Quality Characteristics and Antioxidant Activities of Yanggaeng added with Pleurotus eryngii Powder (새송이버섯 분말 첨가 양갱의 품질 특성 및 항산화 활성)

  • This study investigated the quality characteristics and antioxidant activities of Yanggaeng prepared with different ratios of Pleurotus eryngii powder: 0, 3, 6, and 9%. According to the results, the moisture content and pH increased with the addition of Pleurotus eryngii powder, but the Brix decreased. As the amount of Pleurotus eryngii powder increased, the lightness (L) increased, while the redness (a) and yellowness (b) decreased. The hardness of the control groups was lower than the Pleurotus eryngii powder groups. The total polyphenol content was influenced by the addition of Pleurotus eryngii powder. As the amount of Pleurotus eryngii powder increased, DPPH, reducing power, and metal chelating effect increased significantly. The results of a consumer acceptance test showed no significant differences in color between the controls and two groups (3% and 6%), and no significant differences in smell, taste, and overall acceptability between the controls and groups with 3% Pleurotus eryngii powder. Therefore, up to 3% Pleurotus eryngii powder can be incorporated into Yanggaeng to improve the quality and antioxidant potential.

  • The interaction of cadmium (II) ion with quercetin, qurecitrin and (+)-catechin was investigated in aqueous solution at various pH. The flavonoid/cadmium stochiometries for cadmium (II) binding to flavonoid have been determined by UV-visible spectroscopy. 1:1 Cd(II)-Flavonoid complex had a maximum absorbance and showed the bathochromic shift of the long-wavelength band of the UV-vis spectra in the alkaline pH, that occurs upon complexation, is due to a ligand-tometal charge transfer. The optimal concentration of Cd(II)-flavonoid complexes showed that complexation reaction could be classified in the following way: 55.27 ${\mu}M$ catechin > 54.72 ${\mu}M$ quercetin > 53.52 ${\mu}M$ quercitrin at the chelating site level. These results suggest that Cd(II)-flavonoid complex has the optimal condition of chelation in 0.2 M $NH_3$ - 0.2 M $NH_4Cl$ (pH 8.0) solution.

A Study on the Properties of the Heavy Duty Rust-Converting Agent used in the Potential Hazard Areas of Fire & Explosion (잠재적 화재.폭발 위험 지역 작업용 녹전환형 중방식 코팅제의 특성에 관한 연구)

  • This study was concerned with the development of a heavy duty rust-converting agent, the function of which is to form metal complex coatings, containing vinyl halide-acrylic terpolymer emulsion, defoamer, emulsifying agent, glass flakes, chelating agent such as gallotannic acid, gallic acid, and pyrogallic acid, and other additives. The resulted emulsion products(Sample No.1~No.5) were characterized through test either in the forms of emulsions, which include Viscosity, Penetration rate, Acidity and Film drying rate test, or in the forms of coated layer on rusty steel substrates by FT-IR, which include hardness, gloss, salt spray, adhesion and flame retardant test. The test results are as follows ; Penetration rate(0.1~0.4 mm/min), Solid content(70%), Acidity (pH 1.8~2.0), Specific gravity(1.30~1.35), Film drying rate(108min, RH 40% ; 150min, RH 80%), Gloss(83~92, incident angle $60^{\circ}$; 88~97, incident angle $85^{\circ}$), Pencil hardness(4H~5H), Adhesion (100/100), Salt spray test(>720Hr), LOI(%) value(38%), Vertical burning test(UL 94-v-l). According to the various performance of specimens show above, the evaluation of the availability of this heavy duty rust-converting agent can be concluded that all the samples(No.1~No.5) are capable of being used in the field of chemical plant and in the hazard areas of fire and explosion potential. It was observed that the properties of sample No.2, especially gloss and hardness, were much better than that of the other samples.

