구기자(Lycii fructus) 추출물이 미생물 생육에 미치는 영향 (The Influence of Lycii fructus Extracts on the Growth and Physiology of Microorganism)

  • 구기자 추출물이 S. cerevisiae D7l과 L. casei KC-TC3165, 그리고 E. coli DH5균의 생육에 미치는 영향을 검토하였다. 그 결과 구기자 추출물은 S. cerevisiae D7l과 L. casei KCTC 3165의 생육을 증가시켰으며 E. coli DH5에 대하여는 약간의 생육저해가 있었다. 구기자methanol 추출물을 첨가하여 미생물의 생육을 측정하였을 때 L. casei KCTC 3165와 S. cerevisiae D7l는 각각 1.0%와 1.5% 첨가구에서 가장 높은 생육 촉진 효과를 보였다. 즉 구기자 추출물은 대장균의 생육은 저해하였으나, 인체에 유익한 균으로 알려진 젖산균에 대하여는 생육을 촉진하는 특성이 있었다. 한편, E. coli DH5는 구기자즙 10%, 그리고 methanol, chloroform 추출물 각각 1.0%첨가시 형태적 이상을 일으켜 길이가 8배 이상이 길어졌다. 구기자의 methanol추출물 중 회분성분이 E. coli DH5균에 이상현상의 원인이 되었음을 확인 하였다. 또한 이들 중 $Na^{+}$, $K^{+}$$Na^{+}$$K^{+}$ 혼합물이 0.135% 이상에서 균의 형태가 길어지는 이상 현상이 있었다.

작약 연작지토양 추출물이 작약 배양세포와 배양묘의 생육에 미치는 영향 (Effects of Continuously Cropped Soil Extracts on Cell Viability and Seedling Growth of Peony(Paeonia lactiflora))

  • This experiment was conducted to investigate the growth inhibition effects caused by continuous cropping soil in peony(Paeonia lactiflora Pallas). The effect of extracts from continuous cropping soil of peony was tested with bio-assay method using callus cells induced from peony filament tissues and seedlings derived from peony zygotic embryos. The cell viability and seedling growth were significantly inhibited by methanol extract in continuous cropping soil. Methanol extract from continuous cropping soil was successively fractionated with solvents such as n-hexane, ethyl acetate, n-butanol and water. The seedling growth was inhibited by ethyl acetate fraction obtained in methanol extract.

  • Gastrodia elata (GE) is a oriental medicinal herb which has been used traditionally for the treatment of various brain diseases including convulsion and epilepsy. In orde r to examine the mechanism of anticonvulsive effect, we treated the methanol extract of GE (500mg/kg, P.0) to the pentylenetetrazole (PTZ)-induced convulsive rats. Methanol extracts of GE significantly inhibited (35%) the convulsion state as well as the level of lipid peroxidation (25%) in the brain. The ether fraction of methanol extracts among the others effectively ibhibited in vitro lipid peroxidation dose dependently ($5.O{\times}10^{-6}{\sim}2.O{\times}1O^{-5}g/ml$). The scavenging effect on hydroxy radicals was found in all the fractions of ether, butanol, and dichloromethane. These results suggest that the anticonvulsive effect of GE is possibly due to the antioxidative effects of the active components in GE.

  • 마이크로웨이브 추출방법과 환류 냉각 추출방법을 비교한 결과, 물과 에탄올의 혼합용매로 추출한 경우 마이크로웨이브 추출 방법에 의하여 추출시간을 단축시키면서 환류 냉각 추출 방법에서와 같은 수준의 가용성 고형분 및 총 폴리페놀 함량을 갖는 곰취 추출물을 얻을 수 있었다. 마이크로웨이브 추출시 최적 마이크로웨이브 에너지는 120∼150 W 였고 추출시간은 4∼8분이 적당하였다. 추출에 사용한 용매들 가운데 에탄올, 메탄올 보다 물 그리고 물과 에탄올 또는 메탄올 혼합용매를 사용한 추출물의 가용성 고형분, 총 폴리페놀 함량 및 항산화 효과가 높은 것으로 나타났다.

Brining Property and Antimutagenic Effects of Organic Chinese Cabbage Kimchi

  • Brining property and antimutagenic effects of organically cultivaged Chinese cabbage kimchi (OC kimchi) and common Chinese cabbage imchi (CC kimchi) were studied. The salt absorption rate of leaves was faster than that of stems of the Chinese cabbages. Due to the large portion of leaf in organic Chinese cabbage, organic Chinese cabbage(OC) was much faster in terms of salt absorption rate than common Chinese cabbage(CC). The antimutagenic effects of methanol extracts of CC kimchi and OC kimchi were studied against aflatoxin B1(AFB1) using Ames test on Samonella typhimurium TA 100 and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) using SOS chromotest. Methanol extract from 6 -day fermented OC kimchi at 15 $^{\circ}C$ showed 80% inhibition rate against the indirect mutage, aflatoxin B1 induced mutagenicit where as that from 6-day fermented CC kimchi at 15 $^{\circ}C$ showed 54% inhibition rate in the Ames test. Methanol extracts from 6-day fermented CC kimchi and OC kimchi showed 27 % and 58 % inhibition rate against direct mutagen , N-methyl-N'-nitro-N-nitrosoguanidine induced mutagenicity, respectively in SOS chormotest, thus OC kimchi exhibited higher antimutagenic activity than kimchi.

