Purine이 생쥐 미성숙난자의 핵성숙에 미치는 영향 II. 미성숙 난자의 제 1극체 방출과 생존성에 미치는 Purine의 효과 (Effects of Purine on Meiotic Maturation of Mouse Immature Oocytes II. Effects of Purine on Extrusion Rates of 1st pb and Viability of Immature Oocytes)

  • In the previous study, we observed that Purine has a time dependent effect in maintaining the oocytes in meiotic arrest, and human fetal cord serum(HFCS) and human mature follicular fluid(HMFF) reverse the GVBD suppressed by purines. And it was reported that purine has a harmful effect on the development of oocytes or embryos, when they were cultured for a long time, in vitro. Therefore this study was performed to investigate the effects of purine on extrusion rates of 1st pb and viability of oocytes cultured for a long time, in vitro. Immature oocytes(GV stage) were collected from ovaries of 25~28 day old ICR mice at 48 hrs after PMSG injection. Cumulus-enclosed and denuded oocytes collected were assigned randomly to one of several culture conditions. Some of the oocytes were cultured in 4mM hypoxanthine for 24hr, and the extrusion rates of 1st pb and viability of the oocytes were assessed at every 12 hrs. In the viability, the oocytes showed granulation, pigmentation of cytoplasm or lysis of 1st pb extruded were regarded as degenerating oocytes. Also some of the oocytes were cultured in hypoxanthine for 12 hrs then the resulting oocytes were transferred to hypoxanthine-free medium and cultured for 12 hrs to determine whether the inhibitory effect of hypoxanthine on the 1st pb extrusion was reversible. The rest of the oocytes were cultured in medium containing hypoxanthine and adenosine for 18 hrs to compare the 1st pb extrusion be attendant upon hte concentration of HFCS or HMFF supplemented. Hypoxanthine suppressed the extrusion of 1st pb and viability of the oocytes significantly, when they were cultured for more than 12 hrs and the harmful effect of hypoxanthine was showed in denuded oocytes, prominently. The suppressive effect of hypoxanthine was reversed by just removal of the hypoxanthine from the cultrue medium. Also there was no difference in reverse the pb extrusion rate suppressed between HFCS and HMFF. The extrusion rate of 1st pb in medium containing adenosine and hypoxanthine was increased in line with the concentration of HFCS or HMFF supplemented. Hypoxanthine suppressed the extrusion of 1st pb and viability of the oocytes significantly, when they were cultured for more than 12 hrs and the harmful effect of hypoxanthine was showed in denuded oocytes, prominetly. The suppressive effect of hypoxanthine was reversed by just removal of the hypoxanthine fromthe culture medium. CT 조영제가 MR 조영제의 이완율에 미치는 영향 (Effects of CT Contrast Medium on the Relaxation Rate of MR Contrast Medium)

  • In MR, the iodine CT contrast medium reduces the T1 and T2 relaxation times of the substance, resulting in a change in signal intensity. This study aimed to measure the relaxation rate of MR contrast medium with or without diluting CT contrast medium and analyzed the effect of CT contrast medium. Undiluted Gadoteridol solution was diluted with saline to prepare MR contrast medium phantoms with various levels of Gadoteridol concentrations. Moreover, undiluted Iomeprol was mixed with the prepared MR contrast medium phantoms at 1:1 ratio to make MR contrast medium phantoms with containing CT contrast medium for the experiment. T1 and T2 mappings were conducted to quantitatively evaluate the relaxation time and relaxation rate of these phantoms. The results showed that the T1 and T2 relaxation time and relaxation rate of MR contrast medium diluted with CT contrast medium were significantly (p<0.05) shorter than those of MR contrast medium not diluted with CT contrast medium. The results of this study imply that, when MR contrast medium shall be used after injecting CT contrast medium, CT contrast medium should be discharged enough. Moreover, it would be desirable to conduct CT test after taking MRI test in order to reduce the effects of CT contrast medium on MR contrast medium.

