This study was conducted to evaluate the effect of PAC(Powder Activated Carbon) retention time on stable operation of high concentration powered activated carbon(HCPAC-MBR) in the treatment of secondary domestic wastewater. The pilot scale HCPAC-MBR system was operated at two different SRTs, 25 days and 100 days. The main drawback of HCPAC-MBR system was the rapid increase of trans-membrane pressure. The increase rate of trans-membrane pressure was proportional to SRT value at constant flux. This result seemed to be caused by reduced amount of EPS adsorbed on the PAC in the reactor by decreasing the SRT of the PAC. The particle size of the PAC was also influenced by SRT. The PAC size was decreased as SRT was increased. The change of particle size could be one reason for the change of trans-membrane pressure. The pore volume in the cake-layer formed on the membrane surface became to be increased by reducing SRT, because the cake-layer was highly composed of the PAC. Therefore, increased pore volume might play a role to reduce the trans-membrane pressure. The removal rate of E260 and TOC was also inversely proportional to SRT value.
This study evaluated the performance of a thermophilic two-phase anaerobic digestion (TTPAD) coupled with membrane process treating garbage leachate. The pilot-scale treatment system is consisted of thermophilic acidogenic reactor (TAR) and thermophilic methanogenic reactor (TMR) coupled with an ultrafiltration (UF) membrane unit. The hydraulic retention time of TAR and TMR were 4 and 20 days, respectively. Effluent TCOD and SCOD of the TTPAD were $25\;{\pm}\;6\;and\;12\;{\pm}\;3$ g/L, respectively, and the corresponding TCOD and SCOD removal efficiencies were 77% and 81%, respectively. Propionate was major acids as 75% in the effluent. Scum formation was not observed in TTPAD, which might be resulted from complete lipid degradation. However, TTPAD was appeared to be sensitive to free ammonia toxicity. The UF membrane was operated with constant pressure filtration at average TMP 1.3 atm. Permeate flux had a range of 15-30 $L/m^2/hr$. With UF membrane, TCOD removal increased from 77% to 93%, and this SS free effluent would be beneficial to subsequent processes such as ammonia stripping.

Techniques for protein refolding from inclusion body are discussed in view of its engineering application to large scale protein purification. Among the techniques, dilution and dialysis are mainly utilized due to simple operation. Membrane reactor, gel filtration chromatography, and continuous tank operation are emerging tools for their process-scale possibility in refolding. Reaction engineering approaches could be used to analyze the kinetic behaviour in the process scale refolding reactor. The kinetic analysis is helpful in the optimization of refolding yield in the refolding reactor.
메조포러스 공극구조를 갖는 광촉매 멤브레인은 다양한 환경기술에 적용될 수 있다. 본 연구에서는 $TiO_2$ 층을 형성시킨 광촉매 반응기용 세라믹 멤브레인을 개발하고 이를 염색용액 처리에 적용하였다. 높은 공극률과 균질성을 지닌 $TiO_2$ 광촉매층을 그라프트 공중합체를 사용하여 제조하였다. 멤브레인은 광촉매 반응기와 멤브레인 여과를 결합시킨 하이브리드 광촉매 반응기에 성공적으로 적용하였다. 실험결과 정렬된 구조의 $TiO_2$ 층이 $Al_2O_3$ 지지체에 형성되었다. $TiO_2$ 층 형성 후 제조된 세라믹 분리막의 순수 투과도는 형성된 광촉매 층 저항으로 감소하였다. 정렬된 구조의 $TiO_2$ 층은 UV 결합 시 5시간 안에 완벽한 염색용액 분해를 달성시킬 수 있었다. 광촉매 멤브레인의 염색용액 분해는 Langmuir-Hinshelwood 흡착 모델로 잘 설명할 수 있었다. 또한 $TiO_2$ 층이 고정화된 세라믹 멤브레인의 model Congo Red에 대한 1차 속도상수는 $Al_2O_3$ 지지체 단독인 경우에 비해 약 6배 정도 큰 값을 나타내었다(0.0081 vs. $0.0013min^{-1}$).
불완전 분리의 실질적인 분리투과 특성을 근거로 하는 모사모델을 이용하여 투과증발 막촉진 에스터화 반응의 실질적인 거동을 연구하였다. 막의 분리효율, 막의 물제거 능력을 모델에 포함시킴으로써 반응모사를 통하여 이들이 막촉진반응에 어떻게 영향을 끼치는지를 체계적으로 관찰할 수 있었다. 막을 통한 비완전분리가 일어날 경우에 초기 반응물 몰비율이 1이 아닐 때 반응 혼합물중 소량으로 존재하는 반응성분이 투과되거나, 혹은 초기 반응물 몰비율이 1인 반응 혼합물중 어느 반응성분이 투과되든지 반응이 완결될 수 없었다. 생성물인 에스터가 물과 함께 일부 투과될 경우 정방향 반응속도를 증가시켜 시간에 따른 반응 전환률을 증가시킨다. 막을 통한 불완전 분리시 막을 통한 투과로 인하여 발생하는 반응혼합물의 부피감소로 인하여 반응성분 뿐 아니라 생성성분의 농도가 증가되는데 이들 농도증가가 반응속도에 상반된 영향을 끼친다; 반응물 농축은 반응속도를 증가시키나 생성물 농축은 역방향의 반응을 촉진시킴으로서 반응속도를 감소시킨다. 반응초기에는 반응물 농축효과가 우세하나 반응이 진행됨에 따라 생성물 농축효과가 우세하여 반응속도를 저하시킴이 모사결과 관찰되었다.
