibre, and acid detergent fibre. Nutritive value of diet and plane of nutrition were also comparable for both the periods. However, the average daily gain and feed conversion ratio (FCR) were improved (p<0.05) for $T_1$ and $T_2$ as compared to $T_0$. The microbial protein supply was also higher (p<0.05) for $T_1$ and $T_2$ for both the periods. There was no effect of supplementation on most blood parameters. However, the triglyceride and low density lipoprotein cholesterol levels decreased (p<0.05) and high density lipoprotein-cholesterol level increased (p<0.05) in $T_2$ as compared with $T_0$ and $T_1$. Supplementation also did not affect the cell mediated and humoral immune response in goats. Conclusion: Tea seed at 2.6% of DMI and TSS at 0.4% DMI can be fed to Gaddi goats to improve growth rate, FCR and microbial protein synthesis.

The effect of Camelina sativa (L.) cv. Crantz Seed Yield according to Seeding Rate and Chemical Fertilization (파종량과 비료 시용이 봄 재배용 카멜리나 종자 생산량에 미치는 영향)

  • The objective of this study was comparison of seed yields according to different seeding rates, seeding time and application of chemical fertilizations in Yanji region of China. The experiment was conducted on commercial farmland at Yanji region in 2013. All the experimental fields were designed following randomized block design with 3 replicates. The plant spacing was applied as $65{\times}1cm$ distance. To check the effect of seeding rate, two different seeding rate, 0.2 kg/0.1ha and 0.5 kg/0.1ha were applied. The higher seed yield was observed in low seeding rate (0.2 kg/0.1ha). The application of fertilizer (mixture of N and P) showed different results following different experimental fields. However, there were little positive effects following fertilizer application into commercial farmland. When we compare seed yields between two different seeding dates, $4^{th}$ May and $29^{th}$ May, the earlier seeding date ($4^{th}$ May) showed higher seed yields. In considering these results, low seeding rate and early seeding time is important for getting high camelina seed yields.

Antioxidant Activity of Grape Seed Ethanol Extract (포도종실 에탄올 추출물의 항산화 활성)

  • To examine the antioxidant activity of grape seed ethanol extract, the antioxidative index (AI) by the active oxygen method (AOM) and peroxide value (POV) of linoleic acids containing the extract at levels of 100, 500, 1000 ppm was measured during storage at 6$0^{\circ}C$ for 12 days. When comparing with BHT, the extract at levels of 500 ppm showed similar or better antioxidant activity (AI: 2.25, POV: 57 meq/kg oil) than that (AI: 1.21, POV:58 meq/kg oil ) of BHT at 200 ppm level. The mixture of 500 ppm of the extract and 500 ppm of ascorbic acid showed intense synergistic antioxidant activity (AI: 6.21, POV: 14 meq/kg oil) compared with using 1000 ppm of the extract only (AI: 3.39, POV 43 meq/kg oil). Also to determine the feasibility of using the extract for natural antioxidant, the oxidative stability of roasted peanut and Ramyon was investigated by measuring the POV of crude oils from the samples stored at 60"C for 18 days. The oxidative stability of roasted peanut and Ramyon seemed to be enhanced by treatment with the extract at level of 1000 ppm, especially with the 1 : 1 mixture of extract and ascorbic acid. This study suggested that grape seed ethanol extract could be used as the natural antioxidant for the improvement of overall oxidation stability of fat containing foods.oods.

Dry Matter Yield and Forage Quality at Mixture of Annual Legumes and Italian ryegrass on Paddy Field (논에서 이탈리안 라이그라스와 두과 사료작물 혼파에 따른 생산성 및 사료가치 비교)

  • This experiment was conducted to compare the dry matter yield and forage quality according to the seed mixture of annual legumes and Italian ryegrass (Lolium multiflorum, IRG) on paddy field at the experimental field of Jeollanam-Do Agricultural Research and Extension Services from 2007 to 2008. The five treatments used in this experiment were IRG single (cv. Kowinearly), IRG+Chinese milk vetch mixture, IRG + Crimson clover (cv. Linkarus) mixture, IRG + Austrian winter pea (Pisum sativum subsp. arvense cv. forage Pea) mixture and IRG + Hairy vetch (Vicia villosa. cv. Oregon Common) mixture. And the mixing ratio of Italian ryegrass + annual legumes were 70 : 30 as based mono seed rate. The dry matter (DM) percentage at harvest was 23.6~26.8%. The DM percentage among IRG single and mixture treatments were similar. The yields of fresh and CP were high in IRG + Forage pea mixtures as a 40,100 kg and 625 kg per ha, respectively (p<0.05). The yields of dry matter were high in IRG + Forage pea and IRG + hairy vetch pea mixtures as a 9,470 kg and 9,500 kg per ha, respectively (p<0.05). But the forage quality did not show difference between Italian ryegrass mono-culture and annual legumes mixture. The Av. $P_2O_5$ in IRG+Forage pea mixture was 78 mg/kg and concentration of K were 0.52~0.88. In conclusion, the mixture of IRG and Forage pea, dry matter yield, protein yield increased, and was effective in enhancing the stability of cultivation.