중소규모 사업장에서의 고혈압 근로자에 대한 보건관리 효과 (Effect of Health Services on Workers with Hypertension in Small and Medium Sized Enterprises)

  • The purpose of this study was to evaluate the effect of health services on workers with hypertension in small and medium sized enterprises (20-300 employees). One hundred seventy three workers among 296 workers with hypertension were responded to the questionnaire on general and occupational characteristics, health habits and anti-hypertensive medications. Blood pressure was obtained while seated using a calibrated standard manometer before and after 4 months health services. Statistical analysis was performed using $X^2$-test and logistic regression analysis. The results were as follows ; 1. No significant differences were noted in variables of smoking, glucose intake and sleeping time, while significant differences were showed in variables of exercise time, frequency and amount of drinking and intake of salt, protain, lipid, calcium, vegetable and fruits between groups which was provided and not provided health services. 2. The rate of anti-hypertensive medication was increased and the blood pressure was decreased significantly in group which was provided health services compared with those of group which was not provided. 3. With the logistic regression analysis, significant variables affecting blood pressure change were smoking and anti-hypertensive medication. The results of this study suggested that health services on workers with hypertension in small and medium sized enterprises was effective, and the role of health professionals for health promotion movement in enterprises was important.

중쇄지방산염 함유 Eudispert hv 하이드로겔의 인슐린 직장 흡수증대효과 (Enhancement of Rectal Absorption of Insulin in Eudispert hv Hydrogels Containing Medium Chain Fatty Acid Salts in Rats)

  • The purpose of the present study was to investigate the effect of medium chain fatty acid salts, reported as enhancers in insulin nasal absorption, on the rectal absorption of insulin in rats. The serum glucose and remained insulin level in perfusate were measured after rectal recirculation of insulin with or without sod. laurate, sod. caprate and sod. caprylate in situ. The addition of sod. laurate or sod. caprate reduced serum glucose concentration considerably. Sod. caprate (1.0%) showed the greatest promoting effect on the decrement of serum glucose. Eudispert hv hydrogels containing insulin with medium chain fatty acid salts were, thereby, prepared and evaluated. The release rate of insulin from Eudispert hv hydrogels was reduced with an increase in the content of Eudispert hv, and was raised with increasing NaOH concentration. Ten percent Eudispert hv hydrogels were offered for the rectal administration of insulin. The addition of 1.0% sod. caprate reduced serum glucose concentration remarkably after rectal administration of 10% Eudispert hv hydrogels containing insulin. The level of glucose decrement was greater by 30% compared to subcutaneous administration of insulin solution. From the above findings, Eudispert hv hydrogels would be used as useful rectal delivery systems of insulin.

Aminolysis of Y- Substituted Phenyl Benzenesulfonates in MeCN: Effect of Medium on Reactivity and Reaction Mechanism

  • Second-order rate constants for nucleophilic substitution reactions of 2,4-dinitrophenyl benzenesulfonate 1a with a series of alicyclic secondary amines in MeCN have been measured spectrophotometrically and compared with those reported previously for the corresponding reactions performed in aqueous medium to investigate the effect of medium on reactivity and reaction mechanism. The amines employed in this study are found to be more reactive in the aprotic solvent than in $H_2O$. The reactions of 1a in MeCN result in a linear Br${\o}$nsted-type plot with ${\beta}_{nuc}$ = 0.58, which contrasts to the curved Br${\o}$nsted-type plot reported previously for the corresponding reactions performed in the aqueous medium (i.e., ${\beta}_2$ = 0.86 and ${\beta}_1$ = 0.38). Accordingly, it has been concluded that the reaction mechanism changes from a stepwise mechanism to a concerted pathway upon changing the medium from $H_2O$ to MeCN. Reactions of Y-substituted phenyl benzenesulfonates 1a-c with piperidine in MeCN result in a linear Br${\o}$nsted-type plot with ${\beta}_{lg}$ = -1.31, indicating that expulsion of the leaving group is significantly more advanced than bond formation in the transition state. The trigonal bipyramidal intermediate ($TBPy^{\pm}$) proposed previously for the reactions in $H_2O$ would be highly unstable in MeCN due to strong repulsion between the negative charge in $TBPy^{\pm}$ and the negative dipole end of MeCN. Thus, destabilization of $TBPy^{\pm}$ in MeCN has been concluded to change the reaction mechanism from a stepwise mechanism to a concerted pathway.

