A Study of the Inhibiton Effect of Cd(II), Cu(II) & Zn(II) to the Biodegradation of Linear Alkylbenzene Sulfonate (Linear Alkylbenzene Sulfonate의 생분해에서 Cd(II), Cu(II) 및 Zn(II)의 저해효과에 대한 연구)

  • The standardized activated sludge for the biodegradation test of anion surfactants has been produced from the collected microorganisms in the soil and the wastewaters treatment plant. The activated sludge was kept under control of the pH, dissolved oxygen, microorganisms and inoculated the basal medium flasks with LAS and LAS mixed with heavy metals [Cd(II), Cu(II), Zn(II)]. Based of results, the inhibition effect(%) of heavy metals in LAS biodegradation were 1. All 1% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 0.1mg/l, respectively 2. All 1${\sim}$10% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 1mg/l, respectively 3. All 10${\sim}$40% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 10mg/l, respectively 4. All 30${\sim}$65% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 100mg/l, respectively And toxicity order of heavy metals to the microorganisms in LAS biodegradation were Cd>Cu>Zn in low concentration(0.1${\sim}$1mg/l)and Cd>Zn>Cu in high concentration(10${\sim}$100mg/l).

Adsorption Characteristics of the Heavy Metals, Cd(II) and Pb(II) Ions, on the Si-Immobilized Ulva pertusa (실리카고정 구멍갈파래에 대한 중금속 Cd(II), Pb(II)이온의 흡착 특성)

  • Adsorption characteristics of heavy metal ions, Cd(II) and Pb(II), on eastcoast-living algae, ulva pertusa, has been studied in our experiment. The Maximum adsorption amount of Cd(II) and Pb(II) ions on 1 g of the ulva pertusa were 2.3 mg, 3.1 mg in alkaline and 2.0 mg, 2.8 mg in acidic solution. However 3.4 mg, 7.3 mg in alkaline and 3.1 mg, 6.5 mg in acidic solution were shown on the Si-immobilized ulva pertusa in the same condition. Thus, Si-immobilized ulva pertusa adsorbs more amount of heavy metals, Cd(II), Pb(II), than the ulva pertusa, and more effective absorbent in alkaline. Furthermore, more amounts of Pb(II) ion were absorbed compare to Cd(II) ion in our work. Recovery ratio of Cd(II) and Pb(II) ions on the ulva pertusa were 55.0~61.0%, 59.7~66.8% respectively and 87.6~97.5%, 83.5~99.3% on the Si-immobilized ulva pertusa.

  • Hypocotyl explants of flowering cabbage were precultured on MS medium without kanamycin and then cocultured with Agrobacterium tumefaciens LBA4404;;pGA875 harboring insect resistantce proteinase inhibitor II(PI-II) gene in MS liquid medium adjusted pH 5.5 for 72hr. These explants were transferred to MS medium containing 20 mg/L kanamycin, 500 mg/L carbenicillin, and 1 mg/L BA. The explants were subsequently subcultured every 2 weeks. After 4 weeks of subculture, kanamycin-resistant shoots were obtained from selection medium. Leaves of putative transformants survived on MS selection medium containing 30 mg/L kanamycin. Incoporation of the PI-II gene into flowering cabbage was confirmed by PCR analysis of genomic DNA. Southern blot analysis showed that ECL-labeled probe for PI-II gene was hybridized to the expected amplified genomic DNA fragment of about 500 by from transgenic flowering cabbage.

  • Coupling of tetracycline (Tc) to dithiocarbamate chitosan(DTCC) via chelate bond was carried out in the presence of Mg (II) ion by one- and two-bath process. In one-bath process, DTCC was treated with Mg(II)-Tc solution. In two-bath process, DTCC was treated with Mg(II) to produce DTCCMg(II), which was isolated and treated in turn with Tc solution. The effect of pH and temperature on binding of Tc, was investigated varying the pH of the solution from 2.6 to 3.2 at 10 and $30^{\circ}C$. Binding of Tc was not affected greatly by such variations in pH and temperature. Amount of Tc bound to 1 gm of matrix reached to 180 mg. Release of Tc from DTCC-Mg(II)-Tc was investigated by batch and flow method, and the amount of Tc released against time followed by flow method exhibited near linear relationship. DTCC-Mg(II)-Tc showed very prolonged antimicrobial activity compared to that of free Tc when it was tested against Escherichia coli TG-1 and Bacillus subtilis NA-1.

