Interaction of Antitubercular Drug (II) - Drug Interaction of Rifampicin and Isoniazid (항결핵약물의 상호작용 (II) - 리팜피신과 이소니아짓의 약물상호작용)

  • Rifampicin is an indispensable drug along with isoniazid for the control of tuberculosis and is usually prescribed as the combination of rifampicin and isoniazid. This paper is attemtped to investigate the interaction of rifampicin and isoniazid. Isoniazid was administered orally at a dose of 30mg/kg of rabbits pretreated with rifampicin 7.5mg/kg, 15mg/kg, and 30mg/kg, respectively twice daily for 9 days. The results are as follows: The blood level and relative bioavailability of isoniazid were decreased significantly (p<0.05) by rifampicin at a dose of 15mg/kg and 30mg/kg. The renal clearance of total isoniazid and ratio of its metabolites to isoniazid were increased significantly (p<0.05) by rifampicin at a dose of 15mg/kg and 30mg/kg. It seemed to be due to enzyme induction by rifampicin. Elimination rate constant ($\beta$) of isoniazid was increased and half life ($t_{1/2$\beta}$) was decreased by rifampicin pretreatment. Dosage regimen of isoniazid after long term administration of rifampicin should be adjusted carefully.

A Diagnostic Study of Pulsed Plasma Process for Reactive Deposition (반응성 증착용 펄스 플라즈마 공정의 진단)

  • A real-time monitoring of an immersed antenna type inductively coupled plasma (ICP) was done with optical emission spectroscopy (OES) to check the reports that sputtered atom density is decreasing as the ICP power is increased. At 10 mTorr pressure of Ar, Mg was sputtered by a bipolar pulsed power supply into 2 MHz ICP which has an insulator covered 2.5 turn antenna. Emitted light was collected in two different positions: above the target and inside the ICP region. With 100 W of Mg sputtering power, the intensities of Mg I (285.06 nm), Mg II (279.48 nm), Ar I (420.1 nm) were increased constantly with ICP power from 100 W to 600 W. At 500 W, the intensity of $Mg^+$ exceeded that of Mg under PID controlled discharge voltage of 180 V. The ratio of Mg II/Mg I was increased from 0.45 to 2.71 approximately 6 times.

Characteristics of manganese removal by ozonation: Effect of existing co-ion and optimum dosage (오존을 이용한 용존성 망간 제거 특성: 공존이온의 영향 및 최적주입량)

  • This study is focused on manganese (Mn(II)) removal by ozonation in surface water. Instant ozone demand for the water was 0.5 mg/L in the study. When 0.5 mg/L of Mn(II) is existed in water, the optimum ozone concentration was 1.25 mg/L with reaction time 10 minutes to meet the drinking water regulation. The ozone concentration to meet the drinking water regulation was much higher than the stoichiometric concentration. The reaction of soluble manganese removal was so fast that the reaction time does not affect the removal dramatically. When Mn(II) is existed with Fe, the removal of Mn(II) was not affected by Fe ion. However As(V) is existed as co-ion the removal of Mn(II) was decreased by 10%. Adding ozone to surface water has limited effect to remove dissolved organic matter. When ozone is used as oxidant to remove Mn(II) in the water, the existing co-ion should be evaluated to determine optimum concentration.

Textural and Genetic Implications of Type II Xenoliths Enclosed in Basaltic Rocks from Jeju Island (제주도 현무암에 포획된 Type II 포획암: 성인과 조직적 특성)

