Echinostoma macrorchis (Digenea: Echinostomatidae): Metacercariae in Cipangopaludina chinensis malleata Snails and Adults from Experimental Rats in Korea

  • A survey was performed to know the infection status of echinostome metacercariae in Cipangopaludina chinensis malleata snails from Korea. Total 75 snails collected in 5 localities, i.e., Imsil-gun, Jeollabuk-do, Hwasun-gun and Shinan-gun (Aphae and Jido), Jeollanam-do, and Jinju-si, Gyeongsangnam-do, were examined for metacercariae by the artificial digestion method. Infection rates of metacercariae were 80.0%, 66.7%, 100%, 60.0%, and 73.3%, and their densities were 39, 32, 183, 19, and 30 per snail infected, respectively. The metacercariae were round, $105-118{\times}105-118{\mu}m$ in size, with a thin cyst wall, collar spines on the head collar, and excretory granules in 2 canals of excretory tube. Adult flukes were elongated, ventrally curved, and $5,167{\times}939{\mu}m$ in average size. Head collar distinct, bearing 45 collar spines with 5 end groups on each side. Oral sucker subterminal, pharynx well developed, and esophagus somewhat short. Cirrus sac well developed, with a saccular seminal vesicle, and ventral sucker very large. Ovary elliptical and on the median line of the body. Testes tandem and slightly lobed. Eggs operculated, elliptical, and $90-103{\times}55-60{\mu}m$ in size. By scanning electron microscopy, the head collar was prominent with 45 collar spines resembling horns of younger stags. Scale-like tegumental spines were densely distributed on the body surface between the head collar and ventral sucker. Conclusively, it has been first confirmed that the life cycle of E. macrorchis is indigenously maintained in Korea, and C. chinensis malleata snails are popularly infected with the metacercariae of this echinostome.

Infection Status of Estuarine Fish and Oysters with Intestinal Fluke Metacercariae in Muan-gun, Jeollanam-do, Korea

  • The source of human infection with intestinal flukes was surveyed in estuarine fishes, including the dotted gizzard shad, common sea bass, common blackish goby, redlip mullet, black sea bream, and oyster collected from Muan-gun, Jeollanam-do, Korea during August and September 2007. Collected fishes and oysters were artificially digested in pepsin-HCl solution and examined under a stereomicroscope. In 36 shads (Konosirus punctatus) and 20 basses (Lateolabrax japonicus) examined, Heterophyopsis continua metacercariae were found in 58.3% and 100%, and their average numbers were 12.0 and 6.3 per infected fish, respectively. In 34 gobies (Acanthogobius flavimanus) examined, metacercariae of H. continua were detected in 79.4%, Stictodora lari in 97.1%, and Acanthotrema felis in 92.1%, and their average numbers were 45.8, 189.3, and 235.3 per infected fish, respectively. In 37 redlip mullets (Chelon haematocheilus), Heterophyes nocens metacercariae were found in 56.8%, Pygidiopsis summa in 94.6%, and Stictodora fuscata in 45.9%, and the average metacercarial densities were 17.4, 31.3, and 35.1 per infected fish, respectively. In 30 black sea breams (Acanthopagrus schlegeli) and 45 oysters (Crassostrea gigas) examined, no metacercariae were detected. From the above results, it has been confirmed that the dotted gizzard shad, common sea bass, common blackish goby, and redlip mullet from Muan-gun, Jeollanam-do, Korea are infected with the metacercariae of heterophyid flukes.

Infections of Digenetic Trematode Metacercariae in Wrestling Halfbeak, Dermogenys pusilla from Bangkok Metropolitan Region in Thailand

