Contact-Independent Cell Death of Human Microglial Cells due to Pathogenic Naegleria fowleri Trophozoites

  • Free-living Naegleria fowleri leads to a fatal infection known as primary amebic meningoencephalitis in humans. Previously, the target cell death could be induced by phagocytic activity of N. fowleri as a contact-dependent mechanism. However, in this study we investigated the target cell death under a non-contact system using a tissue-culture insert. The human microglial cells, U87MG cells, co-cultured with N. fowleri trophozoites for 30 min in a non-contact system showed morphological changes such as the cell membrane destruction and a reduction in the number. By fluorescence-activated cell sorter (FACS) analysis, U87MG cells co-cultured with N. fowleri trophozoites in a non-contact system showed a significant increase of apoptotic cells (16%) in comparison with that of the control or N. fowleri lysate. When U87MG cells were co-cultured with N. fowleri trophozoites in a non-contact system for 30 min, 2 hr, and 4 hr, the cytotoxicity of amebae against target cells was 40.5, 44.2, and 45.6%, respectively. By contrast, the cytotoxicity of non-pathogenic N. gruberi trophozoites was 10.2, 12.4, and 13.2%, respectively. These results suggest that the molecules released from N. fowleri in a contact-independent manner as well as phagocytosis in a contact-dependent manner may induce the host cell death.

국내 분리 오제스키병 바이러스 접종자돈의 병리학적 관찰 (Pathological observation on the piglets experimentally infected with Aujeszky′s disease virus isolated in Korea)

  • Pathological studies were performed on the five piglets experimentally infected with Aujeszky's disease virus(pseudorabies), NYJ isolate, isolated from the naturally infected pigs in Korea: two piglets were inoculated intramuscularly, two piglets intranasally, and one piglet subcutaneously at the dose of 1$m\ell$ per animal with the 105.5 $TCID_50$/0.1ml titer. Clinical signs included dyspnea, high fever(>$41^{\circ}C$), anorexia, vomiting, diarrhea or constipation, ataxia, circling movement, posterior paralysis, intermittent convulsion, and coma followed by death although some variations by age and inoculated routes were observed. Gross features included multiple necrotic foci in the liver, congestion and hemorrhage in the lymph nodes and spleen, petechial hemorrhage in the kidney, hemorrhagic pneumonia, marked meningeal congestion, severe sub meningeal hemorrhage in the spinal cord, excessive cerebrospinal fluid retention, and muscular necrosis at the inoculated area. Microscopically, non suppurative meningoencephalitis with gliosis and perivascular cuffing in CNS, ganglioneuritis, necrohemorrhagic splenitis, necrotic hepatitis, tonsillitis and rhinitis, hemorrhagic or interstitial pneumonia, and non-suppurative myositis in the injected area were observed. Eosinophilic intranuclear inclusion bodies were found in a variety of tissues the including the liver, kidney, adrenal gland, spleen, lymph nodes, tonsil, and lung. Ultrastructurally, virus particles were confirmed in nucleus and cytoplasms of pneumocytes around the necrotic areas.

Naegleria fowleri Lysate Induces Strong Cytopathic Effects and Pro-inflammatory Cytokine Release in Rat Microglial Cells

  • Naegleria fowleri, a ubiquitous free-living ameba, causes fatal primary amebic meningoencephalitis in humans. N. fowleri trophozoites are known to induce cytopathic changes upon contact with microglial cells, including necrotic and apoptotic cell death and pro-inflammatory cytokine release. In this study, we treated rat microglial cells with amebic lysate to probe contact-independent mechanisms for cytotoxicity, determining through a combination of light microscopy and scanning and transmission electron microscopy whether N. fowleri lysate could effect on both necrosis and apoptosis on microglia in a time- as well as dose-dependent fashion. A $^{51}Cr$ release assay demonstrated pronounced lysate induction of cytotoxicity (71.5%) toward microglial cells by 24 hr after its addition to cultures. In an assay of pro-inflammatory cytokine release, microglial cells treated with N. fowleri lysate produced TNF-${\alpha}$, IL-6, and IL-$1{\beta}$, though generation of the former 2 cytokines was reduced with time, and that of the last increased throughout the experimental period. In summary, N. fowleri lysate exerted strong cytopathic effects on microglial cells, and elicited pro-inflammatory cytokine release as a primary immune response.

