국내에서 분리된 독소생산 Microcystis 속과 조류 대발생 시기에 대청호, 충주호, 용담호, 소양호, 및 의암호에서 채취한 시료에서 조류 독소인 microcystins의 생합성에 관여하는 mcyA 유전자의 염기서열 다양성을 분석하였다. GenBank에 등록된 mcyA 염기서열과 비교한 결과, 국내 호소에서 분리된 Microcystis 속의 mcyA 유전자 염기서열은 매우 낮은 다양성을 나타내었다. 환경 시료 분석 결과, 2-3종류의 clone이 전체의 87-100%를 차지하였으며, Anabaena 속이나 Planktothrix 속의 mcyA 유전자와 유사한 염기서열은 발견되지 않았다. 이러한 결과로 볼 때, 대상 호소에서 microcystins는 주로 Microcystis 속에 의해 생산되며, 독소 생산에 관여하는 mcyA 유전자의 염기서열은 보존되어 있는 것으로 판단된다.
Microcystis sp.의 부패과정에서 수중으로 용출되는 AOM 특성과 염소 이들에서의 disinfection by-products (DBPs) 생성특성을 조사하였다. 수중으로 용출된 EOM과 cell + IOM에서의 염소 DBPs 생성특성을 조사한 결과, EOM은 보관기간 초기부터 지속적으로 증가하였으나 cell + IOM의 경우는 급격한 감소경향을 나타내었으며, 생성된 DBPs 중 HAAFP가 가장 높은 생성비율을 나타내었다. 또한, 이 때의 DBP 구성종들의 변화를 살펴본 결과, HAA 구성종들의 경우는 EOM에서는 di-HAA 구성종들의 비율은 점점 감소하였고 tri-HAA 구성종들의 비율은 점점 증가하였다. 그러나 cell + IOM의 경우는 EOM의 경우와는 반대 경향을 나타내었다. 또한, HAN 구성종들의 경우는 EOM과 cell + IOM 모두 di-HAN 구성종들의 생성비율이 월등히 높았다.
An abnormal mono-specific bloom of the cyanobacterium Microcystis aeruginosa had developed at a specific location (transitional zone, monitoring station of Hoenam) in Daecheong Reservoir from middle of July to early August, 2001. The maximum cell counts during the peak bloom reached 1,477,500 cells/mL, which was more than 6~10 times greater than those at other monitoring sites. The hypothesis of this study is that the timing and location of the algal bloom was highly correlated with the local environmental niche that was controled by physical processes such as hydrodynamic mixing and pollutant transport in the reservoir. A three-dimensional, coupled hydrodynamic and ecological model, ELCOM-CAEDYM, was applied to the period of development and subsequent decline of the bloom. The model was calibrated against observed water temperature profiles and water quality variables for different locations, and applied to reproduce the algal bloom event and justify the limiting factor that controled the Microcystis bloom at R3. The simulation results supported the hypothesis that the phosphorus loading induced from a contaminated tributary during several runoff events are closely related to the rapid growth of Microcystis during the period of bloom. Also the physical environments of the reservoir such as a strong thermal stratification and weak wind velocity conditions provided competitive advantage to Microcystis given its light adaptation capability. The results show how the ELCOM-CAEDYM captures the complex interactions between the hydrodynamic and biogeochemical processes, and the local environmental niche that is preferable for cyanobacterial species growth.
방류수내 남조류 세포의 밀포와 종조성 및 독소의 계절별, 일일 변화를 일본 중심에 위치하고 있는 Suwa호의 수문에서 1998년 5월부터 10월까지 조사하였다. Microcystis의 높은 세포밀도가 M. ichthyoblabe가 우점했던 7월과 M. viridis가 우점종으로 나타난 9월에 관찰되었다. Microcystis의 종조성과 총 세포밀도는 3종류의 microcystin의 정성${\cdot}$정량적인 변화와 관련이 있었다. M. ichthyoblabe가 우점했던 7월 동안에는 MC-RR과MC-LR만이 검출되었으나 M. aeruginosa의 개체수 증가와 함께 M. viridis가 우점종으로 나타난 8월부터 10월까지는 MC-YR을 포함한 3종류의 microcystin 모두 검출되었다. 방류수내 Microcystis의 세포밀포와 microcystin의 함량의 일 변화는 바람의 일 변화에 큰 영향을 받았다. 아침에 비해 오후에 바람이 강하게 부는 풍속주기의 조건에서 Microcystis의 세포밀도와 독소의 함량은 아침에 증가하고 오후에 감소하는 경향을 나타냈다. 본 연구의 결과들은 수문이나 취수탑에서 방류시기의 조절은 부영양 호수에서 물을 취수하여 이용함에 있어 원수내 조류의 생물량을 저감할 수 있는 유효한 방법임을 제시한다.