  • This study aimed to investigate metacercarial infections in the wrestling halfbeak, Dermogenys pusilla, collected from Bangkok metropolitan region of Thailand. A total of 4,501 fish from 78 study sites were commonly examined with muscle compression and digestion methods (only head part of fish) during September 2017 to July 2018. The overall prevalence of metacercarial infection was 86.1% (3,876/4,501 individuals), and the mean intensity was 48.9 metacercariae per fish infected. Four species, i.e., Posthodiplostomum sp., Stellantchasmus falcatus, Cyathocotylidae fam. sp., and Centrocestus formosanus, of digenetic trematode metacercariae (DTM) were detected. The prevalences were 65.8%, 52.0%, 2.1%, and 1.2%, respectively and their mean intensities were 23.1, 51.6, 1.4, and 3.2 per fish infected, respectively. The seasonal prevalences were 81.0% in winter, 87.8% in summer and 87.4% in rainy, and the mean intensities were 38.9, 46.6, and 55.2 metacercariae per fish infected, respectively. Conclusively, it was confirmed that the wrestling halfbeak play the role of second intermediate hosts of 4 species of digenetic trematodes including S. falcatus and Posthodiplostomum sp. in Bangkok metropolitan region. And then the metacercariae of C. formosanus and Cyathocotylidae fam. sp. are to be first found in the wrestling halfbeak by this study.

  • 폐홉충(Paragonimus westermani) 초기 감염시의 숙주 면역 기전을 알아보기 위하여 폐흡충 피낭유충을 백서에 감염시켜 주별(주별)로 항혈청을 분리하고 등일 백서에서 복강 대식세포(homologous rat peritoneal macrophage)를 얻은 다음, 이들과 폐흡충 탈낭유충 및 숙주 조직 이행중인 유약충으로 effector system을 조작하여 폐흡충 유충에 대한 항체 의존성 대식세포 공격기전을 관찰하였다. 백서 복강 대식세포는 항혈청 출현 시에만 폐흡충 탈낭유충에 대한 세포부착 반응(cell adherence reaction)을 보였고 이 반응에는 보체가 관여하지 않았다. 폐흡충 감염 1주에서 8주까지의 백서 항혈청 중 2주째에 분리한 항쳔청 실험군에서 100%의 세포부착 반응을 나타내었고 사멸된 충체를 관찰할 수 있었으며, 세포부착 반응은 탈낭 유충기에서만 나타나는 반면 조직이행 유약충에서는 전혀 그 반응을 인정할 수 얼어 폐흡충 감염에 따른 항체의존성 숙주 세포 매개독성(antibodydependent cell·mediated cytotoxicity)은 발육단계 별 특이성을 보였다. 한편, 대식세포의 공격에 의하여 사멸된 폐흡충 탈낭유충의 형태학적 특징은 대식세포와 유충 사이에 형성되는 fuzzy material, tegumental syncytium 내의 미세구조의 변성 및 syncytium과 근육층을 연결하는 tubular tunnel의 형성 등이었다.

Larval Gnathostomes and Zoonotic Trematode Metacercariae in Fish from a Local Market in Yangon City, Myanmar

  • A survey was performed to investigate the infection status of zoonotic helminth larvae in fish from a local market of North Dagon District in Yangon City, Myanmar. A total of 486 fish in 13 species were collected 8 times from December 2015 to December 2019. All fish were transported under ice to a laboratory in Korea and examined for helminth larvae using artificial digestion method. Larval gnathostomes and metacercariae of more than 8 zoonotic trematode species, i.e., Opisthorchis viverrini, Haplorchis taichui, H. pumilio, H. yokogawai, Centrocestus spp., Stellantchasmus falcatus, Pygidiopsis cambodiensis, and Procerovum sp., were detected. Larval gnathostomes were found in 58 (16.0%) out of 362 fish of 6 species, with mean intensity of 2.8 per fish infected. Metacercariae of O. viverrini were detected in 10 (2.9%) out of 349 fish of 5 species, with mean intensity of 16.9 per fish infected. Metacercarial prevalences of 4 intestinal flukes, H. taichui, H. pumilio, H. yokogawai, and Centrocestus spp., were 16.8%, 26.0%, 12.5%, and 15.0% in the positive fish species, respectively, and mean metacercarial intensity was 63.3, 26.8, 86.2, and 8.7 per fish infected. Metacercariae of S. falcatus and P. cambodiensis were detected only from the mullet, Chelon macrolepis. Metacercariae of Procerovum sp. were found in Channa striata and Anabas testudineus. Collectively, it was confirmed that the fish were infected with gnathostome larvae and metacercariae of O. viverrini and intestinal flukes in Yangon City, Myanmar.

