Antimicrobial Activities of Medicinal Herbs and Seaweeds Extracts Against Microorganisms Isolated from the Rice Warehouses (미곡창고 오염균주에 대한 천연추출물의 항균활성)

  • In order to prevent the quality deterioration of rice from microbial infection, we investigated antimicrobial activities of Morun albalinne, Glycyorrhiza uralensis, Sargassum siliquastrum, and Ecklonin cava against isolated microorganisms from the paddy and warehouses. The major types of the bacteria grown in the paddy and rice warehouses were Sphingomonas paucimobilis, Arthrobacter atrocyaneus, and Bacillus spp. such as Bacillus cereus Additionally Deuteromycetes, Aspergillus spp. and Penicillum spp. were considered as major contaminant microorganisms in the paddy and rice warehouses. As results of the paper disc assay against the isolated microorganisms, the ethanol extracts with Morun albalinne, Glycyorrhiza uralensis, Sargassum siliquastrum, and Ecklonia cava showed effective antimicrobial activities. Especially, Morun albalinne showed the strongest growth inhibition on the isolated bacteria at $0.0025{\sim}0.0075%$.

The Effect of Silver Diamine Fluoride on Salivary Biofilm (Silver diamine fluoride가 타액 생물막에 미치는 영향)

  • Silver diamine fluoride (SDF) is an effective and efficient agent for arresting dental caries. It can be useful in treating children with behavioral or medical limitations. The purpose of this study was to evaluate the antimicrobial effect of SDF by using salivary biofilm. Pellicle-like saliva coated structure was prepared by using unstimulated saliva. For developing cariogenic biofilm, Streptococcus mutans was added to the mixture of pooled saliva and inoculated into a saliva coated glass or chamber. SDF was applied to cariogenic biofilm to evaluate the antimicrobial effect of SDF. As time passed, total bacteria and S. mutans were reduced after application of SDF (p < 0.000). Confocal laser scanning microscope also showed the increment of the ratio of dead cell. As a result of experiment using enamel and dentin of primary teeth, it was confirmed that the growth of cariogenic biofilm was inhibited when the SDF was treated (p = 0.029 each). This study showed excellent anti-microbial effect of SDF. And anti-caries effect in clinical practice can be expected.

Sterilization of Yakju(Rice Wine) Using a Batch-type High Voltage Pulsed Electric Field System (고전장펄스를 이용한 약주의 회분식 살균)

  • Yakju(rice wine) was sterilized with high-voltage pulses of short time on a batch pulsed electric field(PEF) system. The initial microbial counts of Yakju were 7.52 X $10^4\;CFU/mL$ for total aerobes, 2.20 X $10^4\;CFU/mL$ for lactic acid bacteria and 7.08 X $10^4\;CFU/mL$ for yeasts. The pH, acidity and electric conductivity of Yakju were 3.36, 0.462% and 1.24 mS/cm, respectively. Yakju was treated with 2-250 of pulses exponential-wave formed electric pulses under the field strength of 12.5-25 kV/cm. The critical strengths of the electrical field for the sterilization of Yakju were 7.5 kV/cm for total aerobes, 8.5 kV/cm for lactic acid bacteria and 6.5 kV/cm for yeasts. Logarithmic survival rates decreased linearly at low pulse number, but curvilinearly at high pulse number. The PEF sterilization kinetics of Yakju could be analysed by In s = In A-k In (n) and the sterilization rate constant increased with electric field strength and the size of target microorganisms. No changed in pH, acidity, and the growth of microorganisms were found in the PEF treated Yakju during the storage for 6 weeks at both $4^{\circ}C$ And $30^{\circ}C$.

Changes in Functional Constituents and Stability of Green Tea Beverage during Different Storing Conditions (저장조건에 따른 녹차음료의 기능성성분 변화 및 안정성)

  • Preservation stability of the extract from dried green tea produced in Jeju, extracted at $55^{\circ}C$ for 15 min and bottled in PET container after filtration and then settled at $4^{\circ}C$, indoor at room temperature and outdoor, respectively, was estimated. Color L-value was almost constant but color of the extract became darkened during storage, especially in indoor storage. Main catechins of green tea extract were (-)-epigallocatechin, (-)-epigallocatechin gallate, epicatechin, (+)-catechin, epicatechin gallate in order, and the content of epigallocatechin was 53.8%. Nevertheless total catechin content was not changed in cold storage, and $4.7\sim10%$ of total catechins were reduced in outdoor or indoor storage after 14 weeks. Caffeine content were increased but total polyphenols were not changed largely during storage. Election donating abilities (EDA) showed more than 60% without influence of storage conditions and periods. Nitrite scavenging abilities (NSA) showed more than 90% at pH 1.2, but those were decreased gradually as pH increased, and then disappeared at pH 6.0. Color, polyphenols, antioxidant activities of extract were not so changed for more than 3 months. Microbial growth was not shown during same periods, due to heat treatment of the extracts at $95^{\circ}C$ for 10 min.

