New Record of Scolecenchelys fuscogularis (Anguilliformes: Ophichthidae) Leptocephali from Korea, as Revealed by Morphological and Molecular Analyses

  • Three leptocephali (22.2, 22.7, 56.0 mm in total length) collected from the East/Japan Sea were identified by morphological and genetic analyses as belonging to the genus Scolecenchelys (Anguilliformes, Ophichthidae). Morphologically, the specimens were characterized by 148-158 myomeres, 10 gut swellings, dorsal fin origin above middle of the body, and 6 postanal melanophores between the anus and the caudal margin. An analysis of an 849-base pair 12S rRNA sequence of mitochondrial DNA showed that sequences are concordant with those of adult Scolecenchelys fuscogularis (genetic distance = 0.001). Furthermore total number of myomeres is consistent with the total number of vertebrae in adult S. fuscogularis. This study provides the first description of the morphological characteristics of S. fuscogularis leptocephali and their variations with size. The Korean name of S. fuscogularis is "Ga-neun-mul-baem", established by Ji et al. (2012).

  • Scolecenchelys borealis was first added to the Korean fish fauna based on one leptocephalus specimen (total length (TL) 59.2 mm) collected from Dokdo, in the East Sea. Recently, however, Scolecenchelys borealis has been treated as a junior synonym of Scolecenchelys aoki, necessitating review of the scientific name S. borealis. This study describes the morphological characteristics of S. aoki based on an adult specimen (TL 230.0 mm) collected from the sea south of Korea and compares its mitochondrial DNA 12S rRNA sequence with that of the S. borealis leptocephalus. The adult S. aoki is characterized by several morphological features: vertebral formula 59-55-135; preanal lateral pores 55; infraorbital pores two; teeth on upper jaw visible externally when the mouth is closed; the dorsal fin originates just behind the anus; both jaws and vomerine teeth are conical, arranged in two rows. An analysis of an 886-base paire sequence of the 12S rRNA showed that adult S. aoki was concordant with the S. borealis leptocephalus (sensu Ji et al., 2012), indicating that both are the same species.

A Simultaneous Analytical Method for Duplex Identification of Porcine and Horse in the Meat Products by EvaGreen based Real-time PCR

  • A duplex real-time polymerase chain reaction (PCR) based assay for the detection of porcine and horse meat in sausages was designed by using EvaGreen fluorescent dye. Primers were selected from mitochondrial 12S rRNA and 16S rRNA genes which are powerful regions for identification of horse and porcine meat. DNA from reference samples and industrial products was successfully extracted using the GIDAGEN® Multi-Fast DNA Isolation Kit. Genomes were identified based on their specific melting peaks (Mp) which are 82.5℃ and 78℃ for horse and porcine, respectively. The assay used in this study allowed the detection of as little as 0.0001% level of horse meat and 0.001% level of porcine meat in the experimental admixtures. These findings indicate that EvaGreen based duplex realtime PCR is a potentially sensitive, reliable, rapid and accurate assay for the detection of meat species adulterated with porcine and horse meats.

Two New Records of Juvenile Oedalechilus labiosus and Ellochelon vaigiensis (Mugiliformes: Mugilidae) from Jeju Island, Korea, as Revealed by Molecular Analysis

  • Eighteen specimens of juvenile Mugilidae were collected in October 2012 from the southern coastal waters of Jeju Island, and identified based on analysis of their mitochondrial DNA16S rRNA sequences. Seventeen specimens of Oedalechilus labiosus and a single specimen of Ellochelon vaigiensis were found, constituting a new record for these species among Korean ichthyofauna. O. labiosus is identified by the angle at the posterior end of its mouth, which contains a round notch, a darkish dorsal margin of the pectoral fin, the presence of 33-36 lateral line scales, and 23-24 vertebrae. E. vaigiensis is identified by dark dorsal and pectoral fins, the presence of 26 lateral line scales, and 25 vertebrae. The proposed Korean name for Oedalechilus is 'Sol-ip-sung-eo-sok' and that for Ellochelon is 'Nup-jeok-ggo-ri-sung-eo-sok'. The proposed Korean names for the species are 'Sol-ip-sung-eo' and 'Nup-jeok-ggo-ri-sung-eo' for O. labiosus and E. vaigiensis, respectively. We present a key for identification of the Mugilidae family of species from Korea, and include these two newly recorded species.