Antioxidant Activity and Protection from DNA Damage by Water Extract from Pine (Pinus densiflora) Bark

  • Water extract from Pinus densiflora (WPD) was investigated for its antioxidant activity and its ability to provide protection from DNA damage. A series of antioxidant assays, including a 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay, a reducing power assay, a metal-chelating assay, a superoxide radical scavenging assay, and a nitrite scavenging ability, as well as a DNA damage protection assay were performed. Total phenolic content was found to be 211.32 mg Tan/g WPD. The extract scavenged 50% DPPH free radical at a concentration of 21.35 ${\mu}g/mL$. At that same concentration, the reducing power ability of WPD was higher than that of ${\alpha}$-tocopherol. The extract chelated 68.9% ferrous ion at the concentration of 4 mg/mL. WPD showed better nitrite scavenging effect at the lower pH. Meanwhile, WPD exhibited a strong capability for DNA damage protection at 1 mg/mL concentration. Taken together, these data suggest water extract from Pinus densiflora could be used as a suitable natural antioxidant.

Chelation of Thallium (III) in Rats Using Combined Deferasirox and Deferiprone Therapy

  • Thallium and its compounds are a class of highly toxic chemicals that cause wide-ranging symptoms such as gastrointestinal disturbances; polyneuritis; encephalopathy; tachycardia; skin eruptions; hepatic, renal, cardiac, and neurological toxicities; and have mutagenic and genotoxic effects. The present research aimed to evaluate the efficacy of the chelating agents deferasirox (DFX) and deferiprone (L1) in reducing serum and tissue thallium levels after the administration of thallium (III), according to two different dosing regimens, to several groups of Wistar rats for 60 days. It was hypothesized that the two chelators might be more efficient as a combined therapy than as monotherapies in removing thallium (III) from the rats' organs. The chelators were administered orally as either single or combined therapies for a period of 14 days. Serum and tissue thallium (III) and iron concentrations were determined by flame atomic absorption spectroscopy. Serum and tissue thallium (III) levels were significantly reduced by combined therapy with DFX and L1. Additionally, iron concentrations returned to normal levels and symptoms of toxicity decreased.

In Vitro and Cellular Antioxidant Activity of a Water Extract of Saururus chinensis

  • The water extract of Saururus chinensis was investigated for oxygen radical absorbance capacity (ORAC), reducing capacity, metal chelating activity, and intracellular antioxidant activity using HepG2 cell. When 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) was used for the generation of peroxyl radicals in vitro, S. chinensis extract (SC-E) showed the strong and concentration-dependent scavenging activity through donating protons which could be explained by its reducing property. When hydroxyl radicals were generated in vitro through the addition of $Cu^{2+}$ and $H_2O_2$, SC-E demonstrated the antioxidant activity depending on its concentration. In HepG2 cell model, most of intracellular oxidative stress generated by AAPH was efficiently removed by SC-E. However, when $Cu^{2+}$ without $H_2O_2$ was used as an oxidant in the intracellular assay, SC-E partially reduced the oxidative stress caused by $Cu^{2+}$ in cellular antioxidant activity assay system. These results indicate that SC-E could be utilized for the development of functional foods as antioxidant resource in the near future.

Antioxidative and Antimutagenic Characteristics of Melanoidin Related Products (Melanoidin의 항산화성 및 항돌연변이원성)

  • Melanoidins, as brown colored polymers, are formed through the diversified reaction systems of Maillard type and other reactions. Melanoidins are important components in relation to food quality and also are known to have antioxidative, mutagenic and antimutagenic activities. Since these aspects have been extensively reviwed elsewhere, only the recent studies regarding to their antioxidative and antimutagenic activities are discussed in this review. Even though their mechanisms are not clearly identified, melanoidins or specific fractions isolated from their mixtures have shown varied antioxidative activities depending on the reaction systems and reaction conditions. Those activities presumely are derived from the complex functional properties of hydrogen / electron donors and metal chelating power, which are originated from their reductone structure and others. It is considered that pyrolysate and other mutagens are formed by the given conditions in some cases during browning reaction, whereas melanoidins and their fractions have antimutagenic effects on chemical and other mutagens. There are positive correlationship among the color intensity, antioxidative activity and antimutagenicity of melanoidins or their fractions. These suggest that the antimutagenicity of melanoidins could be attributed to their antioxidative properties, however, it might also be due to other factors, because the relevant responses for antimutagenicity are very complicate and not clear. Accordingly, further studies are required to determine the actual acitivities and mechanisms involved in antioxidation and (anti)mutagenicity of melanoidins by reaction systems / conditions and by the isolated fractions. And also, additional studies are needed to evaluate the applications of melanoidins and their relevant effects to food and human health.

