Incidence of Major Plant-parasitic Nematodes in Main Producing Areas of Strawberry in Korea (국내 딸기 시설재배 주산단지 토양 내의 주요 식물기생선충 감염 현황)

  • To investigate the incidence of plant-parasitic nematodes from plastic-house fields of strawberry, 170 soil samples were collected from the main producing area of strawberry in Korea from 2014 to 2015. Based on the investigation, root-lesion nematode (RLN; Pratylenchus spp.) and root-knot nematode (RKN; Meloidogyne spp.) are the most important pests in plastic-house fields of strawberry with 57% and 36% incidences, respectively. Among the collected fields, 96 and 63 fields were infected RNL and RKN, respectively with 44 fields (38.8%) being detected at both nematode. High degree of incidence of both nematodes were observed in the regions of Jinju, Gyeonnam Province, Korea. For identification of species level, phylogenetic and distance analysis were performed using RLN and RKN isolates generated from the regions of Jinju. The results indicated that the RLN and RKN isolates were Pratylenchus vulnus and Meloidogyne hapla, respectively. This study showed that P. vulnus and M. hapla could be most dominant species in plastic-house fields of strawberry. This data will provide basic knowledge for development of effective control strategies.

Comparison of the Inhibitory Effects of Nematicides on Nematode Populations in a Regional Vinyl Plastic House (지역별 시설재배지에서 식물기생선충의 살선충제에 대한 밀도억제 효과 비교)

  • To assess the efficacy of nematicides for the control of plant-parasitic nematodes in fruit and vegetables fields, soil samples were collected from a cucumber field at Gongju; from strawberry fields at Buyeo, Nonsan, and Jinju; and from a melon field at Gocksung in Jeonnam Province, Korea. Plant-parasitic nematodes were separated from each soil sample and identified. The susceptibilities of the nematodes to abamectin 1.68% SC, cadusafos 3% GR, dazomet 98% GR, fosthiazate 30% SL and BA12011 SL were examined under laboratory and field conditions. The average population density of plant-parasitic nematodes was generally reduced after the treatment with nematicides; however, there was increase in the population of Pratylenchus spp. in soil after treatment with fosthiazate at Buyeo and Gocksung. Furthermore, there were increased populations following treatment with abamectin, cadusafos, and dazomet at Gocksung. The control effects of BA12011 treatment on plant-parasitic nematodes were confirmed to be similar to those of the other 4 nematicides evaluated, although its control effect was higher than that of fosthiazate in cucumber-growing soil at Gongju. The effects of nematicide treatment on egg mass formation in each of the collected soils differed according to the region of soil origin. Abamectin was effective in reducing the degree of egg mass formation in Buyeo and Jinju soil, whereas BA12011 was effective in Nonsan and Gocksung soil. Dazomet was found to inhibit egg mass formation in Gongju soil. To evaluate the effect of the newly developed nematicide, BA12011, experiments were conducted in a cucumber-growing greenhouse. The average population densities of Meloidogyne spp., Pratylenchus spp., and Helicotylenchus spp. after the first treatment were reduced to a greater extent than after the second treatment. It is thus suggested that early nematicide treatment is important for effective control of plant-parasitic nematodes.

Attachment of Pasteuria penetrans Endospores to Meloidogyne spp. Juveniles Affected by Temperatures and the Nematode species (선충기생세균(Pasteuria penetrans) 내생포자의 뿌리혹선충(Meloidogyne spp.) 유충 부착에 대한 온도와 선충종의 영향)

