GA3 처리 왜당귀의 종자 발아, 유묘 생육 및 유용성분 특성 분석 (Effect of GA3 Treatment on Seed Germination, Seedling Growth and Useful Component Content of Angelica acutiloba (Siebold & Zucc.) Kitagawa)

  • 왜당귀[Angelica acutiloba (Siebold & Zucc.) Kitagawa]를 대상으로 종자 발아 및 유묘 생육, 유용성분 함량과 관련된 GA3 처리 효과를 분석하여 왜당귀 재배시 기초자료를 제공하고자 하였다. 왜당귀 종자의 수분흡수율은 침지 2시간이 경과한 시점에서 62.07%를 나타냈고, 48시간이 경과한 시점에서 122.15%로 가장 높은 값을 나타냈다. 왜당귀 종자의 적정 발아온도는 10℃에서 25℃로 확인되었으며, 5℃에서는 발아가 진행되지 않았고, 30℃에서는 GA3 농도가 증가함에 따라 발아율이 유의하게 증가하였다. 또한 유묘 생육은 GA3 처리농도가 높아질수록 양호하였으나 유용성분 함량의 경우 실험구에 따른 유의성이 확인되지 않았다. 이러한 결과는 약용 및 식용으로 활용성이 높은 왜당귀에 대한 최적 재배와 생산기술 정립에 기초자료로 활용할 수 있을 것으로 판단된다.

율무의 조명나방 방제체계 (Control System of Asian corn borer, Ostrinia furnacalis (Guenee) in Coix lacryma-jobi L. var. ma-yuen Stapf)

  • 본 연구는 율무 주산단지인 경기도 북부의 연천 지역에서 율무에 크게 피해를 주는 조명나방에 대한 발생 생태 조사 및 방제적기에 관한 연구를 수행한 결과는 다음과 같다. 율무 조명나방은 연천지역에서 성충 발생은 년간 $2{\sim}3$ 회였고 1, 2, 3 화기 발생최성기는 각각 5월 하순${\sim}$6월 중순, 7월 하순${\sim}$8월 상순, 8월 하순이었으며, 율무 조명나방에 의한 줄기 피해는 $7{\sim}8$월부터 시작하여 10월에 가장 심하였다. 조병나방에 대한 방제약제는 할로스린유제가 93.9%로 방제효과가 가장 높았으며, 효과적 방제적기는 할로스린유제를 7 월하순 fb 8 월 상순 , 8월상순 fb 8월 중순 2회 처리시 방제가가 92.4, 93.5% 로 높았으며 10a당 수량은 무처리구 216kg에 비하여 각각 81%, 82% 증수하였다.

지치의 생육시기 및 재배지역별 시코닌계 색소 함량 및 항산화 활성 (Investigation of Shikonin Pigments and Antioxidant activity of the Roots from Lithospermum erythrorhizon according to the Different Growth Stages and Areas of Cultivation)

  • This study was conducted to evaluate the quality variation of Lithospermum radix on the pigment contents and antioxidant activities according to different growth stages and areas of cultivation. Acetylshikonin contents showed the tendency to decrease gradually from the middle of July (0.28%) to the end of August (0.05%) and then the content was increased again to the end of October (0.25%). Shikonin content was detected as small amount of about 0.009% during the period. The weight of plants was increased from the end of September to the end of October and showed the highest value as 19.8 g on October 25. ROS scavenging activity was the highest in the early of October as $IC_{50}$ value of 0.11 ${\mu}g/mL$. Lithospermum radix of September showed lower ROS scavenging activities than those of other growth stages as $IC_{50}$ value of 1.02 and 0.49 ${\mu}g/mL$ on September 9 and September 27, respectively. Among 17 areas cultivated Lithospermum radix, 10 areas (59%) showed 0.05-0.10% of acetylshikonin contents and 3 areas (18%) were measured to contain 0.16-0.26% of acetylshikonin.

Population structure and regeneration of Zanthoxylum armatum DC. in Salyan, Nepal

