Synthesis of d- and l-Form of $^{99m}Tc$-HMPAO, and Comparison of Brain Uptake ($^{99m}Tc$-HMPAO의 광학이성체 d-, l-Form의 합성과 뇌섭취율 비교)

  • Purpose: $^{99m}Tc$-HMPAO is a radiopharmaceutical for imaging cerebral blood flow. HMPAO (RR, SS)-4.8-diaza-3,6,6,9-tetramethylundecan-2,10- dione bisoxime) has three stereoismers such as, meso-. d-, and l-HMPAO. Techentium complexes of meso-HMPAO and d,l-HMPAO are known to have different in vivo brain uptakes. In this study, enantiomers of HMPAO (d-HMPAO and l-HMPAO) were separated from d,l-HMPAO. These enantiomers were labeled with $^{99m}Tc$ and the biodistribution studies were performed in mice. Materials and Methods: An intermediate imine product was produced from 2,3-butanedione monooxime and 2,2-dimethyl-1,3-propanediamine (54% yield) and was reduced into a mixture of three isomers (35% yield). The meso-isomer was separated from d,l-mixture by repeated fractional crystallization (11 % yield). The d- and l-enantiomers were subsequently separated by co-crystallization with optical isomers of tartaric acid (25% and 5% yield. respectively). Each enantiomeric HMPAO was labeled with $^{99m}Tc$ by reacting with $SnCI_2{\cdot}2H_2O\;and\;^{99m}Tc$-pertechnetate. Biodistribution study was performed 1 hr after tail vein injection to ICR mice. Results: Radiochemical purities of each compound were over 80%. In biodistribution study. the brain uptakes of d,l- d- and l-form were 1.34, 1.12 and 1.67% ID/g, respectively. In case of l-lsomer the brain uptake was higher (1.5 fold) than d-isomer. Conclusion: We successfully purified each enantiomeric HMPAO. In biodistribution study of stereoismers of $^{99m}Tc$-HMPAO in mice, l-HMPAO may show better brain image than d,l-HMPAO which was supplied in a commercial kit.

Preparation and Characterization of Jochung, a Grain Syrup, with Apple (사과 첨가 조청의 제조 및 특성)

  • This study was performed to investigate the effect of apple and maltitol as ingredients on the quality of Jochung, a grain syrup. Four kinds of Jochung products were prepared from steamed-rice, apple juice, heated-apple sarcocarp (at $70^{\circ}C$, 60 min), and a mixture (sarcocarp : maltitol=5:1, w/w) by saccharifying (at $55^{\circ}C$, 8 hrs) with a malt (100 g/500 g rice), mixing the ingredients (steamed-rice : ingredient=5:5, w/w), filtering, and heating the filtrate (at $95^{\circ}C$, 2 hrs): product (A) with apple juice added before saccharified, product (B) with apple juice added after saccharified, product (C) with heated-apple sarcocarp added after saccharified, and product (D) with the mixture added after saccharified. The product (D) had the lowest pH value ($4.60\pm0.01$) of any other products. The contents of reducing sugar and total phenolic compound were the highest in the product (A) among all the products, which comprised $68.10\pm6.71$% and $7.36\pm0.85$ mg/g, respectively, resulting in good quality. The solidity and the dextrose equivalence had the highest value in the product (B) and the product (C), respectively. The malic acid content ($4.10\pm0.02$%) of the product (D) was the highest of any other organic acids identified by HPLC. Hunter L, a, and b values of the product (D) were the highest compared to other products. In sensory evaluation, the product (A) had generally higher score in all sensory attributes. It was concluded from the chemical and sensory evaluation that adding the apple juice before saccharified might be an effective method for manufacturing good quality rice-Jochung.