  • A greenhouse soil infested with an obligate nematode parasitic bacterium, Pasteuria penetrans, was used to test the effect of temperatures on the endospore attachment to root-knot nematode, Meloidogyne arenaria, juveniles (J2). Freshly hatched J2s were inoculated to the soil in petri dish and incubated under different temperatures of $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, and $35^{\circ}C$ for 7 days. The endospore attachment rates were 100% in all the temperatures, while the number of endospores attached per J2 was highest in $25^{\circ}C$ with 28.3 endospores/J2 followed by 20.2, 18.6, and 13.6 in $30^{\circ}C$, $20^{\circ}C$, and $35^{\circ}C$, respectively. When the soil was pre-treated under different temperatures before the J2 inoculation, the endospore attachment rates significantly decreased from 60% in room temperature to $25^{\circ}C$, 31.7, 8.3, 5.0, and 0% after the soil incubation in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$ for 10 days, respectively. The endospore numbers attached per J2 were 3.5, 4.3, 1, 1, and 0 when the soil was pre-treated in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$, respectively, which were lower than 5.3/J2 of room temperature treated soil. The P. penetrans isolate in the soil showed nematode species-specific endospore attachment characteristics with 100% attachment rate only on M. arenaria J2s while the rates were 0% on M. hapla and M. incognita J2s.

Effect of Bacillus spp. Having Gelatin Decomposing Activity on Root-knot Nematode, Meloidogyne incognita on Pepper (젤라틴 분해력이 있는 Bacillus균의 고추 고구마뿌리혹선충에 미치는 영향)

  • This study investigated the decomposing activities on gelatin and egg sac of root-knot nematode (Meloidogyne incognita). Ten Bacillus isolates (KRB-1~10) isolated from the soils of Mt. Naejang. Among them, KRB-5, 9, and 10 showed decomposing activities, and identified as Bacillus subtilis KRB-5, Bacillus amyloliquefaciens KRB-9, and KRB-10 by 16S rRNA sequence analysis, respectively. Under pot experiments using pepper, 100-fold diluted culture broth of three isolates reduced the number of egg sac on roots collected 60 days and 90 days and increased the growth of aerial part compared with the group only treated with Meloidogyne incognita. Especially, the group treated B. subtilis KRB-5 was superior in the growth of pepper. These results suggest that the B. subtilis KRB-5 can be used to control the root-knot nematode on pepper.

Nematodes Associated with Medicinal herbs (약용작물거생선충에 관한 연구)

  • To investigate identification of species distribution state, population density of plant parasitic nematodes and damaged state by parasitism of nematodes with soil samples taken from major cultivating area of medicinal herbs in Kyungbuk districts are summarized as follow; total species belonging to genera in families were identified from 43 different medicinal herbs. The dominant nematode species on medicinal herbs was Meloidogyne hapla by 55.6% of field infection. Damaged state by parasitizm of Root-knot nematode, M. hapla, M. incognita are severed 15 medical plant except P. japonica, infected field were in 54.5~88.0%, and highered in population densities of 2nd larvae, Gall, Egg sac. Percentage of yield-decrease were 57.8% in A. gigas, 49.1% in P.japonica. Lighten effect of M. spp. by cropping system were lowered in population density in Rice+Paeony than Paeony+Paeony in paddy field, and also lowered in Sesame+Paeony or one year fallowing after harvest Paeony than Red pepper+Pseony, Paeony+Paeony in upland field.

Field Application of Egg and Larval Parasitic Fungi and Chemicals for Controlling Root-knot Nematodes on Some Medicinal Herb (몇종의 선충천적 진균과 화학약제를 이용한 약용작물 뿌리혹선충 방제효과 검토)

