Studies on Chromatographic Behaviors of the Metal Complexes by Various Eluents (용리제의 종류에 따른 금속착물의 크로마토그래피적인 거동에 관한 연구)

  • We examined eluting behaviors for cationic complexes with the eluting cations. Retention volumes of hydrophilic complexes when eluted by the high concentration of aqueous KCl, NaCl and $NH_4Cl solutions, have been increased in the order NH_4^+ > Na^+ > K^+$, but in the low concentration, retention volumes of hydrophilic complexes bave been increased in the order $Na^+ > NH4^+ > K^+$. On the order hand, retention volumes of hydrophobic complexes when eluted by aqueous KCl, NaCl and $NH_4Cl solutions, have been increased in the order NH_4^+ > Na^+ > K^+$ in the whole concentration range. And we examined eluting behaviors for cationic complexes with the eluting anions. Hydrophilic complexes were associated more strongly with chloride ion among halide ions. However, hydrophobic complexes were associated more strongly with iodide ion. Sulfate ion, a highly hydrated anion, was associated more strongly with hydrophilic complexes than with hydrophobic complexes.

Neutron Activation Analysis of Cadmium Deposition in Hair and Animal Tissues (동물체모 및 장기 중 카드뮴의 방사화 분석)

  • Rats were ingested in drinking water 600mg/L of cadmium chloride solution during 3 months, then the distribution of Cd in major organs and hair were determined by neutron activation analysis. The results were as followings. 1. After administration for 24 hours using $^{115m}Cd$ as tracer, the distribution of blood was 0.03%, kidney 2.99% and liver 3.50% to determine with whole body counter. 2. Cd metal was rapidly excreted with kidney through blood and their accumulation appeared in liver and hair. 3. The comparative data to determine using neutron activation analysis. the content of cadmium of major organs in rats ingested of $CdCl_2$ during 3 month were shown to increase significantly both hair and liver. Above facts, hair samples were able to use as the diagnostic index to evaluate the accumulation of cadmium in liver.

  • Cyclone electrowinning method was used to recover tin to metal in hydrochloric acid solution. The effects of flow rate, current density, tin concentration, and hydrochloric acid concentration on the electrowinning of tin were investigated. As the flow rate increased and the concentration of tin and hydrochloric acid decreased, the tin recovery and current efficiency increased. As the current density increased, the recovery rate increased but the current efficiency decreased. Tin can be effectively recovered at flow rates of 18 L/min., $3A/dm^2$, 2.5 g/L Sn, and 5 wt.% HCl.

In Vitro Determination of Dengue Virus Type 2 NS2B-NS3 Protease Activity with Fluorescent Peptide Substrates

  • The NS2B-NS3(pro) polyprotein segment from the dengue virus serotype 2 strain 16681 was purified from overexpressing E. coli by metal chelate affinity chromatography and gel filtration. Enzymatic activity of the refolded NS2B-NS3(pro) protease complex was determined in vitro with dansyl-labeled peptide substrates, based upon native dengue virus type 2 cleavage sites. The 12mer substrate peptides and the cleavage products could be separated by reversed-phase HPLC, and were identified by UV and fluorescence detection. All of the peptide substrates (representing the DEN polyprotein junction sequences at the NS2A/NS2B, NS2B/NS3, NS3/NS4A and NS4B/NS5 sites) were cleaved by the recombinant protease NS2B-NS3(pro). No cleavage was observed with an enzymatically inactive S135A mutant of the NS3 protein, or with a modified substrate peptide of the NS3/NS4A polyprotein site that contained a K2093A substitution. Enzymatic activity was dependent on the salt concentration. A 50% decrease of activity was observed in the presence of 0.1M sodium chloride. Our results show that the NS3 protease activity of the refolded NS2B-NS3(pro) protein can be assayed in vitro with high specificity by using cleavage-junction derived peptide substrates.

  • We demonstrated a crack-free ${\alpha}-Ga_2O_3$ on sapphire substrate by horizontal halide vapor phase epitaxy (HVPE). Oxygen-and gallium chloride-synthesized Ga metal and HCl were used as the precursors, and $N_2$ was used as the carrier gas. The HCl flow and growth temperature were controlled in the ranges of 10~30 sccm and $450{\sim}490^{\circ}C$, respectively. The surface of ${\alpha}-Ga_2O_3$ template grown at $470^{\circ}C$ was flat and the root-mean-square (RMS) roughness was ~2 nm. The full width at half maximum (FWHM) values for the symmetric-plane diffractions, were as small as 50 arcsec and those for the asymmetric-plane diffractions were as high as 1,800 arcsec. The crystal quality of ${\alpha}-Ga_2O_3$ on sapphire can be controlled by varying the HCl flow rate and growth temperature.

  • Titanium and iron are closely related in nature, although titanium is the ninth most abundant element in the Earth's crust. Iron in titanium ores must be removed for use as feedstocks in the manufacture of titanium dioxide pigments and pure $TiCl_4$ for metal titanium. In this study, various beneficiation processes of ilmenite for production of $TiO_2$ have been reviewed and compared. Most of these processes involve a combination of pyrometallurgy and hydrometallurgy. These beneficiation processes of ilmenite generate considerable quantities of wastes primarily in the form of iron salt, iron oxide and acidic effluents. Therefore, it is important that recovery of acid value from waste and conversion of iron bearing waste to useful materials for development of new beneficiation processes of ilmenite.

