Nephrotoxicity Assessment of Cephaloridine using Rat Renal Proximal Tubule Suspension (랫트의 신장 근위곡세뇨관 현탁액을 이용한 Cephaloridine의 신장독성 평가)

  • Rat renal proximal tubule suspension was prepared from adult male Sprague Dawley rat (250-300g) by mechanical (non-enzymatical) method and evaluated as a pontential model for mechanistic studies and early screening of nephrotoxicity, using anionic antibiotics (cephaloridine). Cephaloridine (CPL) produced an increase in LDH release into media. This release results from decrease a proximal tubule cell viability and subsequently increase the permeability of cell viability and subsequently increase the permeability of cell membrane. Since loss of intracellular potassium and ATP into media is the sign of disruption of cell membrane, especially basolateral membrane (BLM), CPL induced proximal tubule cell compromise also appear be associated with BLM, maybe $Na^+-K^+$ ATPase. Also seen was significant depression in brush border membrane (BBM) ALP activity and no significantly increase in BBM GGT activities. The inhibition of typical anion, PAH accumulation (especially, CPL 5 mM) and cation, TEA (especially, 4hours incubation) were seen dose dependently. This is because of CPL accumulation in renal proximal tubule and increase of cytotoxicity.

  • The effects of kanendomycin on the potassium permeability in the rabbit erythrocyte membrane are investigated and the results are summarized as follows. 1. Kanendomycin causes the efflux of $K^+\;and\;influx\;of\;Na^+$ across the rabbit erythrocyte membrane. 2. Osmotic resistance of kanendomycin treated erythrocytes is diminished. This diminution of osmotic resistance is more pronounced by increasing concentration of kanendomycin and longer incubation time. But osmotic resistance is rather increased in the presence of lower concentration of kanendomycin. 3. Cysteine and glutathione have little influence on $K^+$ efflux induced by kanendomycin. 4. EDTA has no effect on the increase in $K^+$ outflux by kanendomycin while PCMB augments slightly on $K^+$ outflux. 5. Kanendomycin inhibits $Ca^{++}$ binding competitively to rabbit erythrocyte membrane fragments.

  • The purpose of this paper is the evaluation of a permeability and blood compatibility for silicone/polypropylene combined membrane. Despite the overall good performances of polypropylene membrane, its long-lasting usage for artificial lung has been limited by serum leakage. In order to overcome this problem, we have newly fabricated polydimethylsiloxane(silicone)/polypropylene combined membrane(SPCM). SPCM has been proved to be serum leakage free in hours experimental. It has shown good long-lasting gas transfer and durability features.

  • In this study, the simulation was performed for the CO removal process using a composite membrane in DME production. The composite membrane, PEI-PDMS (polyetherimide- polydimethyl siloxane) manufactured by Airrane Co. Ltd., was used in the modeling through a commercial simulation design program, PRO/II with PROVISION 9.2 by Invensys. To simulate the process, the permeability constants of each of the pure component from Airrane Co. Ltd. were determined by regression analysis from the experimental data. The required separation membrane area and utility cost in the CO removal process were obtained using a chemical process simulator and composite membrane with a compatible permeability constant.

  • The design theory for nanoporous filtration membranes needs to be established. The present study shows that the performance and technical advancement of nanoporous filtration membranes are determined by the fundamental parameter I (in the unit Watt1/2) which is formulated as a function of the shear strength of the liquid-pore wall interface, the radius of the filtration pore, the membrane thickness, and the bulk dynamic viscosity of the flowing liquid. This parameter determines the critical power loss on a single filtration pore for initiating the wall slippage, which is important for the flux of the membrane. It also relates the membrane permeability to the power cost by the filtration pore. It is shown that for biological cellular membranes its values are on the scale 1.0E-8Watt1/2, for mono-layer graphene membranes its values are on the scale 1.0E-9Watt1/2, and for nanoporous membranes made of silica, silicon nitride or silicon carbonized its values are on the scale 1.0E-5Watt1/2. The scale of the value of this parameter directly measures the level of the performance of a nanoporous filtration membrane. The carbon nanotube membrane has the similar performance with biological cellular membranes, as it also has the value of I on the scale 1.0E-8Watt1/2.

