Structure and Isolation of Antioxidative Substance Derived from Phaeodactylum tricornutum (Phaeodactylum tricornutum으로부터 항산화성 물질의 분리 및 구조)

  • The antioxidative activity of marine microalgae, Phaeodactylum tricornutum (P. tricornutum) of Bacillariophyceae, was determined by measuring radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The chloroform fraction of P. tricornutum showed strong antioxidative activity, The potential antioxidative activity of factions extracted with mixture solution of organic solvents was detected in dichloromethane : methanol (2 : 1) fraction, This fraction was further purified by preparative thin layer chromatography (PTLC) and repeated reverse-phase HPLC. On the basis of chemical and spectoscopic evidence from results obtained by UV, FT-IR, EIMS and NMR, the compound purified from P. tricornutum was identified as zeaxanthin.

A Comparison of Sampling and Analytical Methods for Airborne Isocyanates (공기중 이소시아네이트류의 측정 및 분석방법에 관한 비교연구)

  • This study was performed to evaluate accuracy and precision of filter method and impinger method for analyzin airborne isocyanates in mixture (2, 6-TDI, HDI, 2, 4-TDI, MDI). Filter method was performed using the OSHA Method 42 and impinger method using the NIOSH Method 5521. The samples were analyzed by high performance liquid chromatography-ultraviolet detector (HPLC-UVD). After the optimum operating conditions for each method were investigated, samples with various concentration levels were quantified at the conditions. The precision was expressed by the pooled coefficient of variation(C.V.) and the accuracy by overall accuracy. The results are summarized as follows: 1. The optimum condition of filter method was determined at 35/65 ACN/buffer (0.01 M ammonium acetate) in mobile phase. And in case of impinger method, it was at 30/70 ACN/buffer(0.2 M sodium acetate). The effect of concentrations of acetate on the separation of the peaks was not significant, but, the effect of ACN/buffer ratio was significant. 2. The correlation coefficients for the two methods were above 0.9 in all isocyanate compounds. Average recovery efficiencies for 2, 6-TDI, HDI, 2, 4-TDI and MDI in filter method were 92.4%, 102.6%, 87.3% and 101.0%, respectively. Those in impinger method were 106.6%, 106.7%, 99.0% and 103.6%, respectively. As a result, the recovery efficiency of impinger method was higher than those of filter method in analyzing isocyanate compounds. 3. The pooled coefficients of variations of the methods were slightly higher than expected. The overall accuracies of the methods were within $\pm 25%$ for each isocyanate compound. Since these results satisfy NIOSH criteria, the accuracy of the experiment is appropriate. 4. As seen above, impinger method is more efficient than filter method. But, there are many disadvantages in impinger method. Therefore, solid sorbent such as a glass fiber filter must be developed in order to have the high efficiency not less than that of impinger method in the future.

  • Funology is a compound word with fun and technology which means the products or activity carrying fun and technology at the same time. This is one of the key words that can explain the culture of post-digital generation amongst the post-digital age. The purpose of this study was to provide the creative ideas to develop the fashion designs showing funology concept for the 21st century. As a background of funology trend, general ideas about the post-digital age and the post-digital generation were reviewed. First, funology designs among the industrial products and the funology concepts in the advertisement were examined. And the ways of expression and the meanings of funology fashion designs were extracted by analyzing funology fashion between 2000 S/S and 2008 F/W. The external expression ways of funology fashion in the post-digital age were as follows: parody of popular images, inharmony by the exaggeration or transformation, trompe l'oeil, mixture of the styles shown in the children's wear and the objects of children, patterns with childlike or animation characters, and the graffiti art. The internal meanings contained in the funology fashion were 1) fantasm showing surrealistic funology that was based on the people's pursue of fantasy free from the everyday's severe routine 2) memorism showing nostalgic funology that was based on the pursue of vanishing fear of reality and going back to the childhood. To sum up, fashions showing funology were expressed in various ways in the post-digital age. This will provide the great inspirations for creating the new coming fashions and efficient healing contents for the cold-hearted emotions of nowaday's people.

