Ecklonia cava is popular in Korea as a marine functional materials. E. cava is generally collected and used on the coast of Jeju Island. However, the continuous use of collected natural E. cava may be limited because difficult to secure throughout the year and may be exposed to environmental pollution. Jeju magma seawater (MSW) was known to be significant advantages such as safety, cleanness, stability, and functional improvement. Attempts have been reported on application of MSW to the culturing of macro- and microalgae and showed improved results. Thus, the objective of the present study was to explore the anti-melanogenesis activity of brown seaweed E. cava (E. cava cultured with MSW [MSWE]) extract cultured in tanks with MSW of Jeju Island to evaluate the possibility of cosmeceutical industrial application. MSWE extract showed the higher polyphenolic and dieckol contents than natural E. cava (NE) extract. Anti-melanogenesis activity of MSWE extract and NE extract are tested and compared using tyrosinase and dihydroxyphenylalanine (DOPA) oxidation inhibition assay. MSWE extracts evidenced more effective tyrosinase and DOPA oxidation inhibition activity than that of the NE extracts and the commercial whitening agent, arbutin. MSWE extracts also markedly inhibited melanin synthesis and decreased the expression of melanogenesis-related protein in ${\alpha}$-melanocyte stimulating hormone-stimulated B16F10 melanoma cells without cytotoxicity. These results suggest that MSW cultivation process would be more effective in releasing bioactive compounds with whitening effect from seaweed such as E. cava at an industrial scale.

This study was performed to assess the antioxidant activities and whitening effects of Sorbus commixta HEDL cortex on melanin synthesis. The whitening effects of Sorbus commixta HEDL cortex water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effects of Sorbus commixta HEDL cortex water extracts on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effects of Sorbus commixta HEDL cortex onto free radical generation were determined by measuring DPPH and hydroxyl radical scavenging activities. Our results indicated that Sorbus commixta HEDL cortex water extracts effectively inhibited free radical generation. In DPPH radical scavenging activity, Sorbus commixta HEDL cortex water extracts had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Sorbus commixta HEDL cortex suppressed the expression of tyrosinase, TRP-1 and TRP-2 in B16BL6 melanoma cells. These results show that Sorbus commixta HEDL cortex inhibited melanin production on the melanogenesis. The underlying mechanism of Sorbus commixta HEDL cortex on whitening activity may be due to the inhibition of tyrosinase activity and tyrosinase, TRP-1, TRP-2 expression. We suggest that Sorbus commixta HEDL cortex may be contain new natural active ingredients for antioxidant and whitening cosmetics.

In this study, we investigated the application of an extract from Ephedra sinica which has been composed of traditional Korean medicine as a whitening ingredient. The extract of Ephedra sinica which was obtained from the mixture of methanol and water (1:1) the inhibitory effect of tyrosinase. Then, Ephedra sinica was extracted by two different solvents. One was water and the other was methylene chloride. Only, the water extract of Ephedra sinica showed the inhibitory effort of tyrosinase; the anti-tyrosinase activity with $0.2\%$ of the water extract was $60.6\%$. But the extract of Ephedra sinica in methylene chloride fraction showed little inhibitory effect on tyrosinase. The inhibitory effect of the concentrated water extract of Ephedra sinica was tested on L-DOPA auto-oxidation and melanin synthesis in B-16 melanoma. In L-DOPA auto-oxidation, $0.5\%$ of the concentrated water extract showed $87\%$ of inhibition of L-DOPA auto-oxidation and the $0.75\%$ concentrated Ephedra sinica extract in wafer fraction inhibited $98.8\%$ of that. In melanin synthesis of B-16 melanoma, the concentrated water effect of Ephedra sinica inhibited $70.2\%$ or $79.9\%$ of inhibitory effect on that at the concentration of $0.05\%$ or $0.075\%$, respectively. For verifying the skin whitening effect of the concentrated water extract of Ephedra sinica in vivo, we performed the clinical test of that. The research showed the significant skin whitening effect of a cream containing $0.5\%$ Ephedra sinica extract and the statistical analysis showed a significant difference (p < 0.05) between sample (containing $0.5\%$ Ephedra sinica extract) and placebo after 10 weeks.