Comparative Study on Antioxidant Activity and Multi-drug Resistance Reversing Activity in Korean Colored Soybean Cultivars

  • The purpose of this study is to analyze antioxidant activity and multidrug resistance reversing activity in several Korean colored soybean (Glycine max Merr.) cultivars. Antioxidant activity of methanol extracts from colored soybean cultivars was evaluated by TBARS (thiobarbituric acid reactive substances), DPPH (1, 1-diphenyl-2-picrylhydrazyl) and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) methods. By means of TBARS, cultivar "Jeonnam #1" showed the highest activity until 7 days, and followed by "Black #1", "Jinyul" and "Black #3", showing lower activity than that of BHT (butylated hydroxytoluene). Methanol extracts of all cultivars proved that DPPH radical scavenging activity is dose-dependent. Methanol extract from cultivar "Jeonnam #1" showed highest DPPH radical scavenging activity, and followed by cultivars "Black #1". MDR (multi-drug resistance reversing) activity, however, showed the highest effect in "Black #3" and the lowest "Black #1" cultivar. These results suggest that seed colors of soybean may play an important role in antioxidant activity and MDR activity.

Evaluation of Antimicrobial Activity of the Methanol Extracts from 8 Traditional Medicinal Plants

  • The methanol extract of 12 medicinal plants were evaluated for its antibacterial activity against Gram-positive (5 strains) and Gram-negative bacteria (10 strains) by assay for minimum inhibitory concentration (MIC) and minimum bacterial concentration (MBC). The antibacterial activity was determined by an agar dilution method (according to the guidelines of Clinical and Laboratory Standard Institute). All the compounds (12 extracts) of the 8 medicinal plants (leaf or root) were active against both Gram-negative and Gram-positive bacteria. Gram-negative showed a more potent action than Gram positive bacteria. The MIC concentrations were various ranged from $0.6\;{\mu}g/ml$ to $5000\;{\mu}g/ml$. The lowest MIC ($0.6\;{\mu}g/ml$) and MBC ($1.22\;{\mu}g/ml$) values were obtained with extract on 4 and 3 of the 15 microorganisms tested, respectively.

  • Herpes simplex viruses(HSV) are one of the most common infectious virus of man. Though chemotherapies and antibiotics against HSV have been developed in many countries, but anti-HSV agents were not satisfactory to mankind by their toxic reaction and side effects. In order to search for anti-HSV agents from Korean traditional prescriptions, we extended the number of specimens. Both methanol extract and boilling water extract of the Korean traditional prescriptions were screened to detect anti-HSV activities by MTT assay. Korean traditional prescriptions showing anti-HSV activities as methanl extracts were Paekyopsan, Chesupwilyungtang, Yongdamsagantang, and prescription 11. Four methanol extracts showing anti-HSV activities were freationated by hexane and their efficacies were tested. Hexane freationations of Paekyopsan, Chesupwilyungtang, and prescription 11 showed in anti-HSV activities both haxane and methanol fractionation.

녹즙추출물의 아질산염 소거능에 대한 연구 (Nitrite Scavenging Effect of Methaol Fraction Obtained from Green Yellow Vegetable Juices)

  • The objectives of this study were to investigate biological activities such as nitrite scavenging effect of water and methanol fractions obtained from each green juice(Angelica keiskei, kale, carrot) under different pH conditions, and antioxidant vitamin contents in green juice made of green yellow vegetables. The result shows that nitrite scavenging effect of each green juice was 14.0~91.2% in model system. The methanol fraction provided higher effect than the water fraction. The nitrite degradation was the highest at pH 1.2. Nitrite scavenging effect of methanol fraction obtained from kale juice was 91.2% at pH 1.2 which was the highest among tested samples in this experiments. Consequently, extracts of kale juice were shown to provide the greatest functional properties among the vegetables tested. Contents of vitamine E and C in the extracts of kale were higher than the others.

Inhibitory Effects of Kimchi Extracts on Carcinogen-induced Cytotoxicity and Transformation in C3H/10T1/2 Cells

  • Inhibitory effects of kimchi extracts on arcinogen-induced cytotoxicity and transformation in C3H/10T1/2 cells were studied. The methanol extract (500㎍/ml) of fresh (unfermented kimchi), and 3-week-fermented kimchi (properly ripened kimchi at 5℃) inhibited 3-methylcholanthrene (MCA)-induced cytotoxicity in C3H/10T1/2 cells by 84 and 99%, respectively. The inhibitory effect of 3-week-fermented kimchi was higher than that of the fresh kimchi at same test condition. The methanol soluble fraction, and haxane extract of 3-week fermented kimchi also surpressed the cytotoxicity of FC3H/10T1/2 cells mediated by 7,12-dimethylbenz[a]anthracene(DMBA) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). Furthermore, MCA-induced transformation of C3H/10T/1/2 cells was significantly inhibited by the methanol soluble fraction of 3-week fermented kimchi. With these results, we suggest that kimchi might have anticarcinogenic effect in part due to inhibition of carcinogen-induced cytotoxicity and transformation of C3H/10T/1/2 cells.
