부자(附子)의 우울증 억제효과에 관한 연구 (Anti-depressant Effect of the Extracts of Aconitum carmichaeli)

  • Objectives : The water and methanol extracts of Aconitum carmichaeli(Aconiti Tuber Preparat) were investigated for their anti-depressant effects. Methods : In this study, reserpine-induced hypothermia test, tail suspension test and hot plate test. Additionally, the brain monoamine oxidase activity was determined in vivo. Results: In the reserpine-induced hypothermia test, both extracts suppressed the fall of body temperature compared to the control group in a dose-dependent manner, suggesting the inhibition on hypothermia. In the tail suspension test, the methanol extract dose-dependently reduced the duration of immobility by 28.4% at a dose of 1 g/kg compared to control group, which is more effective than the water extract. In the hot plate test, the water extract and methanol extract increased the jump latency time compared to the control group, showing the inhibition rate of 198% and 182%, respectively, at a dose of 1 g/kg. Methanol extracts potently inhibited the brain monoamine oxidase activity in an in vivo assay compared to the control group, showing 84.6% inhibition, but the water extract revealed very weak activity. Conclusions : Above results suggested that the extract of Aconitum carmichaeli can be useful for the prevention and treatment of depression.

천연약용식물의 활성산조종에 의한 산화적 손상의 보호 효과 (Protective Effect of Natural Medicinal Plants against Oxidative Damage Induced by Reactive Oxygen Species)

  • In order to evaluate anti -oxidant activities and protective effect against oxidatve damage, DPPH radical scavenging activity and lipid peroxidation inhibitory activity were measured among methanol extracts prepared from natural medicinal plants. Fourteen natural medicinal plants which were reported to have anti -oxidative or anti-inflammatory effects were selected based on our previous report. In addition to the total methanol extracts, n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions were prepared from each total extract. DPPH radical scavenging assay was performed against 14 total extracts and all samples showed dose-dependent activities in various extent. Among those, 6 samples, methanol extracts of Euryale ferox, paeonia suffruticosa, Areca catechu var. dulcissima, Cinnamomun cassia, Alpinia katsumadai and Betula platyphlla var. japonica showed IC$\sub$50/ value lower than 6.0 $\mu\textrm{g}$/ml. The highest DPPH radical scavenging activity was found in ethylacetate fraction of paeonia suffruticosa with IC$\sub$50/ value of 1.1 $\mu\textrm{g}$/ml. Analysis of lipid peroxidation inhibitory activity on hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast (V79-4) cells revealed that the highest inhibitory effect was observed in methanol extract of Betula platyhpylla var. japonica. Lipid peroxidation inhibitory activity was observed as a dose-dependent manner in all samples used in this study. Among fraction samples, ethylacetate fraction of Alpinia katsumadai had the strongest inhibitory activity with IC$\sub$50/ value of 0.9 $\mu\textrm{g}$/ml.

Screening of Korean Traditional Medicines and Medicinal Herbs for Antimicrobial Activity Against Pathogenic Bacteria

  • This study screened 76 Korean traditional patent medicines and 58 medicinal herbs for antimicrobial activity against pathogenic bacteria. Among the prescriptions methanol and hot water extracts of Samsoum, Chujunghwang-yont'ang, and Sashinhwan showed antimicrobial activities against E. coli and Chujunghwang-yont'ang showed the strongest antimicrobial activity. Among the medicinal herbs, hot water and methanol extracts of Terminalia chebula, Caesalpinia sappan, and Coptis chinensis showed the strongest antimicrobial activities against Escherichia coli O157, Staphylococcus aureus, Shigella flexnery, Shigella sonnei and Salmonella typhimurium. An interesting characteristic of the traditional medicines such as Samsoum, Chujunghwang-yont'ang, and Sashinhwan was that both methanol and hot water extracts were active. Chujunghwang-yont'ang exhibited the strongest antimicrobial activities against the microorganisms tested. Water and methanol extracts of Terminalia chebula, Caesalpinia sappan, and Coptis chinensis showed the strong of inhibitory effects on the growth of all microorganisms tested.

