The change of soil microbial populations was studied at the burnt areas of Mt. Jirisan in Hddong-Gun, Kyungsangnam-Do, where the fire had occurred on Oct. 21 in 1997. On the first day of the fireextinguished, the microbial density (CFUIg dry soil) was investigated at the surface, 5 cm, 10 cm, 20cm, and 30 cm depth of soils. Bacteria at the surface and 5cm depth of burnt sites were estimatedwith the low density level of $10^2$ CFW/g soil comparing to the $10^6$ CFUIg soil of the neighboring unburntsites. Actinomycetes of burnt sites were completely disappeared at the surface, and were estimatedwith the low density level of $10^3$ CFUig soil at thc 5 cm depth comparing to the 10"CFUigsoil at the depth of unburnt sites. Fungi wcrc not isolated at the surface and 5 cm depth at all.However, the rarest lire was not found to decrease the microbial populalions at the lower depths than10 cm. In addilion, the recovery or soil microbial populations following the fire was bimonthly investigatedat the surrace and 5 cm depth. Most of microbial densities at the burnt sites were greatlyincreased two months after the fire, being enough to he compared with the neighboring unburnt sites.However, actinomycetes only at the surface of burnt sites still were estimated the low density level of$10^4$ CFUig soil 4 months after the fire comparing to the $10^6$CFIg soil of unburnt sites.TEX>CFIg soil of unburnt sites.

This study was conducted to determine the fermentative characteristics of wheat bran inoculated with a starter culture of direct-fed microbes as a microbial wheat bran (DMWB) feed additive. Wheat bran was prepared with 1% (w/w, 0.5% Lactobacillus plantarum and 0.5% of Saccharomyces cerevisiae) starter culture treatment (TW) or without starter culture as a control (CW). Those were fermented under anaerobic conditions at $30^{\circ}C$ incubation for 3 days. Samples were taken at 0, 1, 2, and 3 days to analyze chemical composition, microbial growth, pH, and organic acid content. Chemical composition was not significantly different between CW and TW (p > 0.05). In TW, the number of lactic acid bacteria and yeast increased during the 3 days of fermentation (p < 0.05) and the population of lactic acid bacteria was significantly higher than in CW (p < 0.05). After 3 days, the number of yeast in TW was $7.50{\pm}0.07log\;CFU/g$, however, no yeast was detected in CW (p < 0.05). The pH values of both wheat bran samples decreased during the 3 days of fermentation (p < 0.05), and TW showed significantly lower pH than CW after 3 days of fermentation (p < 0.05). Contents of lactic acid and acetic acid increased significantly at 3rd day of fermentation in TW. However, no organic acids were generated in CW during testing period. These results suggest that 3 days of fermentation at $37^{\circ}C$ incubation after the inoculation wheat bran with starter culture makes it possible to produce a direct-feed with a high population of lactic acid bacteria at more than $10^{11}CFU/g$.
Ecological characteristics of microorganisms in tideland soils were studied by investigation of microbial diversity and population. Twenty soil samples were taken at surface, 10, 20 and 30 cm depth each. Bacteria, actinomycetes and fungi were isolated on each selective isolation medium containing different concentration of NaCl. Actinomycetes were the most isolated from soil samples taken at 10 cm depth and isolated by humic acid-vitamin (HV) medium without sea water or salt. Twenty nine strains of actinomycetes were isolated at surface soil and 74, 39, 37 strains were at 10, 20, and 30 cm depth, respectively. All these isolates were analysed and grouped by random amplified polymorphic DNA (RAPD)-PCR analysis. Many of the isolates were clustered into Microtetraspora and Pseudonocardia. Fungal isolates were highly distributed at the surface soil and isolated well on potato dextrose agar (PDA) medium with sea water. Bacterial isolates were higly distributed at surface soil and isolated well by nutrient medium without sea water or salt. Soil samples taken at 10 cm depth showed the highest microbial diversity and population.

