바이오매트 형성에 의한 산성광산배수 내 중금속 유출질량 제거효율 (Removal Efficiency of Heavy Metals in Acidic Mine Drainage from Microbial Mats)

  • This research investigated to reduce mass of heavy metals in AMD(acid mine drainage) by microbial mats formed on the channel bed. As, Cd, Cu, Fe, Mn and Zn components were monitored in water and microbial mats, at three points (AMD1, AMD2 and AMD3), in a total of six times. Average daily discharge mass of heavy metals was highest in July, Fe component contained more than 76% of total discharge mass. Discharge mass of heavy metals of AMD and heavy metal contents in microbial mats decreased with downstream at channel. Heavy metal components that average daily discharge mass is over 0.5 kg were Fe, Cu and Zn, and they were highest in July. Average removal efficiency of heavy metals in AMD was highest about 21% in Fe, this microbial mats were due to form from precipitation of Fe component in AMD by aerobic iron bacteria. Relative content for As component in microbial mats than AMD was over 16 times, this As components were due to absorb at iron oxide and iron hydroxide on the surface of microbial mats.

Tube Dilution Technique과 Ager Plate Smear Method에 의한 키토산의 MPSA 항미생물성 (A Study on the Antimicrobial Activity of Chitosan on the MRSA by Tube Dilution Technique and Agar Plate Smear Method)

  • Three different types of chitosan were prepared from red crab shells to study anti-microbial activity of chitosan on pathogenic bacteria, MRSA(Methicillin-resistant. Staphylococcus aureus), Water-insoluble chitosan, whose degree of deacetylation is kept over 90% and molecular weights are 20,000, 500,000, 150,000, 80,000, and 40,000, respectively. Water-soluble chitosan, whose degree of deacetylation is about 48% and molecular weights are 200,000 and 80,000. Water-soluble chitosan, whose degree of deacetylation is 82% and molecular weight is 3,900. The anti-microbial activities of three types of chitosan were investigated by Tube Dilution Technique(TDT) and Agar Plate Smear Method(APSM). And the following conclusions are made ; Chitosan having 5 different types of M.W chitosan (over 90% deacetylation) showed similar anti-microbial activities at over 0.05% concentration. Especially, chitosan having M.W 40,000 150,000 showed the excellent anti-microbial activity. The anti-microbial activity of chitosan was enhanced when the chitosan/acetic add solution was aged for 7days. The anti-microbial activity of chitosan was only shown at chitosan/acetic acid solution. The anti-microbial activity was not detected in chitosan solution dissolved in neutral pH water. Therefore, it can be concluded that the anti-microbial activity was due to NH3+ cationic ion of chitosan in acidic aqueous solution.

균주를 접종하여 제조한 청태전 차의 관능적 특성과 생리활성 효과 (Sensuous Characteristics and Physiological Activity of Cheongtaejeon Tea Produced with the Inoculation of Microbial Strains)

  • This study was conducted to restore our traditional Cheongtaejeon tea and to develop the special products. We inoculated Cheongtaejeon tea with lactobacilli (Lactobacillus plantarum CHO 25) and the mixed microbial strains (L. plantarum CHO 25 + Saccharomyces cerevisiae + Bacillus amyloliquefaciens CHO 104). We also examined the sensuous characteristics and physiological activity of Cheongtaejeon tea which was produced by the inoculation of microbial strains. The external appearance of Cheongtaejeon teas were not significant among the teas which were produced with or without the inoculation of L. plantarum CHO 25 and the mixed microbial strains. The taste of the tea increased most in Cheongtaejeon tea which was produced without the inoculation of microbial strains. The taste and liking of Cheongtaejeon tea which was inoculated with Aspergillus oryzae and Aspergillus niger decreased significantly, and it was not suitable to drink. Total phenolics compound contents, total flavonoid contents and DPPH ($\alpha,\alpha$-diphenyl-$\beta$-picryl-hydrazyl) radical scavenging activity of Cheongtaejeon tea extracts increased much more in the order of that produced with the inoculation of L. plantarum CHO 25, control and that with the mixed microbial strains. However, nitrite radical scavenging activity in 1,000 mg/L Cheongtaejeon tea hot water extracts were in the order of the control (94.4%), the inoculation of L. plantarum CHO 25 (93.6%) and the mixed microbial strains (91.1%). Overall results indicated that the sensuous characteristics increased most in Cheongtaejeon tea which was produced without the inoculation of microbial strains, and those physiological activities in tea with the inoculation of L. plantarum CHO 25.

