Effect of Agrimonia Pilosa Ledeb. Extract on the Growth of Food-Borne Pathogens (선학초 추출물이 식중독 유발세균의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effect of the Agrimonia pilosa Ledeb. extracts against food-borne pathogens. First, the Agrimonia pilosa Ledeb. was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Agrimonia pilosa Ledeb. was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Agrimonia pilosa Ledeb. extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The petroleum ether extracts of Agrimonia pilosa Ledeb. showed the highest antimicrobial activity against Pseudomonas aeruginosa. The synergistic effect has been found in combined extracts of Agrimonia pilosa Ledeb. and Perillae folium as compared to each extracts alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Agrimonia pilosa Ledeb. against Bacillus Cereus and Salmonella Enteritidis. The petroleum ether extract of Agrimonia pilosa Ledeb. showed strong antimicrobial activity against Bacillus Cereus at the concentration of 4,000 ppm. The 4,000 ppm of petroleum ether extract from Agrimonia pilosa Ledeb. retarded the growth of Bacillus Cereus more than 24 hours and Salmonella Enteritidis up to 36 hours. The petroleum ether extracts of Agrimonia pilosa Ledeb. has been shown the antimicrobial effect against Bacillus Cereus and Salmonella Enteritidis. (Korean J Nutrition 38(2): 112~116, 2005)

Antimicrobial Effect of Citrus unshiu Markovich Extracts on Food-Borne Pathogens (청피 추출물이 식중독 유발 미생물의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effects of the Citrus unshiu Markovich extracts against food-borne pathogens. First, the Citrus unshiu Markovich was extracted with methanol at room temperature, and fractionation of the methanol extracts from Citrus unshiu Markovich was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Citrus unshiu Markovich extracts was determined using the paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Citrus unshiu Markovich showed the highest antimicrobial activity against Bacillus cereus and Shigella sonnei. The ethyl acetate extract of Citrus unshiu Markovich showed strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. The 5,000 ppm of ethyl acetate extract from Citrus unshiu Markovich retarded the growth of Bacillus cereus more than 24 hours and Shigella sonnei up to 24 hours. The ethyl acetate extracts of Citrus unshiu Markovich have shown an antimicrobial effect against Bacillus cereus and Shigella sonnei.

Antioxidant Activities of Citrus unshiu Extracts obtained from Different Solvents (추출용매에 따른 진피 추출물의 항산화 활성)

  • In this study, the total polyphenol content, electron donating ability (EDA) and inhibitory activity of glutathione S-transferase (GST) of freeze-dried Citrus unshiu extracts were examined. The Citrus unshiu extracts was obtained from four solvents such as ethyl acetate, acetone, methyl chloride and methanol, to evaluate its functional properties. Total polyphenol contents were measured in the two different extracts, and the extracts were screened for their potential antioxidant activities using tests such as electron donating ability (EDA), glutathione S-transferase (GST). The total polyphenol contents of Citrus unshiu extracts were $928.48{\pm}1.19{\mu}g\;GAE/mL$ in ethyl acetate (EA), $886.03{\pm}0.44{\mu}g\;RE/mL$ in acetone (AC), $413.08{\pm}1.39{\mu}g\;GAE/mL$ in methylene chloride (MC), $12,648.60{\pm}0.56{\mu}g\;GAE/mL$ in methanol (MeOH), respectively. Also, the total polyphenol contents of EtOH Citrus unshiu extracts were $664.64{\pm}0.74{\mu}g\;GAE/mL$ in EA, $702.67{\pm}0.85{\mu}g\;RE/mL$ in AC, $429.64{\pm}0.61{\mu}g\;GAE/mL$ in MC, $16,108{\pm}0.73{\mu}g\;GAE/mL$ in MeOH, respectively. The total polyphenol contents were significantly difference (p<0.05) between the solvents. The electron donating ability of Citrus unshiu extracts were $62.80{\pm}0.36%$ in EA, $97.43{\pm}0.51%$ in AC, $52.20{\pm}0.30%$ in MC, $97.63{\pm}0.46%$ in MeOH, respectively. Also, the electron donating ability of EtOH Citrus unshiu extracts were $51.49{\pm}0.26%$ in EA, $63.17{\pm}0.31%$ in AC, $67.68{\pm}0.55%$ in MC, $96.18{\pm}0.41%$ in MA, respectively. The electron donating ability were significantly difference (p<0.05) between the solvents. The inhibitory activity of glutathione S-transferase in Citrus unshiu extracts were $76.22{\pm}0.65%$ in EA, $31.73{\pm}0.48%$ in MC, $97.48{\pm}0.56%$ in MeOH, respectively. Also, inhibitory activity of glutathione S-transferase in EtOH Citrus unshiu extracts were $75.54{\pm}0.55%$ in EA, $73.53{\pm}0.38%$ in MC, $48.70{\pm}0.46%$ in MeOH, respectively. The inhibitory activity of glutathione S-transferase were significantly difference (p<0.05) between the solvents. These results indicated that the Citrus unshiu extracts is a high-valued food ingredient and the extraction with methanol will be useful as a nutritional source with natural antioxidant activities. Considering high consumer demand beneficial health effects, Citrus unshiu extracts can be utilized to develop functional food health- promoting and natural antioxidant agents.

