Thrombin-induced Migration and Matrix Metalloproteinase-9 Expression Are Regulated by MAPK and PI3K Pathways in C6 Glioma Cells

  • Glioblastoma multiforme is one of the most common and aggressive tumors in central nervous system. It often possesses characteristic necrotic lesions with hemorrhages, which increase the chances of exposure to thrombin. Thrombin has been known as a regulator of MMP-9 expression and cancer cell migration. However, the effects of thrombin on glioma cells have not been clearly understood. In the present study, influences of thrombin on glioma cell migration were examined using Boyden chamber migration assay and thrombin-induced changes in MMP-9 expression were measured using zymography, semi-quantitative RT-PCR, and Western blotting. Furthermore, underlying signaling pathways by which thrombin induces MMP-9 expression were examined. Thrombin-induced migration and MMP-9 expression were significantly potentiated in the presence of wortmannin, a PI3K inhibitor, whereas MAPK inhibitors suppressed thrombin-induced migration and MMP-9 expression in C6 glioma cells. The present data strongly demonstrate that MAPK and PI3K pathways evidently regulate thrombin-induced migration and MMP-9 expression of C6 glioma cells. Therefore, the control of these pathways might be a beneficial therapeutic strategy for treatment of invasive glioblastoma multiforme.

Migration Trajectories of North Korean Defectors: Former Returnees From Japan Becoming Defectors in East Asia

  • From 1959 to 1984, over 93,000 Koreans moved to North Korea from Japan as part of a repatriation project conducted during this time. Among them were people who had escaped from North Korea and immigrated to Japan and South Korea as well as the descendants of such people. This research examines the immigration trajectories of North Korean defectors related to the repatriation project and its effects on international relations in East Asia in a migration systems context. Specifically, it focuses on 26 North Korean defectors who have connections with Japan and settled in Japan and South Korea. It argues that the migration pathways of North Korean defectors linked with the repatriation project have been constructed with the cooperation of and amidst conflict between East Asian countries. To respond to the situation, North Korean defectors used their connections with Japan in amicable relations between Japan and China. However, after the relations went sour, defectors turned to informal transitional networks. If these strategies were unavailable, the defectors faced difficulties, unless they received social or capital support from the destination countries. After entering the destination country, those who settled in Japan have experienced different situations due to the inconsistency in administrative proceedings, while those in South Korea have been treated equally as other defectors. In this sense, some defectors have faced precarious situations in their immigration.

A Study on Definition and Types of Migration Path of Multiple Generation Technology: Case of Customers' and Suppliers' Migration Paths in Semiconductor Technology (다세대 기술 이동경로(Migration path)의 정의 및 종류에 대한 연구: 반도체 기술의 고객 및 공급자 이동경로 사례)

  • The migration path of a multiple-generation technology that occurs during a technological substitution by a new technology is important to understanding the phenomenon of technological substitution, and can contribute to understanding the process of technological innovation. This research defines the concept of migration path and develops a model of the types of migration paths by multiple dimensions (actor, generation, and time) in a multiple-generation technology. Based on a literature review and tracking of migration paths according to multiple dimensions, the definitions and types of migration paths were provided, and the accuracy of the model was verified based on a case study of the semiconductor industry. The migration paths of suppliers are modeled with three types (switching, leapfrogging, and new entrance paths), and the migration paths of customers are modeled with four types (switching, leapfrogging, new entrance, and diffusion paths) in a multiple-generation technology. This research will be useful for understanding the migration paths in the phenomenon of technological substitution, and can be applied to other industries in addition to the semiconductor industry, including various actors. In addition, suppliers and customers can understand technological substitution and can establish a technology strategy against their competitors.

Establishment and Characterization of MTDH Knockdown by Artificial Micro RNA Interference - Functions as a Potential Tumor Suppressor in Breast Cancer

  • Background: Considerable evidence suggests that metadherin (MTDH) is a potentially crucial mediator of tumor malignancy and an important therapeutic target for simultaneously enhancing chemotherapy efficacy and reducing metastasis risk. Inhibition of MTDH expression by RNA interference has been shown in several previous research, but silencing MTDH expression by microRNA (miRNA) interference in breast cancer has not been established. In the present study, we investigated the role of MTDH-miRNA in down-regulation of proliferation, motility and migration of breast carcinoma cells. Methods: Expression vectors of recombinant plasmids expressing artificial MTDH miRNA were constructed and transfected to knockdown MTDH expression in MDA-MB-231 breast cancer cells. Expression of MTDH mRNA and protein was detected by RT-PCR and Western blot, respectively. MTT assays were conducted to determine proliferation, and wound healing assays and transwell migration experiments for cell motility and migration. Results: Transfection of recombinant a plasmid of pcDNA-MTDH-miR-4 significantly suppressed the MTDH mRNA and protein levels more than 69% in MDA-MB-231 breast cancer cells. This knockdown significantly inhibited proliferation, motility and migration as compared with controls. Conclusions: MTDH-miRNA may play an important role in down-regulating proliferation, motility and migration in breast cancer cells, and should be considered as a potential small molecule inhibitor therapeutic targeting strategy for the future.