Rare Earth as a Feed Additive for Broiler Growth (육계의 성장 촉진을 위한 사료 첨가제로서의 희토(稀土))

  • In order to study the effect of feeding rare earth(RE) on growth of broiler chicks, feed conversion ratio, and probable harmfulness of feeding high levels of RE, two feeding trials with broiler chicks were conducted; one using a commercial broiler compound feed and the other using a self mixed feed excluding any growth stimulating feed additives. The the first trial used three hundred sixty of one day old Cobb broiler chicks for six levels of dietary supplementation of RE : 0, 100, 200, 400, 800 and 1,600 mg/kg. There were significant effect of RE stimulating broiler chick growth and improving feed conversion ratio, The dietary supplementation of RE at a level of 100 mg/kg was the best of all and increased body weight gain by 8.9% (p<0.05). Chicks fed RE at a level of 1,600 mg/kg grew as good as chicks fed feeds without RE and did not show my abnormalities. The second feeding trial was conducted in the same manner as the first trial using Ross broiler chicks and self-mixed experimental diets supplemented with RE at levels of 0, 50, 100 and 150 mg/kg. Differently from Results of the first trial, body weight gain and feed conversion ratio of were the best at the level of 50 mg/kg RE supplementation(p<0.05). In this trial all the birds fed RE showed significantly lower ratio of abdominal fat against live weight than those of the control group birds. Overall, it can be said that dietary supplementation of RE will improve broiler growth and feed conversion ratio and the proper dietary level would be $50\sim100mg/kg$. These results suggested that the proper level of RE for broiler chicks would be 50 to 100 mg/kg and its effectiveness is varied depending upon RE mixture. There appears abdominal fat of broiler chicks is decreased by feeding RE but further investigation is in need.

  • In order to elucidate the effect of phenobarbital sodium(PB) and 3-methylcholanthrene(3-MC) on metabolism in vitro and toxicity of $^{14}C$-carbofuran in rat, they were administered by the chemicals, alone or in combination, and their survival ratios and metabolites were investigated. The $LD_{50}$(96 hrs) value of carbofuran to rats was 6.9 mg/kg. The toxicities of the major metabolites were in the decreasing order of 3-hydroxycarbofuran, 3-ketocarbofuran, 3-hydroxycarbofuran phenol and were much lower than that of the parent compound. When the rats were orally administered by the dose of carbofuran alone, 8.4 mg/kg, the survival ratio was 0%, whereas that was raised up to $60{\sim}80%$ with 20 mg/kg of PB or 3-MC, and 100% with 60 mg/kg of PB or 3-MC. Their metabolism in vitro occurred in the microsomal fraction. In case of carbofuran alone, the major metabolite was 3-hydroxycarbofuran. When carbofuran with PB or 3-MC, on the other hand, was treated, it was 3-ketocarbofuran. In addition, when the co-factor(NADP+G-6-P+G-6-P-DG) was added to the microsomal fraction(phase I system), and a mixture of NADPH+GSH to the 105,000g supernatant(phase II system) taken by carbofuran alone, each metabolites were produced by the maximum levels, respectively. In case of the carbofuran treatment with PB or 3-MC, the microsomal fraction of phase I system produced the maximum levels of metabolites, as in the treatment of carbofuran alone, whereas the 105,000g supernatant supplemented with the co-factor NADPH+FAD(phase II system) was brought about the maximum production of metabolites. The ratio of the formation of metabolites was 2 to 3 times higher in the combined treatment of carbofuran with PB or 3-MC than in the treatment of carbofuran alone.

Effect of Aeration Rates on Emissions of Oxygen and Sulfur compound gases during Composting of Dairy Manure (우분(牛糞) 퇴비화시(堆肥花時) 공기주입률(空氣注入率)이 산소 및 황화합물 가스 배출(排出)에 미치는 영향(影響))