The Effect of Calcium Level on Microbial Phytase Activity and Nutrient Balance in Swine

  • Three barrows weighing 45.0 kg, fitted with simple T-cannulas in both the duodenum and terminal ileum, were assigned to diets in a $3{\times}3$ Latin Square design experiment to determine the effect of two calcium levels (0.8% vs 0.4%) on phytase activity and nutrient balance in growing pigs. The control diet contained 0.8% calcium, with no added inorganic phosphorus (0.45% total phosphorus) and no added phytase. The two additional experimental diets contained microbial phytase (750 phytase units/kg) and supplied either 0.8% or 0.4% calcium. With added microbial phytase, ileal and total tract digestibility of rotal phosphorus were improved by 20.9 and 13.8 percentage units, respectively (p=0.01). The apparent duodenal and ileal digestibility of phytate phosphorus were increased by 51.8 and 49.7 percentage units (p=0.01). Lowering dietary calcium in the presence of microbial phytase increased the digestibility of phytate phosphorus by an additional 10.9 (p=0.001) and 5.7 percentage units for duodenal and ileal digestibility, respectively. Supplementation with microbial phytase significantly reduced fecal excretion of nitrogen and phosphorus and increased the percentage of these nutrients retained by the pig. Lowering dietary calcium further increased the percentage of dietary phosphorus retained. Overall, reducing dietary calcium appeared to increase the effectiveness of added microbial phytase in degrading phytate phosphorus. As a result, care should be taken to avoid high levels of dietary calcium when supplementing swine diets with microbial phytase.

  • A phospholipase C (PLC) inhibitor (MT267-2B) was isolated from the culture broth of actinomycetes isolate MT2617-2 by the extraction with n-butanol and column chromatographic techniques. The molecular weight of the inhibitor was 1057, by the spectroscopic analyses of IR, $^{13}C$-and $^{1}H$-NMR and ESI-MS. The chemical structure of MT2617-2B was found to be a macrolide compound consisted of a hemiketal ring, polyhydroxyl and polymethyl groups, which had a malonate and guanidine group as its side chain. MT2617-2B produced its two isomers having the same molecular weight by standing in methanol solution at room temperature. Therefore, MT2617-2B was identified as copiamycin and niphithricin A, macrolide antibiotics. The values of $IC_{50}$ against PLC-${\gamma}$1 and PLC-${\beta}$1 were 25 and 50${\mu}$g/ml, respectively. MT2617-2B had antimicrobial activities against Staphylococcus aureus and Candida albicans, but not against Escherichia coli.

The relationship between odd- and branched-chain fatty acids and microbial nucleic acid bases in rumen

  • Objective: This study aims to identify the relationship between odd- and branched-chain fatty acids (OBCFAs) and microbial nucleic acid bases in the rumen, and to establish a model to accurately predict microbial protein flow by using OBCFA. Methods: To develop the regression equations, data on the rumen contents of individual cows were obtained from 2 feeding experiments. In the first experiment, 3 rumen-fistulated dry dairy cows arranged in a $3{\times}3$ Latin square were fed diets of differing forage to concentration ratios (F:C). The second experiment consisted of 9 lactating Holstein dairy cows of similar body weights at the same stage of pregnancy. For each lactation stage, 3 cows with similar milk production were selected. The rumen contents were sampled at 4 time points of every two hours after morning feeding 6 h, and then to analyse the concentrations of OBCFA and microbial nucleic acid bases in the rumen samples. Results: The ruminal bacteria nucleic acid bases were significantly influenced by feeding diets of differing forge to concentration ratios and lactation stages of dairy cows (p<0.05). The concentrations of OBCFAs, especially odd-chain fatty acids and C15:0 isomers, strongly correlated with the microbial nucleic acid bases in the rumen (p<0.05). The equations of ruminal microbial nucleic acid bases established by ruminal OBCFAs contents showed a good predictive capacity, as indicated by reasonably low standard errors and high R-squared values. Conclusion: This finding suggests that the rumen OBCFA composition could be used as an internal marker of rumen microbial matter.

