Amino Acid Composition and Relationship of the Five Venerid Clams (Mollusca, Bivalvia) in Korea (한국산 백합과(科) 5종의 아미노산 조성 및 유연관계)

  • Amino acid composition and relationship of the commercially valuable five Korean Veneridae, Protothaca jedoensis, Ruditapes philippinarum, Meretrix lusoria, Saxidomus purpuratus and Cyclina sinensis were compared. The major amino acids of five species in Veveridae clams were ureanine, taurine, proline, glycine, alanine and arginine. A similarity of amino acid between C. sinensis and M. lusoria was highest (0.94) and lowest (0.52) for between C. sinensis and P. jedoensis, respectively.

  • 1. The auther studied the interrelationship between the sensibility of heart of fishes to neuro-hormones (epinephrine, norepinephrine and acetylcholinie) and 5-hydroxytryptamine and the differentiation of nervous system in the heart . Attempts were also made to develop the similarity and systematic relationship of inter-genus and inter-species. 2. Judging from the reactions to neurohormones, it is considered that inter-genus of locah (Misgurnus mizolepis GNTHER) and eel (Anguilla japonic TEMMINEK & SCHJEGEL) are more closely related than that of Crucian (Carassius carassius L. ). 3. As the sensibility of locach to 50 jhdroxytryptamine is more sensitive over 10, 000 times than that of a Mollusca, Meretrix lusoria RODING, it is very excellent material for the bioassay of 5-jydroxytryptamine.

  • The life history of a trematod fluck, Himasthla kusasigi was studied on the morphological characters of metacercariae, its infection rate in the clam and the contamination experiment to the herring gull, Larus crassiostris. The size of the metacercariae was $210{\sim}230{\times}220{\mu}m$, excysted metacercariae was $420{\times}160{\mu}m$ and it had 31 collar spines. The ventral sucker($112{\times}100{\mu}m$) was located just below the central part of the body. Esophagus was narrow and long, the intestine was branched in the upper part of the ventral sucker and extended to the hind part of the body. Excretory bladder without the infected part was located in the hind part of the body. The branched excretory tube was filled with the small granules and extended to the pharynx area. The infection rate of the metacercariae varied from place to place. In Naecho do and Puan the rates were $98.4\%$ and $95.9\%$ respectively. The infected number of Himasthla kusasigi per clam was positively proportional to the size of the clam. The adult fluckes developed from the metacercariae were obtained from the experimented herring gull. The metacercariae emerge from their cysts in the stomach of the herring gull, then they migrate it the intestine and 25 days after, most of them were found in the intestine. The size of the adult was $4.8{\sim}7.5{\times}0.22{\sim}0.24mm$, oral sucker; $68{\sim}120{\times}70{\sim}120{\mu}m$, pharynx: $80{\times}50{\mu}m$, ventral sucker: $630{\sim}680{\times}610{\sim}680{\mu}m$. The head collar $48{\sim}75{\times}10{\mu}m$ had 31 collar spines. The isolated trematod fluck from the samples was classified as a Himasthla kusasigi by the morphological characteristics of the adult flock and its metacercariae. We oberved that the second intermediate host of Himasthla kusasigi was hard clam, Meretrix lusoria, while its final host was herring gull, Larus crassiostris and Tringa ochropus.

Effect of Temperature on Induced Sexual Maturation of the Hard Clam, Meretrix petechiails (Lamarck) Broodstock (말백합, Meretrix petechiails (Lamarck) 어미의 사육수온에 따른 성 성숙 유도)

  • Artificial induction of maturation by heating of the hard clam, Meretrix petechiails (Lamarck) broodstock was investigated from 25 May to 30 June in 2008. We experimented maturation by keeping the breeding water temperature at 20, 25 and $30^{\circ}C$ and as the natural water temperature and found the following results. In case of the female, maturation was the quickest in the experiment group of $30^{\circ}C$ and the maturation tended to be slower as the water temperature was lower. The similar trend was displayed among the male. At experiment completion, maturation induction rate was found to be the highest at 46.0% in the experiment group of $30^{\circ}C$ followed by 35.0% in the experiment group of $15^{\circ}C$, 18.0% in the experiment group of natural water temperature and 12.0% in the experiment group of $20^{\circ}C$. During the period of the experiment, the survival rate was of 90% or more in all groups.