Improved immune-enhancing activity of egg white protein ovotransferrin after enzyme hydrolysis

  • Ovotransferrin (OTF), an egg protein known as transferrin family protein, possess strong antimicrobial and antioxidant activity. This is because OTF has two iron binding sites, so it has a strong metal chelating ability. The present study aimed to evaluate the improved immune-enhancing activities of OTF hydrolysates produced using bromelain, pancreatin, and papain. The effects of OTF hydrolysates on the production and secretion of pro-inflammatory mediators in RAW 264.7 macrophages were confirmed. The production of nitric oxide (NO) was evaluated using Griess reagent and the expression of inducible nitric oxide synthase (iNOS) were evaluated using quantitative real-time polymerase chain reaction (PCR). And the production of pro-inflammatory cytokines (tumor necrosis factor [TNF]-α and interleukin [IL]-6) and the phagocytic activity of macrophages were evaluated using an ELISA assay and neutral red uptake assay, respectively. All OTF hydrolysates enhanced NO production by increasing iNOS mRNA expression. Treating RAW 264.7 macrophages with OTF hydrolysates increased the production of pro-inflammatory cytokines and the phagocytic activity. The production of NO and pro-inflammatory cytokines induced by OTF hydrolysates was inhibited by the addition of specific mitogen-activated protein kinase (MAPK) inhibitors. In conclusion, results indicated that all OTF hydrolysates activated RAW 264.7 macrophages by activating MAPK signaling pathway.

  • A protective oxide layer forms on the material surfaces of a Nuclear Power Plant during operation due to high temperature. These oxides can host radionuclides, the activated corrosion products of fission products, resulting in decommissioning workers' exposure. These deposited oxides are iron oxides such as Fe3O4, Fe2O3 and mixed ferrites such as nickel ferrites, chromium ferrites, and cobalt ferrites. Developing a new chemical decontamination technology for domestic CANDU-type reactors is challenging due to variations in oxide compositions from different structural materials in a Pressurized Water Reactor (PWR) system. The Korea Atomic Energy Research Institute (KAERI) has already developed a chemical decontamination process for PWRs called 'HyBRID' (Hydrazine-Based Reductive metal Ion Decontamination) that does not use organic acids or organic chelating agents at all. As the first step to developing a new chemical decontamination technology for the Pressurized Heavy Water Reactor (PHWR) system, we investigated magnetite dissolution behaviors in various HyBRID inorganic acidic solutions to assess their applicability to the PHWR reactor system, which forms a thicker oxide film.

Study on the Enzyme of Basidiomycetes(I) -The Effects of Iron Ions on the Light-Induced Mitochondrial $F_0F_1-ATPase$ of Lentinus edodes- (담자균류의 효소에 관한 연구(I) -표고버섯 중의 광감응성 Mitochondrial $F_0F_1-ATPase$의 철이온 효과-)

  • The effects of the iron ions for the light-induced mitochondrial $F_0F_1-ATPase$ of Lentinus edodes was studied. The enzyme activity was stimulated up to 202% by 0.1 mM $Fe^{2-}$ ion, but was inhibited by $Fe^{3+}\;and\;Mg^{2+}$. In the presence of 0.5 mM $Mg^{2+}$, the activity also increased 32% by 0.1 mM $Fe^{2+}$ ion, and decreased to a similar extent by $Fe^{3+}$ ion than by only $Fe^{3+}$ ion. Also, the activity was inhibited 53% by 5.0 mM $Fe^{2-}$ ion in the presence of 0.5 mM $Mg^{2+}$ ion and various concentration of $Fe^{3+}$ ion(mM). These results showed that $Fe^{2+}$ strongly stimulated the enzyme activity and its role for the enzyme was independent of $Mg^{2+}$ ion, but was dependent of $Fe^{3+}$ ion. From inactivation of the enzyme by addition of metal chelating agent, EDTA, it is suggested that the enzyme is to be metalloenzyme. The optimal pH and temperature of the enzyme in the presence of 0.1 mM $Fe^{2+}$ was 7.6 and $63^{\circ}C$, respectively.
