Distribution of Alcohol-tolerant Microfungi in Paddy Field Soils

  • Ethanol treatment method was attempted for the selective isolation of ethanol-tolerant fungi from two sites of rice paddy fields around Seoul area. The vertical and seasonal fluctuation of the fungal population were also investigated. The ethanol-tolerant fungi were Talaromyces stipitatus, T. flavus var. flavus, T. helicus var. major, Eupenicillium javanicum, Emericellopsis terricolor, Pseudourotium zonatum, Aspergillus flavus, Cladosporium cladosporioides, Penicillium frequentans, P. janthinellum, and P. verruculosum. The most dominant species isolated by this method was T. stipitatus. It was found that the numbers of fungal species and colony forming units(CFUs) of ethanol-tolerant fungi were higher in Ascomycota than in Deuteromycota. A particular tendency appeared the highest CFUs in autumn, but lower in spring and winter. T. stipitatus was the dominant species of ethanol tolerant microfungi. This result would suggest that membrane lipid composition of ethanol-tolerant fungi isolated from the soils may play on important role in the ethanol tolerance.

  • A total of 82 genera and 271 species of soil-inhabiting fungi including saprobic, nematode-trapping, and arbuscular mycorrhizal but plant pathogenic fungi published hitherto in South Korea are listed with the information on geographic location, habitat, vegetation when available, and relevant literatures.

Molecular Identification of Endophytic Fungi Isolated from Needle Leaves of Conifers in Bohyeon Mountain, Korea

  • Fungal endophytes are microfungi that live in plants without causing apparent symptoms of infection. This study was conducted to identify endophytic fungi isolated from leaves of coniferous trees in Bohyeon Mountain of Korea. We collected leaves of two species of coniferous trees, Pinus densiflora and Pinus koraiensis, from 11 sites in the study area. A total 58 isolates were obtained and identified using molecular and morphological characteristics. Four species of endophytic fungi were isolated from P. densiflora: Lophodermium conigenum, Leotiomycetes sp., Septoria pini-thunbergii, and Polyporales sp., while two fungal species were isolated from P. koraiensis: Eurotiomycetes sp. and Rhytismataceae sp. The most frequently isolated species were L. conigenum and S. pini-thunbergii.

  • 한국 토괴균의 계속적인 연구로서, 분리된 균류중 Aspergillus 8종과 microfungi 5종인 Alternaria alternata, Chrysosporium pannorum, Doratomyces microsporus, Trichoderma koningii, T. viride에 관하여 보고하였다. 이들 동정된 균류 중 아직까지 한국에서 미기록된 것으로는 Chrysosporium pannorum, Doratomyces microsporus, Trichoderma koningii, T. viride 등의 4종이었다. 또한 완전연대인 Emericella nidulans var. nidulans에 관하여 보고하였다.

Morphology and Phylogeny of Two Novel Species within the Class Dothideomycetes Collected from Soil in Korea

  • Two fungal strains (KNU-JJ-1827 and KNU-JJ-1829) belonging to the class Dothideomycetes were discovered from Jeju Island, Korea during this investigation of soil microfungi. Strain KNU-JJ-1827 showed fewer conidial septations, larger conidiogenous cells, and smaller conidia compared to the previously identified closest species of Didymocyrtis. Strain KNU-JJ-1829 revealed the similar characteristics of the nearest certain species of the genus Parathyridaria with the production of conidiogenous cells and conidia, because no asexual morphs were detected from the closest type strain Parathyridaria rosae. The novelty of the strains was also confirmed by analyzing molecular data using internal transcribed spacer regions and 28S rDNA. The molecular phylogeny also strongly support the detailed description and illustration for each proposed species as Didymocyrtis septata sp. nov. (KNU-JJ-1827) and Parathyridaria ellipsoidea sp. nov. (KNU-JJ-1829) isolated from soil in Korea.

Molecular and Morphological Characterization of Two Novel Species from Soil and Beetles (Dorcus titanus castanicolor) in Korea

  • Two fungal strains belonging to Ascomycota were discovered in Gyeonggi-do, Korea, during this investigation of soil microfungi and microbiota of insects. The strain KNUF-20-047 produced milky white on the back and a milky creamy center to white toward the margin on the front side of colonies. Conversely, the closest Xenoacremonium falcatus displayed a pale luteous to luteous center, white toward margins on the front side, and pale luteous or luteous pigment on the back side, whereas X. recifei produced white colonies. The conidiophores of KNUF-20-047 were slightly larger than those of X. falcatus, and the conidia were distinct from X. recifei. Strain KNUF-20-NI-005 produced light brown to subhyaline conidiophores up to 56.0 lm tall, whereas Rhinocladiella anceps displayed golden to dark brown conidiophores up to 350 lm. Strain KNUF-20-NI-005 also produced larger conidia than R. anceps but smaller than R. coryli and R. fasciculata. Moreover, the molecular phylogeny strongly supports the detailed description and illustration of each proposed species to be designated as Xenoacremonium minutisporum sp. nov. and Rhinocladiella terrigenum sp. nov. in Korea.