The Effect of Hierarchy Culture on Clan Leadership and Organizational Commitment of Export-Driven SMEs

  • Purpose: The purpose of this study examines the mediating effect of clan leadership in the relationship between hierarchy culture and organizational commitment. Most previous research focused on the relationship between organizational culture and organizational performance or organizational culture and job satisfaction. There are few empirical studies that focus on organizational commitment data because it is difficult to collect in many cases of export-driven small and medium sized enterprises. However, this research measures affective commitment, continuance commitment, and normative commitment differently than previous research, which is mostly focused on the hierarchy culture, clan leadership, and organizational commitment measurements. Research design, data, methodology: Conceptual research model is based on the studies of Cameron and Quinn (2011), and Gungor and Sahin (2018). The model is designed with three constructs such as hierarchy culture, organizational commitment, and clan leadership. The monitor culture and coordinator culture are as proxy for the hierarchy culture. The affective commitment, continuance commitment, and normative commitment are as proxy for the organizational commitment. And also the facilitator leadership and mentor leadership are as proxy for the clan leadership. Based on three hundred cases such as export-driven small and medium sized enterprises (SMEs), this study verify the hypothesis. Hypothesis was analyzed with the structural equation modeling. Results: In case of export-driven small and medium sized enterprises (SMEs), clan leadership acts as a mediator in the relationship between hierarchy culture and organizational commitment. In case of export-driven small and medium sized enterprises (SMEs) with high organizational commitment, clan leadership acts as a mediator in the relationship between hierarchy culture and organizational commitment. In case of export-driven small and medium sized enterprises (SMEs) with low organizational commitment, clan leadership did not act as a mediator in the relationship between hierarchy culture and organizational commitment. Conclusions: By controlling for the mediating effect of clan culture, this study have improved the academic contributions as well as policy and practical implications through empirical study of clan leadership that affect organizational commitment in the fields of hierarchy culture. In addition, this study means that the mediating effects on the variables of clan leadership were examined.

Bacillus subtilis EK11로부터 Protopectinase 생산을 위한 배지성분의 영향 (Effect of Medium Composition on Protopectinase Production from Bacillus subtilis EK11)

  • Protopectinase (PPases) are heterologous group of enzymes that degrade pectin from the insoluble protopection which is constituent of the middle lamella and primary cell wall of higher plants by restricted depolymerization. From the previous report[6], enzymatically separated plant cells, which are produced from plant tissues by PPases treatment, showed well-conserved cellular components with their rigid cell wall and this characteristic is applicable to preparation of novel food material. The purpose of this study is to investigate the effect of medium composition of PPase production from Bacillus subtilis EK11 which was selected as a PPase producer. Various carbon sources and concentrations on PPase production were studied and corn starch at 0.7% was the most effective for production of PPase. Among the nitrogen sources, yeast extract was the most effective for PPase production and the effect of (NH4)2SO4 was notable as inotganic nitrogen source. Inorganic compounds such as KH2PO4, K2HPO4, Na3-citrate.2H2O and MgSO4 were optimized for PPase production. PPase activity was inhibited by the adition of Ba2+ or Zn2+. The optimal medium for PPase production was devised: 0.7% corn starch, 0.3% yeast extract, 1.4% KH2PO4, 0.6% K2HPO4, 0.1% Na3-citrate.2H2O and 0.02% MgSO4. PPase production by using the optimum medium was carried out with shaking cultivation at 37$^{\circ}C$. The maximum PPase activity of 256unit/ml could be obtained after the cultivation for 48hrs. The activity was increased about 2.2timesthan the activity, 112 unit/ml, in basal medium.