  • Background: Zanthoxylum armatum is one of the 30 prioritized medicinal plants for economic development of Nepal with a high trade value. Understanding the ecology of individual species is important for conservation and cultivation purposes. However, relation of ecological factors on the distribution and populations of Z. armatum in Nepal remain unknown. To address this knowledge gap, an attempt has been made to study the population structure, distribution, and regeneration potentiality of Z. armatum. Vegetation sampling was conducted at six different localities of Salyan district along the elevation range of 1,000 m to 2,000 m. Results: Altogether 50 plant species belonging to 44 genera under 34 families were found to be associated with Z. armatum. Significantly higher species richness was found at Rim (1,400-1,700 m) and Chhatreshwori (1,800-2,000 m) and lower at Kupinde (1,600-1,800 m). The highest population density of Z. armatum was at Kupinde (1,600-1,800 m) with a total of 1,100 individuals/ha. and the lowest at Chhatreshwori (1,800-2,000 m) with 740 individuals/ha. Based on the A/F value (Whitford index), it can be said that Z. armatum has random distribution in the study area. The plants were categorized into seedlings, saplings and adults based on plant height and the status of natural regeneration category determined. The regeneration potentiality of Z. armatum in the study area was fair with the average seedlings and saplings densities of 150 and 100 individuals/ha. Respectively. A Shannon-Weinner index mean value of 2.8 was obtained suggesting high species diversity in the study area. Conclusions: The natural distribution and regeneration of Z. armatum is being affected in the recent years due to anthropogenic disturbances. Increasing market demand and unsustainable harvesting procedures are posing serious threat to Z. armatum. Thus, effective conservation and management initiatives are most important for conserving the natural population of Z. armatum in the study area.

Components of Nutraceutical Value in Physalis minima

  • It is of utmost importance to feed the current world population by improving agricultural production with newer varieties of food crops, but what is still more important is to add nutrition into the food. Some of the plants, which are currently growing in the wastelands, contain certain phytochemicals which add to their neutraceutical and health value. These plants contain secondary metabolites which enhance the over all metabolic functions of the body. Withasteroids are one of such phytochemicals. These chemicals are almost exclusively found in plants of the Solanaceae family; one of which, Physalis minima, contains several with asteroids. The aerial parts and roots of P. minima have been found to contain several steroids, identification of which is been discussed in this paper. These with asteroids contribute to the functional value since incorporation of withanolides in the diet may prevent or decrease the growth of tumors in humans.

한약탕제찌꺼기 발효퇴비 처리에 따른 복분자과실의 생리활성 (Effect of Composts Fermented with Korean Medicinal Herb Wastes on Physiological Activity of Rubus coreanus Miquel (Bokbunja))

  • 본 연구는 한약탕제 찌꺼기를 친환경 퇴비자원으로서 재활용하기 위하여 발효과정을 거친 퇴비(MHWC: medicinal herb waste compost)와 계분퇴비(PMC: poultry manure compost) 및 이를 혼합한 퇴비(MHWC+PMC, 1:1)를 식재 2년생 복분자 포장에 무처리(0), 20, 40 Mg/ha로 처리하여 꽃이 첫 개화한 시기를 기점으로 15, 20, 25 DAF(day after flowering)를 수확시기로 하여 수확한 복분자 과실의 생리활성을 조사하여 비교한 결과는 다음과 같다. 복분자의 총 페놀성 화합물 함량은 15 DAF 과실에서와는 달리 20, 25 DAF 과실에서 유의성 있는 증가를 보였다(p<0.05). 수확시기별로는 25>20>15 DAF 순이었고, 그 함량은 40 MHWC Mg/ha가 가장 높았다. Flavonoid 함량변화는 총 페놀성 화합물 함량변화와 유사하였다. 전자공여효과는 15, 20 DAF 과실에서는 차이가 없었지만, 25 DAF 과실에서는 낮은 효과를 나타내었다. 아질산염 소거효과는 pH 1.2>4.2>6.0 순으로 pH 의존적인 경향이었으며 15, 20 DAF 과실보다 25 DAF 과실이 저하되었다. Tyrosinase 저해효과는 MHWC 처리구가 가장 우수하고 처리량에 따라 저해효과가 증가되었으며 15, 20 DAF 과실은 25 DAF 과실보다 낮은 효과를 보였다. Xanthine oxidase 저해활성은 25 DAF 과실의 MHWC 처리구에서 가장 높았고, PMC 및 MHWC+PMC처리구와 UC와의 차이는 인정하기 어려웠다. 이상의 결과에서 MHWC 처리에 따른 복분자 과실의 생리활성이 증가경향을 보여 친환경퇴비자원으로서 가치가 있는 것으로 사료되었다.