Antioxidant and $\alpha$-glucosidase Inhibition Activity from Different Extracts of Zanthoxylum schnifolium Fruits (산초 (Zanthoxylum schnifolium)열매 추출물의 항산화 및 $\alpha$-Glucosidase 저해 활성)

  • This study was performed to the activity of antioxidant and anti-diabetic from Zanthoxylum schnifolium fruit. The Zanthoxylum schnifolium fruits were extracted with water, 100% methanol, 100% ethanol, 70% methanol and 70% ethanol. The activities of each extracts were measured by antioxidant tests, such as total phenolic contents, total flavonoid contents, hydroxyl radical activity, DPPH radical scavenging activity, reducing power and $\alpha$-glucosidase inhibition activity assay. The water extract represented the highest activity in the antioxidants properties in vitro, including hydroxyl radical activity, DPPH radical scavenging activity, reducing power. Furthermore, water extract also showed significantly high total phenolic contents but the 100% methanol extract showed high total flavonoid contents estimated as $196.48{\mu}g/mg$. The $IC_{50}$ values for $\alpha$-glucosidase inhibition activity of the 100% ethanol and 100% methanol extracts were $275.66{\mu}g/mg$ and $261.44{\mu}g/mg$. Our results indicated that the Zanthoxylum schnifolium fruit expected to be useful as antioxidants.

Cytotoxic Activity of Methanol Fractions and Solvent Extracts from Houttuynia cordata $T_{HUNS}$ (IX) on Various Cancer Cells (어성초 용매추출물과 메탄올 분획물의 암세포주에 대한 세포독성)

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata T/sub HUNB/ extracts on A549 (lung cancer), MDA-MB231 (breast cancer), SNU-C4 (colon cancer) and B16 (mouse melanoma) cell lines. We have determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The 150 ㎍/㎖ concentration of methanol extract (63.81 %) of Houttuynia cordata T/sub HUNB/ was shown significantly antitoxic activity on A549 cell lines. The order of cytotoxicity fractions of methanol from Houttuynia cordata T/sub HUNB/ extracts against cancer cell lines in vitro is as follows : hexane fraction layer > chloroform fraction layer > ethyl acetate fraction layer > buthanol fraction layer > water fraction layer. These results suggest that the hexane fraction of methanol extract from Houttuynia cordata T/sub HUNB/ extract may be a valuable choice for the development of antitumor agents.

Anticancer Effects of Organic Chinese Cabbage Kimchi

  • The anticancer effect of methanol extracts from common Chinese cabbage kimchi(CC kimchi ) and organically cultivated Chinese cabbage kimchi (OC kimchi) was studied on the cell growth, MTT assay and SRB assay using AGS human gastric cancer cells. Methanol extracts from CC kimchi and OC kimchi exhibited the anticancer activites in vitro and in vivo. Methanol extract from 6 day-fermented CC kimchi and OC kimchi inhibited the growth of AGS cells by 55.2 and 60.7% , respectively. At MTT assay an dSRB assay, 6 day-fermented OC kimchi showed higher inhibition rate (MTT : 42%, SRB : 61%) than 6 day-fermented CC kimchi(MTT : 33%, SRB : 52%). Methanol extracts from 6-day fermented CC kimchi and OC reduced the tumor formation and prolonged the life span of sarcoma-180 cell injected Balb.c mouse. OC kimchi treated group resulted in the smaller tumor weight of 4.58$\pm$0.32g compared th the CC kimchi group of 5.40$\pm$0.78g and the control group of 7.50$\pm$0.54g and OC kimchi treted group (25.3 days) lived longest among control (20.2days ) and CC kimchi(23.5days) treted groups.