  • This experiment used the enclosed bench-scale reactors was conducted to find out optimal aeration rate for reducing the emission of odors and producing the good-quality compost with the mixture of dairy manure and rice straw. The reactors with gas sampler were aerated at four different rates of 0.09, 0.18, 0.90 and $1.79l\;min^{-1}kg^{-1}$dry solids for 574 hours. The oxygen content within composting pile instantly decreased after aeration. Oxygen limitation(below 15%) in the treatments of $0.90l\;min^{-1}kg^{-1}$ and less was exponentially negative relationship with aeration rates and in the range of 35 to 300 hours after aeration. However, the treatment of $1.79l\;min^{-1}kg^{-1}$ didn't show the oxygen limitation. The oxygen consumption rate and the cumulative amount of oxygen consumed by different aeration rates was ranged in $0.80{\sim}1.57O_2g\;h^{-1}\;kg^{-1}VS^{-1}$, $460{\sim}900O_2g\;kg^{-1}VS^{-1}$, respectively, and they were high in the order of 0.90, 1.79, 0.18, $0.09l\;min^{-1}kg^{-1}$. The maximum oxygen consumption rate was estimated in the range of $1.2{\sim}1.3lmin^{-1}kg^{-1}$. The emission concentrations of sulfur compounds such as hydrogen sulfide, sulfur dioxide and methylmercaptan were remarkably high in the initial composting time. Then they were rapidly decreased with the passing of composting time and clearly with increasing aeration rates. Their average concentrations were in the range of 0.03~2.18, 0~0.50, $0.07{\sim}3.38mg\;kg^{-1}$, respectively and high in the order of methylmercaptan, hydrogen sulfide, and sulfur dioxide. Concentrations of sulfur compounds emitted from composting showed exponentially negative relationship at 1% statistically with the oxygen concentration. It was estimated that hydrogen sulfide and methylmercaptan suddenly increased in the level of 5% oxygen concentration and below, that they were little emitted in 15% and over but sulfur dioxide was emitted in the level of 20% oxygen.

Phytotoxicity Inducing Factors and Its Safening Methods for Benzenesulfonylurea Compound KSC-13906 (Benzenesulfonylurea계 화합물(化合物) KSC-13906의 약해발생요인(藥害發生要因) 및 경감방법(輕減方法))

  • KSC-13906 [Erythro N-{(4,6-dimethoxypyrimidin-2-yl)aminocarbonyl} -2-(2-fluoro-1-hydroxy-n-propyl) benzenesulfonamide, US Patent 5,461,025] was investigated how can control phytotoxicity fluctuation and what a good method apply to new rice herbicide. The growth inhibition was observed when the rice plants was transplanted at a shallow depth(0 - 1cm) and leaching was low(0 - 1cm/ day) from the paddy soil. KSC-13906 appeared to move readily down into the paddy soil with water by 3cm depth in the soil column(${\phi}$ 10cm) filled with loamy sand soil under 3cm/day of leaching condition. Artificial control releasing pattern, designed as treated with KSC-13906 of 9 or 18g ai/ha either at a once or daily treated dividing volume of 1/20, 1/25 and 1/30 of the total volume, increased the safety of KSC-13906 to direct seeded and transplanted rice. The safety of KSC-13906 was also enhanced when KSC-13906 was mixed with dymron. For example, the mixture of KSC-13906 and dymron effectively reduced injury of direct seeded rice plants at 18 and 500g ai/ha, respectively, treated 7 days after transplanting. However, combination of KSC-13906 and several herbicides didn't show any synergistic effetct on herbicidal activity and safening effect on rice. However, the combination of KSC-13906+dymron (9~12+250~500g ai/ha) or KSC-13906+mefenacet+dymron(9+250+250g ai/ha) controlled almost all weeds in paddy field without causing any injury to rice and thus the combination would successfully be used as an oneshot herbicide in rice culture.

