Optimization of Red Pigmentation and Effect of the Metabolites Produced by Monascus Strains on Microbial Inhibition and Colorization in Processed Ham (Monascus 균주의 적색색소 생산 특성과 육제품에서의 항균 및 착색 효과)

  • In this study, we tested possibility of replacing nitrite salts, which were always added during the meat product processing, with the metabolites produced by antimicrobial and red pigment producing Monascus strains. We have already shown that Monascus No. 116 strain has the highest antimicrobial activity among the strains isolated from Ang-Khak. Monascus isolate No. 229 was chosen due to its outstanding red pigment producing ability. The red pigment production by No. 229 was highest in the medium containing 8% sucrose, 2% yeast extract, 0.1% K2HPO4, 0.5% MgSO4. Optimum pH and temperature for the red pigment production were pH 6.2 and 3$0^{\circ}C$, was found in spot or Rf value 0.54 on TLC plate using ethyl acetate-acetone-water (4:4:1, v/v/v) as development solvent system. Isolate No. 116 and No. 229 were cultured in a optimal condition for the antimicrobial activity and red pigmentation. The culture concentrates were applied in situ to the production of instantly processed ham. Mixed application of 89 ppm Na-nitrite and 300 ppm of culture broth concentrate of Monascus isolate No. 116 and 500 ppm of red color produced by Monascus isolate No. 229 showed similar results with the single application of 94 ppm Na-nitrite. These results confirmed that the antimicrobial activity and red pigment of Monascus strains might be valuable to replace Na-nitrite salt in meat processing.

Quality Characteristics and Anti-Oral microbial Activity of Jelly Using Bamboo (Phyllostachys nigra var. henonis) Leaves Extract (죽엽 추출액을 이용한 젤리 품질평가 및 구강세균에 대한 항균효과)

  • This study examined the quality characteristics and anti-oral microbial activity of bamboo leaf jelly prepared with different 5 levels (0, 10, 20, 30, and 40%) of bamboo (Phyllostachys nigra var. henonis) leaf extract. The sugar contents of bamboo leaf jelly were increased significantly by increasing the level of bamboo leaf extract. The luminance and Hunter's a values of the jelly samples increased with increasing bamboo leaf extract, but the 40% bamboo leaf jelly had the lowest Hunter's b values. The hardness, adhesiveness, gumminess, and chewiness increased significantly with increasing bamboo leaf extract. Among the mechanical properties, only the flavor of the jelly with 30 and 40% bamboo leaf extract were increased significantly. The extract of bamboo leaves had strong antimicrobial activity against S. mutans, S. sobriuns, P. gingivalis, and P. intermedia at a concentration of 40%. These results suggest that bamboo leaf extract can be useful in the production of high quality jelly.

Sensory and Microbial Characteristics of a Rice Processed Snack using Sweet Pumpkin Powder for Infants (단호박 분말 첨가 영아용 쌀 가공 스낵의 관능적 미생물적 품질 특성)

  • The sensory characteristics of a rice processed snack for 6 to 12 month old infants were investigated. The microbiological safety of all samples was also tested. The moisture content of snack of the SW snack was 6.33 significantly higher than in the other snacks (p<0.05). The sensory intensities of RW were highest in glossiness brightness, uniformity, sweet aroma, savory flavor, sweet flavor, chewiness and crispiness. In the; consumer acceptance test, RW showed significantly higher glossiness, uniformity, sweet aroma, savory flavor, sweet flavor, rancid, chewiness, adhesiveness, crispiness and overall acceptance than the other snacks (p<0.05). The aerobic plate counts were negative in all the samples tested. E. coli and pathogenic microorganisms tests were also negative or not detected, showing that all the samples tested were microbiologically safe according to the food code which applies to food manufacturers. Overall, potentially high value products of rice processed snacks for 6 to 12 month old infants were developed and tested They are expected to be utilized and in the competitive and growing infant food industry.

