The Effects of Egg White Powder Addition on Acid Production by Lactic Acid Bacteria and Quality of Curd Yogurt (난백분말의 첨가가 호상요구르트에서 젖산균의 산생성과 요구르트의 품질에 미치는 영향)

  • A curd yogurt was prepared from milk added with skim milk powder or egg white powder(EWP). The effects of EWP addition on growth and acid production by lactic acid bacteria in milk were studied. The effects of EWP addition on quality of curd yogurt in point of apparent viscosity, sensory property and volatile aroma compounds were also investigated. Addition of PAT markedly stimulated acid production by lactic acid bacteria and stimulating effect of EWP on acid production was proportional to the amount of addition. Apparent viscosity of curd yogurt added with EWP was generally higher than that of control. Curd yogurt fermented with L. acidophilus showed thixotropic characteristics. Sensory property of curd yogurt added with EWP was evaluated as better than reference sample and sample added with EWP at 2% level was evaluated as better than other samples. Acetone, ethanol, diacetyl, butanol and acetoin were detected by gas chromatographic analysis.

The Influence of Food Hydrocolloids on Changes in the Physical Properties of Ice Cream

  • This study was carried out to investigate the effect of hydrocolloids on the changes in physical properties of a model ice cream. The model ice cream contained water, sugar, skin milk powder, com oil, and 4 different hydrocolloid stabilizers (gelatin, pectin, hydroxyethylstarch, locust bean gum), was manufactured in a batch type freezer. The following physical characteristics of ice cream were examined: flow behavior, overrun, air cell size, ice crystal size, and melt resistance. With regard to flow behavior, all of aged mixes had a lower apparent viscosity relative to the mix before aging, and ice cream mix containing locust bean gum had the highest viscosity. Air cell size was observed to range from 20 to $38\;{\mu}m$, and ice cream with locust bean gum showed the largest size. There was an inverse correlation between overrun and air cell size. The ice crystal sizes of all samples ranged from 25 to $35\;{\mu}m$. Ice cream with added pectin contained the smallest ice crystal size, which was significantly difference from other stabilizers (p<0.05), and resulted in superior melt resistance with increased melting time compared to other samples.

  • A study of mammary function in relation to glucose metabolism of first lactation Bali cows on grass-legume diets was carried out using 12 primiparous cows (initial BW $263.79{\pm}21.66kg$) for 16 weeks starting immediately post calving. The animals were randomly allocated into 4 dietary treatment groups R1, R2, R3 and R4, receiving from the last 2 months of pregnancy onwards, rations based on a mixture of locally available grass and legume feed ad libitum. On a DM basis R1 contained 70% elephant grass (PP, Penicetum purpureum) plus 30% Gliricidia sepia leaves (GS), R2 was 30% PP plus 25% GS supplemented with 55% Hibiscus tilliacius leaves (HT, defaunating effect), R3 and R4 were 22.5% PP+41.25% GS+11.25% HT+25% concentrate, with R4 supplemented with zinc-diacetate. TDN, CP and zinc contents of the diets were 58.2%, 12.05% and 18.3 mg/kg respectively for R1, 65.05%, 16.9% and 25.6 mg/kg respectively for R2, 66.03%, 16.71% and 29.02 mg/kg respectively for R3 and 66.03%, 16.71% and 60.47 mg/kg respectively for R4. Milk production and body weights were monitored, an energy and protein balance trial conducted, overall glucose kinetics parameters assessed, mammary blood flow (MBF) and metabolite arteriovenous differences (${\Delta}AVs$) measured to get uptake data and mammary performance relationships. Parameters of glucose kinetics at peak lactation or during dry condition were not affected by ration quality. Glucose pool size, space of distribution and flux increased by 61.77, 62.26 and 82.08%, respectively, during lactation compared to the dry period. Mean glucose flux of lactating Bali cows was $5.52mg/min.kgBW^{0.807}$ which resembles the range of values of temperate dairy cows. Calculation showed that glucose requirements for maintenance, milk lactose and fat-glycerol synthesis, and the formation of NADPH reached 461.69 g for a yield of 1 kg/d or equal to 320.62 mg/min, which was less than the average glucose flux of lactating Bali cows of 481.35 mg/min. Mammary blood flow (MBF) values ranged from 56 to 83 l/h for the different treatments and the ratio MBF per kg milk produced improved from av. 1540 l/kg for R1 to av. 967 l/kg for R4 treated cows. Mammary glucose uptake ranged from 6.27 to 12.03 g/h or 120 to 140 g/kg milk. Glucose uptake was mass-wise 2 to 4 times the amount secreted as lactose, which indicated values less than the calculated mammary glucose needs and that little lactose was synthesized. The excess glucose taken-up was used for other metabolic processes. Linear relationships between metabolite ${\Delta}AVs$ and arterial blood plasma concentration [A] showed that in Bali cows triglycerides (TG), phenylalanine (Phe) and tyrosine (Tyr) have high coefficients of determination, i.e. 0.77, 0.81 and 0.69, respectively. For glucose, the relationship is quadratic with an $R^2$ value of 0.49. It was concluded that lactose synthesis was inadequate, which led to a speculation that milk yield could be improved by increased lactose synthesis.