Storage Quality of Sulhyang Strawberries as Affected by High O2 Atmosphere Packaging (고산소 환경기체조절 포장조건에 따른 설향 딸기의 저장 중 품질변화)

  • The storage quality of fresh Sulhyang strawberries packaged under modified atmospheres was investigated to examine the effect of high $O_2$ on the fruit. Fresh strawberries were packed into PP trays and top-sealed with PET/PP film. Initial gas compositions inside the packages were varied with air, 40% $O_2$/60% $N_2$, 60% $O_2$/40% $N_2$, and 80% $O_2$/20% $N_2$. Sealed packages in PE film bags with air and perforated PP trays were also used as another treatment and control, respectively. Quality attributes and viable cell counts of pathogenic bacteria were assessed during storage at $5^{\circ}C$ for 12 days. High $O_2$ concentration showed no significant effects on the physicochemical and microbial qualities of strawberries. Fruit packaged in PE film bags with 6-15% $O_2$ and 7-9% $CO_2$ during storage had the lowest viable cell counts of inherent microorganisms among the treatment samples. Growth of pathogenic bacteria was suppressed in perforated packages where molds occurred frequently. In an overall sensory aspect, the PE film packages exhibited higher scores than the others at the end of storage period. The experimental results suggested that gas-permeable film packaging with an appropriate combination of $O_2$ and $CO_2$ rather than gas-barrier tray packaging with an initially high $O_2$ concentration would be suitable for improving the storability of strawberries.

Quality characteristics of blended tea with added pan-firing Rhus verniciflua seeds and changes in its antioxidant activity during storage (볶은 옻씨를 첨가한 차 음료의 품질특성 및 저장 중 산화방지 활성의 변화량 측정)

  • While manufacturing blended tea, the temperature for pan-firing Rhus verniciflua seed (RVS) is important. The pan-firing duration was fixed to 15 minutes according to a sensory test. We then prepared four types of blended teas with varied ratios of RVS; all the four types showed similar pH, $^{\circ}Bx$, and hunter's color values with no significant difference. According to the sensory test, A-type tea blended with RVS was the most preferred. The antioxidant activity of A-type blended tea was evaluated during storage. The changes in the total polyphenol, total flavonoid, and antioxidant activity by DPPH and ABTS assays were monitored during storage at 4, 25, and $40^{\circ}C$ for 28 days. After 28 days of storage, values for total polyphenol content, total flavonoid content, and antioxidant activity were similar to the initial values. Moreover, microbial growth was not observed during that period.

Impacts of Elevated $CO_2$ on Algal Growth, $CH_4$ Oxidation and $N_2O$ Production in Northern Peatland (이탄습지에서 이산화탄소의 농도가 조류의 증식, 메탄 산화 및 아산화질소 생성에 미치는 영향)

  • Effects of elevated carbon dioxide ($CO_2$) on soil microbial processes were studied in a northern peatland. Intact peat cores with surface vegetation were collected from a northern Welsh fen, and incubated either under elevated carbon dioxide (700 ppm) or ambient carbon dioxide (350 ppm) conditions for 4 months. Higher algal biomass was found under the elevated $CO_2$ condition, suggesting $CO_2$ fertilization effect on primary production, At the end of the incubation, trace gas production and consumption were analyzed using chemical inhibitors. For methane ($CH_4$ ), methyl fluoride ($CH_3F$) was applied to determine methane oxidation rates, while acetylene ($C_2H_2$) blocking method were applied to determine nitrification and denitrification rates. First, we have adopted those methods to optimize the reaction conditions for the wetland samples. Secondly, the methods were applied to the samples incubated under two levels of $CO_2$. The results exhibited that elevated carbon dioxide increased both methane production (210 vs. $100\;ng\;CH_4 g^{-1}\;hr^{-1}$) and oxidation (128 vs. $15\;ng\;CH_4 g^{-1}\;hr^{-1}$), resulting in no net increase in methane flux. For nitrous oxide ($N_2O$) , elevated carbon dioxide enhanced nitrous oxide emission probably from activation of nitrification process rather than denitrification rates. All of these changes seemed to be substantially influenced by higher oxygen diffusion from enhanced algal productivity under elevated $CO_2$.

