미숙과 복분자 추출물의 상체비만 개선 효과: 무작위배정, 이중눈가림, 위약-대조 임상연구 (Effects of Unripe Rubus Coreanus Extract on Upper Body Obesity: A Randomized, Double-Blind, Placebo-Controlled Clinical Trial)

  • Objectives Previously, we demonstrated that a 5% ethanol extract of unripe Rubus coreanus (5-uRCK) and ellagic acid has hypocholesterolemic and antiobesity activity in high-fat diet-fed animals. Therefore, we conducted a clinical study on the anti-obesity effect of 5-uRCK in 140 Korean adults (aged 19-70 years, body mass index (BMI) ≥25 kg/m2). Methods The participants were randomly assigned to two groups and were administered the placebo (n=70) or 5-uRCK extract (800 mg, n=70) daily for 12 weeks. The subjects were instructed to maintain their usual dietary intake and normal physical activity. Anthropometrics, body composition (dual-energy X-ray absorptiometry), and blood parameters were compared between the two groups. Results Average body weight and BMI did not show any significant changes; however, changes in upper body fat mass between the two groups was significantly different, as determined using analysis of covariance adjusted for baseline of percent body fat and drinking units. Moreover, Although not significant, the trunk fat mass and percent trunk fat tended to decrease after 5-uRCK supplementation. There was no significant difference in other laboratory biomarkers between the two groups. All safety parameters were within normal ranges in the both group. Conclusions In the present study, 5-uRCK significantly reduced upper body fat in obese subjects after 12 weeks, which indicates that it may help improve android-type obesity. Furthermore, the supplement is safe and well tolerated.

The responsibility of C-terminal domain in the thermolabile haemolysin activity of Vibrio parahaemolyticus and inhibition treatments by Phellinus sp. extracts

  • The thermolabile haemolysin (tlh) of Vibrio parahaemolyticus (Vptlh) from V. parahaemolyticus is a multiple-function enzyme, initially describes as a haemolytic factor activated by lecithin and phospholipase A2 enzymatic activity (Shinoda, 1991; Vazquez-Morado, 2021; Yanagase et al., 1970). Until now, the tlh structure has hypothesized including N-terminal and C-terminal domain, but what domain of the Vptlh structure does the haemolytic activity has not been refined yet. In this study, a 450-bp VpTLH nucleotide sequence of the entire Vptlh gene encoded the C-terminal domain cloned firstly to examine its responsibility in the activity of the Vptlh. The C-terminal domain fused with a 6-His-tag named the His-tag-VpC-terminal domain was expressed successfully in soluble form in the BL21 (DE3) PlysS cell. Remarkably, both expression and purification results confirmed a high agreement in the molecular weight of the His-tag-VpC-terminal domain was 47 kDa. This work showed the His-tag-VpC-terminal domain lysed the erythrocyte membranes in the blood agar and the phosphate buffered saline (0.9%) media without adding the lecithin substrate of the phospholipase enzyme. Haemolysis occurred at all tested diluted concentrations of His-tag-VpC-terminal domain (p < 0.05), providing evidence for the independent haemolytic activity of the His-tag-VpC-terminal domain. The content of 100 ㎍ of the His-tag-VpC-terminal domain brought the highest haemolytic activity of 80% compared to that in the three remaining contents. Significantly, the His-tag-VpC-terminal domain demonstrated not to involve the phospholipase activity in Luria-Bertani agar supplemented with 1% (vol/vol) egg yolk emulsion. All results proved the vital responsibility of the His-tag-VpC-terminal domain in causing the haemolytic activity without the required activation by the phospholipase enzyme. Raw extracts of Phellinus igniarus and Phellinus pipi at 10-1 mg/mL inhibited the haemolytic activity of the His-tag-VpC-terminal domain from 67.7% to 87.42%, respectively. Hence applying the His-tag-VpC-terminal domain as a simple biological material to evaluate quickly potential derivatives against the Vptlh in vivo conditions will accessible and more advantageous than using the whole of the Vptlh.

Quality Improvement of Crude Glycerol from Biodiesel Production Using Activated Carbon Derived from Krabok (Irvingia malayana) Seed Shells

