Synthesis and Evaluation of Cytotoxicity of Novel Arylsulfonylimidazolidinones Containing Sulfonylurea Pharmacophore

  • Design and synthesis of novel 4-phenyl-1-arylsulfonylimidazolones 3 and 4-phenyl-3-arylsulfonylimidazolones 4 and evaluation of their cytotoxic activity against eleven human cancer cell lines and two murine leukemia cell lines in vitro were performed. As a result, a series of 4-phenyl-1(N)-arylsulfonylimidazolones (3) has been found to be the potential anticancer agent. Compounds 3b, 3c, and 3d exhibit strong activity as indicated by their $IC_{50}$ values 0.39, 3.19, $0.31{\mu}g/mL$ against A549 and 0.80, 0.48, $0.0007{\mu}g/mL$against SK-Mel-2, respectively. These compounds also possess much more potent activity (10-1000 times) than LY186641 against eleven other cell lines.

Combination of Classifiers Decisions for Multilingual Speaker Identification

  • State-of-the-art speaker recognition systems may work better for the English language. However, if the same system is used for recognizing those who speak different languages, the systems may yield a poor performance. In this work, the decisions of a Gaussian mixture model-universal background model (GMM-UBM) and a learning vector quantization (LVQ) are combined to improve the recognition performance of a multilingual speaker identification system. The difference between these classifiers is in their modeling techniques. The former one is based on probabilistic approach and the latter one is based on the fine-tuning of neurons. Since the approaches are different, each modeling technique identifies different sets of speakers for the same database set. Therefore, the decisions of the classifiers may be used to improve the performance. In this study, multitaper mel-frequency cepstral coefficients (MFCCs) are used as the features and the monolingual and cross-lingual speaker identification studies are conducted using NIST-2003 and our own database. The experimental results show that the combined system improves the performance by nearly 10% compared with that of the individual classifier.

  • The alkaloid fuyuziphine was isolated from the whole plant of Fumaria indica. It had inhibitive effect against spore germination of some plant pathogenic fungi (Collectotrichum sp., C. gloeosporioides, C. falcatum, Curvularia maculans, C. lunata, Erysiphe cichoracearum, Helminthosporium pennisetti, Oidium erysiphoides, Ustilago cynodontis, Alternaria chieranthi, A. mel-ongenae, A. brassicicola and A. solam). Curvularia lunata, Oidium erysiphoides, Alternaria brassicicola and A. solani did not germinate at 750 and 1000 ppm and Colletotrichum gloeosporioides, C. falcatum, Curvularia maculans were inhibited at 1000 ppm for 24 hr incubation. Germination of most fungi was significantly inhibited at $100{\sim}750ppm$.

  • A bioassay-guided fractionation of the whole extract of Viscum album (a parasitic plant : Loranthaceae) led to the isolation of two triterpenoidal components, oleanolic acid (1), ${\beta}$-amyrin acetate (2), homoflavoyadorinin B (3) as well as large quantity of free fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble part and BuOH soluble part of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro, whereas the remaining water soluble part exhibited a poor inhibition. Numerical simulation of fully nonlinear sloshing waves in three-dimensional tank under random excitation

  • Based on the fully nonlinear velocity potential theory, the liquid sloshing in a three dimensional tank under random excitation is studied. The governing Laplace equation with fully nonlinear boundary conditions on the moving free surface is solved using the indirect desingularized boundary integral equation method (DBIEM). The fourth-order predictor-corrector Adams-Bashforth-Moulton scheme (ABM4) and mixed Eulerian-Lagrangian (MEL) method are used for the time-stepping integration of the free surface boundary conditions. A smoothing scheme, B-spline curve, is applied to both the longitudinal and transverse directions of the tank to eliminate the possible saw-tooth instabilities. When the tank is undergoing one dimensional regular motion of small amplitude, the calculated results are found to be in very good agreement with linear analytical solution. In the simulation, the normal standing waves, travelling waves and bores are observed. The extensive calculation has been made for the tank undergoing specified random oscillation. The nonlinear effect of random sloshing wave is studied and the effect of peak frequency used for the generation of random oscillation is investigated. It is found that, even as the peak value of spectrum for oscillation becomes smaller, the maximum wave elevation on the side wall becomes bigger when the peak frequency is closer to the natural frequency.

  • To evaluate the antitumor activity and antimetastatic effects of Kamicheungyeolhaedog tang(KCHT), studies were done experimentally. The results were obtained as follows: 1. KCHT extracts exhibited a significant cytotoxicity against A549, SK-MEL-2, SK-OV-3, and B16-BL6 cell lines. 2. KCHT extracts showed significant inhibitoty effect on DNA topoisomerase I 3. The T/C% was 145.8% in KCHT treated group in S-180 bearing ICR mice. 4. KCHT extracts exhibited efficient affect adhesive effect of A549, B16-BL6 cell to complex extracellular matrix. 5. In vitro neovascularization assays, angiogenesis was insignificantly inhibited in KCHT treated group as compared with control group. These results suggested that KCHT extracts might be usefully applied for prevention and treatement of cancer.