마른김(Pyropia spp.) 가공 공정 경과에 따른 미생물 오염도 분석 (Analysis of Microbial Contaminants and Microbial Changes during Dried-laver Pyropia spp. Processing)

  • We investigated the levels of microbial contaminants and microbial hazards during dried-laver processing. We analyzed 321 samples obtained from 18 dried-laver Pyropia spp. manufacturing facilities, including water, swab-, and processing samples as well as final products. The levels of microbial contaminants, including viable cell counts (VCC) and coliform bacteria, increased as processing progressed. The sanitary indicator bacterium, Escherichia coli, was not detected in the final products although VCC levels were high, generally exceeding 5 log CFU/g. We also investigated changes in microbial contaminants at each processing step. Both VCC and total coliform dramatically increased after 4 days of continuous processing, indicating that microbial contaminants originated, mainly, from cross contamination during processing.

논과 밭 토양에서 토층간 미생물 군집의 차이 (Variation of Microbial Community Along Depth in Paddy and Upland Field)

  • 인지질 지방산을 분석하여 특정 미생물군의 수직적 분포와 토층간 미생물 군집 패턴을 조사하였다. 경북 농업기술원에 위치하고, 질소, 인산, 가리의 화학비료만 장기 연용한 논과 밭 포장에서 15 cm 깊이까지 토양을 채취하였다. 인지질 지표 지방산을 주요인 분석으로 분석하여 토양 미생물 군집을 분석한 결과 논과 밭 토양의 미생물 군집은 뚜렷하게 구분되었으며, 토층간 차이보다 논과 밭의 차이가 더 컸다. 논보다 밭은 토층이 깊어짐에 따라 미생물 군집이 급격하게 변하였는데, 미생물 군집 측면에서 밭보다 논의 표층이 더 두껍다고 볼 수 있다. cyclopropyl/monoenoic precursor 비율과 전체 포화지방산/전체 불포화 지방산 비율은 토심이 깊어짐에 따라 증가하였는데, 이는 토심이 깊어질수록 탄소원과 통기가 부족하기 때문에 일어나는 현상으로 보인다. 대체로 표토는 그램음성균, 곰팡이 등의 상대적 비율이 높고 토심이 깊어질수록 세균과 방선균의 상대적 비율이 높아졌다.

한국 전통 된장의 구강미생물에 대한 항균효과 (Anti-microbial Activity of Korean Fermented Soybean Paste (Doen-jang) Against Oral Microbes)

  • This study was performed to investigate the anti-microbial activity of extract from Korean fermented soybean paste (doen-jang) against 16 types of oral microbes, and to determine the minimum inhibition concentration (MIC) of the extract for three major microbes causing human oral diseases (Streptococcus mutans, Porphyromonas gingivalis, and Candida albicans). The extract was prepared using ethyl acetate and it was treated with the oral microbes at a concentration of 5.00 mg/ml (0.5%). The anti-microbial activity and MIC were measured using broth dilution method. Significant reduction of microbial activities of 16 types of oral microbes occurred when the soybean paste extract was added to the broth compared to the control (p<0.01), and striking inhibition (more than 99%) was observed in ten types. S. mutans, which causes dental caries, showed MIC at a concentration of 1.25 mg/ml for the extract. P. gingivalis, which causes adult periodontal disease, showed MIC at a concentration of 2.50 mg/ml for the extract. C. albicans, which causes denture stomatitis and angular stomatitis, showed MIC at a concentration of 20 mg/ml for the extract. These results indicate that ethyl acetate extract of doen-jang showed strong anti-microbial effect against 16 types of oral microbes, and the anti-microbial effect of the extract against oral microbes was stronger against bacteria than against fungi. The anti-microbial effect might be possibly enhanced by the fermentation of soybeans.

Occurrence of Toxigenic Fusarium vorosii among Small Grain Cereals in Korea

  • Fusarium graminearum species complex (FGSC) causes Fusarium head blight in small grain cereals. To date, four species (F. graminearum, F. asiaticum, F. boothii, and F. meridionale ) belonging to FGSC frequently occur in Korean cereals. In addition, we first reported the occurrence of additional species (F. vorosii ) within FGSC, which was isolated from barley, corn, and rice in Korea. Phylogenetic analysis of the Fusarium isolates of this group using combined multigene sequences confirmed species identification. Moreover, the macroconidia produced by these isolates were morphologically similar to those of the F. vorosii holotype. Chemical analysis indicated that the F. vorosii isolates produced various trichothecenes such as nivalenol and deoxynivalenol with their acetyl derivatives along with zearalenone. Pathogenicity tests demonstrated that all of the F. vorosii isolates examined were pathogenic on barley, corn, and rice with variation in aggressiveness. This study is the first report of F. vorosii in Korean cereals, their pathogenicity towards barley and corn, and their ability to produce trichothecenes and zearalenone.