수화현상의 대표적 원인 생물인 남조류의 M. aeruginosa와 대조구로서 녹조류인 C. zofingiensis를 인공 배양하여 분말세라믹 투여에 따른 선택적 조류 제거 가능성에 대해 실험하였다. 0.05 mm의 분말세라믹을 투여에 따른 M. aeruginosa 및 C. zofingiensis의 침전효율은 각각 82와 63%였으며, 입자크기 0.1과 1 mm의 경우에서 는 M. aeruginosa는 각각 69와 34%, C. zofingiensis는 각각 52와 44%의 침전효율을 나타냄으로써, 분말세라믹 처리에 따른 침전효율의 차이를 나타내는 것을 확인하였다. 또한 Microcystis sp.에 의한 수화현상이 빈번한 수역의 현장시료를 대상으로 분말세라믹 처리에 따른 조류 제거효과를 실험한 결과 입자크기 0.05 mm, 투여농도 $1\;g\;L^{-1}$의 조건에서 24시간 후 약 67%의 Microcystis sp. 제거효과를 나타내었다. 분말세라믹의 수화현장에 적용가능성 여부를 확인하기 위하여 소규모 enclosure에서 분말세라믹 처리 농도에 따른 Microcystis sp. 제거효과 실험한 결과 입자크기 0.05 mm,투여농도 $1\;g\;L^{-1}$의 조건에서 24시간 후 약 69%의 Microcystis sp.의 제거효과를 나타내었으며, 이러한 연구결과로 보아 Microcystis sp.에 의해 수화가 발생한 현장에 분말세라믹을 적용하여 독성 남조류를 효율적으로 제거할 수 있을 것으로 기대된다.
This study investigated the removal characteristics, Microcystin decomposition and generation of by-products when aqueous Microcystis sp. is oxidized by ozone. The concentration of Microcystin (MC) in aqueous solution has been found by HPLC analysis to decrease continuously by ozonation after the initial, abrupt increase. The kinetic constant of the decomposition of MC-RR and -LR were 0.0596 and 0.0243, respectively. This means that removal efficiency of MC-RR by its oxidative decomposition is preferable compared with that of MC-LR. On the other hand, it has been found that the decomposition product, TOC, exhibits the continuous decrease in the concentration by further ozonation, while DOC and UV-254 increase temporarily until 10 minutes before the decrease. Furthermore, the GC/MSD analysis has revealed that the ozonation of Microcystis sp. for 100minutes affords five kinds of aldehydes, six kinds of alcohols, and trans-1, 2-dimethyl-cyclopropane.

This study examines the effect of pH, temperature, metal ion concentration and culture density on metal biosorption by the nuisance cyanobacterium Microcystis aeruginosa. Ni biosorption was higher at pH 9.2 than at neutral and acidic pH. In contrast the biosorption of Cu and Zn was maximum at pH 7.0. However, biosorption of Zn was difficult to measure at pH values 9.2 and 10.5, owing to the formation of insoluble complexes. All the test metals (Cu, Zn, and Ni) showed maximum biosorption rate at low culture densities of 40 mg dry wt $1^{-1}$. The biosorption of Cu, Zn, and Ni was maximum at $40^{\circ}C$. However, no worthwhile difference in Zn and Ni sorption was noticed at 4 and $29^{\circ}C$ as compared to $40^{\circ}C$. Of these three metals used Microcystis showed a greater binding capacity ($K_{f}$ value=0.84, Freundlich adsorbent capacity) and accelerated biosorption rate for Cu under various environmental conditions. Fitness of mathematical models on metal biosorption by Microcystis confirmed that the biological materials behave in the same way as physical materials. These results suggest that before using a biosorbent for metal recovery, the environmental requirements of the biosorbent must be ascertained.
Miceocystis aeruginosa에 의한 심한 수화가 발생한 1996년 여름동안 공산호에서 식물플랑크톤의 일수직분포와 물리.화학적 요인을 조사하였다. 최대 현존량은 M. aeruginosa가 우점으로 나타난 표층에서 나타났으며, 수심에 따라 점차 감소하였다. M. aeruginosa개체군의 높은 밀도가 이른 아침과 저녁에 표층에 축척되었으나 일몰 후에 급격히 감소하였다. 현존량의 대부분은 수심 5m 내에 분포하였고, 현존량의 일변화가 표층에서 명확하게 나타났으나 수직이동은 나타나지 않았다. 표층에서 현존량의 일변화는 바람에 의한 수평적 이동의 결과로 추측되었다.
This study is focused on the antimicrobial activity of cyanobacteria Microcystis aeruginosa by the reduction and oxidation reaction of copper and zinc alloy metal fiber filter. Cu/Zn ion is easily makes radicals with molecular hydroperoxide. Especially, hydroperoxide radical shows strong toxicity to the strains. Plasma membrane causes conformational change when hydroperoxide radical binds to plasma membrane. Elution of copper ion from copper and zinc alloy metal fiber is detected in the cyanobacteria solution as 0.5 ppm, and that of zinc ion is 0 ppm respectively. Zinc ion is figured to form a hydroxide in the cyanobacteria solution and precipitated to form a sludge. The concentration of chlorophyll-a in the cyanobacteria solution was proved to be the index of antimicrobial level of Microcystis aeruginosa.
Cyanobacteria regulate their buoyancy in response to changing environmental conditions. This process is essential for cyanobacterial development and can account for their dominance in eutrophic waters in summer. The present investigation was conducted to understand the 24-hour vertical distribution of cyanobacteria and water quality characteristics in Lake Daecheong. Water samples were collected and analyzed at depth intervals of 2 or 3 m and at an interval of three hours for a day on August 28, 2001 and September 24, 2002. In 2001 the accumulated standing crop of Microcystis spp. from surface to a depth of 6 m was 94.9%. Microcystis spp. showed no vertical migration below the thermocline. Microcystis spp. had maximum density near the surface, but shifted to 2 m depth at 2 p.m. A dense population of Anabaena spp. accumulated near the surface from 2 to 5 p.m. in 2002.
