Fluorescence Resonance Energy Transfer in Calf Thymus DNA from a Long-Lifetime Metal-Ligand Complex to Nile Blue

  • We extended the measurable time scale of DNA dynamics to submicrosecond using a long-lifetime metal-ligand complex, $[Ru(phen)_2(dppz)]^{2+}$ (phen=1,10-phenanthroline, dppz=dipyrido[3,2-a:2',3'-c]phenazine) (RuPD), which displays a mean lifetime near 350 ns. We partially characterized the fluorescence resonance energy transfer (FRET) in calf thymus DNA from RuPD to nile blue (NB) using frequency-domain fluorometry with a high-intensity, blue light-emitting diode (LED) as the modulated light source. There was a significant overlap of the emission spectrum of the donor RuPD with the absorption spectrum of the acceptor NB. The F$\ddot{o}$rster distance ($R_0$) that was calculated from the spectral overlap was $33.4\;{\AA}$. We observed dramatic decreases in the steady-state fluorescence intensities of RuPD when the NB concentration was increased. The intensity decays of RuPD were matched the closest by a triple exponential decay. The mean decay time of RuPD in the absence of the acceptor NB was 350.7 ns. In a concentration-dependent manner, RuPD showed rapid intensity decay times upon adding NB. The mean decay time decreased to 184.6 ns at $100\;{\mu}M$ NB. The FRET efficiency values that are calculated from the mean decay times increased from 0.107 at $20\;{\mu}M$ NB to 0.474 at $100\;{\mu}M$ NB concentration. The use of FRET with a long-lifetime metal-ligand complex donor is expected to offer the opportunity to increase the information about the structure and dynamics of nucleic acids.

Effects of Metal Anion Complexes in Electrolyte on the Properties of Anodic Oxide Films on ADC12 Al Alloy

  • The anodization of ADC12 aluminum alloy was investigated in the metal anionic acid media. Anodic oxide films containing foreign elements were formed on ADC12 Al alloy by anodization in the anion complex solution. Furthermore, the rough surface and cracks were considerably smoothened by the deposit of metal anions. When the size of metal anion was small, relatively large amount of metal anions was loaded in anodic films. Existence of $MoO_3$, $TiO_2$ and MgO was confirmed by XPS. According to the results of Tafel analysis, Mo oxide represented the most noble anti-corrosion potential due to $MoS_2$ formation. Corrosion current densities were generally higher than that of pristine anodic oxide without anion complexes.

The Treatment of Heavy Metal-cyanide Complexes Wastewater by Zn$^{+2}$/Fe$^{+2}$ Ion and Coprecipitation in Practical Plant (II) (아연백법 및 공침공정을 이용한 복합 중금속-시안착염 폐수의 현장처리(II))

  • Industrial wastewater generated in the electroplating and metal finishing industries typically contain toxic free and complex metal cyanide with various heavy metals. Alkaline chlorination, the normal treatment method destroys only free cyanide, not complex metal cyanide. A novel treatment method has been developed which destroys both free and complex metal cyanide as compared with Practical Plant(I). Prior to the removal of complex metal cyanide by Fe/Zn coprecipitation and removal of others(Cu, Ni), Chromium is reduced from the hexavalent to the trivalent form by Sodium bisulfite(NaHSO$_3$), followed by alkaline-chlorination for the cyanide destruction. The maximum removal efficiency of chromium by reduction was found to be 99.92% under pH 2.0, ORP 250 mV for 0.5 hours. The removal efficiency of complex metal cyanide was max. 98.24%(residual CN: 4.50 mg/L) in pH 9.5, 240 rpm with 3.0 $\times$ 10$^{-4}$ mol of FeSO$_4$/ZnCl$_2$ for 0.5 hours. The removal efficiency of Cu, Ni using both hydroxide and sulfide precipitation was found to be max. 99.9% as Cu in 3.0 mol of Na$_2$S and 93.86% as Ni in 4.0 mol of Na$_2$S under pH 9.0$\sim$10.0, 240 rpm for 0.5 hours. The concentration of residual CN by alkaline-chlorination was 0.21 mg/L(removal efficiencies: 95.33%) under the following conditions; 1st Oxidation : pH 10.0, ORP 350 mV, reaction time 0.5 hours, 2nd Oxidation : pH 8.0, ORP 650 mV, reaction time 0.5 hours. It is important to note that the removal of free and complex metal cyanide from the electroplating wastewater should be employed by chromium reduction, Fe/Zn coprecipitation and, sulfide precipitation, followed by alkaline-chlorination for the Korean permissible limit of wastewater discharge, where the better results could be found as compared to the preceding paper as indicated in practical treatment(I).

The Fluorescence Study on the Inducing Orientation of 4-Biphenyl Acetonitrile Adsorbed on Metal Colloids (금속콜로이드 표면에 흡착된 4-Biphenyl Acetonitrile의 흡착배향 유도에 관한 형광 연구)

  • The fluorescence study was performed to see whether the adsorption orientation of 4-biphenyl acetonitrile(BPAN) on metal colloids can be changed by forming an inclusion complex with $\alpha$-cyclodextrin($\alpha$-CD). The fluorescence quenching was observed with increasing temperature to confirm the direct adsorption of BPAN to the Au and Ag colloidal surfaces. BPAN adsorbed on the metal colloids formed inclusion complex with $\alpha$-CD regardless of the kinds of metal colloids. The formation constants, 32 $M^{-1}$ and 13 $M^{-1}$ for Au and Ag colloids respectively, were obtained with Benesi-Hildebrand plot. The molecules adsorbed on both the Au and Ag colloidal surfaces behaved similarly to each other, leading to the conclusion that the orientation of BPAN adsorbed on the metal colloids can be modified with $\alpha$-CD.