  • SDS-polyacrylamide gel electrophoresis for the proteins obtained from the plasma, Scapharca subcrenata and suapharca broughtonii, and for the proteins of muscles of several species in Bivalvia was performed. The protein patterns of plasma showed little difference between S. subcrenata and S. broughtonii in lower molecular weight weight proteins. However, the protein patterns of muscles of other species, which wre used in this study, were more shown in the lower molecular weights than the higher molecular weights in difference. Thus it is thought be an interesting fact. The protein band of blood corpuscles, 17,800 dalton, was not appeared in S. broughtonii, but this band was appeared in S. subcrenata. Henceforth this is the significantly important difference in these two species. But the protein patterns obtained from muscles of the species did not show a difference in a range of molecular weights between 10,000 and 100,000 daltons. Meanwhile, several protein bands obtained from Meretrix lusoria were similar to those of Mercenaria stimpsona. Hence, in this study, 6 protein bands which exist all species in Bivalvia and 4 characteristic protein bands in S. subcrenata and S. broughtoniionly, were investigated. And in four species of Eulamellibranchia, two protein bands in comon and the characteristic band of 23,000 dalton which is belong to Meretrix lusoria and Mercenaria stimdsona, were found. The molecular weights of the characteristic protein patterns, which are contained in each species, were measured and compared.

EFFECTS OF SUSPENDED SILT AND CLAY ON THE MORTALITY OF SOME SPECIES OF BIVALVES (이매패류의 폐사에 미치는 현탁부이의 영향)

  • Effects of suspended silt and clay on the survival of the rearing bivalves, Meretrix lusoria, Cyclina sinensis and Mactra veneriformis were studied. During sixty-five days of experimental cultures in different concentrations of suspended silt and clay and in different time of immersions, mortalities of the bivalves were checked every day, and the rates of shell movement and oxygen consumption were measured. In general, the higher the concentration of silt and clay and the longer the immersion time, the earlier the occurence, and the higher the rate of the mortalities of the experimental bivalves. M. veneriformis was subjected to earlier mortality than the others, and the period of $50\%$ mortality was the shortest of the three species (37 days in 1,000 ppm), and longer was that of C. sinensis(42 days). Mortality of M. lusoria occured latest and $50\%$ mortality was seen at the immersion time of 50-51 days. Particularly, M. lusoria showed no mortality in the sea water with 100 ppm of suspended silt and clay, and even in the concentrations of 500 or 1,000 ppm they were able to survive without mortality only if the immersion time was short (12-18 hours per day). M. veneriformis and C. sinensis were also able to survive without mortality when the immersion time was short (12 hours per day) in low concentration (100 ppm). Shell movements of experimental bivalves varied depending upon species. In the case of M. veneriformis the shell was opened continuously and C. sinensis opened their shells frequently, while M. lusoria maintained their shells closed in any experimental concentrations of sea water with suspended silt and clay. Total metabolic activity of M. veneriformis was found to be highest while that of M. lusoria showed the lowest. Little difference of oxygen consumption in excised gill tissue was shown between the control group and the experimental groups. Consequently, it may be stated that the mortality results from a immersion in sea water with high concentration of suspended silt and clay for long-time although the survival rates of the experimental bivalves depend also upon the species, physiological conditions, concentration of suspended silt and clay and immersion time. The survival percentage of bivalve y in relation to the time (day) of immersion X in sea water of suspended silt and clay was found to be: $$M.\;lusoria\;(1,000\;ppm):\;y=7.7\times10^9\;\chi^{-4.77}\;(500\;ppm):\;y=259\chi^{-0.26}$$ $$C.\;sinensis\;(1,000\;ppm):\;y=-21\chi+936\;(\chi<44),\;y=-0.65\;\chi+35\;(\chi>44)$$ $$(500\;ppm):\;y=4.4\times10^5\;\chi^{-2.27}$$ $$M.\;veneriformis\;(1,000\;ppm):\;y=-18\chi+716\;(\chi<39),\;y=-0.89\chi+39\;(\chi>39)$$

Current Status of Genome Research in Phylum Mollusks (연체동물 유전체 연구현황)

  • The availability of fast and inexpensive sequencing technology has enabled researchers around the world to conduct many genome sequencing and expressed sequence tag (EST) projects of diverse organisms. In recent years, whole genome projects have been undertaken to sequence ten species from the phylum Mollusca. These include Aplysia californica, Lottia gigantea, Crassostrea virginica, Spisula solidissima, Mytilus californianus, Biomphalaria glabrata, Crepidula fornicata, Elysia chlorotica, Lottia scutum and Radix balthica. Additionally, complete mitochondrial genomes of 91 mollusks have been reported. In Korea, EST projects have been conducted in nine mollusk species that include Nesiohelix samarangae, Pisidium (Neopisidium) coreanum, Physa acuta, Incilaria fruhstorferi, Meretrix lusoria, Ruditapes philippinarum, Nordotis gigantea, Crassostrea gigas and Laternula elliptica. Finally, the mitochondrial genome projects from the Pacific Oyster (Crassostrea gigas) and the rock shell (Thais clavigera) have been conducted and reported. However, no systemic mollusk genome project has so far been conducted in Korea. In this report, the current status and research trends in mollusk genome study in Korea will be discussed.