  • Interaction of dietary Magnesium, Calcium and Polyunsaturated fatty acid(vegetable oils)on 3-Hydroxy-3-Methylglutaryl Co-A reductase activity was studied for a period of 30 days using isocalories and isonitrogenous as a basal diet . The subject rabbits were divided into 18 feeding groups. The results are summarized as follows: 1. The ratio of ${\alpha}_1-lipoprotein$ par lipalbumin is 0.34 for control group, 0.38 the highest group fed 0.1 Mg(II) 10ml plus perilla oil and basal diet, the lowest 0.25 group fed 0.1M Mg(II) 5ml plus sesame oil and basal diet. 2. The ratio ratio of ${\alpha}_2-lipoprotein$ per lipalbumin is 0.64 for control group. 0.95 as the highest for the group fed 0.1M Ca(II) 15ml plus sesame oil and basal diet, 0.1M Ca(II) 5ml plus perilla oil and basal diet. 3. The ratio of ${\beta}-lipoprotein$ per lipalbumin is 0.71 for control group, the highest 0.81 for the groups fed 0.1M Mg(II) 10ml plus sesame oil and basal diet, the lowest 0.37 for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 4. In serum triglyceride, control group was 129.5mg%, the highest 155.4mg% for the group fed 0.1M Ca(II) 5ml plus sesame oil and basal diet, the lowest 85.7mg% for the group fed 0.1M Mg(II) 10ml plus soybean oil and basal diet. 5. In serum cholesterol, control group was 96.7mg%, the highest 152.5mg% for the group fed 0.1M Ca(II) 10ml plus sesame oil and basal diet, the lowest 80.5mg% for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 6. In case of HMG-CoA reductase activity, control group was 0.95, the highest 0.98 for the group fed 0.1M Ca(II) plus soybean oil and basal diet. 7. Interaction between metal(II) ions and polyunsaturated fatty acid(vegetable oil) are soybean oil>sesame oil>perilla oil, for Mg(II). soybean oil>perilla oil>sesame oil, for Ca(II). Therefore, it is invetigated that the interaction between metal ion and polyunsaturated fatty acid is the higher, the cholesterol level is the lower, and HMG-CoA reductase activity is increased.

  • This dissertation was to research how some metal level within SipJeonDaeBo - Decoction, one of oriental prescriptions, influence Sprague-Dawley animals. 1. In level of non hazardous heavy metal control group detected $44.925{\pm}18.468\;mg/kg$, experiment I group $39.917{\pm}12.772\;mg/kg$, experiment II group $49.525{\pm}33.484\;mg/kg$. Besides in total concentration control group searched $18.082{\pm}7.395\;mg/kg$, experiment I group $16.068{\pm}5.128\;mg/kg$, experiment II group $19.977{\pm}13.443\;mg/kg$. There was no statistical significance but hazardous metal gets more level in the experilnent group than in the control group. 5. In level of hazardous metal within brain control group searched $0.145{\pm}0.056\;mg/kg$, experiment I group $$0.167{\pm}0.030\;mg/kg, erperiment II group $0.172{\pm}0.123\;mg/kg$. In level of non hazardous heavy metal control group detected $6.488{\pm}0.965\;mg/kg$, experiment I group $7.290{\pm}0.588\;mg/kg$, experiment II group $7.010{\pm}1.627\;mg/kg$. Besides in total concentration control group searched $2.683{\pm}7.395\;mg/kg$, experiment I group $3.017{\pm}0.238\;mg/kg$, experiment II group $2.908 {\pm} 0.711\;mg/kg$. Therefore there was no statistical significance. 6. In level of hazardous metal within bone control group searched $8.172{\pm}5.195 \;mg/kg$, experiment I group $9.128{\pm}4.143\;mg/kg$, experiment II group $9.401{\pm}6.924\;mg/kg$. There is statistical significance(p<0.05). In level of non hazardous metal control group detected $94.065{\pm}36.035\;mg/kg$, experiment I group $147.563 {\pm}79.939\;mg/kg$, experiment II group $142.730{\pm}77.374\;mg/kg$. Besides in total level control group searched $48.530{\pm}16.523\;mg/kg$, experiment I group $64.502{\pm}31.078\;mg/kg$, experiment II group $62.733 {\pm}34.641\;mg/kg$. Therefore there was no statistical significance. 7 In the correlative research as to how each metal influences to ingestion Cd and Co searched 0.954 and Pb and Ni -0.0884 from kidney. Co and Cd was 0.995 and Zn and As -0.190 from liver. Co and Cd were 0.995 and Zn and Cu -0.393 from brain. Co and Cd were 0.998 and Zn and Mn -0.206 from bones