Optimization of spray drying conditions of soft persimmon and milk mixture using response surface methodology (반응표면분석법을 이용한 홍시와 우유혼합물의 최적 분무건조 조건)

  • The purpose of this study was to determine the optimum spray drying conditions of soft persimmon latte using response surface methodology that is a statistical procedure used for optimization studies. A central composite design was applied to investigate the effects of independent variables, inlet temperature ($X_1$), air flow rate ($X_2$), and feed flow rate ($X_3$), on responses such as yield, water absorption index, and total phenolic compounds. Statistical analysis revealed that independent variables significantly affected all the responses. A maximum yield of 8.11 g was obtained at $90^{\circ}C$ of $X_1$, 51.82 mL/min of $X_2$ and 7.00 mL/min of $X_3$. A minimum water absorption index of 0.58 was obtained at $101^{\circ}C$ of $X_1$, 60.00 mL/min of $X_2$ and 17.00 mL/min of $X_3$. A maximum total phenolic compounds of $298.02{\mu}g/mL$ was obtained at $90^{\circ}C$ of $X_1$, 43.33 mL/min of $X_2$ and 17.00 mL/min of $X_3$. In conclusion, the best spray drying conditions were as follows: $X_1$, $90^{\circ}C$; $X_2$, 53 mL/min; $X_3$, 17 mL/min. Under those optimal conditions, the powder's yield (7.46 g), water absorption index (0.54), and the content of total phenolic compounds ($294.75{\mu}g/mL$) were estimated.

  • This research is focused on an improvement of additional value of high grade limestone. To obtain the basic data of precipitated calcium carbonate(PCC), studies of physical properties of limestone, calcination and hydration characteristics, the characteristics to manufacture quick lime, hydrated lime, ground calcium carbonate and precipitated calcium carbonate were performed. In the carbonation process, formation of rombohedral must be kept under $10^{\circ}C$ for reaction. Although the temperature of reaction of lime milk was limited under $30^{\circ}C$ for a colloidal PCC manufacture, over $50^{\circ}C$ for spindle type PCC. The recommended reaction conditions for colloidal PCC are $20^{\circ}C$ of reaction temperature, 4% of $Ca(OH)_2$ concentration, 1000rpm of stirring rate and 200ml/min of $CO_2$ gas flow rate.

A Study on the Optimum Conditions for Preparation of Calcium hydrogenphosphate Dihydrate by Box-Wilson Experimental Design (Box-Wilson 실험계획에 의한 연마용 인산일수소칼슘의 최적 제조조건 추구 및 안정화)

  • An abrasive, calcium hydrogen phosphate dihydrate (DCPD), was synthesized in a Box-wilson experimental design by reactions between phosphoric acid and milk of lime, and calcium chloride and sodium phosphate solutions, and stabilized with TSPP and TMP. The optimum conditions for preparation of DCPD from phosphoric acid with milk of lime were such as; reaction temp.; $51.9^{\circ}C$, conc. of lime; 25.9%, conc. of phosphoric acd; 77.9%, drying temp.; $60.2^{\circ}C$ and final pH; 6.46. The physico-chemical and pharmaceutical properties of DCPD were showed as follows: glycerin absorption value(68 ml/100g), whiteness(99.5%), particle size(10.9 nm), pH(7.8), and set test(pass). XRD and SEM of DCPD indicated a monoclinic system crystallographically. $N_2$ adsorption isotherm curve by BET showed non porous type II form. The micromeritic parameters of DCPD showed that surface area was $3.27{\sim}4.6\;cm^{2}/g$ and pore volume, pore area and pore radius were negligible. The rheogram of the toothpaste containing DCPD showed pseudoplastic flow with yield value of 321, and thixotropic behavior forming hysteresis loop. These results meet the requirements as abrasive standard, and sythesized DCPD is expected as a good dental abrasive such as a high quality grade in practice.

Performance of Holstein cows subjected to different cooling sessions during subtropical summer

  • Objective: This study aimed to determine the effect of different cooling sessions (CSs) as a water conservation strategy on physiological, and production responses and welfare in Holstein Friesian cows during subtropical summer in Pakistan. Methods: Twenty-one cows were subjected to three CS in a completely randomized design. The treatments were: i) eleven hours continuous cooling with sprinklers - control (CNT), ii) four CS, and iii) two CS. The CNT represented the practices of the commercial dairy farms in the area, while the other CSs were used as water reduction strategies. Each CS lasted for 1 h with a 12 min cycle (3 min water on and 9 min off) with a sprinkler flow rate of 1.25 L/min. Results: The average temperature humidity index of the shed and the outside open area were 81.9 and 82.5, respectively. The results showed that both physiological responses were highest in the 2CS group followed by the CNT and the 4CS (p = 0.001). The CNT and 4CS groups had similar milk yield (p = 0.040). The 4CS group had more lying and eating times than the CNT and 2CS groups (p = 0.000). The cortisol level in the 2CS group was 2.0 and 2.2 ㎍/dL more than the CNT and the 4CS groups, respectively (p = 0.000). Conclusion: In conclusion, the 4CS was more efficient in cooling the cows and had better welfare, as it yielded similar milk yield, and better physiological responses than the CNT despite using 90% less water.