간단한 조작으로 시안화물을 신속하게 계측할 수 있는 센서의 개발을 목적으로 하여 미생물과 산소전극을 이용한 시안센서의 개발을 연구하였다. 산소전극에 S. cerevisiae를 흡착 고정화한 막을 부착하여 막형 시안센서를 제작하고 시료용액중의 시 안이온농도를 O~1.00ppm이 되게 시안을 첨가하면 서 전류값 변화를 살펴본 결과 시안농도 0.10~1.00ppm의 범위에서 응답이 확인되었으며 고정화 직후의 호흡활성은 약 하루정도 유지되었다. S. ceremSlae를 고정화한 키토-펼 HP-5020을 사용하여 반응기형 시안센서를 제작하고 막형 시안센서와 동일하게 시안이온농도 O~1.00ppm 용액의 전 류값 변화를 살펴본 결과 0.10~ 1.00ppm의 범위에서 응답이 확인되었으며 고정화 직후 90%의 호흡활 성이 16일간 유지되었다. 반응기형 시안샌서의 반응기 크기의 영향을 검토 한 결과, 반응기의 최적 체적이 존재한다는 사실을 알 수 있었다.
연속식 2단계 막(MWCO 10,000, MWCO 5,000)반응기를 이용하여 어피젤라틴 가수분해물 제조를 위한 최적 가수분해조건과 막반응기 장치에서 효소활성, 효소안정성에 미치는 인자 및 가수분해물의 생성량에 대하여 검토하였다. pH-drop법으로 젤라틴 분해에 대한 효소활성은 pronase E가 가장 높았고, 다음이 trypsin이었다. 1단계 막반응기에서 trypsin으로 어피젤라틴의 최적 가수분해조건은 반응온도 $55^{\circ}C$, pH 9.0, 효소농도 0.1 mg/ml, 기질 대 효소비 100(w/w), 반응부피 600 ml, 유출속도 6.14 ml/min였으며, 이때 가수분해도는 79%였다. 2단계에서 1단계 젤라틴 가수분해물을 pronase E로 분해할 경우는 반응온도 $50^{\circ}C$, pH 8.0, 효소농도 0.3 mg/ml, 기질 대 효소비 33(w/w)였으며 그 이외의 조건은 1단계와 동일하였고, 이때의 가수분해도는 80% 이상이였다. 막을 통한 효소의 누출량은 1단계에서 전체 효소량 중 작동시간 5시간에서 11.5%였으나 5시간 이후에는 거의 누출되지 않았으며, 2단계에서는 작동시간 4시간에서 9.0%가 누출되었다. 막반응기의 기계적인 전단웅력에 의한 효소활성저해는 1단계 및 2단계에서는 작동시간 3시간까지 34% 및 18%가 각각 감소되었으며, 이때 막을 제거시킨 반응기에서 효소활성 감소는 23% 및 10%가 각각 저해되었다. 그러나 효소누출과 가수분해물의 생산량 사이에는 명백한 상호작용은 나타나지 않았다. 1단계 및 2단계 막반응기의 각 최적조건하에서 부피대체율 8배까지의 가수분해물의 생산량은 각각 334 mg 및 250 mg으로 2단계 보다 1단계의 효소 mg당 생산량이 약 1.3배 높았다.
본 연구에서는 막결합형 연속회분석공정(MSBR)을 이용한 크루즈선 오 폐수처리장치 개발을 위하여 기초 실험을 수행하였다. MSBR 공정을 평가하기 위하여 SBR, MBR, MSBR system을 Lab scale로 제작하여 처리 효율과 크루즈선이라는 특수환경과의 접목성을 비교 운영하였으며, 검토 결과 MSBR 공정은 처리효율과 장치 운영면에서 선박이라는 특수한 환경에 가장 적합한 공정으로 평가되었다. MSBR의 system의 BOD 제거 효율은 99%, COD 제거 효율은 98%, SS 제거 효율은 99%로 나타내었으며 유기물, 영양염류, 병원성 미생물 처리에 있어 모두 안정적인 효율을 나타내며 IMO의 규제 기준을 모두 만족하였다.
To compare the antioxidative activities of fish skin and bovine skin gelatin hydrolysate, gelatin hydrolysates from Alaska pollack and bovine skin were prepared by various enzymatic hydrolysis methods (1st step, Alcalase; 2nd step, pronase E; 3rd step, collagenase) using a continuous three-step membrane reactor. The molecular weight distributions of the 1st, 2nd and 3rd step hydrolysates were 7∼10 kDa, 2∼5 kDa and 0.7∼0.9 kDa, respectively. The antioxidative activity of fish skin gelatin hydrolysate was stronger than that of bovine skin gelatin hydrolysate, and in particular, both of 2nd step hydrolysates showed more antioxidative activity than hydrolysates of any other step. The optimum antioxidative activity concentration of the 2nd step hydeolysates of fish and boving skin were 1% (w/w) in a linoleic acid water-alcohol emulsion. In cultured cells exposed to t-butyl hydroperoxide (t-BHP), the 2nd step hydrolysate of fish skin gelatin delayed cell death most. These results suggest that the antioxidative activity of fish skin gelatin hydrolysate is higher than that of bovine skin gelatin hydrolysate because of their different amino acid contents.