  • This research investigated the preparation of activated carbon derived from Krabok (Irvingia malayana) seed shells to improve the quality of crude glycerol obtained during biodiesel production. The activated carbon was prepared using a dry chemical activation method with NaOH, utilizing an innovative biomass incinerator. The results revealed that the resulting KC/AC-two-step exhibited favorable physicochemical adsorption properties, with a high surface area of 758.72 m2/g and an iodine number of 611.10 mg/g. These values meet the criteria of the industrial product standard for activated carbon No. TIS 900-2004, as specified by the Ministry of Industry in Thailand. Additionally, the adsorption efficiency for methylene blue reached an impressive 99.35 %. This developed activated carbon was then used to improve the quality of crude glycerol obtained from biodiesel production. The experimental results showed that the KC/AC-two-step increased the purity of crude glycerol to 73.61 %. In comparison, commercially available activated carbon (C/AC) resulted in a higher crude glycerol purity of 81.19 %, as analyzed by the GC technique. Additionally, the metal content (Zn, Cu, Fe, Pb, Cd, and Na) in purified glycerol using KC/AC-two-step was below the standards for heavy metals permitted in food and cosmeceuticals by the Food and Drug Administration of Thailand and the European Committee for Food Contact Materials and Articles. As a result, it can be inferred that Krabok seed shells have favorable properties for producing activated carbon suitable as an adsorbent to enhance crude glycerol purity. Furthermore, the improved crude glycerol from this research has potential for various industrial applications.

Home-made 요구르트와 시판 중인 요구르트에서 분리한 젖산균의 기능적 특성 조사 (Studies on the Functional Properties of Lactic Acid Bacteria Isolated from Home-made Yogurt and Commercial Yogurt)

  • 본 연구는 home-made 요구르트에서 분리한 Lactobacillus casei SK-7과 시중에서 판매되고 있는 요구르트에서 분리한 Lactobacillus bulgaricus YK-11의 기능성을 비교 분석하기 위해 실시하였다. 먼저 분리세균인 SK-7과 YK-11의 생리화학적 특성조사를 진행하였다. 이들 균주는 16S rRNA 염기서열을 이용한 계통유전학적 분석방법으로 동정하여, 각각 L. casei SK-7와 L. bulgaricus YK-11로 명명하였다. 16S rRNA 염기서열을 바탕으로 SK-7과 YK-11의 유전적 계통수를 작성하였다. 72시간 동안 SK-7과 YK-11 배양에서 lactic acid와 acetic acid의 생산, 그리고 pH 변화를 조사하였다. 배양기간 동안 L. casei SK-7과 L. bulgaricus YK-11의 몇가지 기능성을 조사하였다. L. casei SK-7과 L. bulgaricus YK-11 배양상등액은 주어진 nitrite를 각각 93.9%와 88.2% 소거하였다. SK-7 and YK-11 배양상등액의 항산화능은 각각 62.6%와 54.9%였으며, ${\beta}$-galactosidase 활성은 14.9 units/mg과 13.1 units/mg로 측정되었다. 항미생물능력은 SK-7과 YK-11 배양의 20배 배양농축액으로 조사되었으며, SK-7은 이 조사에 사용된 모든 미생물에서 명백한 것으로 관찰되었으나, YK-11에서는 관찰되지 않았다. 부가적인 기능성 연구가 이루어져야 되겠지만, 본 연구에서는 home-made 요구르트에서 분리된 젖산균의 기능성은 시판요구르트에서의 기능성과 비교하여 우수한 것으로 조사되었다.

부유 및 퇴적의 분체 조건이 화재폭발 특성에 미치는 영향 (Effect of Powder Condition on the Fire and Explosion Characteristics of Suspended and Deposited Dusts)

  • 동일 분체특성의 분진이 평균입경, 농도, 분진조건(부유 또는 퇴적) 변화에 따른 화재폭발 위험성을 조사하였다. 이를 위해 20L분진폭발시험장치, 열중량분석장치, 연소속도시험장치(UN시험법)를 사용하였다. 4종 분진(Sugar, Mg, Al, Zr)의 입경이 서로 다른 8개 분진 시료에 대하여 부유 분진의 폭발특성과 화염전파속도(FPV), 그리고 퇴적분진의 화염확산속도(FSV)를 조사하였다. 부유 분진 조건에서 Mg 및 Al 분진은 입경이 감소하면 폭발 위험성이 증가하였지만, Sugar는 입경 변화에 따른 폭발 위험성의 영향이 거의 나타나지 않았다. 부유 분진의 화염전파속도(FPV)는 마이크로 범위에서의 입경 변화보다 마이크로에서 나노로 입경이 감소하면 크게 증가하였다. 퇴적층의 화염확산속도(FSV)는 수평면(기울기 0°)보다 경사면(기울기 30°)에서 증가하는 경향을 나타냈으며, 경사면(기울기 30°) 퇴적층 조건에서는 상방 전파가 하방 전파보다 높게 나타났다.