Identification of MgII Absorbers in the Quasar Lines of Sight

  • Large area infrared surveys are often accompanied with follow-up optical spectroscopic surveys that has a significant legacy value even for other areas of research. Using these spectral database, we have performed a search for MgII absorption lines in the optical spectrum of background quasar. Over the ~4deg2 of AKARI North Ecliptic Pole survey field and Spitzer First Look Survey field, 18 and 16 MgII absorber systems are identified respectively. The redshift range for the background quasars was 1.0<$z_{qso}$<3.4, while the redshift range for the absorber was 0.6<$z_{abs}$<1.6. Galaxies responsible for MgII absorptions are identified in the deep optical images (CFHT r-band), yet the identification still remains ambiguous for 60% of the systems due to the limited image depth and the source crowdedness. The impact parameter ranges 20-60kpc, and the rest-frame equivalent width of MgII absorption ranges $0.7-4{\AA}$. The most critical part in the identification of MgII absorber galaxies is the existence of deep optical images in addition to the high S/N quasar spectrum with R>3000.

THE CHROMOSPHERIC ACTIVITY ON V711 TAU (V711 TAU의 채층활동)

  • The relationship between Mg II emission line and light variation of V711 Tau has been derived to investigate the chromospheric activity on V711 Tau. First, a shape of an optical light curve was compared with that of ultraviolet constructed from the IUE low resolution spectra. Second, the intensities of Mg II k emission lines have been reduced from IUE high resolution spectra. The intensity of Mg II k line was compared with brightness of the UV light curve at given phase. The Mg II line intensity is maximum at the phase $O.^{P}4$ where the light is minimum. The evidence of chromosperic activity is indicated by the intensity variation of the MgII emission line with orbital phase for V711 Tau.

STELLAR MAGNETIC ACTIVITY MEASURE BASED ON IUE MG II H+K EMISSION LINES OF MAIN-SEQUENCE G STARS

  • Stellar magnetic activity is important for formulating the evolution of the star. To represent the stellar magnetic activity, the S index is defined using the Ca II H+K flux measure from the Mount Wilson Observatory. Mg II lines are generated in a manner similar to the formation of Ca II lines, which are more sensitive to weak chromospheric activity. Mg II flux data are available from the International Ultraviolet Explorer (IUE). Thus, the main purpose of this study was to analyze the magnetic activity of stars. We used 343 high-resolution IUE spectra of 14 main-sequence G stars to obtain the Mg II continuum surface flux and Mg II line-core flux around 2,800 Å. We calculated S index using the IUE spectra and compared it with the conventional Mount Wilson S index. We found a color (B - V ) dependent association between the S index and the Mg II emission line-core flux. Furthermore, we attempted to obtain the magnetic activity cycles of these stars based on the new S index. Unfortunately, this was not successful because the IUE observation interval of approximately 17 years is too short to estimate the magnetic activity cycles of G-type stars, whose cycles may be longer than the 11 year mean activity cycle of the sun.

Growth and Flowering of Cut Spray Chrysanthemum 'Gama' by Daminozide (스프레이 절화국화 '가마'의 daminozide에 의한 생육 및 개화반응)

  • This study was conducted to improve commercial quality of a cut spray chrysanthemum 'Gama' bred in Korea by foliar application of daminozide (DMZ), suppressing excessive elongation of peduncle caused by high temperature in summer season production. Applications were made at three floral bud developmental stages (I, II, and III), and concentrations used were 0, 500, 1000, and $2000mg{\cdot}L^{-1}$. Both cut flower length and peduncle length showed the greatest suppression of elongation by DMZ treatments at stage II. DMZ also gradually reduced values of those in a concentration-dependant manner. Stem thickness and flower diameter reduced compared to control as DMZ was sprayed at later stage and those two characteristics decreased in a concentration-dependant manner. The number of paralleled flowers neighboring apical part of whole plant showed the best results at stage I and II. As for DMZ concentration, those increased effectively at $500-1,000mg{\cdot}L^{-1}$. Among all combinations, $1,000mg{\cdot}L^{-1}$ DMZ at stage II recorded the most paralleled flowers and showed 24.2% increment as compared to control. The number of fully expanded flowers and total floral buds also showed the highest values using $1,000mg{\cdot}L^{-1}$ DMZ at stage II than the other treatments. In cut spray chrysanthemum cultivar 'Gama' bred in Korea, foliar-spraying with $1,000mg{\cdot}L^{-1}$ DMZ at stage II most effectively suppressed excessive peduncle elongation, parallelled flower cluster arrangement, and increased the number of flowers in summer. However, this combination affected negatively the other commercial qualities, reducing cut flower length, stem thickness, and flower diameter. Therefore, we recommended that foliar-spraying stage and DMZ concentration was stage I and $1,000mg{\cdot}L^{-1}$, respectively, in a practical culture for 'Gama'.
