  • The effects of substrate concentration, enzyme concentration, reaction temperature, and water content were investigated in intramolecular esterification. This study used cyclohexane as organic solvent, power lipase as enzyme, and benzyl alcohol and octanoic acid as substrate. The initial reaction rate was found to be proportional to enzyme concentration; followed Michaelis-Menten equation for octanoic acid; and was inhibited by benzyl alcohol . The observed initial reaction rate first increased, then decreased with increasing reaction temperature, giving rise to the maximum rate at 20$\circ$. The drop in the reaction rate at higher temperature was to partition equilibrium change of substrate between organic solvent and hydration layer of enzyme molecule in addition to the deactivation by enzyme denaturation. Water layer surrounding enzyme molecule seemed to activate in organic solvent and the realistic reaction was done in the water layer. In the enzymatic reaction in organic solvent, the initial reaction rate was influenced by partition quilibrium of substrate, so the optimum condition of substrate concentration, enzyme concentration, reaction temperature, and water content would give a good design tool.

Effects of Intravenous Administration of Taurocholate on Hepatic Catechol-O-Methyltransferase Activity in Common Bile Duct Ligated Rats

  • Possible mechanism of decreased catechol-O-methyltransferase (COMT) activity in cholestatic rat liver was studied. Hepatic and serum COMT activities were determined from the experimental rats with common bile duct ligation (CBDL). The Michaelis-Menten constants in this hepatic enzyme were also measured. The activities of cytosolic, mitochondrial and mircosomal COMT as well as their Vmax values were found to be decreased significantly in CBDL plus taurocholic acid (TCA) injected group than in the control group, such as CBDL alone groups. However, their Km values in the experimental groups did not vary. Serum COMT activity increased slightly in the CBDL plus TCA injected group than in the control group. The above results suggest that TCA represses biosynthesis of the COMT in the liver. The elevated activity of the serum COMT is believed to be caused by the increment of membrane permeability of hepatocytes upon TCA mediated liver cell necrosis.

  • The possible mechanisms of increased aryl sulfotransferase (AST) isozymes activities in cholestatic rat liver were studied. Hepatic AST-I, II and -III, IV activities were determined from the experimental rats with common bile duct ligation (CBDL). The Michaelis-Menten constants in these hepatic enzymes were also measured. The activities of mitochondrial AST-I, II and -III, IV, and microsomal AST-III, IV as well as their Vmax values were found to be increased significantly in CBDL plus taurocholic acid (TCA) injected group than in the control group, such as CBDL alone groups. However, their Km values in the experimental groups did not vary. The results suggest that TCA stimulates biosynthesis of the AST in the liver.

  • Arthrobacter simplex (ATCC 6946) was cultured, induced and immobilized in acrylamide polymer. The characteristics of the immobilized whole-cell enayme were studied using hydrocortisone as the substrate. The enzyme activity was increased during the incubation of the gel particle in 0.5% peptone media. The ennzyme reaction kinetics of the Δ'-dehydrogenase (3-oxosteroid Δ'-oxydo reductase, E. C. 1.3.99.4) foliowed the Michaelis-Menten type. Km and Vm values were different significantly after immobilization of the cell. The optimum pH and temperature were changed, too. Nitrogen sources such as casitone, peptone or tryptone were good media for the enzyme reaction. And there was no need to add cofactors of the enzyme in the pre-sence of energy sources used in the test. The effect of metal ions on the enzyme activity was insignificant. Organic solvents were used increase the substrate concentration and there was no optimum solvent concentration depending on the substrate concentration.