Nickel Ion Adsorption Behavior of Ceriporia lacerata Isolated from Mine Tailings in Korea

  • In the present study, surface of laccase producing Ceriporia lacerata was modified using 4-bromobutyryl chloride and polyethylenimine. The modified biomass was freeze dried and utilized as a biosorbent for the removal of Ni(II) from aqueous solution. The physicochemical properties of the biosorbent were analyzed using scanning electron microscopy and Fourier transform infrared spectroscopy. Batch experiments were carried out as a function of contact time (0-60 min), pH (2 to 8), adsorbent dosage (25-150 mg), and initial Ni(II) concentration (25-125 mg/L). The results indicate that surface modified biosorbent effectively adsorbed (9.5 mg/0.1 g biomass) Ni(II) present in the solution. The equilibrium adsorption data were modeled with different kinetic and isotherm models. The Ni(II) adsorption followed pseudo-first-order kinetics (R2 = 0.998) and Langmuir isotherm (R2 = 0.994) model. Hydroxyl and carbonyl functional groups present in biomass play a major role in the adsorption of Ni(II). The adsorbed Ni(II) from the biosorbent was successfully desorbed (85%) by 1M HCl. The results of the study indicate that the surface modified C. lacerate biomass could be used for the treatment of Ni(II) contaminated ground waters.

A study on characteristics of influent and effluent pollutants in public sewage treatment works combined with industrial wastewater and landfill leachate (공공하수처리시설에서 수질오염물질 유입 및 배출 특성 고찰 - 산업폐수 및 매립지 침출수 연계처리 시설을 중심으로 -)

  • In this study, we investigated influent and effluent water pollutants in 53 Public Sewage Treatment Works (PSTWs) where industrial wastewater or landfill leachate is combined four times for two years from 2014 to 2015. Also, we analyzed the characteristics of heavy metals and volatile organic carbons at influent and effluent of these PSTWs caused by sewage treatment combined with industrial wastewater or landfill leachate. As a result, six heavy metals such as barium, copper, iron, manganese, nickel and zinc, and four volatile organic carbons (VOCs) including phenols, di(2-)ethylhexyl phthalate (DEHP), formaldehyde and toluene were observed above detection limits in most of PSTWs. Also, it was revealed that six heavy metals such as hexavalent chromium, mercury, cadmium, chromium, nickel and selenium, and four VOCs including 1,1-dichloroethylene, vinyl chloride, naphthalene, and epichlorohydrin were observed more frequently according to precipitation. As a result of reviewing the monitoring data on "Water Quality Monitoring Networks" in lower watersheds of PSTWs, both heavy metals and VOCs were below detection limits, indicating that the effluent water had little influence on the watershed. Nevertheless for the better management of influent and effluent pollutants in PSTWs, it is necessary to establish the advanced management plans for water pollutants in PSTWs, which include a list of priority substances management, monitoring plans, and guidelines for industrial wastewater and landfill leachate combined in PSTWs.

Desalting of papermaking tobacco sheet extract using selective electrodialysis

  • The inorganic components in tobacco sheet extract have significant influence on the sensory taste of the cigars and the harmful component delivery in cigarette smoke. To identify the contributions of the divalent inorganic components on harmful components delivery in cigarette smoke, a self-made selective electrodialysis was assembled with monovalent ion-selective ion exchange membranes. The influences of current density and extract content on the desalination performance were investigated. Result indicates that the majorities chloride, nitrate, and sulfate ions were removed, comparing with 50-60% of potassium and only less than 10% of magnesium and calcium ions removed in the investigated current density. The permselectivity of the tested cations across the Selemion CSO cation exchange membranes follows the order: $K^+>Ca^{2+}>Mg^{2+}$. A current density of $15mA/cm^2$ is an optional choice by considering both the energy consumption and separation efficiency. When the extract contents are in the range of 7%-20%, the removal ratios the potassium ions are kept around 60%, while the removal ratios of the calcium and magnesium ions fluctuate in the range of 16-27% and 8-14%, respectively. The tobacco smoke experiments indicated that the divalent metal ions have dual roles for the harmful component delivery in cigarette smoke. The divalent potassium and calcium ions were unfavorable for the total particulate matter emission but beneficial to decrease the HCN delivery in the mainstream cigarette smoke. The selective electrodialysis is a robust technology to decrease the harmful component delivery in cigarette smoke.

Phytotoxic effects of mercury on seed germination and seedling growth of Albizia lebbeck (L.) Benth. (Leguminosae)

  • A study was conducted to determine the phytotoxic effect of mercury on seed germination and seedling growth of an important arid legume tree Albizia lebbeck. The seeds germination and seedling growth performance of A. lebbeck responded differently to mercuric chloride treatment (1 mM, 3 mM, 5 mM and 7 mM) as compared to control. Seed germination of A. lebbeck was significantly (p < 0.05) affected by mercury treatment at 1 mM. Root growth of A. lebbeck was not significantly affected by mercury treatment at 1 mM, and 3 mM. Shoot and root length of A. lebbeck were significantly (p < 0.05) affected by 5 mM concentration of mercury treatment. Increase in concentration of mercury treatment at 5 mM and 7 mM significantly (p < 0.05) reduced seedling dry weight of A. lebbeck. The treatment of mercury at 1 mM decreased high percentage of seed germination (22%), seedling length (10%), root length (21.85%) and seedling dry weight (9%). Highest decrease in seed germination (51%), seedling (34%), root length (48%) and seedling dry weight (41%) of A. lebbeck occurred at 7 mM mercury treatment. A. lebbeck showed high percentage of tolerance (78.14%) to mercury at 1 mM. However, 7 mM concentration of mercury produced lowest percentage of tolerance (51.65%) in A. lebbeck. The seed germination potential and seedling vigor index (SVI) clearly decreased with the higher level of mercury. Plantation of A. lebbeck in mercury-polluted area will help in reducing the burden of mercury pollution. A. lebbeck can serve better in coordinating in land management programs in metal contaminated areas. The identification of the toxic concentration of metals and tolerance indices of A. lebbeck would also be helpful for the establishment of air quality standard.