  • In this paper, we propose an efficient dynamic workload balancing strategy which improves the performance of high-performance computing system. The key idea of this dynamic workload balancing strategy is to minimize execution time of each job and to maximize the system throughput by effectively using system resource such as CPU, memory. Also, this strategy dynamically allocates job by considering demanded memory size of executing job and workload status of each node. If an overload node occurs due to allocated job, the proposed scheme migrates job, executing in overload nodes, to another free nodes and reduces the waiting time and execution time of job by balancing workload of each node. Through simulation, we show that the proposed dynamic workload balancing strategy based on CPU, memory improves the performance of high-performance computing system compared to previous strategies.

  • In the network function virtualization environment, dynamic changes in network traffic will lead to the dynamic changes of service function chain resource demand, which entails timely dynamic adjustment of service function chain resource configuration. At present, most researches solve this problem through virtual network function migration and link rerouting, and there exist some problems such as long service interruption time, excessive network operation cost and high penalty. This paper proposes a dynamic adjustment method of service function chain resource configuration for the dynamic changes of network traffic. First, a dynamic adjustment request of service function chain is generated according to the prediction of network traffic. Second, a dynamic adjustment strategy of service function chain resource configuration is determined according to substrate network resources. Finally, the resource configuration of a service function chain is pre-adjusted according to the dynamic adjustment strategy. Virtual network functions combination and virtual machine reusing are fully considered in this process. The experimental results show that this method can reduce the influence of service function chain resource configuration dynamic adjustment on quality of service, reduce network operation cost and improve the revenue of service providers.

Chloroquine Exerts Anti-metastatic Activities Under Hypoxic Conditions in Cholangiocarcinoma Cells

  • Intra-tumoral hypoxia is an environment that promotes tumor cell migration, angiogenesis and epithelial-mesenchymal transition that accounts for a major mechanism of metastasis. Chloroquine potentially offers a new therapeutic approach with an 'old' drug for effective and safe cancer therapies, as it exerts anti-metastatic activity. We investigated the inhibitory effect of chloroquine on cholangiocarcinoma (CCA) cell migration under cobalt chloride ($CoCl_2$)-stimulated hypoxia. We showed that chloroquine suppressed CCA cell migration under hypoxic-mimicking conditions on exposure to $100{\mu}M$ $CoCl_2$. Moreover, chloroquine stabilized the protein level of prolyl hydroxylase domain proteins (PHD-2) but reduced the levels of hypoxic responsive proteins such as hypoxia-inducible factor (HIF-$1{\alpha}$) and vascular endothelial growth factor (VEGF). It also suppressed epithelial mesenchymal transition (EMT) by increasing the ratio of E-cadherin to N-cadherin under hypoxic conditions. In conclusion, chloroquine can inhibit hypoxia-stimulated metastasis via HIF-$1{\alpha}$/VEGF/EMT which may serve as a useful additional strategy for CCA therapy.

Negotiations in the Gendered Experiences of Transpinay Entertainers in Japan

  • Among Filipino entertainers in Japan, trans women (transgender women) or transpinay (Filipino trans woman) entertainers remain understudied compared to cisgender women. Though the number of entertainers has declined, transpinay entertainers remain relevant as transgender issues continue to be salient globally. This study explains the gendered experiences of the transpinay migrants, particularly in entertainment work and their relationships, which are different from cisgender Filipino women entertainers' experiences. Using grounded theory and drawing on concepts of performativity in interactions to analyze the narratives of transpinay entertainers, I delve into how transpinay entertainers negotiate their gender and migrant identities as they establish professional and personal relationships. Moreover, the transpinays' entertainment work is a significant contributing factor to their sense of belonging in Japan, as they form relationships with colleagues, clients, and partners who support them and, thereby, sustain their lifestyles as trans women. The transpinay entertainers' flows of migration between Japan and the Philippines reveal that they embrace various aspects of social remittances and use them to their advantage to create and enhance their transpinay identity in Japan. By examining the transpinays' migrant experiences, this study aims to elucidate the gendered experiences of transpinay entertainers, which involve significant negotiations in their migration pathways notably different from cisgender Filipino women entertainers, reveal resilience.