  • The adsorption characteristics of heavy metal ions $(Ni^{2+}$, $Zn^{2+}$) on domestic activated carbon which manufactured from the wood, waste rubber and coal has been studied. The following conclusions were obtained by investigating the effect of pH, halides, cyanide and temperature on the absorption of heavy metal ions and by determining absorption rate and absorption isotherms. When heavy metal ions were adsorbed on activated carbon in the pH range from 6 to 7, treated heavy metal ions were almost come to the equilibrium and recovered in the form of complex anions such as $MX_4^{2- }$and $M(CN)_4^{2-}$ in an hour. The absorption isotherms of heavy matel ions were well fitted in Freundlich's equation. Heavy metal complex anions on activated carbon were easily eluted by using 0.1N HCl solution. These results suggest that the complex anions have unexpectedly strong affinity for the surface of activated carbon.

A High Frequency Complex Modulation Method of the Electronic Ballast for Metal Halide Lamps (메탈 할라이드 램프용 전자식 안정기의 고주파 복합 변조법)

  • This paper presents an electronic ballast using a novel complex modulation method for the metal halide lamp. The proposed modulation method, which has a modulating signal of swept complex frequency, can eliminate the acoustic resonance more effectively than the conventional modulation method, which has a modulating signal of constant frequency. For the purpose of future application specific integrated circuits (ASIC). the controller of the proposed ballast has been designed only with erasable programmable logic devices (EPLDs), but without a microprocessor. In this paper, detailed proposed modulation schemes are described and experimental results on the proto type 150W metal halide lamp ballast with the proposed modulation method ate discussed.

Spectroscopic characterization of N,N'-bis(salicylidene)pentane-1,3-diamine nickel(II) complex

  • The $N_2O_2$ tetradentate Schiff base ligand, N,N'-bis(salicylidene)pentane-1,3-diamine (Salpn), coupled with 1:2 concentration ratio of 1,3-diaminopentane and salicylaldehyde was used to produce a series of macrocyclic Nikel(II) complexes. In the metal complexation, it was observed that Salpn macrocyclic ligand can adopt more than a metal ion giving an unique multinuclear metal complexes including Ni(II)Salpn and $Ni(II)_3(Salpn)_2$. Characteristic IR ${\upsilon}(M-O)$ peaks for Ni(II)Salpn and $Ni(II)_3(Salpn)_2$ were observed to be $1028cm^{-1}$ and $1024cm^{-1}$, respectively. Characteristic UV-Vis absorption ${\lambda}_{max}$ peaks for $Ni(II)_3(Salpn)_2$ were observed to be 241nm and 401 nm. Structural characterization of $Ni(II)_3(Salpn)_2$ by NMR exhibits that the salicylidene ring moiety has two different resonance signals originated from the magnetically asymmetric diligand and trinuclear bis complex. Complete NMR signal assignments and characterizations elucidating structural features of $Ni(II)_3(Salpn)_2$ were described in detail.

Heavy Metal Concentrations in the Soil of Yoecheon Industrialized Complex Area (여천공단 주변 토양의 중금속함량에 관한 연구)

  • This study was focused on the investigation for the heavy metal contamination of soils derived from Yoecheon Industrial Complex in Chonnam Province. Total 201 soil samples including farmland, paddy, forest and playground soils were collected, extracted by 0.1N HCl and analyzed for Cd, Cu, Cr, Fe, Mn and Zn using AAS. Most pH values in soils were weak acidic ranges of 4-6, while some of them were over 8.0. Concentrations of Cd, Cu, Fe and Zn in the soils tend to be relatively high around the Au-Ag mine area, railroad stations and oil station, however, those of Cr, Mn and Pb show high at the northside of Pohang Steel Co (POSCO). Cadmium-Zn couples, and Cd-Zn and Zn-Cr couples have relatively good correlations in the farmland soils and paddy soils, respectively. Concentrations of Cd, Fe and Zn with depth tend to be higher in the top soils than in the bottom soils, but those of Cu and Pb do not show regular variations with depth.

  • infrared(IR) absorption spectra were calculated for the ethyl ester of p-tert-butylcalix[4]arene (1) in the cone conformer and its alkali-metal-ion complex. The vibrational spectra were obtained by restricted Hartree-Fock (RHF) calculations with the 6-31G basis set. The characteristic vibrational frequencies of various C-O and C=O stretching motions of the complexes show that the structure of 1+K+ complex is almost of C4v symmetry compared to 1+Na+ (C2v) analogue. The theoretical results for the host molecule 1 and complex (1+Na+) were compared with the experimental results, and the calculated vibrational frequencies agree well with the features of the experimental spectra.

Surface Analysis and Electrical Properties for Complex with Concentration of Metal ion in LB Ultra-thin Films Using IMI-O Polymer (IMI-O 고분자 LB막의 금속 이온의 착체 농도에 따른 전기특성 및 표면분석)

  • We fabricated an IMI-O polymer containing an imidazole group that could form a complex structure between the monolayer and the metal ions at the air-water interface. Also, the surface analysis and the electrical properties of metal ion complex of Langmuir-Blodgett (LB) films were investigated by using $\pi$-A isotherms. Atomoic force microscopy (AFM), current-voltage (I-V) measurements. In the $\pi$-A isotherms the molecular area was expanded with $Fe^{3+}$ concentration increase. It is considered that the expansion of molecular area is due to electrostatic repulsion between the polymer chains and hydrophobic increase of ionic strength. In the I-V characteristics, it is found that the limiting area has effects on the change of conductivity. And, the dielectric relaxation time decreased for increase of the $Fe^{3+}$ concentration.
