1. The yeasts in the two samples of Nuruk (mold wheat) which one prepared at the College of Agriculture, Choong-Nam University (S) and the other purchased at a market(T), were examined and counted. The yeasts were differe entiated by the market (T), were examined and counted. The yeasts were differe entiated by the TTC(2,3,5-triphenyltetrazolium chloride)agar overlay technique that yields a varied shade of color. The results were : the population of yeasts in 1g of Nuruk S was about $6{\times}10^4$, 56.5% of which were TTC-pink yeasts, 16% TTC-red pink yeasts, 8% were TTC-red yeasts, and 16.5% TTC-white yeasts. In Nuruk T(1g), the number of yeasts accounted for $14{\times}10^4$ and constituted of 42% TTC0pink, 21% TTC-red pink, 23% TTC-red and 9% TTC-white. 2. During the fermentation of Takju (Korean Sake) employing the Nuruk S and T the yeast flora throughout the brewing were observed in 12 hour intervals. TTC pink and red yeasts considered to be the major yeasts were isolated all cultured. The strains ($1{\times}10^5$/ml) were added to the mashes S and T in which pH was adjusted to 4.2 and the change of yeast flora was examined during fermentation.
Examining the microbial populations in the Han River estuary, we conducted this experiment at six sites of the estuarine area, Chollyu-ri, Cholsan-ri, Wolgon-ri, Chogi-ri, Inhwa-ri, and Oepo-ri for 5 months since May 1967. From the results obtained it could be summarized as follows. 1) The salinity of the estuarine water increased in order of the distances from the base point of the Old Han River Bridge to every site of the estuary, and pH of the water, which were between 7. 3 and 8. 1, showing little difference each other in both the sites and dates of experiment. 2) The populations of the general bacteria and coliform group bacteria were highest at the site of Chollyu-ri, and it decreased with the downstreaming of the river water toward Oepo-ri site. As for fungi which have comparatively high tolerance to the salinity, its population was shown hi hest at Chogi-ri. 3) The relationship between the salinity and the number of the general bacteria have indicated that the incresed salinity reduced the growth rates of the bacteria. By this it can be assumed that the fresh-water bacteria decreased due to the sea water as well as its dilution effects. 4) The high pollution of the esturarine water was caused by the increases of inhabitants and water thrown by industries in Seoul and Kyung-In Industrial District as well as the excrements fertilized to the farms.

Constructed wetlands are improvised man-made systems, designed for adopting the principle of natural wetlands for purifying wastewater - the elixir of life. They are used widely as a cost-effective and energy-efficient solution for treating greywater generated from different tertiary treatment sources. It provides an elaborate platform for research activities in an attempt to recycle earth's natural resources. Among the several organic impurities removal mechanisms existing in constructed wetland systems, the earth's active microbial population plays a vital role. This review deals with the recent advancements in constructed wetland systems from a microbiological perspective to (effect/ devise/ formulate) chemical and physical treatment for water impurities. It focuses on microbial diversity studies in constructed wetlands, influence of wetland media on microbial diversity and wetland performance, role of specific microbes in water reuse, removal of trace elements, some heavy metals and antibiotics in constructed wetlands. The impurities removal processes in constructed wetlands is achieved by combined interactive systems such as selected plant species, nature of substrate used for microbial diversity and several biogeochemical effected reaction cycles in wetland systems. Therefore, the correlation studies that have been conducted by earlier researchers in microbial diversity in wetlands are addressed herewith.
Observed trends in climate change, globalization and an aging population have an effect on public health conditions in Korea, prompting a reevaluation of current environmental regulations. In this study, we evaluated the performance of the total coliform (TC) standard, which is the only microbiological standard in the current regulation regime for hot spring water, by estimating correlation with the presence/absence of Legionella, a non-fecal opportunistic pathogen with heat-tolerance. Microbiological data in 7 studies that surveyed Legionella in hot spring waters were subjected to meta-analyses with the odds ratio (OR) as the effect size. The presence/absence of Legionella was significantly correlated to TC levels [OR = 3.1(1.5–6.4, 95% CI), p = 0.002]. Due to there being no direct explanation as to the reason for the occurrence of TC, mesophilic fecal bacteria, being coupled with Legionella presence, the mechanism of the correlation between the two kinds of bacteria was further investigated. Legionella presence was more prevalent with a high heterotrophic plate count [HPC; 4.0(2.2–7.2); p < 0.001] and water temperature [4.3(1.4–13.6), p = 0.011] when the temperature range was <40℃. However, it was reverse-correlated with water temperature when the temperature was >50℃ [0.2(0.1–0.4), p < 0.001]. Therefore, bacterial standing crops in hot spring waters appear to be determined by water temperature in general, and this forces TC and Legionella levels be correlated. In accordance with this relationship, HPC rather than TC reflect the levels of non-fecal contamination better. Therefore, employing HPC as the sole microbiological standard, or adding HPC into the current standard of hot spring water assessment, is suggested as a proactive measure to prevent health issues arising from contamination.