  • 기업의 신기술 개발 역량은 기술 융합 및 발전과 함께 빠르게 변화하고 있는 사업 환경의 치열한 경쟁 속에서 성공적인 제품혁신과 프로세스 혁신을 창출하는 데에 필수적이다. 본 연구는 개방형 혁신의 관점에서 외부 정보 네트워크의 다양성이 중소 벤처기업의 신기술 개발 역량에 미치는 직접 효과와 이러한 직접 효과에 미치는 제품 기획 역량의 매개효과에 대한 연구 모형을 개발한다. 683개의 중소 벤처기업 데이터를 사용하여 최소 자승 회귀 분석을 실시한 통계적 연구모형 검증 결과를 통해, 본 연구는 다음과 같은 2가지의 주요 연구 결과를 제공 한다. 첫째, 중소 벤처기업의 외부 정보 네트워크의 다양성은 신기술 개발 역량에 정(+)의 영향을 미친다. 둘째, 중소 벤처기업의 제품 기획 역량은 외부 정보 네트워크의 다양성이 신기술 개발 역량에 미치는 정(+)의 영향을 완전 매개한다.

Ca$^{2+}$ 농도에 따른 생쥐 초기배의 발생 (The Development of Early Mouse Embryos Depend on $Ca^{2+}$ Concentration)

  • Objective: This study was to determine the effect of different concentration of calcium III medium on the preimplantational development of zygotes and early 2 cell embryos. Methods: Female mice of ICR strain ($5{\sim}8$ weeks old) were superovulated and mated with fertile males. Zygotes or early 2-cell embryos were collected by flushing the oviducts $31{\sim}32$ hours after hCG injection. The embryos were cultured in various concentrations of $Ca^{2+}$ in medium or with EDTA, EGTA and $Ni^{2+}$. Result and Conclusion: Treatment of high concentraion of $Ca^{2+}$ (3.42 mM $(2X){\sim}17.l$ mM (10X)) in medium didn't develop well compared to the control. Low concentrations of $Ca^{2+}$ (0.214mM $(1/8X){\sim}0.855$ mM (1/2X) were deterimental to development beyond 2-cell stage. EDTA, $Ca^{2+}$ chelating agent was treated with ranged concentrations of EDTA (0.014 $mM{\sim}0.107$ mM) to medium contaning 1.71 mM $Ca^{2+}$ showed beneficial effect to development to blastocyst compared to the control. EGTA, extracellular $Ca^{2+}$ chelator, was treated with ranged concentrations of EGTA ($0.014{\sim}0.107$ mM) to the medium contaning 1.71 mM $Ca^{2+}$. There is no significant difference with the control. $Ni^{2+}$ (50 ${\mu}M$), T-type $Ca^{2+}$-channel blocker was treated to medium contaning low concentration of $Ca^{2+}$. It overcame 2-cell block significantly. Rate of degenerated embryos decreased and developmental rate to morula and blastocyst increased more than low $Ca^{2+}$ concentration alone. Further studies are needed for the overcoming effect of 2-cell block by $Ni^{2+}$.

TOE 프레임워크를 활용한 RPA 도입 의도에 미치는 영향 요인 연구 - 중소기업 규모의 조절효과를 중심으로 - (A Study on Factors Affecting the Degree of RPA Patching Using the TOE Framework - Focusing on the Effect of Adjusting the Size of Small and Medium-sized Businesses -)

  • Purpose: By empirically analyzing factors that affect the intention to introduce RPA, we aim to increase understanding of RPA introduction in small and medium-sized businesses and contribute to establishing an effective introduction strategy. The aim is to improve the company's productivity, reduce costs, and strengthen its competitiveness. It also provides policy recommendations for the introduction of RPA. Methods: A survey was conducted to examine whether the technical, organizational, and environmental factors of the TOE framework had an impact on the intention to adopt RPA. We also used stepwise regression analysis to determine whether firm size moderates this relationship. Results: Technical factors, organizational factors, and environmental factors were all found to have a significant impact on small and medium-sized enterprises' intention to adopt RPA. It was confirmed that company size has a moderating effect affecting the intention to adopt RPA. In particular, customer pressure, relative advantage, competitive pressure, age, government support, and the perceived ease of use of RPA was a key determinant of its adoption by small and medium-sized enterprises. Conclusion: This suggests that small and medium-sized businesses should comprehensively consider technical, organizational, and environmental factors when introducing RPA. It is expected to increase understanding of RPA introduction in small and medium-sized businesses, contribute to establishing effective introduction strategies, and contribute to improving company productivity, reducing costs, and strengthening competitiveness.