부산 기장에서 채집된 말미잘의 분자생물학적 방법을 이용한 동정 (Molecular Identification of a Sea Anemone (Cnidaria: Anthozoa: Actiniaria) Obtained in Gijang, Busan)

  • In this study, we tried to identify a sea anemone collected from the coast of Gijang, Busan. The anemone was morphologically similar to species belonging to the genus Anthopleura, but its morphological characteristics did not allow for confirmed identification to species level. Multiple genes from mitochondrial cytochrome oxidase III, 12S and 16S rRNA, and nuclear 18S and 28S rRNA, were amplified for multilocus sequence typing (MLST) analysis using genomic DNA extracted from the sampled anemone and a different primer set. Based on the MLST analysis, the anemone obtained in this study was identified as Anthopleura artemisia. Also, the sequence of internal transcribed spacer-2 was most closely related to A. artemisia, indicating that this single region might be useful for anemone identification. This study shows significance of molecular identification for sea anemones, and will be helpful in studies of sea anemone identification using genotyping-by-sequencing.

마이토콘드리아 유전자 2개를 이용한 대한민국 전라남도 해남군 발생 풀무치 Locusta migratoria (메뚜기목: 메뚜기과)의 계통분석 (Phylogenetic analysis of Locusta migratoria (Orthoptera: Acridae) in Haenam-gun, Jeollanam-do, Korea using Two Mitochondrial Genes)

  • 전라남도 해남군 산이면에 있는 간척지 중 친환경 사료작물 재배지에서 2014년도 8월에 풀무치(Locusta migratoria)가 대발생을 하여, 식량작물과 사료작물에 커다란 피해를 주었다. 오랜 기간 작물에 피해를 주지 않던 migratory locust가 어떠한 원인들에 의하여 대발생하여 해충화 되었는지에 아직까지 모르는 실정이다. 대발생의 원인을 규명하기 위한 연구의 일환으로 해남군 산이면에서 발생한 풀무치의 마이토콘드리아에 존재하는 16S ribosomal RNA와 D-loop의 염기서열을 분석하여 이들과 다른 지역계통간의 유전적인 연관성을 분석하였다. 그 결과 해남군 산이면에서 대량 발생한 풀무치는 북방계통 중 유라시아 대륙지역형과 유전적으로 가장 가까운 것으로 나타났다. 앞으로 2개의 표적 유전자는 국내외 유전적 집단 구조 비교에 활용할 수 있으며 대발생 풀무치의 대발생 원인을 분석하는데 기여할 수 있을 것이다.

한국 남해에서 출현한 돌돔 (Oplegnathus fasciatus)과 강담돔 (Oplegnathus punctatus) 사이의 자연교잡종 (Occurrence of Natural Hybrid between Oplegnathus fasciatus and Oplegnathus punctatus from the South Sea of Korea)

  • 2008년 8월 경남 통영에서 강담돔과 돌돔의 자연교잡종 1개체가 발견되어 형태 및 유전적 특정을 강담돔 및 돌돔과 비교하였다. 형태적으로 본 총은 체측에 선명한 검은색 둥근 점을 많이 가지며 동시에 4개의 연한 가로줄을 가져 강담돔과 비슷하였다. 강담돔 및 돌돔의 계수 계측형질이 거의 일치하였으나, 체장에 대한 배지느러미길이 비에서 자연교잡종(26.7%)은 돌돔(17.2~23.6%)보다 강담돔(26.4%)에 더욱 가까웠다. 미토콘드리아 DNA 16S rRNA 510 bp를 이용한 분자분석에서 자연교잡종은 형태결과와는 달리 강담돔(d=0.020)보다 돌돔(d=0.000~0.010)에 더욱 가까웠다. 본 연구결과를 통해 경남 통영에서 발견된 자연교잡종은 암컷 돌돔과 수컷 강담돔 사이에서 태어난 것으로 추정된다.

