Effect of Supplementation of Fish and Canola Oil in the Diet on Milk Fatty Acid Composition in Early Lactating Holstein Cows

  • This study examined the effects of supplementation of fish oil and canola oil in the diet on milk yield, milk components and fatty acid composition of Holstein dairy cows in early lactation. Eight multiparous early lactation Holstein cows ($42{\pm}12$ DIM, $40{\pm}6kg$ daily milk yield) were fed a total mixed ration supplemented with either 0% oil (Control), 2% fish oil (FO), 1% canola oil +1% fish oil (FOCO), or 2% canola oil (CO) according to a double $4{\times}4$ Latin square design. Each period lasted 3 wk; experimental analyses were restricted to the last week of each period. Supplemental oils were added to a basal diet which was formulated according to NRC (2001) and consisted of 20% alfalfa, 20% corn silage and 60% concentrate. Milk yield was similar between diets (p>0.05), but dry matter intake (DMI) was lower (p<0.05) in cows fed FO diet compared to other diets. Milk fat percentage and daily yield decreased (p<0.01) with the supplementation of fish and canola oil. The daily yield and percentage of milk protein, lactose and solids-not-fat (SNF) were not affected by diets (p>0.05). The proportion (g/100 g fatty acids) of short chain fatty acids (SCFA) decreased and polyunsaturated fatty acids (PUFA) increased (p<0.05) in milk of all cows fed diets supplemented with oil. The proportions of 6:0, 8:0, 10:0 12:0 and 14:0 fatty acids in milk fat decreased (p<0.01) for all diets supplemented with oil, but the proportions of 14:1, 16:0 and 16:1 fatty acids were not affected by diets (p>0.05). The proportion of trans(t)-18:1 increased (p<0.01) in milk fat of cows fed FO and FOCO diets, but CO diet had the highest proportion of cis(c)-11 18:1 (p<0.01). The concentration of t-10, c-12 18:2, c-9 t-11 18:2, 18:3, eicosapentaenoic acid (EPA, 20:5) and docosahexaenoic acid (DHA, 22:6) increased (p<0.05) in FO and FOCO diets in comparison with the other two diets. These data indicate that including fish oil in combination with canola oil significantly modifies the fatty acid composition of milk.

Comparison of miR-106b, miR-191, and miR-30d expression dynamics in milk with regard to its composition in Holstein and Ayrshire cows

  • Objective: Milk composition varies considerably and depends on paratypical, genetic, and epigenetic factors. MiRNAs belong to the class of small non-coding RNAs; they are one of the key tools of epigenetic control because of their ability to regulate gene expression at the post-transcriptional level. We compared the relative expression levels of miR-106b, miR-191, and miR-30d in milk to demonstrate the relationship between the content of these miRNAs with protein and fat components of milk in Holstein and Ayrshire cattle. Methods: Milk fat, protein, and casein contents were determined in the obtained samples, as well as the content of the main fatty acids (g/100 g milk), including: saturated acids, such as myristic (C14:0), palmitic (C16:0), and stearic (C18:0) acids; monounsaturated acids, including oleic (C18:1) acid; as well as long-, medium- and short-chain, polyunsaturated, and trans fatty acids. Real-time stem-loop one-tube reverse transcription polymerase chain reaction with TaqMan probes was used to measure the miRNA expression levels. Results: The miRNA expression levels in milk samples were found to be decreased in the first two months in Holstein breed, and in the first four months in Ayrshire breed. Correlation analysis did not reveal any dependence between changes in the expression level of miRNA and milk fat content, but showed a multidirectional relationship with individual milk fatty acids. Positive associations between the expression levels of miR-106b and miR-30d and protein and casein content were found in the Ayrshire breed. Receiver operating characteristic curve analysis showed that miR-106b and miR-30d expression levels can cause changes in fatty acid and protein composition of milk in Ayrshire cows, whereas miR-106b expression level determines the fatty acid composition in Holsteins. Conclusion: The data obtained in this study showed that miR-106b, miR-191, and miR-30d expression levels in milk samples have peculiarities associated with breed affiliation and the lactation period.