Promotion of Tricholoma matsutake mycelium growth by Penicillium citreonigrum

  • Tricholoma matsutake has been the most valuable ectomycorrhizal fungi in Asia because of its unique flavor and taste. However, due to the difficulty of artificial cultivation, the cultivation of T. matsutake has relied on natural growth in forests. To cultivate the T. matsutake artificially, microorganisms in fairy rings were introduced. In this study, we isolated 30 fungal species of microfungi from the soil of fairy rings. Among them, one single fungal strain showed a promoting effect on the growth of T. matsutake. The growth effect was confirmed by measuring the growth area of T. matsutake and enzyme activities including a-amylase, cellulase, and b-glucosidase. In comparison with control, microfungal metabolite increased the growth area of T. matsutake by 213% and the enzyme activity of T. matsutake by 110-200%. The isolated fungal strain was identified as Penicillium citreonigrum by BLAST on the NCBI database. The Discovery of this microfungal strain is expected to contribute to artificial cultivation of T. matsutake.

한국 논 토양중의 균류에 관한 연구 - III . 자낭균류 - (Fungs flora of paddy fields in Korea. - III. Ascomycetes -)

  • 한국의 두장소의 논토양에서 회석평판법에 의하여 토양균을 분리하였다. 동정된 30종중 14종은 한국에서 미기록종이며 이중 7종은 자낭균류에 속하는 것으로 본 논문에 서술하였다. Talaromyces의 종중에서는 Talaromyces flavus var. flavus, T. panasenkoi, T. stipitauts 그리고 T. trachyspermus가 가장 우세하게 분포되어 있다. 또 특이한 것은 Westerdykella multispora가 현저하게 주목된다. Westerdykella multispora는 많은 격막이 있는 원통형 자낭포자가 32개 포함된 자낭을 가지고 있는 구형 또는 아구형의 pseudothecia를 형성한다. Chaelomium globosum은 cellulose를 분해하는 ascomycetes의 종류로 많은 파상 정모와 ostiole을 가지고 있는 검은 자낭각을 형성한다. 이 자낭각은 레몬형태의 올리브색을 띤 갈색자낭포자 8개를 포함하고 있는 곤봉모양을 하고 있으며 소실성의 자낭을 형성한다. 또 다른 Ascomycete로써 Acremonium an-amorph를 가지고 있는 Emericellopsis terricola로 2곳의 채취장소에서 분리할 수 있었다.

Morphological and Phylogenetic Analyses Reveal a New Species of Genus Monochaetia Belonging to the Family Sporocadaceae in Korea

  • The fungal strain belonging to the genus Monochaetia of the family Sporocadaceae was isolated from hairy long-horned toad beetle (Moechotypa diphysis) during the screening of microfungi associated with insects from Gangwon Province, Korea. The strain KNUF-6L2F produced white, light brown to dirty black surface, and olivaceous green colonies with the higher growth, while the closest strain M. ilicis KUMCC 15-0520T were light brown to brown, and M. schimae SAUCC 212201T light brown to brown toward center. The strain KNUF-6L2F produced shorter (5.7-14.0 ㎛) apical appendages than M. ilicis (6.0-24.0 ㎛), but similar to M. schimae (7.0-12.5 ㎛). Three median cells of KNUF-6L2F were light brown to olivaceous green, whereas brown and olivaceous cells were observed from M. ilicis and M. schimae, respectively. And the strain KNUF-6L2F produced larger conidiogenous cells than M. ilicis and M. schimae. Additionally, phylogenetic analyses based on molecular datasets of internal transcribed spacer (ITS) regions, translation elongation factor 1-alpha (TEF1α), and β-tubulin (TUB2) genes corroborated the strain's originality. Thus, the strain is different from other known Monochaetia species, according to molecular phylogeny and morophology, hence we suggested the new species Monochaetia mediana sp. nov. and provided a descriptive illustration.