Antioxidant Activities of Methanol Extracts and Phenolic Compounds in Asian Pear at Different Stages of Maturity

  • Contents of phenolic compounds in peel, flesh, and core of three Asian pear cultivars, Hosui, Niitaka, and Chuwhangbae, were determined at different stages of maturity. Antioxidant properties of methanol extracts of peels at various fruit maturity stages were also evaluated. Total phenolic content decreased with maturity. Arbutin, chlorogenic acid, and epicatechin were major phenolic compounds in young fruits. Catechin, 4-hydroxymethyl benzoic acid, and caffeic acid were detected in peel and core of immature and mature pears. 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activities of methanol extracts were 16.30 and $15.73\;{\mu}g$ in peel of immature Hosui and Chuwhangbae pears, respectively, and $11.59\;{\mu}g$ in mature Niitaka pears, which was significantly higher than those of other maturity stage in the same cultivar. Inhibitory activities on lipid oxidation of methanol extracts of three cultivars at all maturity stages were similar to that of ${\alpha}$-tocopherol.

돌미나리 메탄올 추출물의 항돌연변이 작용과 암세포증식 억제효과 (The Antimutagenic Activity and the Growth Inhibition Effect of Cancer Cells on Methanol Extracts from Small Water Dropwort)

  • The study was carried out to evaluate the antimutagenic and anticancer effects of small water dropwort. The methanol extracts from small water dropwort significantly reduced the mutagenicity induced by aflatoxin $B_1\;(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Salmonella typhimutium TA 100. Also, the methanol extracts inhibited the growth of AZ-521 human gastric cancer cells and HT-29 colon cancer cells. The chloroform fraction from methanol extracts of small water dropwort inhibited $40\;to\;80\%$ of the mutagenicity by $AFB_1$ in Sal. typhimurium TA 100 by the addition of 2.5 to $10\%$. To separate active compounds, the chloroform fraction was subjected to column chromatography on a silica gel and separated into five fractions. Among the five fractions, fraction 4 showed the highest antimutagenic effect against $AFB_1$ and an anticancer effect in the HT-29 colon cancer cell. As the result of the analysis in GC-MS, 1-napthalene carbonitrile, 5,6,7,8-tetrahydrol and benzene, 1,1'-(1,4-pentadiene-1,5-diyl) bis-,(E,E) were identified potentially from fraction 4.

Bleeding Time Prolongation Effect of Methanol Extract of Viscum album var. coloratum

  • The methanol extract of Viscum album var. coloratum, Korean mistletoe, showed potent prolongation effects on the bleeding time in rats in vivo, and whole blood clotting time and plasma recalcification time in rats ex vivo. The prolongation effect on the bleeding time of Korean mistletoe is comparable to that of Viscum album L., European mistletoe, 185.6% and 176.5%, respectively. However, the water extracts of the both plants did not show any prolongation effects. Platelet activating factor (PAF) receptor binding assay was carried out to elucidate the action mechanisms of the extracts, and both of the methanol extracts did not show any inhibitory activity. The $LD_{50}$ of the methanol extracts of both mistletoes are more than 2 g/kg. These results suggest that the mehtanol extract of Korean mistletoe might be a potential candidate to develop new drug to improve microcirculation.

Antibacterial activity and toxicity of Halymenia durvillei red seaweed from Kayangan island, South Sulawesi, Indonesia

  • This study aimed to determine the antibacterial activity and toxicity of methanol and hexane extracts of Halymenia durvillei red seaweed which were found abundantly in Kayangan island, South Sulawesi. The antibacterial activity of the crude extract was tested against five gram-negative bacteria, namely Escherichia coli, Salmonella typhi, Pseudomonas aeruginosa, Aeromonas hydrophila, and Vibrio harveyi at a dose of 200 g/disk. Extract toxicity was tested on Artemia salina larvae at concentrations of 1,000, 500, 250, 125, 62.5, and 31.25 ㎍/mL. The results showed that the methanol and hexane extracts of H. durvillei had the highest activity against S. thypi and A. hydrophila, respectively, with inhibition zones of 26.2 mm and 21.0 mm. On the other hand, the two extracts did not show activity against E. coli and P. aeruginosa, respectively. The toxicity of the methanol extract of H. durvillei was twice as high as that of the hexane extract with half-maximal inhibitory concentration of 98.24 and 184.21 ㎍/mL, respectively. Thus, the methanol and hexane extracts of red seaweed H. durvillei have the potential as new antibacterial agents respectively against the pathogenic bacteria S. typhi and A. hydrophila, but also have the opportunity to be developed into antitumor herbal compounds.