Photobiomodulation (PBM)-based therapy, which uses a phenomenon in which a light source of a specific wavelength band promotes ATP production in mitochondria, has attracted much attention in the fields of biology and medicine because of its effects on wound healing, inflammation reduction, and pain relief. Research on PBM-based therapy has mainly used lasers and light-emitting diodes (LEDs) as light sources and, despite the advantages of organic light-emitting diodes (OLEDs), there have been only a few cases where OLEDs were used in PBM-based therapy. In this research, the skin-care effect of PBM was analyzed using red (λ = 620 nm), green (λ = 525 nm), and blue (λ = 455 nm) OLED lighting modules, and was compared to the PBM effect of LEDs. We demonstrated the PBM-based skin-care effect of the red, green, blue OLED lighting modules by measuring the increase in the amount of collagen type-1 synthesis, the inhibition of melanin synthesis, and the suppression of nitric oxide generation, respectively.

In this study, the various whitening activities of Inonotus obliquus were assessed for potential use as functional cosmetic materials. When measuring electron donating ability to confirm the antioxidant ability of I. obliquus extract, increased activity was observed as the concentration increased, and it was found an outstanding antioxidant capacity of 82.1% at a 1,000 ㎍/ml concentration. Also, the tyrosinase inhibitory effect, related to a whitening effect, was found to have inhibitory activity that increased in a concentration-dependent manner. The results of verifying the viability of melanoma cells (B16F10) using an MTT assay showed cell viability of more than 80% at concentrations below 100 ㎍/ml. Therefore, cell-related experiments were conducted at 25, 50, and 100 ㎍/ml concentrations. By measuring protein expression related to melanin synthesis via treating B16F10 cells with I. obliquus extract, it was confirmed that protein expression was inhibited in all factors, depending on the concentration. TRP-1 and MITF appeared by 40.1% and 64.2% amount of expression, respectively, at 100 ㎍/ml concentrations, and tyrosinase and TRP-2 were verified as having better protein expression inhibition than arbutin. In measuring mRNA expression related to melanin synthesis by treating B16F10 cells with I. obliquus extract, it was confirmed that mRNA expression was suppressed as the concentration increased. Accordingly, it was confirmed that I. obliquus extract has excellent whitening activity and could be used as a cosmetic material.

In order to develop a new depigmenting agent, extracts were obtained from 60 native plants and their antimelanogenic activities were screened by evaluating the inhibitory effect on tyrosinase which is a major enzyme responsibles for the melanin synthesis. The extracts of Trichosanthes kirilowii fruits, Phyllostachys bambusoides inner films (BIF), Clerodendrum trichotomum leaves, and Acer okamotoanum leaves showed relatively high inhibitory effect on tyrosinase and their $IC_{50}$ values were $50{\sim}100{\mu}g/mL$. The extract of BIF inhibited melanin synthesis of B16F10 melanoma cells by 52%, which was the highest among those of various extracts. Furthermore, the effect of BIF extract is 10% higher than that of arbutin (42%), a popular depigmenting agent in Korea. Ten compounds having antimelanogenic activity were isolated from the BIF extract by solvent extraction and chromatography. These compounds were identified as phenolic derivatives: SM701, SM702, SM703, and BPR211 were hydroquinone derivatives; SM707 a gallic acid derivative; SM704, SM705, SM706, SM708 and SM709 ferulic acid derivatives. The free radical scavenging activities of these compounds were measured and compared to those of hydroquinone and vitamin C. The $SC_{50}$ values scavenging 50% DPPH of SM702 and SM709 were $60{\sim}70{\mu}M$ similar to that of hydroquinone and those of SM701 and SM708 were $30{\sim}40{\mu}M$ slightly lower than that of vitamin C. These results suggest the presence of components having high antioxidant activity in the BIF extract. The SM709, identified as 1,2-O-diferulylglycerol, inhibited the activities of tyrosine hydroxylase and dopa oxidase by 18 and 60%, respectively. The SM709 also inhibited the melanin synthesis of B16F10 melanoma cells by 62% and this was the highest antimelanogenic activity among those obtained from the various purified compounds. Therefore, antimelanogenic activity of the BIF extract was concluded to be due to both inhibition of DOPA oxidase and antioxidant activity.