Optimization of spray drying conditions of soft persimmon and milk mixture using response surface methodology (반응표면분석법을 이용한 홍시와 우유혼합물의 최적 분무건조 조건)

  • The purpose of this study was to determine the optimum spray drying conditions of soft persimmon latte using response surface methodology that is a statistical procedure used for optimization studies. A central composite design was applied to investigate the effects of independent variables, inlet temperature ($X_1$), air flow rate ($X_2$), and feed flow rate ($X_3$), on responses such as yield, water absorption index, and total phenolic compounds. Statistical analysis revealed that independent variables significantly affected all the responses. A maximum yield of 8.11 g was obtained at $90^{\circ}C$ of $X_1$, 51.82 mL/min of $X_2$ and 7.00 mL/min of $X_3$. A minimum water absorption index of 0.58 was obtained at $101^{\circ}C$ of $X_1$, 60.00 mL/min of $X_2$ and 17.00 mL/min of $X_3$. A maximum total phenolic compounds of $298.02{\mu}g/mL$ was obtained at $90^{\circ}C$ of $X_1$, 43.33 mL/min of $X_2$ and 17.00 mL/min of $X_3$. In conclusion, the best spray drying conditions were as follows: $X_1$, $90^{\circ}C$; $X_2$, 53 mL/min; $X_3$, 17 mL/min. Under those optimal conditions, the powder's yield (7.46 g), water absorption index (0.54), and the content of total phenolic compounds ($294.75{\mu}g/mL$) were estimated.

  • We measured cloud points of Poly(methylmethacrylate) (PMMA) in various solvents using the high-pressure variable volume view cell apparatus. The solvents used for dissolving PMMA were chlorodifluoromethane (HCFC-22), dimethylether (DME), 1,1,1-trifluoroethane (HFC-143a), 1,1-difluoroethane (HFC-152a) and 1,1,1,2-tetrafluoroethane (HFC-134a), and the effect of $CO_2$ concentration on the phase behavior of $PMMA+HCFC-22+CO_2$ system and $PMMA+DME+CO_2$ system was observed. PMMA was dissolved well in HCFC-22 from about 340 K, 5MPa and in DME from about 300 K, 28MPa. However, PMMA was not dissolved at all up to 423.15 K, 160MPa in the other fluorine compound such as HFC-l43a, HFC-152a and HFC-134a. PMMA+HCFC-22, $PMMA+HCFC-22+CO_2$ and PMMA+DME systems exhibit the lower critical solution temperature (LCST) behavior, however, $PMMA+DME+CO_2$ system exhibits the upper critical solution temperature (UCST) behavior. In the $CO_2$ mixture, the cloud point pressure of PMMA was increased dramatically proportional to the amount of $CO_2$ added, and from this result, it was known that $CO_2$ could be used as an antisolvent for fabricating PMMA nano-particles. And the cloud point of PMMA could be controlled by changing the concentration of $CO_2$.

Physicochemical characteristics and volatile flavor compounds of produced mixture wine with kiwi and permission fruits using wild yeast, Saccharomyces cerevisiae Y28 (야생 효모 Saccharomyces cerevisiae Y28을 이용하여 제조한 참다래-대봉감 혼합과실주의 이화학적 특성 및 향기성분)

  • The study was aimed to investigate the mixing ratio of kiwi and persimmon juices for the production of good quality wine by Saccharomyces cerevisiae Y28. Firstly, the optimum condition of rapidase treatment for the kiwi and persimmon juices was established, thereafter various mixing ratio (10:0, 9:1, 8:2, 7:3, 6:4, 5:5) of kiwi and persimmon was investigated regarding physiochemical properties and flavor compounds of wine. As the result, the optimum conditions were obtained as 0.3% rapidase for 1 h in kiwi and 0.3% rapidase for 3 h in persimmon. According to higher ration of persimmon, the pH of wines increased from 3.69 to 3.77, while the acidity of wines decreased from 2.07% to 1.51% at 14 days fermentation. The ranges of brix and reducing sugar in wines were decreased which ranges around 9.6 to 8.8 and 6.07 to 6.90 g/L, respectively, after fermentation. Major organic acid in wines were identified as tartaric acid, malic acid, and citric acid. A small amount of free sugar such as sucrose and glucose were detected in wines, but fructose was completely absent. The soluble phenolic contents were decreased that ranges around 1.00 to 1.25 g/L, in contrast, browning degree were increased ranges around 0.212 to 0.412 after fermentation. The major flavor components were identified as ethyl acetate and hydrazine, and 1,1-dimethyl. Importantly, phenylethyl alcohol was detected from the all wines that have a typical rose like flavor. But sensory test results and preference of kiwi-persimmon (7:3) mixing wine was better than the other wines.