Effects of Antioxidant Supplementation on Antioxidant Status and PHA-Stimulated Interleukin-2 Production by Peripheral Blood Mononuclear Cells in the Elderly Women

  • This study was done to investigate effects of antioxidant supplementation on phytohemagglutinin (PHA) -stimulated interleukin-2 (IL-2) production by peripheral blood mononuclear cells (PBMCs) in elderly women. This study was designed as a placebo-controlled, single-blinded, randomized intervention trial. Twenty four elderly women aged over 60 years, visitings social welfare center in Seoul were divided into 3 groups, placebo (n = 8), vitamin C supplemented (n = 8) , and vitamin E supplemented (n = 8) groups. Experimental groups were given either 1000mg of L-ascorbic acid or 400 ill of d- $\alpha$-tocopherol for 4 weeks. There was no significant difference in antioxidant vitamins intakes and their plasma levels among pre-intervention groups. Plasma vitamin C or E levels was significantly increased after vitamin C or E sup-plementations. The increases of plasma thiobarbituric acid-reactive substance (TBARS) levels in the placebo group were significantly higher than those of the supplemented 2 groups. There were no significant differences in the changes of plasma IL-2 level between pre- and post-intervention among the 3 groups. However there was a significant increase in PHA­stimulated IL-2 production by PBMCs after 4-week vitamin E or vitamin C supplementation. Particularly, vitamin E supplemented group showed a higher PHA-stimulated IL-2 production than vitamin C supplemented group. These results indicate that vitamin E or vitamin C supplementation might enhance mitogen-stimulated cytokine production by immune cells, which could be one of the factors to improve health status in the elderly.

항산화제가 bleomycin에 의해서 유발되는 랫트 폐장병변에 미치는 영향에 관한 연구 (The effect of antioxidant on pulmonary lesions induced by bleomycin in rats)

  • The purpose of this study was to investigate the effect of antioxidants (vitamin E, selenium, and coenzyme $Q_{10}$) on the bleomycin-induced pulmonary lesions in male rats. Sprague-Dawley male rats were divided into 4 treatment groups ($T_1$, $T_2$, $T_3$, $T_4$) and 4 control groups ($C_1$, $C_1$, $C_3$, $C_4$). The treatment groups of rats weie given a single intratracheal dose of bleomycin (1.5 units/rat) and control groups of rats were given a single intratracheal dose of normal saline (0.15ml/rat). The rats in the $T_1$ group and $C_1$, group were dosed with normal saline (0.5ml/kg/day), the rats in the $T_2$ group and $C_2$ group were dosed with vitamin E (50mg/kg/day), the rats in the $T_3$ group and $C_3$ group were dosed with sodium selenite (3mg/kg/day) and the rats in the $T_4$ gronp and $C_4$ group were dosed with coenzyme $Q_{10}$ (2.5mg/kg/day) intraperitoneally for 7 days or 14 days, respectively. Animals were killed at 7th and 14th day after dosing with bleomycin or saline. The results obtained were as follows: 1. Lung wet weight of treatment groups of rats was increased significantly while body weight gain of them was decreased significantly in comparison with that of control groups of rats (p<0.01). 2. The ratio(%) of lung wet weight to final body weight of treatment groups of rats was increased significantly in comparison with that of control groups of rats (p<0.01). 3. The main histopathological findings of lungs observed in rats at 7th day after dosing with bleomycin were proliferation of the type II alveolar epithelial cells and fibroblasts, increased invading of macrophages into lesions, round cell infiltration and perivascular edema. 4. Lung fibrous tissues were markedly increased in rats observed at 14th day after dosing with bleomycin. 5. Pumonary lesions observed in rats dosed with bleomycin and antioxidants(vitamin E, selenium, coenzyme $Q_{10}$) were not significantly different from those of rats given bleomycin alone.

게르마늄 시용에 따른 녹두 생육 및 부위별 Ge 함량 변화 (Effects of Germanium(Ge) on Growth, Yield and Ge Content of Mungbean)

  • 본 연구는 게르마늄 함유 녹두 생산을 위해 게르마늄의 효과적인 처리방법과 이행특성 등을 구명하고자 수행하였다. 1. 액상 게르마늄(7 mg/l) 개화기 엽면살포구 1, 2차 수확 종실의 게르마늄 함량은 각각 38.7, $14.1{\mu}g/kg$로 입상 게르마늄(7 mg/kg)을 기비와 함께 시용하는 것보다 각각 2.5, 2.3배 많다. 2. 게르마늄 3.5, 7, 14, 28 mg/l 를 개화기 엽면살포구 1, 2차 수확 종실의 게르마늄 함량은 각각 $14.9{\sim}77.8{\mu}g/kg,\;6.9{\sim}26.7{\mu}g/kg$로 엽면살포 농도에 비례하여 많았으나 1차 수확 종실이 2차 수확 종실에 비해 $2.2{\sim}4.1$배 많다. 반면에 무처리구 1, 2차 수확 종실의 게르마늄 함량은 각각 1.9, $3.2{\mu}g/kg$이었다. 3. 게르마늄 7 mg/l를 2회나 3회 엽면살포시 $1{\sim}3$차 수확 종실의 게르마늄 함량은 $20{\mu}g/kg$ 이상으로 일정 수준의 게르마늄 축적이 가능하였다. 4. 게르마늄 $96{\mu}g/kg$이 함유된 녹두 종실에서 자엽의 게르마늄 함량은 $138{\mu}g/kg$으로 종피보다 단위 중량당 79% 많았다. 5. 본 연구에 이용된 게르마늄의 제형, 엽면살포 농도 및 횟수에 따른 녹두 생육과 수량은 유의차가 없다.