Melanin pigments are abundantly distributed in mammalian skin, hair, eyes, and nervous system. Under normal physiological conditions, melanin protects the skin against various environmental stresses and acts as a physiological redox buffer to maintain homeostasis. However, abnormal melanin accumulation results in various hyperpigmentation conditions, such as chloasma, freckles, senile lentigo, and inflammatory pigmentation. Tyrosinase, a copper-containing enzyme, plays an important role in the regulation of the melanin pigment biosynthetic pathway. Although several whitening agents based on tyrosinase inhibition have been developed, their side effects, such as allergies, DNA damage, mutagenesis, and cytotoxicity of melanocytes, limit their applications. In this study, we synthesized 4-chromanone derivatives (MHY compounds) and investigated their ability to inhibit tyrosinase activity. Of these compounds, (E)-3-(4-hydroxybenzylidene)chroman-4-one (MHY1294) more potently inhibited the enzymatic activity of tyrosinase (IC50 = 5.1±0.86 μM) than kojic acid (14.3±1.43 μM), a representative tyrosinase inhibitor. In addition, MHY1294 showed competitive inhibitory action at the catalytic site of tyrosinase and had greater binding affinity at this site than kojic acid. Furthermore, MHY1294 effectively inhibited α-melanocyte stimulating hormone (α-MSH)-induced melanin synthesis and intracellular tyrosinase activity in B16F10 melanoma cells. The results of the present study indicate that MHY1294 may be considered as a candidate pharmacological agent and cosmetic whitening ingredient.
The objective of the present study was to evaluate the skin whitening effect of the extracts, Lycii fructus (LF), Dry-L. foilum (DLF) and Fresh-L. folium (FLF). Tyrosinase inhibition activities was 44% in DLF ethanol extracts at a $500{\mu}g/mL$. When the tyrosinase activities in B16F10 murine melanoma cell were tested, the activities in DLF ethanol extracts was 14% at a $50{\mu}g/mL$ concentration. The protein expression of microphthalmia-associated transcription factor, tyrosinase related protein 1 (TRP-1), TRP-2, and tyrosinase, which are all melanin related factors, showed that LF, DLF and FLF extracts inhibited the protein bio-synthesis in B16F10 melanoma cell. Especially the DLF extract showed greater decrease of protein expressions. Results indicate that the DLF extract tested in the present study had skin whitening activity and can be used as a function a ingredients for food and cosmetic compositions.
Background: Bioactive compounds in plant extracts are able to reduce metal ions to nanoparticles through the process of green synthesis. Panax ginseng is an oriental medicinal herb and an adaptogen which has been historically used to cure various diseases. In addition, the P. ginseng leaves-mediated gold nanoparticles are the value-added novel materials. Its potential as a cosmetic ingredient is still unexplored. The aim of this study was to evaluate the antioxidant, moisture retention and whitening properties of gold nanoparticles (PgAuNPs) in cosmetic applications. Methods: Cell-free experiments were performed to evaluate PgAuNP's antioxidant and moisture retention properties and inhibition activity on mushroom tyrosinase. Furthermore, in vitro cell cytotoxicity was evaluated using normal human dermal fibroblast and murine B16BL6 melanoma cells (B16) after treatment with increasing concentrations of PgAuNPs for 24 h, 48 h, and 72 h. Finally, in vitro cell assays on B16 cells were performed to evaluate the whitening effect of PgAuNPs through reduction of cellular melanin content and tyrosinase activity. Results: In vitro DPPH radical scavenging assay results revealed that PgAuNPs exhibited antioxidant activity in a dose-dependent manner. PgAuNPs exhibited moisture retention capacity and effectively inhibited mushroom tyrosinase. In addition, 3-(4,5-dimethyl-thiazol-2yl)-2,5-diphenyl tetrazolium bromide results revealed that PgAuNPs were not toxic to human dermal fibroblast and B16 cells; in addition, they significantly reduced melanin content, tyrosinase activity, and mRNA expression of melanogenesis-associated transcription factor and tyrosinase in B16 cells. Conclusion: Our study is the first report to provide evidence supporting that P. ginseng leaves-capped gold nanoparticles could be used as multifunctional ingredients in cosmetics.

Several hydroxycinnamic acid derivatives, p-coumaric acid, ferulic acid, N,N'-dicoumaroylputrescine (DCP), N-p-coumaroyl-N'-feruloyl-putrescine (CFP), and N,N'-diferuloylputrescine (DFP) were isolated and purified from corn bran. To develop the skin whitening agent, we investigated the effects of hydroxycinnamic acid derivatives from corn bran, on melanogenesis. CFP and DFP inhibited melanin synthesis in a dose dependent manner up to 44.7 ${\pm}$ 6.0 %, and 58.5 ${\pm}$ 3.1 % at a concentration of 50 ${\mu}g/mL$, respectively. The intracellular tyrosinase activity decreased about 42.5 ${\pm}$ 14.6 %, and 9.0 ${\pm}$ 4.4 % at a concentration of 50 ${\mu}g/mL$ of CFP and DFP, respectively. Our results suggest that inhibitory effects of hydroxycinnamic acid derivatives on melanogenesis are due to the inhibition of the intracellular tyrosinase activity. These results indicate that these hydroxycinnamic acid derivatives from corn bran may be potential natural skin whitening agents.