Investigation of Microplastics from Three Marine Organisms (해양생물 체내 잔류 미세플라스틱 조사)

  • Microplastic pollution in the marine ecosystem has been emerged as a global issue. In this study, we investigated the abundance of microplastics from clam Meretrix lusoria, blood arkshell Scapharca broughtonii, and warty sea squirt Styela clava obtained from a local market in Busan, Korea. The marine organisms were digested in 10% KOH, and were incubated at 40℃ and 150 rpm, for 7 days. The digest was filtered through standard sieve (5 mm, 1 mm, 300 ㎛, and 100 ㎛), and mciroplastics were identified using a light microscope and microFT-IR. The abundance of microplastics of clam, blood arkshell, warty sea squirt was 0.08 items/g, 0.05 items/g, and 0.12 items/g, respectively. The predominant microplastic size was in the range of 100-300 ㎛, occupying 48%, and the predominant type was fiber. The composition of microplastics was mostly rayon, semi-synthetic cellulosic material and polyester, which are main component of fabric and textile. We strongly believe that this preliminary work may provide useful information for the establishment of the standardized analysis method of microplastics ingested by marine organisms.

Change in Community Structure of Shellfish in the Reclaimed Saemangeum Area (새만금 간척사업에 따른 갯벌 패류의 군집구조 변화)

  • Species composition and distribution patterns of shellfish were investigated in Eoeun and Geojeon tidal flat located in the Saemangeum area on the west coast of Korea from May to October 2000. Nineteen species of shellfish were collected in Geojeon tidal flat. The samples in number of individuals included Umbonium thomasi $(90.0\%)$ and Mactra veneriformis $(5.0\%).$ In Eoeun tidal flat, ten species of shellfish were collected and the dominant species in number of individuals were Potamocorbula amurensis $(55.0\%)$ and U. thomasi $(18.6\%).$ These results were compared with previous studies conducted before the beginning of reclamation in Saemangeum. Compared with the results from 1988, a change In species composition was observed. Laternula flexuosa and Nuttallia olivacea appeared, while Mactra chinensis and Coelomactra antiquata disappeared in the deposition area in Geojeon tidal flat. In the erosion area of Eoeun tidal flat, M. veneriformis and Meretrix lusoria appeared, while Cyclina sinensis disappeared. Based on a cluster analysis, the shellfish community in Eoeun tidal flat was classified into three station groups based on sediment types. Geojeon tidal flat was also classified into three station groups. The distribution of shellfish in the Saemangeum area was closely related to the sediment types.

The Action of Extract of Aconitum koreanum R. Raymond on Isolated Clam Heart (우리나라 백부자의 적출 조개 심장운동에 대한 작용)

  • Korean aconitum (Aconitum koreanum R. Raymond) as one of the botanical crude drugs which pertain to helleboraceae has been extensively applied in Chinese medicine during the past decades. It has been particularly used in immortal tonic among the folk remedies in China, however, its general uses comprehend diuresis, cardiotonic, analgesia, neuralgia, gout and, furthermore, even neoplastic effect. The components of aconitum have been acknowledged as aconitine, mesaconitine, hypaconitine, aconine and so on. The main ingredient, aconitine has the advantage of causing the atrial fibrillation, but, its pharmacological research has not been fully elucidated. Although there are many reports with regard to the pharmacological effects on the motility of several animal hearts, their conclusions have not been regretfully coincided yet. The authors hereby paid attention to this point of view and made experiment to examine the relationship between the alcohol extract of Korean aconitum and the motility of the isolated clam heart, making the use of several drugs related to the heart such as serotonin, acetylcholine, pilocarpine, physostigmine, barium chloride, procaine and quinidine. The cardiac movement of the isolated clam (Meretrix lusoria) heart in the standard sea water solution was recorded with the electric kymograph according to the Magnus method. The results of the experiment are as follows. 1. The motility of the isolated clam heart represents the tendency of gradual inhibition in proportion to the concentration of AK-A $10^{-4}$, $5{\times}10^{-4}$, and $10^{-3}$. 2. The cardiac inhibitory effect of AK-A $10^{-3}$ antagonizes the motility of the isolated clam heart pretreated with serotonin $10^{-6}$. 3. The cardiac inhibitory effect of AK-A $10^{-3}$ antagonizes the systolic state appealed by barium chloride $10^{-3}$. 4. The systolic state caused by quinidine $10^{-4}$ is not inhibited by AK-A $10^{-3}$.