한국산(韓國産) 왕대나무의 현존량(現存量)과 토양(土壤) 미세균류상(微細菌類相) (The Standing Crops and Soil-borne Microfungal Flora of Phyllostachys reticulata in Korea)

  • 본(本) 연구(硏究)는 예산지역(禮山地域)(A)과 광산지역(光山地域)(B)의 왕대나무(Phyllostachys reticulata)의 현존량(現存量)과 그 토양(土壤) 미세균류상(微細菌類相)을 조사(調査)한 것이다. 왕대나무의 죽림밀도(竹林密度)는 예산지역(禮山地域)이 17,250본(本)/ha이며 광산지역(光山地域)이 14,780본(本)/ha으로서 예산지역(禮山地域)이 16.1%가 많았다. 양지역(兩地域)의 환경요인(環境要因)은 B지역(地域)의 생장기간(生長期間)의 평균기온(平均氣溫)이 A지역(地域)보다 $1.5{\sim}2^{\circ}C$가 높았고 토양온도(土壤溫度)도 $1{\sim}2^{\circ}C$가 높았으며 토양내(土壤內)에 함유(含有)되어 있는 전실소(全室素), 인산(燐酸) 및 유기물질량(有機物質量)도 약간(若干)많았다. 또 B지역(地域)에서는 낙엽량(落葉量)과 부식량(腐植量) 그리고 죽림내(竹林內)의 식생수량(植生數量)도 많았으며 죽림지(竹林地)에서 환원(還元)되는 각종(各種) 유기물(有機物) 분해(分解)에 관여(關與)하는 미세균류(微細菌類)도 Mortierella elongata, Mucor circinelloides, Aspergillus japonicus, Penicillium waksmani and Trichoderma lignorum등의 5종(種)이 더 많았다. 온도(溫度)는 죽림내부(竹林內部)로 들어 갈수록 낮았고 습도(濕度)는 높았다. 죽림내(竹林內)의 상대조도(相對照度)의 비율(比率)은 A지역(地域)이 4.19% B지역(地域)이 2.7%로서 하단분(下端部)에서는 모두 광합성작용(光合成作用)을 할 수 있는 능력(能力)이 상실(喪失)되었으나 조도(照度)가 약(弱)할수록 표토(表土)근처에서 서식(棲息)하는 미세균류(微細菌類)는 오히려 활동력(活動力)을 강(强)하게 하는 것으로 생각한다. 생산구조도(生産構造圖)에서 B지역(地域)의 광합성부(光合成部)의 최대량(最大量)이 대나무 지상부(地上部)의 상단(上端)에 위치(位置)하고 있어서 높은 생산량(生産量)을 유지(維持)하는데 효과적(效果約)인 구조(構造)였다고 생각된다. A,B지역(地域) 죽림(竹林)에서 $D^2H$, $w_s,\;w_b$, 및 $w_l$ 상대생장식(相對生長式)을 유도(誘導)한 결과(結果)는 다음과 같다. A지역(地域) $logw_s=0.5262\;logD^2H+1.9546$ $logw_b=0.6288\;logD^2H+1.5723$ $logw_l=0.5181\;logD^2H+1.8732$ B지역(地域) $logw_s=0.5433\;logD^2H+1.8610$ $logw_b=0.1630\;logD^2H+2.3475$ $logw_l=0.4509\;logD^2H+2.0041$ 상기(上記)한 식(式)을 적용(適用)하여 10a당 현존량(現存量)을 조사(調査)한 결과(結果) A지역(地域) $w_s=1128.83kg,\;w_b=689.05kg,\;w_l=926.69kg$ 으로 $w_e=2744.57kg$이었고, B지역(地域) $w_s=1206.66kg,\;w_b=679.92kg,\;w_l=1112.51kg$으로 $w_t=2999kg$이였다. 따라서 A,B양지역간(兩地域間)에 있어서 $D^2H,\;w_s,\;w_b$, 및 $w_l$의 현존량(現存量)을 비교한 결과(結果)(t-test), $D^2H,\;w_s,\;w_b,\;w_l$에서는 유의차(有意差)가 인정되었으나 $w_b$는 유의차(有意差)가 없었다. 토양생(土壤生) 미세균류(微細菌類)를 조사(調査)한 결과(結果) 158균주(菌株)를 분리(分離)하고 55종(種)을 동정(同定)하였다. 그 중 A지역(地域) 50종(種), B지역(地域) 55종(種)으로 양지역(兩地域)의 우점종(優占種)들은 다음과 같다. Trichoderma viride, Penicillium janthinellum, P. commune, Aspergillus oryzae, A. niger, A. gigantus, A. fumigatus, Mortierella ramaniana, var. anguliFPora, Mucor hiemalis와 Zygorhynchus moelleri. 이상(以上)의 결과(結果)에 의(依)하면 토성(土性)이 좋고 토양양료(土壤養料) 및 토양생(土壤生) 미세균류(微細菌類)의 증가(增加) 그리고 생육기간(生育期間)의 온도(溫度)가 왕대나무의 생장(生長)이나 임상식물(林床植物)의 종(種)과 양(量)을 증가(增加)시킨 것으로 나타났고 왕대나무의 현존량(現存量)과 환경요인(環境要因)과의 상관관계(相關關係)는 이들의 모든 요인(要因)이 상호연관(相互連關)을 갖고 복잡(複雜)하게 작용(作用)한 것으로 보며 더욱 죽림밀도(竹林密度)가 중용(重要)한 인자(因子)로 작용(作用)한 것같다.