Fatty acids composition and lipolysis of Parmigiano Reggiano PDO cheese: effect of the milk cooling temperature at the farm

  • Objective: The aim was to study the influence of cooling milk at 9℃ at the farm versus keeping it at 20℃ on Parmigiano Reggiano cheese lipolysis. Methods: A total of six cheesemaking trials (3 in winter and 3 in summer) were performed. In each trial, milk was divided continuously into two identical aliquots, one of which was kept at 9℃ (MC9) and the other at 20℃ (MC20). For each trial and milk temperature, vat milk (V-milk) and the resulting 21 month ripened cheese were analysed. Results: Fat and dry matter and fat/casein ratio were lower in MC9 V-milk (p≤0.05) than in MC20. Total bacteria, mesophilic lactic acid and psychrotrophic and lipolytic bacteria showed significant differences (p≤0.05) between the two V-milks. Regarding cheese, fat content resulted lower and crude protein higher (p≤0.05) both in outer (OZ) and in inner zone (IZ) of the MC9 cheese wheels. Concerning total fatty acids, the MC9 OZ had a lower concentration of butyric, capric (p≤0.05) and medium chain fatty acids (p≤0.05), while the MC9 IZ had lower content of butyric (p≤0.05), caproic (p≤0.01) and short chain fatty acids (p≤0.05). The levels of short chain and medium chain free fatty acids (p≤0.05) were lower and that of long chain fatty acids (p≤0.05) was higher in MC9 OZ cheese. The principal component analysis of total and free fatty acids resulted in a clear separation among samples by seasons, whereas slight differences were observed between the two different milk temperatures. Conclusion: Storing milk at 9℃ at the herd affects the chemical composition of Parmigiano Reggiano, with repercussion on lipolysis. However, the changes are not very relevant, and since the cheese can present a high variability among the different cheese factories, such changes should be considered within the "normal variations" of Parmigiano Reggiano chemical characteristics.

Pelleting in Associated with Sodium Monensin Increases the Conjugated Linoleic Acids Concentration in the Milk of Dairy Cows Fed Canola Seeds

  • To evaluate the effects of the pelleting and the addition of sodium monensin on production, the chemical and lipid composition of milk and butter physical characteristics, 4 Holstein dairy cows (135 days of lactation) with an average milk production of 14.7 kg/d, were supplemented with a concentrate containing ground canola seeds. The cows were assigned to a $4{\times}4$ Latin square design with a $2{\times}2$ factorial arrangement of treatments: i) ground maize, soybean meal, mineral and vitamin supplements, and ground canola seeds (CG); ii) CG concentrate with 31.5 mg of monensin added per kg of dry matter (DM); iii) CG pelleted concentrate; iv) CG concentrate with monensin addition pelleted. There was no difference in milk production and composition. The addition of monensin increased milk concentration of polyunsaturated fatty acids (PUFA), the PUFA/saturated fatty acids (SFA) ratio, and omega 6. The pelleting increased the concentration of monounsaturated fatty acids, the PUFA/SFA ratio, and the omega 6/omega 3 ratio, but decreased the concentration of SFA. The association between pelleting and the addition of monensin increased the concentration of conjugated linoleic acids by 46.9%. The physical characteristics of butter were not affected by the evaluated diets. We concluded that the concentrate with 31.5 mg of monensin added per kg DM basis combined with the pelleting improves the lipid composition of milk from Holstein cows that are on pasture and supplemented with ground canola seeds, without changing the production, milk composition, and spreadability of butter.