  • 34 strains were isolated from dyeing wastewater in order to improve treatment efficiency of dyeing wastewater containing PVA. Two strains of them were finally selected through the PVA degrading test, and identified as Microbacterium barkeri LCa and Paenibacillus amylolyticus LCb. As a result, optimal conditions for microbial growth and PVA degradation were 30$^{\circ}C$, neutral pH, starch as a carbon source, and peptone as a nitrogen source. And it was concluded that these two strains have good ability for PVA degradation. And 90% over PVA was degraded by single culture as well as a mixed culture of 2 different strains.

  • The biodegradable and biocompatible poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), a copolymer of microbial polyester, was fabricated as nanofibrous mats by electrospinning. Image analysis of the electrospun nanofibers fabricated from a 2 wt% 2,2,2-trifluoroethanol solution revealed a unimodal distribution pattern of fiber diameters with an observed average diameter of ca. 185 nm. The fiber diameter of electrospun fabrics could be controlled by adjusting the electro spinning parameters, including the solvent composition, concentration, applied voltage, and tip-to-collector distance. Chondrocytes derived from rabbit ear were cultured on a PHBV cast film and an electrospun PHBV nano-fibrous mat. After incubation for 2 h, the percentages of attached chondrocytes on the surfaces of the flat PHBV film and the PHBV nanofibrous mat were 19.0 and 30.1 %, respectively. On the surface of the electrospun PHBV fabric, more chondrocytes were attached and appeared to have a much greater spreaded morphology than did that of the flat PHBV cast film in the early culture stage. The electro spun PHBV nanofabric provides an attractive structure for the attachment and growth of chondrocytes as cell culture surfaces for tissue engineering.

Liquid Culture Enhances Protoplast Formation from the Auxotroph (Ser-) of lentinula edodes

  • The optimal conditions for the production and regeneration of the protoplasts from Lentinula edodes were studied. Protoplast formation from the mycelia of L. edodes which were cultured in liquid medium showed a significantly high yield compared with that of the mycelia which were cultured on cellophane covered agar media. A mixture of Novozyme 234 (15 mg/ml) and Cellulase Onozuka R10 (10 mg/ml) in 0.6 M mannitol (pH 4) was optimal lytic enzyme for the protoplast release. The optimal incubation time and mycelia age were 3.5-4 hours at $30^{\circ}C$ and 6-8 days, respectively. Regeneration frequency was 0.18% plated onto a medium containing 0.6 M sucrose, and 0.08% plated onto a medium containing mannitol. But hardly any regeneration was observed in the media containing NaCl, KCl, or $MgSO_{4}$ More than 90% of the protoplasts contained nuclei and the nucleus number per protoplast was 1.1. The DNA content per nucleus was 5.1 pg. The diameter of the protoplast was $3-5{\mu}m$ and it had a well defined cell structure.

  • Glutaraldehyde is known as an ideal preservatives for pericardial heterograft, and many laboratories used this chemicals for preparing tissue valves, pericardial patches and MVOP [monocusp ventricular outflow patch] so we tried to find out the appropriate concentration and ingredients of the Glutaraldehyde for the preparing bovine pericardium. We selected 50 calves, aged about 2 years, and procured their pericardia. These were divided 6 groups such as fresh group, treated with only antibiotics, treated with Glutaraldehyde 0,5%, 0.625 %, 0.75 %, and 0.875 %, and our experiments included microbial culture test, tensile strength measurement and microscopic examination. On microbial culture, there were no growth on 1 week and 4 weeks after preparation with all kind of Glutaraldehyde, but on 4 weeks after only antibiotics treatment [Penicillin, Streptomycin, Kanamycin, Amphotericin -B] E.coli and candida albicans were observed. On tensile strength test, 0.625 % and 0.75 % Glutaraldehyde were revealed as the best preservatives for bovine pericardium and compared to other commercial products they kept more desirable tensile strength. On light and electron microscopic examinations, Glutaraldehyde treated pericardia had much regular and compact collagen fibers and preserved more normal structures, but there were no difference between the different concentration of the Glutaraldehyde. We concluded that 0.625% and 0.75 % Glutaraldehyde were the best concentration for preservation of bovine pericardium in our experiment.

