Effects of Four Different Feeds on Larval Weight and Survival Rate of Protaetia brevitarsis seulensis (4가지 다른 사료를 흰점박이꽃무지 유충에게 급여시 유충의 체중과 생존율에 미치는 영향)

  • This study evaluated the effect of four different feeds on the larval weight and survival rate of Protaetia brevitarsis seulensis (PBS) over five weeks. Four different types of feed (self-fermentation medium, RM medium, Samsung livestock medium, and bean-curd dregs medium) were given to third-instar larvae of PBS. Weight changes and survival rates of PBS larvae were measured weekly for five weeks. During the experimental period PBS larvae showed the highest growth in Samsung livestock medium, followed by bean-curd dregs medium, RM medium, and self-fermentation medium. Survival rate of PBS larvae were the lowest in bean-curd dregs medium, and then in the Samsung livestock medium. Using the Samsung livestock medium can shorten the delivery period of PBS larvae by approximately 40 days based on the larva shipped on average 75 days. Therefore, this study provides information and data on insect farming techniques useful for insect industrialization.

  • With the acceleration of China's urbanization process, the economic development and market potential of small and medium-sized cities have been constantly enhanced, with the urbanization competition having been caught in the vortex of homogenization. However, small and medium-sized cities are exposed to limited resources and funds, and therefore need to be more flexible and innovative in the strategy implementation, while urban brand communication is one of the key factors to promote the competitiveness of cities. Taking the small and medium-sized cities in China as the research objects, this study, based on the domestic and foreign theories about the brand communication of small and medium-sized cities, as well as other city-related theories, analyzes textual materials about the current situation and new changes in brand communication of small and medium-sized cities with grounded theory, reflecting on the problems in brand communication in China's small and medium-sized cities. Combining the basic elements of urban brand communication, a model for constructing brand communication strategies for small and medium-sized cities is further proposed, so as to provide the differentiated and distinctive strategies for the construction and communication of urban brands in small and medium-sized cities in China, as well as provide a new perspective and strategy on how to enhance the competitiveness and contribute to the sustainable economic development of small and medium-sized.

Microspore-derived Embryo Formation in Response to Cold Pretreatment, Washing Medium, and Medium Composition of Radish (Raphanus sativus L.)

  • Cold pretreatment, washing medium and composition of nutrient media may have marked effects on microspore embryogenesis. When microspores isolated from radish (Raphanus sativus L. cv. Gwanhun) flower buds were washed with Nitsch & Nitsch (NLN) medium liquid medium containing $130g{\cdot}L^{-1}$ sucrose (NLN-13), yields of microspore-derived embryos were greater than when using B5 liquid medium containing $130g{\cdot}L^{-1}$ sucrose. Microspore viability is known to decrease rapidly with storage; however, in this experiment, microspore viability was maintained for 24 h at $4^{\circ}C$ without media. Among the various medium concentrations used ($0.25{\times}$, $0.5{\times}$, $1.0{\times}$, $2.0{\times}$, and $4.0{\times}$ NLN liquid medium), $0.5{\times}$ NLN liquid medium induced the most efficient formation of microspore-derived embryos. In addition, microspore-derived embryos yields were greater when microspores were cultured in $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$, $0.5{\times}$, and $1.0{\times}$ NLN microelements, compared to medium not supplemented with microelements. In this study, the highest yield of microspore-derived embryos was observed when the microspores derived from flower buds were washed using NLN-13 liquid medium and then cultured on $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$ NLN microelements, followed by incubation at $25^{\circ}C$ for 30 days.

  • To investigate exhausted-medium-induced apoptosis in human corneal epithelial(HCE) cells, this study was performed DNA gel electrophoresis, M30 CytoDEATH staining and FAS-FAS ligand ELISA. SV-40 transfected cells were grown to confluency in culture for 7days. The supernatant was harvested and filtered with $0.22{\mu}m$ filter paper. Fresh HCE cells were exposed to the filtered exhausted medium for 1~2 days. Apoptotic cells were prepared for DNA extraction and run the agarose gel for DNA ladder pattern. M30 CytoDEATH was used a tool for easy and reliable determination of very early apoptosis in HCE cells. The control and exhausted medium were assayed for soluble FAS/FAS ligand protein by ELISA. HCE cells exposed to exhausted medium showed a typical DNA ladder pattern. Sporadic M30 CytoDEATH positive cells were detected among HCE cells exposed to exhausted medium. Soluble FAS/FAS ligand levels were not elevated in the exhausted medium compared to the fresh medium control. This study suggests that possible mechanism of exhausted medium induced apoptosis does not include the FAS-FAS ligand system.