Characterizations of four freshwater amoebae including opportunistic pathogens newly recorded in the Republic of Korea

  • Background: Free-living amoebae (FLA) are widely distributed in freshwater, seawater, soil, and extreme environments, and play a critical role as feeders on diverse preys in the ecosystem. Also, some FLA can become opportunistic pathogens in animals including humans. The taxa Amoebozoa and Heterolobosea are important amoeboid groups associated with human pathogens. However, the identification and habitat of amoebae belonging to Amoebozoa and Heterolobosea remain poorly reported in the Republic of Korea. This study highlights the first record for identification and source of four amoebae including putative pathogens in the Republic of Korea. Results: In the present study, four previously reported FLA were isolated from freshwaters in Sangju Gonggeomji Reservoir (strain GO001), one of the largest reservoirs during the Joseon Dynasty period, and along the Nakdong River, the largest river in the Republic of Korea (strains NR5-2, NR12-1, and NR14-1) for the first time. Microscopic observations and 18S rDNA phylogenetic trees revealed the four isolated strains to be Acanthamoeba polyphaga (strains NR5-2 and NR12-1), Tetramitus waccamawensis (strain GO001), and Naegleria australiensis (strain NR14-1). Strains NR5-2 and NR12-1 might be the same species and belonged to the morphological Group 2 and the T4 genotype of Acanthamoeba. Strain GO001 formed a clade with T. waccamawensis in 18S rDNA phylogeny, and showed morphological characteristics similar to previously recorded strains, although the species' flagellate form was not observed. Strain NR14-1 had the typical morphology of Naegleria and formed a strongly supported clade with previously recorded strains of N. australiensis in phylogenetic analysis of 18S rDNA sequences. Conclusions: On the bases of morphological and molecular analyses, four strains of FLA were newly observed and classified in the Republic of Korea. Three strains belonging to the two species (A. polyphaga and N. australiensis) isolated from the Nakdong River have the potential to act as opportunistic pathogens that can cause fatal diseases (i.e. granulomatous amoebic encephalitis, Acanthamoeba Keratitis, and meningoencephalitis) in animals including humans. The Nakdong River in the Republic of Korea may provide a habitat for potentially pathogenic amoebae, but additional research is required to confirm the true pathogenicity of these FLA now known in the Republic of Korea.

The anti-amoebic activity of Pinus densiflora leaf extract against the brain-eating amoeba Naegleria fowleri

  • Naegleria fowleri invades the brain and causes a fatal primary amoebic meningoencephalitis (PAM). Despite its high mortality rate of approximately 97%, an effective therapeutic drug for PAM has not been developed. Approaches with miltefosine, amphotericin B, and other antimicrobials have been clinically attempted to treat PAM, but their therapeutic efficacy remains unclear. The development of an effective and safe therapeutic drug for PAM is urgently needed. In this study, we investigated the anti-amoebic activity of Pinus densiflora leaf extract (PLE) against N. fowleri. PLE induced significant morphological changes in N. fowleri trophozoites, resulting in the death of the amoeba. The IC50 of PLE on N. fowleri was 62.3±0.95 ㎍/ml. Alternatively, PLE did not significantly affect the viability of the rat glial cell line C6. Transcriptome analysis revealed differentially expressed genes (DEGs) between PLE-treated and non-treated amoebae. A total of 5,846 DEGs were identified, of which 2,189 were upregulated, and 3,657 were downregulated in the PLE-treated amoebae. The DEGs were categorized into biological process (1,742 genes), cellular component (1,237 genes), and molecular function (846 genes) based on the gene ontology analysis, indicating that PLE may have dramatically altered the biological and cellular functions of the amoeba and contributed to their death. These results suggest that PLE has anti-N. fowleri activity and may be considered as a potential candidate for the development of therapeutic drugs for PAM. It may also be used as a supplement compound to enhance the therapeutic efficacy of drugs currently used to treat PAM.

Acanthamoebaculbertsoni와 A. royreba의 가수분해 효소 활성도의 비교 연구 (A Comparative Study on Hydrolase Activities in Acanthamoeba culbeytconi and A. roureba)