Variation in Milk Fatty Acid Composition with Body Condition in Dairy Buffaloes (Bubalus bubalis)

  • Buffaloes usually maintain higher body condition and do not produce milk at the cost of their own body reserves under tropical conditions. The mobilization of body reserves for fulfilling the demands of lactation has been extensively studied in dairy cows while limited work is available on this aspect in dairy buffaloes. Therefore, the present study was conducted to examine variations in milk fatty acid profiles with body condition in Nili-Ravi buffaloes. A total of 24 Nili-Ravi buffaloes within 60 days after parturition, were selected from a private dairy farm in the district of Peshawar. All animals consumed the same diet during the experimental period. A total of 576 raw milk samples were collected for laboratory analysis. The study continued up to 6 months during 2008. Body condition score (BCS), milk yield and composition were recorded once a week. Means for milk fatty acid profile were compared for various levels of BCS. The mean milk yield and fat content were 9.28 kg/d and 5.36%, respectively. The total saturated fatty acids (SFA) were 64.22 g/100 g and the unsaturated fatty acids (UFA) were 35.79 g/100 g. Of the SFA the highest amount was recorded for $C_{16:0}$, followed by $C_{18:0}$, and $C_{14:0}$. The total sum of hypercholesterolemic fatty acids (HCFA, $C_{12:0}$, $C_{14:0}$ and $C_{16:0}$) was 43.33 g/100 g. The concentrations of UFA were greater for moderate BCS followed by poor and highest BCS while SFA showed the opposite trend. The correlation analysis showed that milk yield was negatively affected by BCS and milk fat positively affected, though non-significantly. The present study suggests that Nili-Ravi dairy buffaloes produce similar milk to dairy cows regarding availability of cardioprotective fatty acids, with the highest concentration of $C_{18:1\;cis-9}$. Two HCFA ($C_{12:0}$ and $C_{14:0}$) were associated with higher body condition. Buffaloes with moderate body condition yielded milk containing healthier fatty acids.

  • Changes in total lipid content. total cholesterol content and fatty acid composition of human milk were investigated longitudinaly from 2-5 days to 12 weeks postpartum Milk samples were collected from 19 Korean lactating women at 2-5 days and at 1. 2. 4. 6. and 12 weeks postpartum. The obtained results were as follows : On average the daily energy intake protein intake and fat intake of Korean lactating women was 1812 kcal 72.5g and 29.8g respectively. The composition ratio of energy consis-ting of protein fat and carbohydrate was 16:15:69 The total lipid content increased from 1.39g/이 at 2-5 days to 2.86g/dl at 12 weeks ; while the total cholesterol concentration (mg/g) decreased significantly with time following postpartum. The total unsaturated fatty acids content was higher in colostrum than in mature milk. and the total saturated fatty acids were higher in mature milk. The average DHA content was 0.55% and the P/S ratio of human milk lipids was 0.37.

Preterm Milk의 총지질, 총콜레스테롤 함량 및 지방산 조성 변화에 관한 연구 (Changes in Contents of Total Lipid, Total Cholesterol and Fatty Acid Composition of Preterm Milk during Lactation)

  • Changes in total lipid content, total cholesterol content and fatty acid composition of preterm milk were investigated during early lactation. Milk samples were collected from Korean mothers of 16 premature(34 to 36 weeks gestation age) infants on day 2 to 5(colostrum) and at 6 weeks(mature) postpartum. We estimated the lipid nutrition of preterm milk by comparing with the lipids of term milk. The total lipid content of preterm colostrum was significantly lower than the lipid content of preterm mature milk(p<0.001). Lipid content, determined gravimetrically in colostrum and matured milk, was 1.50g/이 and 3.2g/dl, respectively. Also the total cholesterol content(mg/dl) in preterm milk tends to increase from 14.16mg/dl to 15.20mg/dl, while the total cholesterol(mg/g lipid) concentration higher significantly in colostrum(12.36mg/g) than in mature(5.73mg/g)(p<0.001). The total unsaturated fatty acid contents in preterm milk were higher in colostrum than in mature milk and the total saturated fatty acids were higher in mature milk. The average DHA contents of colostrum and matured milk was 0.64%, 0.53% and the P/M/S ratio of preterm milk were 0.63 : 1.05 : 1.00, 0.47 : 0.79 : 1.00, respectively. Also, $\omega$6/$\omega$3 ratio of preterm milk were 2.35 in colostrum and 5.81 in mature. Therefore, colostrum in preterm milk contained higher amounts of $\omega$3 PUFA than mature milk. The levels of total lipid in preterm milk were higher than term milk. Also, preterm milk is richer in cholesterol, and long chain polyunsaturated fatty acid than term. It appears that the milk secreted by mothers who delivered prematurely differs from milk in several important respect. These components may serve as precursors for membrane, myelin development in the preterm infants. Therefore, it would be necessary to study further into the machanism of how the gestation age might affect to the lipid composition in human milk. These data may provide a basis for better construction of infant formaulas to provide more adequately for the lipid requirements of the Korean premature infant.