  • Identification of the pathogens causing infection is important in terms of patient's health management and infection control. Synovial fluids could be used as clinical samples to detect causative pathogens of prosthetic joint infections (PJIs) using molecular diagnostic assays, therefore, normalization and validation of clinical samples are necessary. Microbial culture is considered the gold standard for all infections, including PJIs. Recently, molecular diagnostic methods have been developed to overcome the limitation of microbial culture. Therefore, guideline for validating clinical samples to provide reliable results of molecular diagnostic assays for infectious diseases is required in clinical field. The present study aimed to develop an accurate validating method of synovial fluid clinical samples using GAPDH gene as an internal control to perform the quantitative PCR TaqMan probe assay to detect pathogens causing PJIs.

Effect of Microbial Products Made of Bacillus stearothermophilus DL-3 on Growth of Chickens and Pigs (Bacillus stearothermophilus DL-3을 사용하여 제조한 미생물 제재가 닭과 돼지의 성장에 미치는 영향)

  • Effect of microbial products made of Bacillus stearothermophilus DL-3 on growth of chickens and pigs was investigated. Two types of microbial product were made in this study. One is the microbial product made of culture broth of B. stearothemophilus DL-3 and rice bran which named as the microbial product A. The other is the microbial product made of culture broth of B. stearothermophilus DL-3, apple pomace, soybean pomace and rice bran which named as the microbial product B. Chickens were divided into three groups and each group was fed with $100\%$ general feed, $90\%$ general feed supplemented with $10\%$ microbial product A or $90\%$ general feed supplemented with $10\%$ microbial product B. The average chicken weight of each group was $41.1{\pm}2.5g,\;41.6{\pm}3.2g\;and\;42.3{\pm}2.9g$ and those after 28 days was $547.7{\pm}91.7g,\;560.1{\pm}17.2g\;and\;562.2{\pm}32.5g$, respectively. The average weight gain for each group was 506.6 g, 518.6 g and 519.9 g, respectively, and weight increases of groups fed with $90\%$ general feeder and $10\%$ microbial product A and B were $2.4\%\;and\;2.6\%$ higher than the group fed with $100\%$ general feed. Pigs were also divided into three groups and each group was fed like chickens. The average weight of each group was $9.3{\pm}1.0kg,\;9.4{\pm}1.1kg\;and\;9.6{\pm}1.0kg$ and those after 37 days was $19.3{\pm}4.1kg,\;20.2{\pm}3.9kg\;and\;20.8{\pm}4.2kg$, respectively. The average Weight gain for each group was 10.65 kg, 10.82 kg and 11.20 kg, respectively, and weight increases of groups fed with $90\%$ general feeder and $10\%$ microbial product A and B were $1.6\%$ and $5.2\%$ higher than the group fed with $100\%$ general feed.

  • For the production of microbial cells from cellulosie materials by cellulore-assimilating bacteria, Cellulomonas flavigena GFB 24-1, isolated by authors, utilization of this organism on some microbiological properties was investigated. The results of these studies were summarized at follows; 1. When the organism was incubated in the growth medium at pH 7.0 for 50 hours, its growth was the most effective and the level of excreted total protein in the menstruum increased continuously during the stationary phase of cell growth. 2. The optimal enzyme activity was observed in the pH region of 5 to 7 and culture period of 40 to 50 hours. 3. The microbial degestibility of cellulosic wastes such as sawdust, rice hull, rice straw, peanut hull and used newspaper was less than 30%, whereas that of cellulose powder was 47.1% and rice straw was digested 77% by NaOH treatment. 4. Bacterial cells incubated in the growth medium were increased up to 8% of sustrate concentration and showed a decrease on further concentration. 5. The production of microbial cells from NaOH treated rice straw was obtained 10.6mg/ml of culture medium.