  • 아메바의 cytolytic process가 주로 가수분해 효소 환성과 관계 깊다는 연구들이 70년대 후반부터 병원성 Entamoeba sp.와 Naegleria sp.에서 보고되었지만 Acanthamoeba sp.에서는 가수분해 효소 환성에 관한 연구는 찾아볼 수 없다. 본 연구에서는 병원성이 없는 것으로 알려진 Acanthamoeba royreba를 대조로 하여 병원성이 있는 A. culbertsonl이 가수분해 효소 활성도를 측정하고 이들을 비교하고자 하였다. A. culbertsoni를 마우스에 감염시켰더니 15일내에 마우스 모두가 사망하였으며, 평균 생존기간은 8.5일이었고, 아베바성 수막뇌염으로 사망했음을 알 수 있었다. 반면에 A. royreba를 감염시킨 마우스에서는 수막뇌염이 발생되지 않았다. CGV배지에서 두 종(種) 아메바를 배양하여 아메바 추출물 및 배양액에서 가수분해 효소의 환성을 측정한 결과 A. rcyreba에 비해 A. culbertsoni 세포의 추출물 및 배양액에서 현저히 활성도가 높은 가수분해 효소는 acid phosphatase, p-N-acetyl galactosaminidase, β-N-acetyl glucosaminidase, α-mannosidase, neutral proteinase, acid proteinase였다. 표적 세포인 CHO 세포에 혼합한 A. culbertsoni는 대조군에 비해 강한 세포독성을 나타내었고 CHO 세포와의 혼합후 48시간 후 80%, 72시간후 95% 이상의 CHO 세포가 사멸하는 것을 알 수 있었다. 반면에 A. royreba를 혼합하였을 때는 별다른 세포독성을 나타나지 않았다. CHO 세포에 혼합한 A. tulbgrtsoni이 가수분해 효소 활성도를 경과 시간별로 측정하였던 바 CGV 배지에서 배 양시간이 지남에 따라 아메바내보다 그 배양액에서 더 높은 효소 활성도를 나타내었던 acid phosphatase, β-N·acetyl galactosaminidase, β-N·acetyl glucosaminidase, α-mannosidase, acid proteinase는 혼합 120시간 동안 아메바내에서 더 높은 효소 활성도를 나타내었으며, neutral proteinase의 활성도는 경과시간이 지남에 따라 아메바내에서는 일정하였지만 그 배양액인 EBSS용액내에서는 점점 높아짐을 알 수 있었다. 이상의 성적으로 보아 마우스에서 수막뇌염을 일으키는 A. cuzbertsoni는 비 병원성인 A. reyreba와 비교할 때 여러 종류 가수분해 효소들의 활성도에 현저한 차이를 나타내고 있음을 알 수 있었다. 또한 표적세포인 CHO세포에 대한 A. cuzbertsoni의 세포독성은 강함을 알 수 있었고, 이 때 몇 종류 가수분해 효소들의 환성도도 높게 나타났으며, 이러한 소견이 A. culbertsoni의 병원성과 관련이 있을 것으로 추측되었다.

자유생활아메바 Naegleria fowleri의 계대감염에 의한 병원성의 변화에 관한 연구 (Changes in the pathogenicity of Naegleria fowleri by serial brain passage in mice)

  • 장기간 시험관내에서 무균적으로 배양하여 병원성이 약화된 자유생활아메바 naegleria fowleri를 마우스에 연속적으로 감염시켜 그 병원성의 증감여부를 관찰하고자 하였다. 체증 18∼22gm의 백색 마우스를 secobarbital로 마취시키고 오른쪽 비강에 Naegleria fowleri를 떨어뜨려 감염시켰다. 시험관내에서 7년이상 CGVS배지에서 계대배양된 0주와 마우스에 감염시켜 뇌조직을 2번 통과시킨 2-1주의 병원성을 비교하였다. 행동둔화, 자극에 민감, 회전운동, 사지마비등 여ㅓ 증상이 나타났고, 뇌의 오른쪽 앞쪽 부위에 심한 염증 및 괴사를 발견하였다. 이러한 증상과 병소는 0주 감염군보다 2-1감염군에서 보다 빨리, 보다 심하게 발생되었음을 관찰하였다. 감염 7일 후부터 감염된 마우스가 사망하기 시작하였으며 2-1주 감염군에서 0주 감염군 보다 생존기간이 짧았으며 감염 13일 후부터 사망한 예에 있어서는 육안적으로 뇌에서 병소가 없었고 폐장에서 염증이 심하였음을 관찰하였다. 마우스에 비강을 Naegleria fowleri를 감염시켜 전형적인 원발성 아메바성 뇌수막염을 발생시킬 수 있었다. 장기간 시험관에서 무균적으로 계대배양된 Naegleria fowleri는 병원성이 약화되어 있었고 이 아메바를 마우스에 연속적으로 감염시켜 뇌조직을 통과시킴으로 다시 병원성이 증강됨을 관찰하였다.