Study on Antitumor Activity of Sobokchukeotang and Kamisobokchukeotang (소복축어탕과 가미소복축어탕이 항암활성에 미치는 영향)

  • This study was attempted to investigate the anti-tumor and anti-metastatic effects of Sobokchukeotang(SBCT) and Kamisobokchukeotang(KSBCT). Cytotoxicity against various cancer cell lines, anti-adhesion, pulmonary colonization, anti-angiogenesis, and T/C% were evaluated. SBCT and KSBCT exhibited no cytotoxicity against HT-1080, A549, SK-OV-3, B16-F10 and SK-Mel-2 cell lines. In inhibitory effect on DNA topoisomerase I, the $IC_{50S}$ were shown $250-500{\;}\mu\textrm{g}/ml$ of SBCT and $62.5-125{\;}\mu\textrm{g}/ml$ of KSBCT respectively. In the in vivo experiments, SBCT(135.98%) and KSBCT(151.92%) apparently increased the life span of mice bearing sarcoma-180. KSBCT significantly inhibited the adhesion of HT-1080 to complex extracellular matrix in a dose-dependent manner in contrast to SBCT. In pulmonary colonization assay by B16-F10, a number of colonies in the lungs were decreased more significantly in KSBCT group than those in SBCT group. In vitro neovascularization and CAM assay, angiogenesis was more significantly inhibited in KSBCT-treated group than in SBCT- treated group. Above results suggests that KSBCT is more effectively applied to prevention and treatment of cancer than SBCT.

The fourth-order predictor-corrector Adams-Bashforth-Moulton scheme (ABM4) and mixed Eulerian-Lagrangian (MEL) method are used for the time-stepping integration of the free surface boundary conditions. A smoothing algorithm, B-spline, is applied to eliminate the possible saw-tooth instabilities. The artificial wave speed employed in MTF (multi-transmitting formula) approach is investigated for fully nonlinear wave problem. The numerical results from incorporating the damping zone (DZ), MTF and MTF coupled DZ (MTF+DZ) methods as radiation condition are compared with analytical solution. An effective MTF+DZ method is finally adopted to simulate the 3D linear wave, second-order wave and irregular wave propagation. It is shown that the MTF+DZ method can be used for simulating fully nonlinear wave propagation very efficiently.

Pycnogenol Supplementation Retards Immune Dysfunction in Murine AIDS (MAIDS) After LP-BM5 Leukemia Virus Infection by Modulating Cytokine Secretion

  • We investigated the effect of pycnogenol (PYC) supplementation on retarding the immune dysfunction of CS7BL/6 mice after murine AIDS (MAIDS) development. Dysfunction of T and B cell mitogenesis from primary cultured splenocytes has been observed with retrovirus infection and PYC supplementation partially recovered the dysfunction of T and B cells. There was an abnormal shift of cytokine pattern with retrovirns infection, which was designated by the decreased secretion of Th1 cytokines and increased secretion of Th2 cytokines. PYC supplementation increased IL-2 and $IFN-\gamma$ secretion and decreased IL-4, IL-6, and $TNF-\alpha$ secretion, but it was not sufficient enough to maintain the normal level of these cytokines. Hepatic vitamin E level was significantly decreased by retrovirns infection, in accordance with increased hepatic lipid peroxidation level, whereas PYC supplementation normalized the hepatic level of vitamin E and lipid peroxidation. This study suggests that PYC supplementation may partially help retard the incidence of symptoms during MAIDS.

Two-Channel Noise Reduction Using Beamforming and DOA-Based Masking (빔포밍 및 DOA 기반의 마스킹을 이용한 2채널 잡음제거)

  • In this paper, we propose a multi-channel speech enhancement algorithm using beamforming and direction-of-arrival (DOA)-based masking. The proposed algorithm enhances noisy speech basically by the linearly constrained minimum variance (LCMV) algorithm and then a mel-scale Wiener filter designed using DOA-based masking is applied to remove still remaining noises. To improve the performance, we optimize the learning rate of the adaptive filters in LCMV and the DOA threshold to detect target speech spectrum. As performance indices, the perceptual evaluation of speech quality (PESQ) score and output SNRs are measured. Experimantal results show that the proposed algorithm outperforms the conventional LCMV beamformer by 0.09 in PESQ score and 5.75 dB in output SNR, respectively.