  • Series of studies were conducted to establish biological and chemical control method for Meloidogyne spp to medical herbs by applied of nematophagous fungi, Arthrobotrys spp, Fusarium spp, and egg parastic fungi, Paecilomyces lilacinus were applied for root-knot nematodes on medicinal herbs, Paeonia albiflora, Codonopsis lanceolata, Cnidium officinale. The results are as follow. In pot experiments, The no. of root gall and egg mass and larvae of Cnidium officinale. The results are as follow. In pot experiments. The no. of root gall and egg mass and larvae of Cnidium officinale, Codonopsis lanceolata, Paeonia japonica lowered in P. Lilacinus treated plots compare to untreated control plots. But A. thaumasia F. oxysporum treated plots were less effective. Effect of egg parasitic fungi and chemical treatment at divided root of Paeonia japonica after sterilized in pot were increased in the fresh weight, root weight, control effect in P. lilacinus treated plots as chemical, Carbo G treated plots compare to untreted control plots. I field experiment, the number of root gall, egg mass and nematode density of Paeonia were also suppressed in P. lilacinus treated plots. It was very effective continuous 2 years and transplanting time on Paeonia japonica infested soil with the M. hapla in field in both region, Chillgok and Euisung treated P. lilacinus as chemical treated plots. Soaking effect of insecticide for Paeonia japonica at diving shoot before transplating in pot were effective for 12hours immersion into 1,000 ppm of Benlate T + Mep Ec, Benlate T + Fenthion EC in pot and field Experiment. In the Examination of fungi activily on P.japonica field 1 year after soil treatment, Number of spore of P. lilacinus were 1,000~1,300 in 3 region except Euisung.

Studies on the Important Nematodes in Korea 1. Some Nematodes Found in the Soil around the Root of Floral Plants in the Green-house (우리나라 농업상 중요한 선충류에 관한 연구 (I) 온실화훼류의 근부에서 발생되는 수종의 선충에 관하여)

  • 최근 농작물에 대한 각종 선충류의 피해가 주목되어 있는 실성에 비추어 필자는 선충류연구의 기초적 과제로서 온실내의 화훼류 근부에서 발견되는 선충류 중 농업상 비교적 중요하다고 인정되는 종류와 그밖에 몇 종류에 대하여 형태적인 특징을 기술하였다. 즉 취급된 종류들은 다음과 같다. 1. Meloidogyne arenaria Chitwood 2. Scutellonema coheni Andrassy 3. Paurodontus gracilis Thorne 4. xiphinema americaum Cobb 5. Rhabditis sp 6. Cephalobus sp 7, Diplogaster sp 그리고 이상 7종류의 기주식물 및 기생정도를 보면 Meloidogyne arenaria Chitwood는 Begonia에 만 기생하였고 그밖에 화훼류에는 기생을 볼수 없었다. Scutellonema choeni Andrassy는 꽃치자, 소펄, 이마리 리스, 제브리나에 가장 많이 발견되었고, Paurodonthus gracilis Thorne는 동백나무, 청목. 지내고사리, 카네이션, 충에서 가장 많이 발견되었다. 그리고 Xiphinema americanum Coob는 산호수, 서향, 코피나무, 시크라맨, 용설란, 동백 등에서만 발견되었으나 그중에 용설란, 동백에서 가장 많이 볼 수 있었다. Rhabditis sp.는 초본류 화훼류의 대부분에서 발견되었으나 그중에도 산세배리아에서 가장 많았다 Diplogaster sp는 대부분의 화훼류에서 발견되었으며 그중에 크로톤에 비교적 많았다. Cephalobus sp.는 전반적으로 발견되었으며 발견정도는 모두 비슷하였다. Monhystera app. , Mononchus spp. , Dorylaimus app. 등도 조사란 과반수의 화훼류들에서 발견되었으나 특히 많이 발견되는 화훼류는 없었다. 이상 선충류중 근류를 형성하여 피해가 가장 주목되는 Mceloidogyne arenaris Chitwood에 대해서 온실내에서 그의 유일한 기주식물인 Begonia의 품종별 기생정도를 비교조사한 결과 semperflorens 품종에 가장 기생률이 높았고 metarica 품종은 중정도였으며 erytholopylla와 argento-guttata 품종들에는 기생이 없었다. 이 점은 매우 의의있는 사실로서 앞으로 더욱 연구할 과제로 삼았다. 끝으로 Sanceberia에서 많이 발견되는 Rhabditis sp를 $18^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$의 동온도별의 incubator에서 사육한 결과 본 종의 생육적온이 $30^{\circ}C$라는 사실을 알았다.