Evidence for Genetic Similarity of Vegetative Compatibility Groupings in Sclerotinia homoeocarpa

  • Vegetative compatibility groups (VCGs) are determined for many fungi to test for the ability of fungal isolates to undergo heterokaryon formation. In several fungal plant pathogens, isolates belonging to a VCG have been shown to share significantly higher genetic similarity than those of different VCGs. In this study we sought to examine the relationship between VCG and genetic similarity of an important cool season turfgrass pathogen, Sclerotinia homoeocarpa. Twenty-two S. homoeocarpa isolates from the Midwest and Eastern US, which were previously characterized in several studies, were all evaluated for VCG using an improved nit mutant assay. These isolates were also genotyped using 19 microsatellites developed from partial genome sequence of S. homoeocarpa. Additionally, partial sequences of mitochondrial genes cytochrome oxidase II and mitochondrial small subunit (mtSSU) rRNA, and the atp6-rns intergenic spacer, were generated for isolates from each nit mutant VCG to determine if mitochondrial haplotypes differed among VCGs. Of the 22 isolates screened, 15 were amenable to the nit mutant VCG assay and were grouped into six VCGs. The 19 microsatellites gave 57 alleles for this set. Unweighted pair group methods with arithmetic mean (UPGMA) tree of binary microsatellite data were used to produce a dendrogram of the isolate genotypes based on microsatellite alleles, which showed high genetic similarity of nit mutant VCGs. Analysis of molecular variance of microsatellite data demonstrates that the current nit mutant VCGs explain the microsatellite genotypic variation among isolates better than the previous nit mutant VCGs or the conventionally determined VCGs. Mitochondrial sequences were identical among all isolates, suggesting that this marker type may not be informative for US populations of S. homoeocarpa.

미네랄 워터를 이용한 블랙티의 효율적 추출 방법 및 추출물의 항산화와 항염 효능 (Efficient Extraction Method of Black Tea using Mineral Water and Antioxidant and Anti-inflammatory Activity of Extract)

  • 본 연구에서는 블랙티에 함유된 고분자 카테킨 성분을 효율적으로 추출할 수 있는 조건을 최적화하였다. 발효도에 따른 고분자 카테킨 함량을 확인하기 위하여 세가지 발효도의 블랙티내 고분자 카테킨 함량을 비교하였다. 발효도가 증가할수록 고분자 카테킨 함량이 증가되며 그중 크기가 가장 큰 데아브론닌(theabrownin) 함량이 크게 증가함을 확인하였다. 또한, 경수인 미네랄 워터를 사용하여 추출시 다른 추출수를 사용했을 때 비해 약 150% 이상 데아브로닌의 추출 햠량이 증가됨을 확인하였다. 최적 조건에서 추출된 블랙티 추출물과 그 추출물내 다량으로 함유된 데아브로닌의 항산화 효능을 ABTS assay를 이용하여 확인하였다. 그 결과, 두 추출물 모두 항산화 효능을 갖고 있으며, IC50 값이 각각 10.60 ppm, 13.21 ppm으로 확인하였다. 또한, 주요 성분인 데아브로닌의 항염 효능을 확인한 결과 UVB 조사에 의해 증가된 IL-8의 mRNA 발현이 데아브로닌에 의해 농도 의존적으로 억제됨을 확인하였다. 또한, 데아브로닌 10 ppm을 처리하면 UVB 조사에 의해 저하된 미토콘드리아의 기능이 대조군 대비 86 ± 1.9%으로 회복되었다. 따라서, 데아브로닌은 UVB 조사에 의해 유발된 미토콘드리아의 손상을 방지할 수 있음을 확인되었다.

Utility of taxon-specific molecular markers for the species identification of herbarium specimens: an example from Desmarestia japonica (Phaeophyceae, Desmarestiales) in Korea

  • Desmarestia japonica (Phaeophyceae, Desmarestiales) was recently established from the Japanese ligulate Desmarestia and is morphologically similar to D. ligulata. This species has been reported only from Japan. However, the taxonomic reports based on additional regional distributions are needed to clarify this taxonomic entity and its species boundaries. Because Desmarestia species have restricted distributions in Korea, we reexamined herbarium specimens of D. ligulata deposited at the National Institute of Biological Resources (South Korea). To improve the amplification efficiency of the polymerase chain reaction and avoid contamination by the DNA of other organisms, we developed taxon-specific molecular markers suitable for DNA barcoding of Desmarestia species. Nuclear ribosomal small subunit RNA (18S rDNA) and mitochondrial cytochrome c oxidase 1 (cox1) regions were selected as target DNA. As a result, both were successfully isolated from herbarium specimens of D. japonica acquired over 10 years. These molecular markers provide useful genetic information for herbarium specimens for which conventional molecular analysis is challenging.