Scavenging Capacities of DPPH and ABTS Free Radicals and Anti-inflammatory Activities of Ethanol Extracts and their Fractions from Sophora tonkinensis

  • The first purpose of this study was to evaluate the scavenging capacity (SC) of DPPH and ABTS free radicals for ethanol extract (STR-E) and its active fractions from Sophora tonkinensis root (STR). Four different fractions from STR-E were prepared by using different types of solvents such as chloroform (STR-E-C), ethyl acetate (STR-E-EA), n-butanol (STR-E-B), and water (STR-E-W). STR-E-C showed the highest value of total phenolic content, while STR-E showed the highest value of total flavonoid and terpenoid content. In STR-E and its four fractions, STR-E-EA showed the strongest SC with the lowest SC50 values of the DPPH radicals and ABTS radicals. The second purpose of this study was to evaluate anti-inflammatory activity in the lipopolysaccharide (LPS)-induced RAW 264.7 macrophages treated with STR-E, STR-E-C, and STR-E-EA, respectively. No cytotoxic effect to RAW 264.7 cells was observed at 20 ~ 25 ㎍/ml of STR-E, 10 ㎍/ml of STR-E-C, and 5 ㎍/ml of the STR-E-EA, presenting cell viability values close to that of the untreated control (100%). STR-E, STR-E-C, and STR-E-EA significantly suppressed the LPS-induced nitric oxide (NO) in a dose-dependent manner. Results of reverse-transcription (RT)-qPCR analysis showed that the peak mRNA levels of IL-1β, TNF-α, iNOS, IL-6, and IL-10 were observed in the LPS-stimulated macrophages at 4 h, 2 h, 12 h, 12 h, and 12 h, respectively. The peak mRNA levels of IL-1β, TNF-α, iNOS, and IL-6 were significantly reduced in the LPS-stimulated macrophages co-treated with 20 ㎍/ml and 25 ㎍/ml of STR-E, respectively. In the case of IL-10, its peak mRNA level slightly increased without statistical significance. Compared with the LPS-stimulated macrophages, the peak mRNA levels of IL-1β, TNF-α, iNOS, and IL-6 reduced in the LPS-stimulated macrophages co-treated with 10 ㎍/ml and 20 ㎍/ml of STR-E-C, respectively. In contrast, the peak mRNA level of IL-10 significantly increased at 8 h. Compared with the LPS-stimulated macrophages, the peak mRNA levels of IL-1β, TNF-α, iNOS, and IL-6 reduced in the LPS-stimulated macrophages co-treated with 5 ㎍/ml and 10 ㎍/ml of STR-E-EA, respectively. In contrast, the peak mRNA level of IL-10 increased at 4 h. Taken together, our data indicated that STR-E, STR-E-C, and STR-E-EA activate macrophages to secrete both pro-inflammatory and anti-inflammatory cytokines.

Effect of LED Irradiation on Growth Characteristids of Ginseng Cultivated in Plastic Film House

  • This experiment was carried out using artificial clay and LED in the plastic film house (irradiation time: 08:00~18:00/day). Seedlings (n = 63 per $3.3m^2$) of ginseng was planted on May 17, 2018. LED was combined with red and blue light in a 3:1 ratio and irradiated with different light intensity. The average air temperature from April to September was $12.3^{\circ}C$ $-26.0^{\circ}C$ and it was the the highest at $26.0^{\circ}C$ in August. The test area where fluorescent lamp was irradiated tended to be somewhat higher than the LED irradiation area. The chemical properties of the test soil are as follows. pH levels was 5.3~5.5, EC levels 0.45~0.52 dS/m and OM levels 33~37%. The total nitrogen content was 0.35~0.47% and the available $P_2O_5$ contents was 13.7~16.0 mg/kg, which was lower than the suitable level of 70~200 mg/kg. Exchangeable cations K and Mg contents were within acceptable ranges, but the Ca contents was $28{\sim}38cmol^+/kg$ levels higher than the permissible level ($2{\sim}6cmol^+/kg$). Germination of ginseng leaves took 8~9 days and the overall germination rate was 70~75%. The photometric characteristics of LED light intensity are as follows. The greater the light intensity, the higher the PAR (Photosynthetic Action Radiation) value, illuminance and solar irradiation. Photosynthetic rate was also increased with higher light intensity was investigated at $1.7{\sim}3.2{\mu}mol\;CO_2/m^2/s$. Leaf temperature ($23.7{\sim}24.8^{\circ}C$) by light intensity was the same trend. The growth of aerial parts (plant height etc.) were generally excellent when irradiated with 3 times the light intensity, the growth of the ginseng aerial parts were excellent as follows. The plant height was 42.6 cm, stem length was 25.2 cm, leaf length was 9.6 cm and stem diameter was 5.0 mm. The growth of underground part (root length etc.) was the same, and the root length was 24.4 cm, the tap root length was 6.0 cm, diameter of taproot was 18.2 mm and the fresh root weight was 17.2 g. There were no disease incidence such as Alternaria blight, Gray mold and Anthracnose. Disease of Damping off occurred 2.2~3.6% and incidence ratio of rusty root ginseng was 14.6~20.7%. Leaf discoloration rate was 13.7~48.9% and increased with increasing light intensity. Ginsenoside content of ginseng by light intensity is under analysis.