  • Ultramafic xenoliths from southeastern part of Jeju Island can be grouped into two types: Type I and Type II. Type I xenoliths are magnesian and olivine-rich peridotite (mg#=89-91), which are commonly found at the outcrop. Most previous works have been focused on Type I xenoliths. Type II xenoliths, consisting of olivine, orthopyroxene and clinopyroxene with higher Fe and Ti components (mg#=77-83) and lower Mg, Ni, Cr, are reported in this study. They are less common with a more extensive compositional range. The studied Type II xenoliths are wehrlite, olivine-clinopyroxenite, olivine websterite, and websterite. They sometimes show ophitic textures in outcrops indicating cumulate natures. The textural characteristics, such as kink banding and more straight grain boundaries with triple junctions, are interpreted as the result of recrystallization and annealing. Large pyroxene grains have exsolution textures and show almost the same major compositions as small exsolution-free pyroxenes. Although the exsolution texture indicates a previous high-temperature history, all mineral phases are completely reequilibrated to some lower temperature. Orthopyroxenes replacing clinopyroxene margin or olivine indicate an orthopyroxene enrichment event. Mineral phases of Type II are compared with Type I xenoliths, gabbroic xenoliths, and the host basalts. Those from Type II xenoliths show a distinct discontinuity with those from Type I mantle xenoliths, whereas they show a continuous or overlapping relation with those from gabbroic xenoliths and the host basalts. Our petrographic and geochemical results suggest that the studied type II xenoliths appear to be cumulates derived from the host magma-related system, being formed by early fractional crystallization, although these xenoliths may not be directly linked to the host basalt.

Nitrogen Mineralization in Soil Amended with Oil-Cake and Amino Acid Fertilizer under a Upland Condition (밭토양 조건에서 유박과 아미노산 비료의 질소 무기화량 추정)

  • The potential of nitrogen mineralization was studied by applying organic fertilizer to soil and incubating at $25^{\circ}C$ for 28 weeks. The organic fertilizers used in this experiment were oil-cake (CF-I, CF-II) and amino acid fertilizer (AAF-I, AAF-II). Accumulated mineralized nitrogen (N) fits the frist-order kinetics during incubation. The N mineralization potential ($N_0$) for organic fertilizers treated soil was highest at AAF-II treatment with a value of 27.71 N mg/100g, then followed by CF-II, AAF-I, CF-I. The pure N mineralization potential ($N_0$ treatment - $N_0$ control) for CF-I, CF-II, AAF-I, AAF-II were 2.55, 5.83, 3.66, 8.57 N mg/100g, respectively. The amount of N mineralized from organic fertilizers applied soil ranged from 46% to 61% of the total N content in organic fertilizer. The half-life ($t_{1/2}$) of organic nitrogen in soil treated with oil-cake and amino acid fertilizer was 17-21 days. Therefore, half of nitrogen contained in oil-cake and amino acid fertilizer was mineralized after 3 weeks application.

Transformation of Lettuce (Lactuca sativa L.) Using Cold Regulated Gene (BN115) (저온 관련 유전자를 이용한 상추 (Lactuca sativa L.)의 형질전환)

  • Explants of lettuce (Lactuca sativa L.) were co-cultivated with Agrobacterium tumifacience GV 3101 strain containing nptII gene and cold regulated gene (BN115) from Brassica napus for transformation. Multiple shoots were obtained from the explants in the selection medium (MS basal medium supplemented with 100 mg/L kanamycin, 500 mg/L carbenicillin, 0.1 mg/L NAA, 0.5 mg/L kinetin) after 3 to 4 weeks of co-culture. The putative transgenic shoots were transferred to rooting medium (1/2 MS basal medium supplemented with 100 mg/L kanamycin and 250 mg/L carbenicillin). The selected shoots were tested with PCR analysis using nptll, BN115 primers whether cold-regulated gene was introduced to genome of the plants. The vir G primers were particularly used to check contamination of Agrobacterium during PCR analysis. The nptII and BN115 primers produced the specific PCR bands in the putative transgenic lines but the vir G primers did not. These results confirmed that the PCR products were not the result of contamination with Agrobacterium. Additionally the Southern analysis of the PCR products and RT-PCR analysis proved that the cold-regulated gene was successfully integrated and transcribed in the putative transgenic lettuce plants.