Fisetin-Mediated Perturbations of Membrane Permeability and Intracellular pH in Candida albicans

  • The antifungal activity of fisetin against Candida albicans is explored, elucidating a mechanism centered on membrane permeabilization and ensuing disruption of pH homeostasis. The Minimum Inhibitory Concentration (MIC) of fisetin, indicative of its interaction with the fungal membrane, increases in the presence of ergosterol. Hoechst 33342 and propidium-iodide staining reveal substantial propidium-iodide accumulation in fisetin-treated C. albicans cells at their MIC, with crystal violet uptake assays confirming fisetin-induced membrane permeabilization. Leakage analysis demonstrates a significant release of DNA and proteins in fisetin-treated cells compared to controls, underscoring the antifungal effect through membrane disruption. Green fluorescence, evident in both the cytoplasm and vacuoles of fisetin-treated cells under BCECF, AM staining, stands in contrast to controls where only acidic vacuoles exhibit staining. Ratiometric pH measurements using BCECF, AM reveal a noteworthy reduction in intracellular pH in fisetin-treated cells, emphasizing its impact on pH homeostasis. DiBAC4(3) uptake assays demonstrate membrane hyperpolarization in fisetintreated cells, suggesting potential disruptions in ion flux and cellular homeostasis. These results provide comprehensive insights into the antifungal mechanisms of fisetin, positioning it as a promising therapeutic agent against Candida infections.

A Study on Irreversible Degradation through OCV Reduction and Recovery Behavior in the Electrochemical Degradation Process of PEMFC Polymer Membrane (PEMFC 고분자 막의 전기화학적 열화과정에서 OCV 감소 및 회복 거동을 통한 비가역적 열화 연구)

  • It is very important to analyze the OCV change behavior during the open circuit potential holding (OCV holding) process, which accelerates the evaluation of the electrochemical durability of the PEMFC membrane. In this study, an empirical formula using the experimental data of three MEAs with different durability was created and compared. The durability evaluation time of the reinforced membrane MEA without radical scavenger inside the membrane was 383 h, and the durability evaluation time of the reinforced membrane MEA with radical scavenger inside the membrane was 1,000 and 1,650 h, respectively. The degradation of the membrane was divided into the reversible degradation that can be recovered by activation and the irreversible degradation that is not recovered. The irreversible degradation of the membrane was indicated by an increase in hydrogen permeability, and the change in hydrogen permeability was similar to the irreversible degradation constant c of all three MEAs. The initiation of irreversible deterioration without recovery is indicated by an increase in hydrogen permeability, and the OCV is not recovered due to an increase in hydrogen permeability, so the slope of the OCV recovery line (ORL) decreases, which can be confirmed by an increase in the constant c value of the empirical formula.

Effect of Zeolitic Imidazolate Framework-7 in Pebax Mixed Matrix Membrane for CO2/N2 Separation (CO2/N2 분리를 위한 Pebax 혼합막에서 Zeolitic Imidazolate Framework-7의 영향)

  • In this study, a mixed matrix membrane was prepared by putting the zeolitic imidazolate framework-7 (ZIF-7) synthesized in Pebax-1657 and Pebax-2533, which are representative poly(ether-b-amide), and the permeability properties of single gas such as N2 and CO2 were investigated. From the gas permeation results, in the case of N2, both the Pebax-1657/ZIF-7 and Pebax-2533/ZIF-7 mixed matrix membranes showed a similar phenomenon in which the permeability decreased with the incorporation of ZIF-7. For CO2 permeability, the tendency was slightly different depending on the type of polymer. In the Pebax-1657/ZIF-7 mixed membrane, the CO2 permeability decreased in the range of 0~3 wt% of ZIF-7, and increased at higher contents. The CO2 permeability of the Pebax-2533/ZIF-7 mixed matrix membrane gradually decreased without increasing the permeability in the range of 0~5 wt% of ZIF-7. Regarding CO2/N2 selectivity, both mixed films showed a tendency to increase with increasing the ZIF-7 content. In particular, Pebax-2533/ZIF-7 5 wt% showed the best gas permeation performance compared to other mixed matrix membrane. This is thought to be because ZIF-7 shows better compatibility with Pebax-2533 than that of Pebax-1657 and also better CO2 selective property.