Analysis of Kinetic Data of Pectinases with Substrate Inhibition

  • Enzyme kinetics data play a vital role in the design of reactors and control of processes. In the present study, kinetic studies on pectinases were carried out. Partially purified polymethylgalacturonase (PMG) and polygalacturonase (PG) were the two pectinases studied. The plot of initial rate vs. initial substrate concentration did not follow the conventional Michaelis-Menten kinetics, but substrate inhibition was observed. For PMG, maximum rate was attained at an initial pectin concentration of 3 g/l, whereas maximum rate was attained when the initial substrate concentration of 2.5 g/l of polygalacturonic acid for PG I and PG II. The kinetic data were fitted to five different kinetic models to explain the substrate inhibition effect. Among the five models tested, the combined mechanism of protective diffusion limitation of both high and inhibitory substrate concentrations (semi-empirical model) explained the inhibition data with 96-99% confidence interval.

  • The catalytic activities of several metalloporphyrin, wherein the porphyrins are TPP(5,lO,l5,20-Tetraphenyl-21H,23H-porphyrin) and (p-X)TPP (X =$CH_{3}O$, $CH_{3}$, F, Cl), are reported for the oxidation of styrene and it's derivatives. The electronic effects of substrates and porphyrins on the catalytic activity of metalloporphyrin containing the transition metal ion such as Mn(III) was discussed. Investigating the correlation between the Michaelis-Menten's rate parameters and the substituent constants, we are going to analyze the influences on the changes of catalytic activity or rate determining step during the processes of the formation and the dissociation of the M-oxo-olefin.

  • Recombinant E. coli JM109(pZH3-5/pMT) harboring manganese transport gene(mntA) and metal sequestering protein, metallothionein(MT), was cultivated to accumulate cadmium in aqueous phase. Bioaccumulation followed Michaelis-Menten type kinetics. Equilibrium isotherm showed Langmuir type isotherm. IPTG induction cell showed fast $Cd^{2+}$ uptake and had higher uptake rate than wild type and no-induced cell. The optimum pH and temperature for $Cd^{2+}$ uptake was 7 and $37^{\circ}C$, respectively. Manganese (0.01M) inhibited the $Cd^{2+}$ accumulation. However, $Cu^{2+}$, $Zn^{2+}$ and $Pb^{2+}$ did not affect the $Cd^{2+}$ bioaccumulation.

Characteristics of Nitrobenzene Degradation by Mycobacterium chelonae Strain NB01

  • A bacterial strain NB01, isolated from wastewater, was found to utilize nitrobenzene (NB) as the sole source of nitrogen, carbon, and energy. The strain was classified as a member of a high G+C Gram-positive group and identified as Mycobacterium chelonae based on an analysis of its 16S rRNA gene sequence. The strain grew on NB with a concomitant release of about 63% of the total available nitrogen as ammonia, suggesting a reductive degradation mechanism. The optimal pH and temperature for degradation were PH 7.0-8.0 and $30^{\circ}C$, respectively. The cell growth was retarded at NB concentrations above 1.8 mM. The degradation of NB followed Michaelis-Menten kinetics within the tolerance range, and the $K_m$ and maximum specific removal rate for NB were 0.33 mM and $11.04\;h^{-1}$, respectively.

QUENCHING OF TYPE II PHOTOSENSITIZER IN THEIR TRIPLET STATES BY $\alpha$-TOCOPHEROL VIA AN ELECTRON TRANSFER REACTION

  • Occurrence of an electron (or H atom equivalent to one electron plus H+) transfer from $\alpha$- tocopherol $\alpha$(TOH) to a number of photosensitizers in theri triplet states were investigated by monitoring the ESR signal of $\alpha$-chromaoxyl radical ($\alpha$(TO.) in ethanolic solutions of $\alpha$TOH and the sensitizers under continuous illumination. Every sensitizer molecule examined, such as protocholorophyllide (Pchl), hematoporphyrin and rose bengal which are generally regarded as efficient type II photosensitizers and thus have long-lived triplet states, was found to actively participate in an electro transfer reaction with $\alpha$TOH even under air-saturation conditions, generating $\alpha$TOH complex as an intermediate in a fashion of Michaelis-Menten type of reaction. For the reaction of $\alpha$TOH with triplet Pchl, the rate law was derived by applying the steady approximation for the binary complex, triplet Pchl-$\alpha$TOH , which turned out to be well consistent with the kinetic data.