산죽잎으로 부터 추출한 항산화물질의 특성 (Characterization of Antioxidants Extracted from Leaves of Sanjook(Sasa boreails var. chiisanensis))

  • 국내산 산죽잎을 methanol로 추출하여 천연 항산화제로서의 이용성을 조사하기 위하여 여러 방법으로 항산화 활성을 측정하고 안정성에 대해 조사하였다. 산죽잎 메탄올 추출물은 0.1 mg/l에서 0.8mg/ml의 범위에서 농도 의존적으로 항산화활성이 증가하였다. 산죽잎 메탄올추출물은 DPPH radical 소거능에 대한 $IC_{50}$ 값은 $583{\mu}g/ml$, SOD 유사활성능에 대한 $IC_{50}$값은 $800{\mu}g/ml$, hydrogen peroxide 소거능에 대한 $IC_{50}$값은 $38{\mug}/ml$이 었으며 대조구로 사용한 BHT의 각각 의 $IC_{50}$값은 $271{\mu}g/ml,{\;}680{\mu}g/ml,{\;}30{\mu}g/ml$이었다. 산죽잎 메탄올추출물의 자동산화 억제능은 상기 농도 범위에서 $55\∼60\%$의 억제효과를 나타내었다. 산죽잎 메탄올추출물의 pH 안정성은 pH 6에서 안정하였으나 산성이나 알카리성으로 갈수록 pH 안정성은 떨어졌다. 산죽잎 메탄올추출물의 열안정성은 $0^{\circ}C$에서 $120^{\circ}C$의 온도범위에서 $80\%$ 이상의 항산화능을 유지하였다.

용매별 정향 추출물의 항산화효과 (Antioxidant Activities of Clove by Extraction Solvent)

  • 본 연구는 정향추출물(물, 메탄올 및 에테르)의 항산화 효과를 리놀레인산 기질 및 리놀레인산 에멀젼기질에서 비교, 검토하였다. 물, 메탄올 및 에테르용매에 따른 정향의 총페놀 함량은 23.1, 42.8, 34.9%로 메탄올>에테르>물 추출물 순이었다. Eugenol 함량은 메탄올 추출물의 경우 6.1%이었고 에테르 추출물 8.1%이었으며 물 추출물에서는 검출되지 않았다. 정향 추출물의 DPPH 소거효과는 물 추출물 경우 36.1%, 메탄을 추출물의 경우 30.9%, 에테르 추출물의 경우 29.7%로 나타났다. 리놀레인산 기질에서의 항산화 효과는 BHT>에테르 추출물> $\alpha$-tocopherol>메탄올 추출물>물 추출물의 순이었다. 리놀레인산 에멀젼 시스템에서의 항산화 효과는 BHT>물 추출물>메탄올 추출물>에테르 추출물> $\alpha$-tocopherol의 순이었다.

국내 자생식물의 항균활성 (Screening of Domestic Plants with Antibacterial Activity)

  • 새로운 유용식물자원개발의 일환으로 80종류(95 시료)의 국내자생식물을 채집하여, 이들 methanol 추출물의 Bacillus subtilis, Staphylococcus aureus, Escherichia coli 및 Vibrio parahaemolyticus에 대한 항균력을 검토하였다. 실험대상 4가지 균주 모두에 항균성을 보인 것은 사철쑥, 지칭개, 뽀리뱅이, 꿀풀, 광대나물 그리고 향나무였으며, 씀바귀, 떡쑥, 애기똥풀, 조팝나무, 냉이 전초 및 동백나무 잎 가지 등을 포함한 8 종류는 적어도 3가지 균주에 대해 항균성을 보였다. 반면에, 라일락 잎은 그람 음성균인 E. coli와 V. parahaemolyticus에서, 꽃다지는 V. parahaemolyticus에서, 얼레지의 인경은 B. subtilis에 대해 선택적인 항균성을 보였다. 특정균주에 대한 항균력을 살펴보면, B. subtilis에 대해서는 얼레지의 인경(18mm)이, S. aureus에 대해서는 조팝나무(16 mm), E. coli에 대해서는 라일락의 잎(18mm) 그리고 V. parahaemolyticus에 대해서는 꽃다지(23mm)가 각각 가장 높은 항균성을 나타내었다. 한편, 비교적 강한 항균활성을 보여준 9종류의 methanol 추출물을 극성에 따라 용매분획하여 얻은 n-hexane, chloroform, ethyl acetate와 물분획물의 B. subtilis와 V. parahaemolyticus에 대한 항균성 실험을 실시한 결과, 실험한 모든 분획물중에서 지칭개의 chloroform 분획물이 가장 높은 항균력 (각각 17mm, 29mm)을 보였다. 뽀리뱅이는 chloroform 분획물에서, 사철쑥, 씀바귀 및 꿀풀은 hexane 분획물에서, 그리고 동백 잎 줄기는 ethyl acetate 분획물에서 각각 상대적으로 강한 항균성을 보였으나, 향나무와 쇠뜨기는 물분획물을 제외한 모든 분획물에서 비교적 고른 항균성을 보였다.