Quantitative Analysis of Ellagic Acid in Nymphaea tetragona Georgi Root by HPLC-UVD (HPLC-UVD를 이용한 수련(Nymphaea tetragona Georgi)의 Ellagic Acid 정량 분석)

  • A high performance liquid chromatography (HPLC) method for the quantitation of ellagic acid in Nymphaea tetragona was developed for the quality control of functional cosmetic ingredient, the extract of N. tetragona. Separation and quantitation were successfully achieved with a Kromasil C18 column ($5{\mu}m$, $250mm{\times}4.6mm$, i.d.) by isocratic elution of a mixture of acetonitrile containing 0.1% trifluoroacetic acid and water containing 0.03% phosphoric acid at a flow rate of 1.0 ml/min. The UV detector was used for the detection and the wavelength for quantitation was set at 254 nm. The presence of ellagic acid in the extract was determined by comparison of retention time and spiking with authentic standard. Analytical results showed good linearity ($R^2=0.99996$) in relatively wide concentration ranges. The R.S.D. for precision test was less than 3.0%. Recovery of the compound was 98.55~101.72% with R.S.D values less than 4.0%. In conclusion, this method has been successfully applied to the determination of ellagic acid in N. tetragona.

Simultaneous Determination of Paeoniflorin and Glycyrrhizin in Sayuk-san by HPLC/DAD

  • A high performance liquid chromatographic (HPLC) method for the simultaneous determination of marker constituents, paeoniflorin and glycyrrhizin was established for the quality control of traditional herbal medicinal preparation, Sayuk-san (SYS). Separation and quantification were successfully achieved with a Waters XTerra RP18 column ($5{\mu}m$, $4.6mm\;I.D.{\times}150mm$) by gradient elution of a mixture of acetonitrile and water containing 0.03% phosphoric acid (pH 2.03) at a flow rate of 1.0 mL/min. The diode-array UV/vis detector (DAD) was used for the detection and the wavelength for quantification was set at 230 nm. The presence of paeoniflorin and glycyrrhizin in this decoction was ascertained by retention time, spiking with each authentic standard and UV spectrum. All two compounds showed good linearity ($r^2$>0.996) in a relatively wide concentration ranges. The R.S.D. for intra-day and inter-day precision was less than 7.3% and the limits of detection (LOD) were less than 55.7 ng. The mean recovery of each compound was $102.3{\sim}111.1%$ with R.S.D. values less than 4.6%. This method was successfully applied to the determination of contents of paeoniflorin and glycyrrhizin in three commercial products of SYS. These results suggest that the developed HPLC method is simple, effective and could be readily utilized as a quality control method for commercial SYS products.

  • Crude oil is complex mixture if thousands of different organic compound formed from a variety of organic materials that are chemically converted under differing geological conditions over long periods of time. Also oil composition varies according to crude source, refining, processing, handling and storage. The oil fingerprint method is application if specific knowledge of petrochemicals and use if sophisticated analytical equipment and techniques to identify the source(s) if oil pollution. KCG currently utilizes four primary analytical techniques: Gas Chromatography (GC), Fluorescence Spectroscopy(FL), Infrared Spectroscopy(IR) and Gas Chromatography mass spectrometer(GC/MS). Of all these techniques, GC technique are most widely used Gas Chromatography is used as a primary analytical method because high reliableness, high separating efficiency and repeatability.

A Study on the Dry Matter Yield and Nutritive Values of Wild Korean Lespedeza (Lespedeza stipulacea Maxim.)

  • This study was conducted to evaluate the dry matter (DM) yield, quality and utilization of nutrients in Korean lespedeza (Lespedeza stipulacea Maxim.) swards as a roughage source. DM yield in various stages, chemical composition, and DM digestibility were observed, and DM intake, digestibility and utilization of nitrogen and energy by Korean native goat were determined. Experimental diets include three treatments: mixture hay (MH) 100%, MH (80%)+wild Korean lespedeza hay (LH) 20% and MH (60%)+LH (40%). As growth stage advanced, the DM yield of Korean lespedeza tended to increase significantly (p<0.05). The mean DM yield of all growing stages was observed as 7,336 kg/ha. Crude protein (CP) content of Korean lespedeza tended to decrease, but fibrous contents tended to increase when the growth stage advanced. The tannin content of Korean lespedeza was the highest at the bud stage (64.7 mg/g). However, it showed a tendency to decrease as the growth stage advanced, and thus it was the lowest at the ripe seed stage (26.8 mg/g) (p<0.05). With increasing level of LH, voluntary DM intake by Korean native goats slightly increased, but no differences were observed between diets. The digestibility of DM, cellular constituents and NDF was slightly higher in LH containing diets than that of MH 100% diet (p<0.05). There was no significant difference in apparently digested N% among all diets, but retained N% and retained N% of the absorbed were higher in MH (60%)+LH (40%) than those of other diets (p<0.05). A difference in the utilization of energy was not detected. In conclusion, Juvenile LH improved the digestibility of nutrients, presumably due to its high CP content and low fibrous compound. Especially, tannin in LH did not affect in DM intake, but increased the nitrogen utilization of Korean native goats. Accordingly, it could be suggested that Korean lespedeza has a potential to be provided as a roughage source for Korean native goats.