  • 한국인(韓國人) 모유(母乳)와 우유(牛乳)에 대(對)해서 GLC법(法)에 의(依)하여 지방산(脂肪酸) 조성(組成)을 구(求)하여 비교(比較)하고 또 채취(採取)한 시료(試料)에 대(對)해서의 일반조성(一般組成)의 치(値)도 기재하였다. 인유(人乳)의 지방산(脂肪酸)은 Butyric acid, Caproic acid 및 Caprylic acid 등(等)의 지방산(脂肪酸)을 거의 함유(含有)하고 있지 않은 점(點)과 우유(牛乳)의 지방산(脂肪酸)은 저급(低級) 지방산(脂肪酸)을 함유(含有)하고 있는 반면(反面) Linol산(酸)의 함량(含量)이 인유(人乳)에 비(比)해서 훨씬 적었다는 점(點)이다.

Milk Yield and Its Fat Content as Affected by Dietary Factors: A-Rewiew

  • Milk yield and its composition is governed by level of nutrition and the composition of diet. Higher concentrate input improves milk yield, whereas its input at moderate levels improves yield of milk fat. High level of dietary protein improves dry matter intake and milk production, however, CP content above 14% has less advantage. Milk yield is enhanced by the feeding of cottonseed and soyabean meal, whereas milk fat increases by the supplementation of cottonseed. Dietary fat increases energy intake, production of milk and milk fat. Quality and quantity of feeds consumed affect fermentation patterns in rumen. Among the rumen metabolites, volatile fatty acids (VFA) content and propionate proportion have been related positively with milk yield, whereas proportion of acetate and butyrate have been related positively with milk fat content. Dietary carbohydrates through the source of sugar, starch, roughage and fibre affect VFA concentration in rumen. Therefore, concentration of volatile fatty acids could be altered to the advantage of consumer through judicious manipulation of diet.

  • The effects of feeding diets with different fatty acids on the composition of fatty acids and vitamin E status in maternal milk & serum and pup's serum were studied. Dietary fats(10% by wt) include on oil(CO), soybean oil(SO), perilla seed oil(PO : about 60% , C18 : 3 $\omega$3) and fish oil(FO : rich in C20 : 5$\omega$3, eicosapentaenoic acid = EPA & 22 : 6$\omega$3, docosahexaenoic acid = DHA), Sprague-Dawley rats weighing 200-250g, were fed experimental diets from pregnancy through lactation period. Maternal milk was obtained by gentle squeezing after 30 minutes of oxytocin(0.2 IU, intraperitoneal) injection. The fatty acid compositions of milk and serum were analyzed at day-2 and day-15. The concentrations of vitamin E in maternal milk and serum and pup's serum were also analyzed. The groups of CO, SO and PO which had no DHA in their diet, contained DHA in their milk, The rations of EPA+DHA/arachidonic acid(AA) were higher in PO group than those in either CO or SO group. This seemed to be due not only to more conversion from C18 : 3$\omega$3 to C20 : 5$\omega$3 and C22 : 6$\omega$3 but also to inhibition of C18 : 2$\omega$6 conversion to C20 : 4$\omega$6. More DHA was found in day-2 milk than in day-15 milk. It was also noted that milk contained more DHA was found in day-2 milk than in day-15 milk. It was also noted that milk contained more DHA than serum and this difference was larger in day-2 than in day-15 milk. Even though the concentrations of vitamin E both in maternal serum and milk were lower in PO and FO groups fed highly unsaturated fat than in CO or SO groups, pup's serum did not show a significant difference among all the experimental groups indicating that the pups man secure their essential nutrients by the biomagnification mechanism.