A biosurfactant-producing microorganism, .Pseudomouos sp. EL-G527 was isolated from activated sludge by enrichment culture when grown on mineral salt medium containing n-hexadecane as a carbon source. The biosurfactant from .Pseudomonar sp. EL-G527 exhibited lesser toxicity to bacterial population than synthetic surfactants and in the biodegradation test, biosurfactant was rapidly degraded and lost its activity as surface active material after 1 day incubation. In this study, the biosurfactant from Pseudomonas sp. EL-G527 was effective surface-active compound, more biodegradable and less toxic to microbial ecosystem than various synthetic surfactants.
Population changes of aerobic bacteria and lactic acid bacteria and changes of pH, acidity, and total sugar content were creased from initial period of fermentation. Lactic acid bacteria Increased during the first one nay rapidly and thereafter they creased slowly thereafter. Acidity increased on the third day of fermentation. Acidity was very low In Kimchi containing both Total sugar content decreased gradually from initial period to late period of fermentation.
The behavior of biological particles in DAF (dissolved air flotation) process was analyzed by employing PBT (Population balance theory). After decline growth phase of activated sludge, the value of the initial collision-attachment efficiency was increased over than 0.35 corresponding relatively high value in the whole life cycle of microorganism. For practical application of DAF as a solid separation process. It is desirable that microbial particles should be operated to perform high solid removal efficiency in biological wastewater treatment.
A series of experiments was conducted to evaluate phytin phosphohydrolysis actlVlty in the rumen and to isolate phytase positive rumen bacteria. Endogenous phytase activity of wheat bran was estimated and compared with that of bacterial phytin phosphohydrolysis. Substantial phytase activity was detected in wheat bran during in vitro rumen incubation. Bacterial phytase activity was suggested not to be high. Only two facultative anaerobes, Klebsiella sp. and Corynebacterium sp. were isolated as phytase producing organisms. These belonged to a minor microbial group in the rumen population. Protozoal fraction showed an initial velocity of phytin phosphohydrolysis 7 times higher than the bacterial fraction.
Bacteriophages (phages) are known determinants of kimchi microbial ecology. Lactobacillus plantarum is related to kimchi over-acidification during the late stages of kimchi fermentation. A phage infecting Lac. plantarum was isolated from kimchi and characterized. The phage population for kimchi in a market was 2.3 log particles/mL, which corresponded to 32% of the bacterial population on a log scale. The isolated phage was designated as ΦLP12116. ΦLP12116 which belonged to the Siphoviridae family and has a very narrow host range, infecting only Lac. plantarum. The phage was stable at a lactic acid concentration of 1.0% and pH 4.0 at 4℃, indicating that it could survive in kimchi. In the kimchi extract broth treated by the phage, the growth of Lac. plantarum KCCM 12116 was inhibited by 2.2 log CFU/mL compared to the growth in non-phage-treated broth. Therefore, this study suggests that the growth of Lac. plantarum, which is known as an acid-producing strain during late fermentation in kimchi, may be controlled using the phage.

Organic amendment practices can influence diversity and activities of soil microorganisms. There is a need to investigate this impact compared with other types of materials. This study was carried out to evaluate the long term effects of chemical and organic fertilizer on soil microbial community in upland field. During the last 11 years green manure, rice straw compost, rapeseed cake, pig mature compost, NPK, and NPK + pig mature compost were treated in upland soil. Organic fertilizer treatment found with high bacterial colony forming units (CFUs) as compared to chemical and without fertilizer treatment. There was no significant difference in the actinomycetes and fungal population. The average well color development (AWCD) value was the highest in green manure and, the lowest in without fertilizer treatment. Analyses based on the denaturing gradient gel electrophoresis (DGGE) profile showed that rice straw compost and pig mature compost had a similar banding pattern while rapeseed cake, NPK, NPK + pig mature compost and without fertilizer treatment were clustered in another cluster and clearly distinguished from green manure treatment. Bacterial diversity can be highly increased by the application of organic fertilizer while chemical fertilizer had less impact. It can be concluded that green manure had a beneficial impact on soil microbial flora, while, the use of chemical fertilizer could affect the soil bacterial communities adversely.
The present study compared the microbial diversity and activity during the application of various bioremediation processes to crude oil-contaminated soil. Five different treatments, including natural attenuation (NA), biostimulation (BS), biosurfactant addition (BE), bioaugmentation (BA), and a combined treatment (CT) of biostimulation, biosurfactant addition, and bioaugmentation, were used to analyze the degradation rate and microbial communities. After 120 days, the level of remaining hydrocarbons after all the treatments was similar, however, the highest rate (k) of total petroleum hydrocarbon (TPH) degradation was observed with the CT treatment (P<0.05). The total bacterial counts increased during the first 2 weeks with all the treatments, and then remained stable. The bacterial communities and alkane monooxygenase gene fragment, alkB, were compared by denaturing gradient gel electrophoresis (DGGE). The DGGE analyses of the BA and CT treatments, which included Nocardia sp. H17-1, revealed a simple dominant population structure, compared with the other treatments. The Shannon-Weaver diversity index (H') and Simpson dominance index (D), calculated from the DGGE profiles using 16S rDNA, showed considerable qualitative differences in the community structure before and after the bioremediation treatment as well as between treatment conditions.