Enterovirus 71-associated hand, foot and mouth diseases with neurologic symptoms, a university hospital experience in Korea, 2009

  • Purpose: Hand-foot-mouth disease (HFMD) is a common viral illness in children, which is usually mild and self-limiting. However, in recent epidemics of HFMD in Asia, enterovirus 71 (EV71) has been recognized as a causative agent with severe neurological symptoms with or without cardiopulmonary involvement. HFMD was epidemic in Korea in the spring of 2009. Severe cases with complications including death have been reported. The clinical characteristics in children with neurologic manifestations of EV71 were studied in Ewha Womans University Mokdong Hospital. Methods: Examinations for EV71 were performed from the stools, respiratory secretion or CSF of children who presented neurologic symptoms associated with HFMD by realtime PCR. Clinical and radiologic data of the patients were collected and analyzed. Results: EV71 was isolated from the stool of 16 patients but not from respiratory secretion or CSF. Among the 16 patients, meningitis (n=10) was the most common manifestation, followed by Guillain-Barre syndrome (n=3), meningoencephalitis (n=2), poliomyelitis-like paralytic disease (n=1), and myoclonus (n=1). Gene analysis showed that most of them were caused by EV71 subgenotype C4a, which was prevalent in China in 2008. Conclusion: Because EV71 causes severe complications and death in children, a surveillance system to predict upcoming outbreaks should be established and maintained and adequate public health measures are needed to control disease.

Naegleyiu fowleri에 대한 단세포군 항체의 생산과 그 특성에 관한 연구 (The production and characterization of anti-Naegleria fowleri monoclonal antibodies)

  • 자유생활 아메바인 Naegleria fowleri는 비강을 통해 인체에 들어와 출혈성 수막뇌염을 일으켜 감염된 사람의 대부분을 일주일 이내에 사망하게 하는 원충이다. 이 실험에서는 N. fowleri에 특이한 단세포군 항체를 만들어 이의 특성 및 그 이용 가능성을 알아보고자 하였다. 7 종류의 단세포군 항체(Nf 1, Nf 2, NE 256, Nf 279, Nf 27, Nf 154, Nf 137)를 제조하였는데 각각의 isotope은 IgGl이 두 종류(Nf 27, Nf 154), IgG3가 1 종류(Nf 137), IgA가 4 종류(Nf 1, Nf 2, Nf 256, Nf 279)이었다. 이들 항체를 N. fwoleri와 다른 종류의 아메바를 항원으로 하여 효소표식 면역검사법을 시행하여 Nf 1 및 Nf 256 항체를 제외한 5종류의 항체가 N. fowleri에 특이한 항체임을 확인하였다. 또한 간접 형광항체법을 통하여 NF 256 항체를 제외한 6 종류의 단세포군 항체가 세포막의 일부에 결합하였음을 관찰하였다. Nf 154 항체를 이용한 immunoperoxidase 염색에서도 세포막의 반응을 관찰하였다. 단세포를 항체가 N. fowleri를 응집시키는지 알아보고자 항체를 N. fowleri 영양형과 반응시켰더니 Nf 27 항 체를 제외한 5 종류의 단세포를 항체에 의해 응집이 일어남을 알 수 있었으며 응집된 덩 양형을 보체로 처리한 후 CGVS배지에서 배양하였더니 영양형의 증식이 억제되었다. 또한 2종류의 단세포군 항체(Nf 2, Nf 154)는 조직 세포(Chinese hamster ovary cell: CHO)에 대한 N.fowleri의 세포독성을 저하시켰다. 단세포군 항채와 반응하는 항원의 분자량을 알아보고자 EITB(Enzyme-linked immunoelectrotransfer blot)를 시행하였는데 Nf 279 항체는 25 kDa 및 28 kDa, Nf 154 항체는 43 kDa, N( 137 항체는 29 kDa에 해당되는 분획에서 항원항체반응을 나타내었다. 이상의 성적을 종합하면 N. fowleri에 대한 7종류의 IgG., IgG3 및 IgA 단세포군 항체를 생산하였다. 그중 Nf 256 항체를 제외한 6 종류는 N. fowleri이 세포막 성분중 28 kDa-43 kDa의 항원과 반응하는 특이한 항체임을 관찰할 수 있었다. 또한 이 단세포를 항체들은 영양형을 응집시키며, 시험관 내에서의 증식을 억제시키고 CHO 세포에 대한 N. fowleri의 세포 독성을 저하시키는 성질을 가지고 있음을 알 수 있었다.