A Study on the Seasonal Color Characteristics of Warm- and Cool-Season Grasses II. Color Characteristics and Life-span of Leaves in Turfgrasses and Cover Plants+ (난지형 및 한지형 지피식물의 엽색변화에 관한 연구 II. 엽색특성 및 엽수명연장)

  • Nitrogen fertilization and cutting practice were studied on turfgrasses and cover plants to investigate the possibility of maintaining green color during the growing season. Research also involved the effect of the nitrogen on a few morphological characteristics of leaf performance elements which might give an information to coloration and life-span of turf leaves. Treatments in the first experiment undertaken on pot included one N level: 350kgN /ha applied as compound fertilizer in split applications of one-half in mid-May and the rest both in late June and August, and four spring-summer cuts: late May, late June, late July and late August. The soil filled in pot a moderately well-drained sandy loam. In the second experiment(field observation) leaf length and width, inflorescence and flowering, and color performance were also investigated. With nitrogen fertilizer applied on turfs, desirable turf color was maintained during a period of poor coloration in specific seasons such as mid-summer for cool season grasses and late fall for warm season grasses comparing to the non-treatment. However, this was not stimulated by cutting treatment to nitrogen status existed. Cutting effect on coloration was more remarkable in both Korean lawngrass and Manilagrass than in cool season turfgrasses such as Italian rye-grass, perennial ryegrass and tall fescue. Especially down-slide of leaf color in cool season turfgrasses could he detected in mid-summer /early fall season ranging up to mid-September. In early November as well as mid-September, Italian ryegrass, perennial ryegrass and tall fes-cue retained a high level of green color as followed by nitrogen application and cutting treatment, and little detectable variation of leaf color notation between cool season turfgrasses was obtained. However, Korean la'vngrass and Manilagrass failed to retain the green color until early November. Color notations in cool season turfgrasses investigated early November on the final date of the experiment ranged from 5 GY 3/1 to 4/8 in 'Ramultra' Italian ryegrass, 'Reveile' perennial ryegrass and 'Arid' tall fescue, but those in Zoysiagrasses were 7.5 YR 4/8 in Korean lawngrass and 2.5 y 5 /6 in Manilagrass. Life-span of leaves was shorter in Italian ryegrass, perennial ryegrass and tall fescue than in beth Korean lawngrass and Manilagrass with and without nitrogen application. In general, leaves appeared in early May had a long life-span than those appeared in late April or mid-June. Nitrogen application significantly prolonged the green color retaining period in perennial ryegrass, Italian ryegrass, Korean lawngrass and Manilagrass, and this was contrasted with the fact that there was no prolonged life-span of leaves emerging in early May and mid-June in tall fescue. SPAD reading values in 48 turfs and cover plants investigated in the field trial were increasing until late June and again decreasing till September. Increasing trends of reading value could be observed in the middle of October in most of grasses. On the other hand, clovers and reed canarygrasses did not restore their color values even in October. Color differences between inter-varieties, and inter-species occurred during the growing season under the field condition implicated that selection of species and /or cultivars for mixture should be taken into consideration. In Munsell color notation investigated in the final date in the middle of November, 32 cultivars belonged under the category of 5 GY and 10 cultivars under the category of 7.5 GY. This was implying that most of cool season turfs and cover plants grown in the center zone of Korean Peninsula which are able to utilize for landscape use can bear their reasonable green color by early or mid-November when properly managed. The applicable possibilities of SPAD readings and Munsell color notation to determine the color status of turfgrasses and cover plants used in this study were discussed.