Detection of Extremely Low Concentration Compound and Adsorption by Activated Carbon (극미량 농도 물질의 측정 및 활성탄 흡착 처리)

  • Since the difficulty of analysis at low concentration and the uncertainty of the removal mechanism for Nitrosodimethylamine (NDMA) have been reported, this study has detected extremely low concentration $^{14}$C-NDMA using the LSC(Liquid Scintillation Counter) and tested NDMA removal by Powdered Activated Carbon(PAC). The results showed the highest correlation over 99% when samples were measured with the mixture ratio of sample to scintillation liquid of 10 : 10 and at the detection time of 10 min. For $^{14}$C-NDMA removal by the PACs(S-A(Sigma-Aldrich co.) and Dj(Daejung co.)) raging from 50$\sim$10,000 mg/L, $^{14}$C-NDMA was removed over 90% by adsorption treatment. In addition, S-A showed twice greater adsorption capacity than that of Dj. However, the required PAC amount for $^{14}$C-NDMA removal was higher than that of other amine compounds.

Improvement of Hydrolysis and Bioavailability of Coprecipitated Products of Coptidis Rhizoma and Scutellariae Radix by β-Glucuronidase (베타-글루쿠로니다제에 의한 황련.황금 공침물의 가수분해 및 생체이용률 증가)

  • During the preparation of decoction from the mixture of Coptidis Rhizoma and Scutellariae Radix, insoluble copreciptate was formed. The coprecipitated product (COP) was composed of berberine and baicalin which was the active ingredient of Coptidis Rhizoma and Scutellariae Radix, respectively. COP was slightly soluble in water and could not be well absorbed after oral administration. This poor bioavailibility might be associated with its poor aqueous solubility. With the purpose of increasing the solubility and bioavailibility of COP, hydrolysis of COP by ${\beta}-glucuronidase$ was carried out. Hydrolyzed products (HOP) of COP were identified and assayed for active ingredients. The partition coefficient study, in situ absorption test, and pharmacokinetic study after oral administration were also performed. COP was found to be consisted of berberine and baicalin with molecular ratio of 1 to 1. This compound was hydrolyzed to berberine and baicalin by ${\beta}-glucuronidase$. The rate of hydrolysis was higher at higher temperature up to $50^{\circ}C$ and higher concentration of ${\beta}-glucuronidase$ up to 2500 unit under our experimental conditions. Baicalein, which is more liphophilic than baicalin, showed greater absorption in small intestine than baicalin did. The plasma concentrations of berberine and baicalein after oral administration of HOP were significantly higer than those of COP. The possible mechanism of increased bioavailibility of berberine and baicalein could be the hydrolysis of COP by ${\beta}-glucuronidase$. On the basis of the above results, it might be said that HOP should be a suitable preparation for increasing the bioavailibility of Coptidis Rhizoma and Scutellariae Radix.

Substrate Utilization Patterns During BTEX Biodegradation by an o-Xylene-Degrading Bacterium Ralstonia sp. PHS1

  • The biodegradation of BTEX components (benzene, toluene, ethylbenzene, o-xylene, m-xylene, and p-xylene) individually and in mixtures was investigated using the o-xylene-degrading thermo-tolerant bacterium Ralsronia sp. strain PHS1 , which utilizes benzene, toluene, ethylbenzene, or o-xylene as its sole carbon source. The results showed that as a single substrate for growth, benzene was superior to both toluene and ethylbenzene. While growth inhibition was severe at higher o-xylene concentrations, no inhibition was observed (up to 100 mg $l^-1$) with ethylbenzene. In mixtures of BTEX compounds, the PHS1 culture was shown to degrade all six BTEX components and the degradation rates were in the order of benzene, toluene, o-xylene, ethylbenzene, and m- and p-xylene. m-Xylene and p-xylene were found to be co-metabolized by this microorganism in the presence of the growth-supporting BTEX compounds. In binary mixtures containing the growth substrates (benzene, toluene, ethylbenzene. and o-xylene), PHS1 degraded each BTEX compound faster when it was alone than when it was a component of a BTEX mixture, although the degree of inhibition varied according to the substrates in the mixtures. p-Xylene was shown to be the most potent inhibitor of BTEX biodegradation in binary mixtures. On the other hand, the degradation rates of the non-growth substrates (m-xylene and p-xylene) were significantly enhanced by the addition of growth substrates. The substrate utilization patterns between PHS1 and other microorganisms were also examined.