Effect of Calcium and Magnesium on the Lipid and Mineral Composition of Serum and Tissues in Cholesterol-fed Rats (칼슘과 마그네슘이 콜레스테롤 첨가식이로 사육한 흰쥐의 혈청과 조직중 지질 및 무기질에 미치는 영향)

  • This experiment was undertaken to investigate the effect of caluium and magnesium on the lipid and mineral composition of serum and tissues in cholesterol-fed rats. GOT of serum was significantly decreased in normal Ca and high Mg supplemented group. Serum cholesterol level was significantly decreased in high Mg supplemented group, whereas that of low Mg supplemented group was significantly increased. Content of total lipid in liver were significantly decreased by supplementation with high calcium of magnesium. However, the content of cholesterol was not affected by supplementation with high calcim and that of low Mg supplemented group was significantly increased. In fatty acid composition of liver, he the order of fatty acid content is as following; oleic acid (18:1), stearic acid(18:0), and palmitic acid(16:0). Levels of Ca, Mg, K and Cu in liver were significantly affected by dietary calcium and magnesium levels. The Mg level of kidney was decreased but Cu level was elevated in cholesterol supplemented group. Levels of Ca, Mg, and Zn in serum were significantly elevated by supplementation with high calcium or magnesium.

  • Embryogenic callus induces from shoot tip and leaf segment of Zanthoxylum piperitum for producing somatic embryogenesis and plant regeneration were cultured in vitro on Murashige and Tucker's(MT) medium treated with casein hydrolysate $NH_4NO_3$, $KNO_3$ and plant growth regulator. The most effective somatic embryogensis was observed in the medium added by two fold $NH_4NO_3$(3300mg/l)+2. 4-D 0.1mg/l and $KNO_3$(3800mg/l)+2.4-D 0.1mg/l. Also, MT medium supplemented with casein hydrolysate 700mg/l added by 2, 4-D 0.1mg/l were effective in obtainingn somatic embryos from embryogenic callus The effect ofm MT medium supplemented with casein hydrolysate without 2, 4-D was lower than that with (3300mg/l) 2, 4-D for the formation of somatic embryos.

Purification of Inositol Triphosphate Kinase from Bovine Brain (소의 뇌로부터 Inositol Triphosphate Kinase의 정제)

  • Inositol 1,4,5-triphosphate($InsP_3$) is a second messenger for mobilizing intracellular $Ca^{2+}$. It can be dephosphorylated by soluble and particulate forms on $InsP_3$ 5-phosphatase, or phosphorylated to produce inositol 1,3,4,5-tetrakisphosphate($InsP_3$) by $InsP_3$ 3-kinase. These enzymes represent possible targets for the regulation of the $InsP_3/InsP_4$ signal. $InsP_3$ 3-kinase which catalyses th ATP-dependent phosphorylation of $InsP_3$ was purified from bovine brain tissue. All operation were carried out at $4^{\circ}C$. Fresh tissure was homogenized and centrifuged. The supernatant was pooled. Proteins were precipitated from 10% polyethylene glycol, and suspended solution was applied to DEAE cellulose column for chromatography. As the result of above procedure, two isozymes of $InsP_3$ 3-kinase, I and II were obtained. Each isozyme was applied to Matriz green gel, Calmodulin-Affigel 15 column and subsequent phenyl-TSK HPLC column. Specific activites(SA) and fold of puriety were observed at each purification step of chromatography. At DEAE cellulose chromatography, SA were I, 0.6 and II, 4.8 nM/min/mg, and folds were I, 17.2 and II, 16.6. At Matrix green gel chromatography, SA were I, 18 and II, 11 nM/min/mg, folds were I, 62.1 and II, 38.0. At calmodulin-Affigel 15 column chromatography, SA were I, 19 and II, 13 nM/min/mg, folds were I, 65.5 and II, 44.8. Finally $InsP_3$ kinase I and II were purified 3,103-fold and 2,310-fold, and SA were I, 900 and II, 670 nM/min/mg, respectively. SDS-polyacrylamide gel electrophoresis elucidated 3 distinct fractions of Mr of 145,000, 85,000 and 69,500 from isozyme I, and 2 distinct fractions of Mr of 79,000 and 57,000 from isozyme II.