A Study on the Wild Exochorda serratifolia for Landscape Horticuitural Cultivation (야생 가침박탈(Exochorda serratifolia)의 조경원예화에 관한 연구)

  • Exochorda serratifolia, broad-leaved shrub has beautiful flowers and is resistant to cold, shade and decease. New this native plant is considered to be worth being exploited as the outstanding plant for landscaping and horticulture. So this study was executed to utilize Exochorda serratifolia as the planting material for landscaping and horticulture through the survey of its habitat environment and the experimennt of its seed physiology and germination, vegetative propagation, culture and utilization, etc.. The results are as follows ; 1. The color of the flower is white, blooming in the early and middle of May and the seeds ripen late in Sep.. 2. The elevation, elevation, gradient and direction of the native habitat were 250m, 20-25$^{\circ}$and northern side respectively. 3. The soil pH of the natural habitat was 5.3 and soil fertility was poor. 4. Exochorda serratifolia appeared as indicator within P. densiflora community and its neighboring species were Q. mongolica, Q. dentata, R. mucronulatum, L. obtusiloba, E. oxyphillus, C. heterolphylla, var thunbergii, etc.. 5. The optimum temperature for seed germination was found at 20$^{\circ}C$ and the longer the stratification period at 5$^{\circ}C$ was, the lower the germination rate was. 6. The treat merits of GA and Kinetin increased the seed germination rate, especially under the dark condition but their high concentrations decreased the seed germination rate. 7. The rooted rate of the greenwood cutting was the highest at IBA 100ppm plot of vermiculite bed but its high concentration decreased the rooted rate conspicuously. 8. In the tissue culture, the each 1.0 ppd plot of NAA, Kinetin and NAA 1.0ppm + Kinetin showed the best growth. And the mixture of NAA 1.0ppm and Kinetin showed better growth than the single treatment of NAA or Kinetin did. 9. Transplanted Exochorda serratifolia showed healthy growth with shaded environmental condition(42.1% light intensity), therefore they can be cultivated as a shade tolerant landscape plant. 10. It was considered that Exochorda serratifolia was applicable to group planting at shade places or under trees in parks, homes, etc..

Optimization of Antimicrobial Activity Against Food-borne Pathogens in Grapefruit Seed Extract and a Lactic Acid Mixture (식품위해미생물에 대한 자몽종자 추출물과 젖산 혼합물의 항균효과 최적화)

  • Response surface methodology (RSM) is frequently used for optimization studies. In the present work, RSM was used to determine the antimicrobial activitiesof grapefruit seed extract (GFSE) and a lactic acid mixture (LA) against Staphylococcus aureus, Bacillus cereus, Escherichia coli, Salmonella typhimurium, Pseudomonas fluorescens, and Vibrio parahaemolyticus. A central composite design was used to investigate the effects of independent variables on dependent parameters. One set of antimicrobial preparations included mixtures of 1% (w/w) GFSE and 10% (w/w) LA, in which the relative proportions of component antimicrobials varied between 0 and 100%. In further experiments, the relative proportions were between 20% and 100%. Antimicrobial effects against various microorganisms were mathematically encoded for analysis. The codes are given in parentheses after the bacterial names, and were S. aureus ($Y_1$), B. cereus ($Y_2$), E. coli ($Y_3$), S. typhimurium ($Y_4$), P. fluorescens ($Y_5$), and V. parahaemolyticus ($Y_6$). The optimum antimicrobial activity of the 1% (w/w) GFSE:10% (w/w) LA mixture against each microorganism was obtained by superimposing contour plots ofantimicrobial activities on measures of response obtained under various conditions. The optimum rangesfor maximum antimicrobial activity of a mixture with a ratio of 1:10 (by weight) GFSE and LA were 35.73:64.27 and 56.58:43.42 (v/v), and the optimum mixture ratio was 51.70-100%. Under the tested conditions (a ratio of 1% [w/w] GFSE to 10% [w/w] LA of 40:60, and a concentration of 1% [w/w] GFSE and 10% [w/w] LA, 70% of the highest value tested), and within optimum antimicrobial activity ranges, the antimicrobial activities of the 1% (w/w) GFSE:10% (w/w) LA mixture against S. aureus ($Y_1$), B. cereus ($Y_2$), E. coli ($Y_3$), S. typhimurium ($Y_4$), P. fluorescens ($Y_5$), and V. parahaemolyticus ($Y_6$) were 24.55, 25.22, 20.20, 22.49, 23.89, and 28.04 mm, respectively. The predicted values at optimum conditions were similar to experimental values.

Interlaboratory Study for Cryptosporidium and Giardia Test Methods in Water (정수장 원생동물 분석방법의 국내 실험실간 교차시험 연구)

  • This interlaboratory study was designed to evaluate protozoan test methods in water and to predict the major causes of deviation of the methods. Each of four laboratories with previous experience of protozoa analysis in water participated, and met the initial performance criteria of EPA 1623 method provided. The protozoan analysis procedure consists of filtrations, concentration, immunomagnetic separation, dyeing (staining) and counting with observation. We tested three different filtration equipments: capsule filter for 10 L of surface water, and high volume (HV) capsule filter and membrane filter for 100 L of finished water. When the recovery of each step of the procedure was evaluated with EasySeed, the commercial stock of each 100 Cryptosporidium and Giardia, immunomagnetic separation and filtration step were the most crucial steps affecting the stability of the recovery, especially for Cryptosporidium. There was no significant difference of recovery among the filtration methods. Recovery of protozoa from source water were evaluated with spiked EasySeed as matrix tests. The recoveries of Giardia increased significantly in the matrix tests compared those in the deionized water. We also applied red stained mixture stocks of Cryptosporidium and Giardia called ColorSeed as internal standards of water sample tests. The recoveries of both EasySeed and ColorSeed in samples tested were within the range of the criteria, however, the Giardia recoveries using ColorSeed decreased significantly. Further optimization study with ColorSeed will be necessary, considering the convenience of using the internal standard without additional sample analysis. The significant factors of the recovery variation were identified as the differences of laboratories as well as water quality and type of the stock for spiking. The results of this study emphasize the importance of the quality assurance program for protozoan analysis lab in water.