Application of a Lateral Flow Immunoassay to Determine Ampicillin Residues in Muscle Tissue of Olive Flounder (Paralichthys olivaceus)

  • Antibiotic Detection Kit (Combination I), a lateral flow immunoassay (LFIA) developed for the detection of antibiotic residues in milk, was utilized for the analysis of antibiotic residues in the muscle tissue of olive flounder. After 60-min treatment by dipping in water dosed with ampicillin (200-g/ton water), the residue depletion of ampicillin was investigated in 25 cultured olive flounder (Paralichthys olivaceus). Muscles of fish were sampled on the 1st, 2nd, 3rd, 4th and 5th day after drug treatment. The concentration of ampicillin in the muscle was determined by LFIA. The absorbance ratio of the sample to the control blank (Bs/Bo) was employed as an index to determine the muscle residues in olive flounder. To investigate the recovery rate, standard solutions were added to muscle samples to give final concentrations in the muscle of 4 and 8 ng/ml. The recovery rates of all spiked samples were > 96% of the spiked value. Ampicillin was detected in the muscle of fish treated with the drug until the 2nd day of the withdrawal period. The present study showed that the LFIA can be easily adopted to predict ampicillin residues in tissue of farmed fishes.

Silibinin Inhibits Cell Growth and Induces Apoptosis through Cell-cycle Arrest in PC-3 Prostate Cancer Cells (인간 전립선 암세포 PC-3 세포에서 Silibinin의 세포주기조절을 통한 세포사멸 유도 효과)

  • Milk thistle (silybum marianum) is a famous dietary supplement widely used in the United States and Europe. Silbinin is a major biologically active compound of milk thistle and has strong antioxidant and radical scavenger activities. Anticancer activities, as well as chemopreventive effects on various cancer cell lines, including prostate, lung, colon, skin, and bladder, have also been reported in silbinin. In the present study, we investigated the anticancer effects of silibinin and apoptosis through cell cycle arrest on prostate cancer cell PC-3. We performed cell viability by MTT assay and western blotting to confirm cell cycle check point proteins such as cyclin A/D1/E and cyclin-dependent kinase (CDK) 2/4/6. To quantify silibinin-induced apoptotic cell death of PC-3, Annexin V and PI double staining was performed by flow cytometry, by which its cell distribution was determined. As a result, silibinin inhibited the cell growth of PC-3 cells in a time- and dose-dependent manner, and its treatment resulted in cell cycle arrest at the G1 phase. Also the level of cell cycle check point proteins (cyclin, CDK) was decreased by silibinin in a dose-dependent manner. Taken together, we suggest that apoptosis of prostate cancer cell line PC-3 induced by silibinin is associated with cell cycle arrest through decrease of cell cycle check point proteins, caspase-3 activation and poly (ADP-ribose) polymerase (PARP) cleavage.

Physicochemical Properties of a Biopolymer Flocculant Produced from Bacillus subtilis PUL-A (Bacillus subtilis PUL-A로부터 생산된 Biopolymer 응집제의 물리화학적 특성)

  • Soybean milk cake (SMC) was used for the solid-state fermentation by Bacillus subtilis PUL-A isolated from soybean milk cake. In the presence of 5% glutamate the maximum production of biopolymer (59.9 g/kg) was performed by fermentation at $42^{\circ}C$ for 24 hr. The recovered biopolymer was consisted of 87% $\gamma$-polyglutamic acid with molecular weight of $1.3{\times}10^6$ dalton and other biopolymer. The biopolymer solution showed the great decrease in consistency below pH 6.0, regardless of the molecular weight of PGA. Biopolymer solution has a typical pseudoplastic flow behavior and yield stress. The consistency of biopolymer solution was greatly decreased by increasing heating time and temperature in acidic condition compared to the alkaline condition. In kaolin clay suspension, the flocculating activity of biopolymer was the highest value with 15 mg/L biopolymer and 4.5 mM $CaCl_2$, but decreased greatly with $FeCl_3$. The flocculating activity of biopolymer was maximum at pH5, but decreased drastically by heating at $60{\sim}100^{\circ}C$. In particular, biopolymer with native PGA showed the efficient flocculating activity compared to that of modified biopolymer containing low molecular weight of PGA.