Antimicrobial Activity of UlGeum (Curcuma longa L.) Extract and Its Microbiological and Sensory Characteristic Effects in Processed Foods (울금 추출물의 항균효과 및 첨가식품의 미생물학적.관능적 특성)

  • In order to develop a natural food preservative, UlGeum (Curcuma longa L.) powder was extracted with ethanol, and its antimicrobial activity was investigated. This ethanol extract showed antimicrobial activity against Bacillussubtilis, Escherichiacoli, and Staphylococcus aureus, and the resulting inhibition zones against the microorganisms by the extract (4 mg/disc) were 11, 10, and 8.5 mm, respectively. To test for further food preservation effects, solutions of the extract were added to rice cakes and noodles at concentrations of 0%, 0.25%, 0.50%, 0.75%, and 1.00%, respectively. Microbial growth was less in the UlGeum ethanol extract-added groups than in the control group. In sensory evaluations, there were significant differences among the groups of rice cakes and noodles in terms of color, taste, flavor, chewiness, and overall acceptability (p<0.05), and the 0.5% added groups received significantly higher scores than the other groups. Furthermore, as storage time increased, the sensory scores of the rice cakes and noodles significantly decreased in all groups (p<0.05). These results suggest that UlGeum is effective for increasing the shelf-life of rice cakes and noodles.

Quality Changes in 'Elliot' Blueberries and 'Sulhyang' Strawberries Packed with Two Different Packaging Materials during Refrigerated Storage (기체투과도가 다른 포장재로 포장한 '엘리오트' 블루베리와 '설향' 딸기의 냉장 저장 중 품질 변화)

  • Blueberries and strawberries are highly perishable and easily contaminated with microorganisms. To maintain the quality of these commodities during refrigerated storage, the effects of two packaging materials as well as passive modified atmosphere packaging on the quality of blueberries and strawberries were investigated. The harvested blueberries and strawberries were first treated with combined non-thermal treatment of aqueous chlorine dioxide and fumaric acid, followed by packaging with polyolefin film (6,000 mL $O_2/m^2{\cdot}24hr{\cdot}atm$ at $24^{\circ}C$) and polyamide/polyamide/polyethylene film (PA/PA/PE, 60 mL $O_2/m^2{\cdot}24hr{\cdot}atm$ at $24^{\circ}C$), respectively. After combined sanitizer treatment, the populations of total aerobic bacteria in blueberries and strawberries were reduced by 2.50 and 1.97 log CFU/g while those of yeast and molds were reduced by 1.95 and 2.18 log CFU/g, respectively, compared with the control. In particular, microbial growth in these samples packed with PA/PA/PE film was reduced during refrigerated storage. In addition, the blueberries and strawberries packed with PA/PA/PE film underwent lower weight loss than those packed with polyolefin film during storage. These results suggest that appropriate packaging with proper gas permeability is necessary to maintain the quality of blueberries and strawberries during refrigerated storage.

Quality Changes of Salted Chinese Cabbages with Electrolyzed Water Washing and a Low Storage Temperature (전해수 세척 및 저장 온도에 따른 절임배추의 품질변화)

  • To extend the shelf-life of salted Chinese cabbages, electrolyzed water (EW) was used to wash raw Chinese cabbages before the salting process (to control microbial growth), and different storage temperatures evaluated (0, 4, and $10^{\circ}C$). A tap water washing group (TW) was used for comparison. The initial total bacterial population was 5.36 log CFU/g in the TW treatment and 3.50 log CFU/g in the EW treatment. The EW treatment decreased bacterial numbers by approximately 2 log CFU/g compared to the TW treatment and kept this initial population number for 32 days at $0^{\circ}C$. The salinity had no difference during storage. In general, several factors (pH, reducing sugars, hardness, and transmittance) decreased over time, and decreased slowly with EW treatment and $0^{\circ}C$ storage. Overall, the salted Chinese cabbages with EW treatment showed lower bacterial populations compared to TW treatment, and when stored at $0^{\circ}C$, delayed decreases in quality.