The status quo of rapid changing into informationization have been realized at considerable level in Korea. Information society is defined as information technology is main key to determine individuals competitiveness, which can solve effectively the side effect result from industrialization. It cleary imply that information technology is the most promising and important industry in 21st century. Therefore, We should seek to foster independent a small and medium enterprise and develop them corresponding to new concept of a small and medium enterprise in 21st century. The main frame of policy should be new economic system, which can contribute establishment of a small and medium enterprise, management innovation. It also attribute a small and medium enterprise to reveal their creative. New economic paradigm in 21st will be expanded with organization, market, technology. So far, a small and medium enterprise has been acknowledge as economic weaker and the one should be protected. However, In 21st century a small and medium enterprise will be considered as active majority or a source of creative. Development of technology to produce a small quantity with variety product and acceleration of knowledge and informationization will result in comparative merits of a small and medium enterprise. Hereby, The role and relative importance of a small and medium enterprise in our economic will getting be larger and It will be developed as the main force to activate the economic. However, Only a small and medium enterprise, which overcome difficulty with active desire and effort to improve their lot can be developed as a competitive enterprise in 21st century in considering themselves to be developed as diversity, active, independent, business by an enterprise.

A New Selective Medium for Detecting Acidovorax avenae subsp. avenae in Rice Seeds

  • A selective agar medium was developed and tested for the isolation of Acidovorax avenae subsp. avenae, the causal bacterial pathogen of bacterial brown stripe, from rice seeds. The new selective agar medium, designated sorbitol pyroglutamic acid agar (SPA) medium, contained 0.5 g of $K_2$HPO$_4$, 3.0 g of Na$_2$HPO$_4$, 2.0 g of D-sorbitol, 0.2 g of L-pyroglutamic acid, 10.0 $m\ell$ of tween 80, 40.0 mg of victoria blue B, 15.0 g of agar, 150.0 mg of ampicillin and 25.0 mg of vancomycin per litter. Colonies of A. avenae subsp. avenae on SPA medium were smooth, round, convex, shiny, blue and 1.5-2.0 mm in diameter 4 days after incubation at 28$^{\circ}C$. Blue colored colony having dark blue zone was typical type of A. avenae subsp. avenae colonies on the medium. Mean recovery of 8 isolates of A. avenae subsp. avenae on the selective SPA medium was 95.8% in comparison to that on KB medium. The saprophytic bacteria were reduced to 97.9% on SPA medium compared to those on KB medium. Most of other rice seedborne bacteria as well as reported pathogenic bacteria were failed to grow on SPA medium. This medium was highly selective for recovering A. avenae subsp. avenae from rice seed samples, and it could be used to enhance the recovery of this bacterium from rice seed samples, which may be contaminated with large numbers of competing microorganisms.

The Effects of MPPA (Mileages/Points to Purchase Amount) Ratio on Consumers' Preference (구매금액 대비 마일리지/포인트의 비율이 소비자 선호에 미치는 영향)

  • Consumers earn a number of points for every purchase and then they can exchange a specified number of points for a desired reward in a typical loyalty program. The immediate payoff of their effort given as points is not the real reward they actually care about. It is merely an instrument (or medium) which has no value in itself. In a real world, consumers frequently choose the option with a bigger medium even though the economic value of the option is not changed by the medium. We call it 'medium effect'. In this study we explored if the size of medium affects on consumers' preferences. For this we controlled the reward options with three types of medium (small, medium, big) and measured the magnitude of preference difference among the three types of reward options. In addition, we manipulated comparability of reward options with wine and gas discount coupon. We confirmed that choosing one of two wines was easier than that of the two gas coupons. 164 respondents were allocated into three experimental groups and one control group. In three experimental groups, the ratios of the focused reward option's medium to the compared reward option's medium were different. For example, the focused reward option has 10 million points whereas the compared reward option has 10 million points for 1 million won purchase amount in the first group. Then each respondent was asked to choose one of two loyalty programs (focused program vs. compared program) in two different conditions (comparability between reward options: easiness vs. difficulty). To compare the medium effects among the experiment conditions we used chi-squares tests. The empirical results show the medium effect increases and then decreases as the ratio of reward mileages/points to purchase amount increases. Additionally, they let us know that comparability of alternatives affects on the medium effects depending on the ratio of reward mileages/points to purchase amount.