Environmentally-friendly Control of Soil Nematode by Crashed-rape (Brassica naptus) seed (유채 종실을 이용한 뿌리혹선충의 친환경적 방제)

  • The present studies were carried out for three years from 2007 until 2009 to control nematode and to increase yield of cucumber by crashed-rape seed application at polyethylene film house. Crashed-rape seed has a lot of glucosinolate. Myrosinase decompose gulcosinolate into isothiocyanate and thiocyanate when crashedrape seed go to decay at soil. Those chemical compounds act on poison to nematode at soil. When the crashedrape seed treated at soil, an amount of thiocyanate at soil was risen up. Thiocyanate of plot treated with 200 and 400 kg crashed-rape seed per 10a was 30 and 40 mg/kg, respectively. Nematode(meloidogyne spp) population at soil was 13 to 17 nematodes per dried soil 300g. Yield of cucumber increased 6 to 15 percent to be compared with control. While, Nematode(meloidogyne spp) population of control plot were 463 nematodes per dried soil 300 g. This level was much higher than 150 nematodes which can be brought about injury to plant. Even if the more an amount of crashed-rape seed application, the higher yield of cucumber and control effect of nematode. Consider economical efficiency, 200 kg of crashed-rape seed per 10a was the most effective. Therefore, we suggest applying 200 kg of crashed-rape seed per 10a to control soil nematode when culture cucumber at plastic film house.

Turfgrass Insect Pests and Natural Enemies in Golf Courses (골프장 잔디 해충과 천적의 종류)

  • - Turfgrass insect pests and natura.l enemies for biological control were investigated to develop pest management effectively in golf courses at several golf clubs. Twenty eight insect pest species of 10 families in 6 orders were collected from golf courses. The zoysiagrass mite, Eriophyes zoysiae and root-knot nematode, Meloidogyne incognita were also collected from zoysiagrass. White grubs of several scarab beetles and cutworms (Agrotis spp.) damaged seriously at most surveyed golf clubs. In addition, bluegrass webworm (Crambus sp.), Japanese lawngrass cutworm (Spodoptera depravata), scale insects, Tipula sp., and ants (Camponitus japonicus, Formica japonica, and Lasins japonicus) damaged turfgrasses directly or indirectly in golf courses. The entomopathogenic nematodes, Heterorhabditis spp., Steinernema glaseri, and S. longicaudum, entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae, and milky disease, Paenibacil/us popil/iae were isolated from white grubs or turfgrass soil as microbial control agents. Besides, dipteran predators, Cophinopoda chinensis, Philonicus albiceps, and Promachus yesonicus and hymenopteran parasitoid, Tiphia sp. were also collected. The P. yesonicus was the most active in golf courses. The root-knot nematode, M. incognita was found from Zoysia japonica, Z. matrella. and Cynodon dactylon.

Screening of Botanicals Against Root Knot Disease Complex in Mulberry (Morus indica L.)

  • For developing an integrated eco-friendly package against root knot disease complex of mulberry caused by the association of Meloidogyne incognita with Fusarium solani and F. oxysporum causing serious loss in terms of leaf yield and quality during cultivation, twenty botanical extracts at 5, 10 & 20% concentrations were screened under in vitro conditions. Among the extracts, Allium sativum followed by Lasownia inermis were found to be effective at 20% concentration against both the virulent fungi and nematode. Both the extracts reduced the mycelial growth of virulent fungi to an extent of 76-100%, inhibited the hatching of nematode eggs by 80-90% and 76-85% larval mortality over the control. The other extracts were found either moderately or poorly effective in reducing the growth of fungi, hatching of nematode eggs and enhancing the mortality of larvae. The two effective botanical extracts, which rated as strong inhibitors against both nematode and virulent fungi, can be utilized in developing an integrated ecofriendly technology for better management of root knot disease complex in mulberry.