To investigate the variations of physicochemical factors and microbial populations in seven stations at water region of Gangjin bay, nutritive salts, water temperature, transparency, suspended solid, salinity, COD, DO, pH, heterotrophic bacteria, fungi and fecal coliform were analysed four times from February to October, 1999. Total fecal coliform and heterotrophic bacterial population during investigation periods ranged 16.1~166.0 CFU/ml and $5.0{\times}10^3$~$13.1{\times}10^3$CFU/ml, respectively. Stational mean values of these bacterial population showed higher densities at the inlet of fresh water inflow than those of other stations. When the aspects of abiotic and biotic parameters measured were analyzed, ecosystem of Gangjin bay was regulated by factors such as inflow of fresh water, nutrient salts, salinity and variation of water temperature during four seasons.
The growth rate of yeast population (Saccharomyces uvarum) cultivated in the Knopp's modified medium (plus various carbon sources) appeared the highest value when the Knopp's minimal medium was treated to 1.5% with disaccharide such as maltose and sucrose. Also the treatment of lactose and raffinose resulted in polulation growth as to the population size in case of maltose and sucrose. However, the gorwth of yeast was not occurred at all when a polysaccharide, such as inulin, was added as carbon source. The growth from of yeast population in Knopp's modified medium are characterized by the fact that log phase continued 100hrs after inoculation and that stationary state phase appeared in general 250hrs after inoculation. Applying the various carbon sources to respiration substrate for yeast cell, the respiration rate of yeast showed the highest value in treatment of maltose and followed in order of raffinose, lactose, glucose, and sucrose. Determined the amount of poly-phosphate and turn over pathway of poly-phosphate according to culture phase of yeast, it is revealed that the yeast synthesized 3 types of poly phosphate (poly-P A,B, and C) and postulated that turn over pathway of poly-phosphate as follows ; Inorganic phosphate is converted into each kind of polyphosphates, and then one part of poly-P-C is converted into poly-P-B, the rest poly-p-C and poly-P-B are converted into poly-P-A. The synthesized poly-phosphate is considered to have a role as energy pool utilizing to synthesis of cellular organic materials. Of the 13 carbon sources used in this experiment, the useful carbon sources for biosynthesis of poly-phosphate and cellular organic materials are confirmed as disaccharide (maltose and sucrose) as well as glucose. Protein synthesis in yeast cell showed the two peaks on 6th and 8th day after inoculation ; nucleic acid on 2nd day (48hrs), carbohydrates on 2nd day (48hrs), and phospholipid on 2nd and 8th day after inoculation, respectively.
Subpopulations of nucleotides that bind specifically to a variety of proteins have been isolated from a population of random sequence RNA/DNA molecules. Roughly one in $10^{13}$ random sequence RNA/DNA molecules folds in such a way as to create a specific binding site for small ligands. Since the development of in vitro selection procedure, more than 50 nucleic acid ligands (aptamers) have been isolated. These molecules are very useful for the study of molecular recognition between nucleic acid and protein/organic compound. In addition to these basic studies this method gives us a dream to produce new drugs against several diseases. We focused on several aptamers which specifically binds to trypsin-like serine proteases (thrombin, human neutrophil elastase, activated protein C and NS3 protease of human hepatitis C virus) and want to introduce their structural characteristics and some functions.
To investigate the electrophoretic karytype of Fusarium oxysporum, intact chromosomal DNA was separated by pulsed-field gel electrophoresis (PEGE). DNA extraction from nulcei, mycelia and protoplasts were compared with one another and with the quantity and the suitability for PFGE separation in agarose gel. As a result, the most useful extracting method for intact DNA was found to be that from protoplasts. By varying the electrophoretic conditions, 8 chromosomal DNA bounds were resolved. Using the Schizosaccharomyces pombe and Saccharomyces cerevisiae as size standards, the size of Fusarium oxysporum chromosomes was estimated to range from approximately 0.6 Mb TO 6.7 Mb, and total genome size was 26.7 Mb. The suitability of electrophoretic karyotyping as a tool for strain characterization is discussed.
There were observed the host-parasite relationship between rice grains and contaminating fungi indicated by the fungal penetration degree in the tested rices. The results were as follows ; 1. The fungal penetration could be observed in the outer layer of the rices but couldn't be seen in the starch portin of the tested rices. 2. The Gram staining method was better than the PAS staining method for the observation of fungi penetration into the rices.
The distribution and annual variation of detergent-degrading bacteria which were isolated from the Han River downstream running through the center of Seoul area were studied. The concentration of dissolved detergent was the lowest, 2.16 ppm at site 1 and it was gradually increased to downstream, the highest, 2.67 ppm at site 4. Population density of detergent degrading bacteria was about $10^2{\sim}10^3 cells/ml$. The density of LAS degrading bacteria was 1.1 - 1.8 times larger than that of SDBS-degrading bacteria. In annual variation the lowest density was appeared on January and the highest on July. In seasonal distribution of LAS-degrading bacteria, annual dominant group was genus Pseudomonas. There were few except genus Pseudomonas in winter and the subdomiannt group in spring and summer was genus Aeromonas and Enterobacteriaceae respectively. In autumn genus Pseudomonas, Aeromonas and Enterobacteriaceae were equally distributed. In SDBS-degrading bacteria, the annual dominant group was also genus Pseudomonas.