Growth of Strawberry Plantlets Cultured in Vitro in the Agar or Commercial Plug Medium as Affected by Ionic Strength (이온강도에 따른 Agar와 공정육묘용 상토에서 기내배양된 딸기 소식물체의 생육)

  • This study was conducted to investigate the effect of supporting material and ionic strength of the MS medium on growth of strawberry plantlets cultured in vitro for the rapid mass production. Explants of $Fragaria$ $ananassa$ 'Houkouwase' in vitro were planted in the agar or Tosilee (commercial plug medium) medium as the supporting material and supplied with 1/4 MS, 1/2 MS or basal (as the control) MS medium in an autotrophic micropropagation. Plant height and root length were significantly greater when they were cultured in 1/2 MS medium as compared to those grown in the agar medium. Also, shoot fresh and dry weights, and leaf area in the 1/2 MS medium were greater than in 1/4 MS or basal MS medium. When plantlets were cultured in Tosilee medium and fed with the basal MS medium, plant height, root length, shoot fresh and dry weights, root fresh and dry weights, and leaf area were promoted and greater than those in plantlets cultured in the agar medium.

Effects of Doneness on the Microbial, Nutritional, and Quality Properties of Pork Steak of Different Thicknesses

  • This study aimed to evaluate the effect of doneness on the microbial, nutritional, and quality characteristics of 1.5 cm- and 2.0 cm-thick pork neck steaks. Pork neck meat was obtained within 24 h after slaughtering, cut into 1.5 cm- and 2.0 cm-thick slices (n=5), packed in LLD-PE wrap, and stored at $4{\pm}2^{\circ}C$ for 7-10 days until aerobic plate counts (APC) reach 5.51-6.50 Log CFU/g. Then, the pork meat was cooked on a frying pan till it was medium-rare, medium, or well-done. The microbial inhibition rates of the 1.5 cm- and 2.0 cm-thick steak in medium-rare state were 58.26% and 51.70%, respectively, whereas it was 100% for medium-done pork steak of either thickness. The total calories of the 1.5 cm- and 2.0 cm-thick well-done pork steaks were 643.61 kcal/100 g and 675.00 kcal/100 g, respectively, which was higher than that in medium-rare and medium-done steaks. The retention ratios for Fe and K in the well-done steak were significantly lower than those in the medium and medium-rare steak of either thickness (p<0.05). The shear force of the medium-rare and medium steak did not differ, whereas that of the well-done steak was significantly higher than that of the medium-rare steak of either thickness (p<0.05). We observed that the well-done pork steak had tough texture, low mineral content, and high calories. Therefore, consumption of medium and medium-rare pork is more beneficial than that of well-done pork.

Effect of MEM Vitamins Supplementation of In vitro Maturation Medium and In vitro Culture Medium on the Development of Porcine Embryos

  • This study was carried out to examine the influence of minimum essential medium (MEM) vitamins supplementation to in vitro maturation medium and in vitro culture medium on the development of porcine embryos. Porcine embryo development was investigated following cultivation in both in vitro maturation and culture medium with the supplementation of MEM vitamins (0, 0.1, 0.2 and 0.4%) using immature oocytes collected from the ovary of prepubertal gilts. Embryo development was observed and the total cell number in each blastocyst generated under the culture conditions was quantified following supplementation of the medium. The embryonic development rate of the blastocyst and hatched blastocyst was higher, but not significantly so, when 0.4% MEM vitamins were supplemented to the in vitro maturation medium of the porcine oocyte. Interestingly, the total number of cells in the blastocyst was significantly higher in the in vitro maturation MEM vitamins supplemented group compared to either the untreated group or the group which had MEM vitamins supplemented to both in vitro maturation and in vitro culture medium simultaneously (p<0.05). Therefore, the supplementation of 0.4% MEM vitamins to the in vitro mature medium has a beneficial effect on the embryonic development of in vitro produced blastocysts derived from the immature porcine oocytes.