Trematode metacercariae and adults in cyprinoid fish from Khun Thale Swamp in Surat Thani province, Thailand

  • The present study aimed to determine the infection status of trematode metacercariae and adults in cyprinoid fish from the Khun Thale Swamp in Surat Thani, Southern Thailand, with epidemiologic and faunistic viewpoints. In 2020, 577 fish in 15 species were collected in the summer (February-April) and rainy (September-November) seasons. Fish were individually examined for trematode metacercariae in the whole body and adults in the gastrointestinal tract using a stereomicroscope. Three species of digenetic trematode metacercariae, i.e., Haplorchis taichui, Haplorchoides mehrai, and Centrocestus formosanus, were detected in the muscle, fin, and/or scale of fish. Two species of adult flukes, including Rohdella siamensis and Helostomatis cyprinorum, were collected in the intestines. The prevalence of overall trematode infections was 32.4% (187/577 fish), which was higher in the rainy season (41.4%; 118/285) than in the summer season (23.6%; 69/292). The metacercariae of H. taichui and H. mehrai were detected in 7 fish species each, and those of C. formosanus were found only in Rasbora toneri. The aspidogastrean trematode R. siamensis (adult) was detected in Babonymus gonionotus. A digenean species, H. cyprinorum (adult), was found in Labiobarbus siamensis and Osteochilus vittatus. The present study has first confirmed that the metacercariae of heterophyid flukes, including H. taichui, H. mehrai, and C. formosanus, and adults of R. siamensis (Aspidogastrea) and H. cyprinorum (Digenea) are infected in some species of the cyprinoid fish from the Khun Thale Swamp in Surat Thani, Thailand.

  • 검은머리물떼새의 서식지로 알려져 있는 서해안 일부 도서지역에서 채집한 참굴에서 참굴큰입흡 충 (Gwmophano쳐esseoi)의 피낭유충 감염상을 조사하였다. 조사 지역은 전라남도 신안군의 압 해도, 충청남도 서천군의 유부도, 전라북도 군산시 옥도면의 선유도, 무녀도, 장자도, 인천광역시 강화군 서도면의 주문도, 볼음도 등 7개 지역이었다. 이 흡충의 고도 유행지인 압해도에서는 1995 년 8월부터 1996년 10월까지 매 월별 피낭유충 감염상을 조사하였다. 총 14차례 중 5차례를 제외하고는 모두 100%의 감염률을 나타내었고 감염된 굴의 월별 평균 피낭유충 감염량은 761-2,077 개 범위이었다. 감염밀도의 계절별 변화양상은 나타나지 않았으나 굴의 무게가 증가합에 따라 감염량도 증가하는 양상을 보였다. 유부도산 굴은 조사한 총 63개에서 54개의 피낭유충이, 선유도산굴은 조사한 30개 중 25개 (83.3%)에서 1-66개 (평균 12.6개)의 피낭유충이 검출되었으며 무녀 도산굴은 조사한 50개 모두에서 3 162개 (평균 53.5개)의 피낭유충이 검출되었다. 주문도산 참굴 100개에서는 총 4개의 피낭유충이 검출되었으나 장자도산 55개와 볼음도산 50개의 굴에서는 피낭유충이 전혀 검출되지 않았다. 이상의 결과로 참굴큰입흡충이 전라남도 신안군 압해도에서는 지금도참굴을 중간숙주로 하여 농후하게 유행하고 있고, 전라북도 군산시 옥도면의 무녀도와 선유도, 충청남도 서천군 장항읍의 유부도, 인천광역시 강화군 서도면 주문도 등이 이 흡충의 새로운 유행지임을 확인하였다.

전남 해남군산 반염수어의 이형흡충류 피낭유충 감염상 (Infection status with the metacercariae of heterophyid flukes in the brackishwater fish from Haenam-gun, Chollanam-do, Korea)