산성(酸性) 신개간전(新開墾田) 토양(土壤)의 염기포화도(鹽基飽和度)와 대두수량(大豆收量) (Effect of the base saturation rate of acidic upland soils on the yield of soybean)

  • 염기포화도(鹽基飽和度)가 40% 미만(未滿)이고 유효인산함량(有效燐酸含量)이 매우 낮은 3개토양(三個土壤)에 대(對)해 염기포화도(鹽基飽和度) 조절(調節) 및 인산시용(燐酸施用)의 효과(效果)를 검토(檢討)하기 위(爲)해 인산(燐酸) 40 ppm 및 80 ppm을 시용(施用)하고 염기포화도(鹽基飽和度)를 무조절(無調節), 60% 및 80%로 조절(調節)하여 대두(大豆)를 공시작물(供試作物)로 pot시험(試驗)을 실시(實施)했든바 다음과 같은 결과(結果)를 얻었다. 1) 유효인산함량(有效燐酸含量)이 매우 적었던 이들 토양(土壤)에 대(對)해 인산(燐酸) 40ppm 및 80ppm 시용(施用)은 효과적(效果的)인 시용량(施用量)이 아니었던 것 같다. 2) 염기포화도(鹽基飽和度)를 60%까지 상승(上昇)시키도록 조절(調節)한 경우(境遇) 무조절(無調節) 비(比)해 현저(顯著)한 수량(收量)의 증가(增加) 및 식물(植物) 중(中)의 염기(鹽基)의 증가(增加)가 있었으나 그 이상(以上)에서는 이런 효과(效果)가 없었다. 6) Ca 포화도(飽和度)와 Mg 포화도(飽和度)의 효과(效果)를 개별적(個別的)으로 살펴볼 때 Ca 포화도(飽和度)는 35~55%로 충분(充分)한 것으로 나타났으나 Mg 포화도(飽和度)는 10~15%의 범위(範圍)에서 직선적(直線的)으로 수량(收量)과 정(正)의 상관(相關)을 나타 내었으므로 우리나라 조건(條件)에서는 Mg의 포화도(飽和度)가 15% 보다 훨씬 높아야 할것이 추측(推測)되었다. 4) 이 점(點)으로 미루어 보아 우리나라의 전(全) 경지(耕地)에는 Mg의 상대적(相對的) 부족(不足)이 있을것이 예상되며 Mg가 포함(包含)되지 않는 석회(石灰)의 일방적(一方的) 시용(施用)은 이를 악화(惡化)할 것이다.

Butachlor의 4종 담수조류(freshwater algae)에 대한 생장영향 (Effects of Butachlor on Growth of Four Freshwater Algae)

  • 우리나라에서 사용되고 있는 벼재배용 제초제인 butachlor의 담수조류에 대한 생장영향을 알아보고자 녹조류 Selenastrum capriconutum, Scenedesmus subspicatus, Chlorella vulgaris 3종과 규조류 Nitzschia palea 1종에 대해 72시간 생장저해시험(growth inhibition test)을 수행하였다. 시험결과 butachlor에 대한 S. capricornutum, S. subspicatus, C. vulgaris, 그리고 N. palea의 생장율 기준 반수영향농도($EC_{50}$)는 각각 0.0022, 0.019, 8.67 그리고 $4.94\;mg\;L^{-1}$이었고, 무영향농도(NOEC)는 0.0008, 0.0016, 5.34 그리고 $2.92\;mg\;L^{-1}$로 나타나 S. capricornutum가 가장 높은 감수성을 보였다. 양성대조물질인 $K_2Cr_2O_7$에 대한 S. capricornutum, S. subspicatus, C. vulgaris 그리고 N. palea의 생장율 기준 $EC_{50}$은 각각 0.91 0.78, 0.85 그리고 0.57 mg $EC_{50}$이었고, NOEC는 각각 0.2, 0.2, 0.2 그리고 0.18 mg $EC_{50}$였다. butachlor를 논에 살포하였을 때 논물에 전량 용해된 후 배수로를 통해 농수로에 배출될 때를 가정한 환경추정농도(Predicted Environmental Concentration; PEC)에 따른 생장저해율을 계산한 결과, S. capricornutum와 S. subspicatus는 각각 100%와 75%의 생장율 저해를 보였으나 C. vulgaris와 N. palea는 영향이 없었다.