Desmutagenic Effect of Extracts Obtained from Seaweeds (해조류의 항돌연변이 효과)

  • Extracts of six seaweeds obtained from solvents such as water, methanol, hexane and ethyl ether have been tested for desmutagenic activities in the Salmonella typhimurium/microsome systems. Water extracts obtained from tangle, sea-tangle, and gompi were moderately effective on the mutagenicity of Trp-P-2 and aflatoxin $B_1$. Methanol extracts of sea-tangle and gompi were effective against Trp-p-2, and all samples of methanol extracts were very effective from 1.0mg to 2.0mg per plate on the mutagenicity of MeIQ and aflatoxin $B_1$. Hexane extracts of tangle, ses-tangle, gompi and sea-straghorn were very effective on the mutagenicity of Trp-p-2, MeIQ and aflatoxin $B_1$ except laver and green laver. Ethyl ether extracts of gompi and sea-straghorn were most effective from 1.0mg to 2.0mg per plate on the mutagenicity of Trp-P-2. Especially, ethyl ether extracts of all samples were great effective on the mutagenicity of MeIQ and aflatoxin $B_1$.

Yam (Dioscorea batatas) Root and Bark Extracts Stimulate Osteoblast Mineralization by Increasing Ca and P Accumulation and Alkaline Phosphatase Activity

  • Yam (Dioscorea batatas) is widely consumed as functional food for health promotion mainly in East Asia countries. We assessed whether yam root (tuber) or bark (peel) extracts stimulated the activity of osteoblasts for osteogenesis. MC3T3-E1 cells (mouse osteoblasts) were treated with yam root extracts (water or methanol) (study I) or bark extracts (water or hexane) (study II) within $0{\sim}10{\mu}g/mL$ during the periods of osteoblast proliferation (5~10 day), matrix maturation (11~15 day) and mineralization (16~20 day) as appropriate. In study I, both yam root water and methanol extracts increased cell proliferation as concentration-dependent manner. Cellular collagen synthesis and alkaline phosphatase (ALP) activity, both the indicators of bone matrix protein and inorganic phosphate production for calcification respectively, were also increased by yam root water and methanol extract. Osteoblast calcification as cell matrix Ca and P accumulation was also increased by the addition of yam root extracts. In study II, yam bark extracts (water and hexane) increased osteoblast proliferation and differentiation, as collagen synthesis and ALP activity and osteoblast matrix Ca and P deposition. The study results suggested that both yam root and bark extracts stimulate osteogenic function in osteoblasts by stimulating bone matrix maturation by increasing collagen synthesis, ALP activity, and matrix mineralization.

The Fibrinolytic Activity of Kimchi and its Ingredients in vivo and in vitro (In vivo와 in vitro에서 김치 및 김치재료의 피브린 분해활성)

  • Fibrionolytic activity(FA) of kimchi in rat and FAs of water and methanol extracts of kimchi ingredient were determined using fibrin plate method to see where the active principles for FA are present in kimchi. Nine Sprague-Dawley per each group were fed diet containing 3, 5 or 10% of freeze dried kimchi for 6 weeks. The FA of plasma obtained from 10% kimchi group showed stronger FA than that of control. FAs of 3 and 5% kimchi group were not noticeable. FA from methanol extract of kimchi was approximately 6 times higher than that from water extract assuming that the active material may be present in a fat soluble fraction. From water extract of kimchi ingredients, dropwort, green onion, radish and baechu showed FA in descending order. And from methanol extracts, red pepper powder, radish, green onion and baechu showed FA in decending order. The Fa of methanol extract of red pepper powder was the most noticeable among samples.

Antimicrobial Effect of Methanol Extracts from Some Medicinal Herbs and the Content of Phenolic Compounds (약용식물 추출물에 대한 항미생물 활성 검색과 폴리페놀 함량)

  • Methanol extracts were prepared from 32 medicinal herbs of the extracts were tested their microbial inhibition activities against food borne pathogens and/or food poisoning microorganisms, food-related bacteria and yeast. Methanol extracts of Cinnamomum cassia, Paeonia suffruticosa, Alnus japonica, Eugenio caryophyllata and Illicium verum exhibited antimicrobial activity for the microorganisms tested, except lactic acid bacteria and yeast. Minimum inhibitory concentrations(MIC) were about 5 mg/mL for Bacillus subtilis, Staphylococcus epidermidis and Pseudomonas aeruginosa. Cell growth of lactic acid bacteria was inhibitied, but greatly on Leuconostoc mesenteroides. The phenolic compound contents were 10.98 mg/g, 10.31 mg/g, 8.55 mg/g and 6.69 mg/g in Thea sinensis, Eugenia caryophyllata, Alnus japonica and Artenisia capillaris, respectively. Antimicrobial activity appeared to be related to phenol compound content in medicinal herbs. The methanol extracts of medicinal herbs could be suitable for the development of a food preservative.