  • 1991년 11월파 12월에 전남 해남군 해남읍 시장에서 구입한 전어(30마리). 농어(20 마리). 숭어(30마리). 문절망둑(30마리) 등에서 이형흡충류 피낭유충 감염상을 알아보고자 하였다. 전어에서는 조사한 30마리 중 20마리(66.7%)에서 긴이형흡충(Hetenphyopsis coninua)의 피낭유충이 총 53개가 검출되었다. 농어에서는 조사한 20마리 중 19마리(95.0%)에서 긴이형흡충 피낭유충이 총 327개(감염 어체 당 평균 17.2개)가 검출되었다. 숭어에서는 조사한 30마리 중 11 마리(36.7%)에서 유해이형흡충(Hetenphyes nocens)의 피낭유충이 총 567개(감염 어체당 평균 51.5개)가 검출되었고. 10마리(33.3%)에서 표주박이형흡충(Fygidiopsis summa)과 자루이형흡충 (Stictodora fuscatum)의 피낭유충이 각각 31개 및 349개가 검출되었다 문절망둑에서는 조사한 30마리 중 3마리에서 유해이형흡충의 피낭유충이 총 96개가 검출되었고, 8마리(26.7%)에서 긴이형흡충의 피낭유충이 총 101개(감염어체당 평균 12.6개)가 검출되었으며 3마리에서 자루이형흡충의 피낭유충이 9개가 검출되었다. 이상과 같이 전남 해남군산 반염수어들이 이형흡충류 피낭유충에 농후히 감염되어 있음을 확인하였다.

A survey of Heterophyes nocens and Pygidiopsis summa metacercariae in mullets and gobies along the coastal areas of the Republic of Korea

  • The infection status of mullets Mugil cephalus (n = 139) and gobies (n = 35) Acanthogobius fIavimanus with metacercariae of Heterophyes nocens and Pygidiopsis summa was examined in 11 western, southern, and eastern coastal areas of the Republic of Korea, using a digestion technique. Heterophyid metacercariae were highly prevalent in mullets from western and southern coastal areas; Shinan-gun (100% for H. nocens and 100% for P. summa), Muan-gun (93% and 100%), Buan-gun (42% and 75%), Seocheon-gun (73% and 53%), Ganghwa-gun (47% and 100%), Sacheon-shi (47% and 77%), and Gangjin-gun (50% and 70%, respectively). Only 1 (10%) of 10 mullets from an eastern coastal area, i.e., Donghae-shi, was positive for P. summa metacercariae. Metacercarial densities were the highest in the trunk of mullets for H. nocens and the gill for P. summa. Gobies from Muan-gun were positive for H. nocens (40%) and P. summa metacercariae (40%), and gobies from Seocheon-gun revealed H. nocens metacercariae (20%). The metacercarial density was remarkably higher in mullets than in gobies. The results revealed that H. nocens and P. summa metacercariae are prevalent in mullets and gobies from coastal areas of the Republic of Korea, and the prevalence and intensity of infection vary according to geographical locality.

Endemicity of Zoonotic Trematode Metacercariae in Fish from Deokcheon-gang (River) in Sancheong-gun, Gyeongsangnam-do, Republic of Korea

  • The endemicity of zoonotic trematode metacercariae (ZTM) was investigated with total 871 freshwater fishes (19 species) from Deokcheon-gang (a branch stream of Gyeongho-gang) in Sancheong-gun, Gyeongsangnam-do, Korea for 3 years (2018-2020). All fishes were examined with the artificial digestion method. The metacercariae of Clonorchis sinensis (CsMc) were detected in 233 (36.3%) out of 642 fish in 11 positive fish species (PFS), and their infection intensity was 27 per fish infected (PFI). Especially, in index fish, Puntungia herzi, of CsMc infection, prevalence was 64.2% and infection intensity was 37 PFI. Metagonimus spp. metacercariae (MsMc) were found in 760 (87.5%) out of 869 fish in 18 PFS and their infection intensity was 228 PFI. In sweet smelt, Plecoglossus altivelis, the prevalence of MsMc was 97.6% and their infection intensity was 3,570 PFI. Centrocestus armatus metacercariae were detected in 209 (29.4%) out of 710 fish in 8 PFS and their infection intensity was 1,361 PFI. Echinostoma spp. metacercariae were found in 293 (42.6%) out of 688 fish in 15 PFS and their infection intensity was 5 PFI. Metacercariae of Clinostomum complanatum and Metorchis orientalis were also detected in 2.7% and 21.2% fish in 4 PFS and their infection intensities were 3.1 and 3.4 PFI respectively. By the present study, it was confirmed that some species of ZTM including CsMc and MsMc are more or less prevalent in fishes from Deokcheon-gang in Sancheong-gun, Gyeongsangnam-do, Korea.