Effect of Soluble-silicate or Chitosan Foliar Spray on Ginseng Cultivated in Blue-white Plastic Film House

  • The experiments were performed in the Jinan (elevation: 300 meters above sea level), Jeollabuk-do. Seedlings (n = 63 per $3.3m^2$) of ginseng cultivar (Cheonpung, Yeonpung) were planted on April 10, 2015. Shading material of plastic film house was blue-white film. Before the Planting seedling, silicate (3 kg/10 a) or chitosan (40 kg/10 a) was fertilized and foliar sprayed on the leaves 1000 times dilution solution once a month from May to September every year. The growth results of 5-year old ginseng surveyed in 2018 are as follows. The average air temperature in the plastic film house was the highest at $26.6^{\circ}C$ and $26.5^{\circ}C$ in July and August, respectively, and the highest temperature was $40.5^{\circ}C$ in July. The maximum daily temperature of $35^{\circ}C$ or more was 30 days, with the average soil temperature being $24.9^{\circ}C$ in August. The chemical properties of the test soil are as follows. pH was 6.4~6.9 level and EC was 0.35~0.46 dS/m. The organic matter content was 33.5~41.4 g/kg, and available-P content was 251.9~306.8 mg/kg. Exchangeable cations contents, such as K, Ca and Mg were all the appropriate ranges. The soil microbial density surveyed by the dilution plate method was 10~50 times higher than that of control (Non-treatment) and actinomycete density was 3~6 times higher. Pathogens of the genus Fusarium by Metagenome analysis decreased 91.3% and 68.2% respectively in the foliar sprayed of chitosan and soluble-silicate. The light intensity (PAR) in the blue-white film plastic film house gradually increased until July and then decereased, with the average of light intensity in March-October was $120.3umol/m^2/s$. The growth of aerial parts such as plant height and stem length was better than non-sprayed group in silicate or chitosan treatments and Yeonpung cultivar was superior to the Cheonpung cultivar. The SPAD value was higher in Yeonpung cultivar foliar sprayed with soluble-silicate. The growth of underground parts such as root length and taproot length were better in chitosan and soluble-silicate treatment than control, especially in Yeonpung cultivar foliar sprayed with chitosan was good in taproot length and taproot diameter, and fresh weight of root was 60.1 g. Ginsenoside contents were 24.9 mg/g and 22.4 mg/g, in the Cheonpung cultivar foliar sprayed with soluble-silicate or chitosan respectively, 28% and 15% higher than control (19.5 mg/g). The incidence of disease by Alteraria panax and Botrytis cinerea was 3~9% and 4~9%, respectively. High temperature damage rate was 3~5%.

Pharmacological Characterization of (10bS)-1,2,3,5,6,10b-hexahydropyrrolo[2,1-a]isoquinoline Oxalate (YSL-3S) as a New ${\alpha}_2$-Adrenoceptor Antagonist

  • ${\alpha}_2$-Adrenoceptor antagonists, which can enhance synaptic norepinephrine levels by blocking feedback inhibition processes, are potentially useful in the treatment of disease states such. as depression, memory impairment, impotence and sexual dysfunction. (10bS)-1,2,3,5,6,10b-Hexahydropyrrolo[2,1-a]isoquinoline oxalate (YSL-3S) was evaluated in several in vitro biological tests to establish its pharmacological profile of activities as an ${\alpha}_2$-adrenoceptor antagonist. Saturation binding assay revealed that$^{3}[H]$rauwolscine bound to the $\alpha$$_2$-adrenoceptors with a Kd value of 6.3$\pm$0.5 nM and a Bmax value of 25l$\pm$39 fmol/mg protein in rat cortical synaptic membranes. Competitive binding assay showed that YSL-3S inhibited the binding of$^3[H]$rauwolscine (1 nM) in a concentration-dependent manner with a Ki value of 98.2$\pm$12.1 nM while it did not inhibit the binding of [$^3$H]cytisine (1.25 nM) to neuronal nicotinic cholinergic receptors. The Ki values of yohimbine, clonidine and norepinephrine for $^3[H]$rauwolscine binding were 15.8$\pm$1.0, 40.1$\pm$5.9 and 40.0$\pm$11.5 nM, respectively. In addition, the binding affinity of YSL-3S for ${\alpha}_2$-adrenoceptors was higher than that of its antipode and the racemic mixture. The functional activity of YSL-3S at the presynaptic ${\alpha}_2$-adrenoceptors was assessed using the prostatic portion of the rat vas deferens. Clonidine inhibited field-stimulated contractions of the vas deference in a dose-dependent manner. The presence of YSL-3S or yohimbine caused a parallel, rightward the dose-response curve of clonidine in a dose-dependent manner, indicating an antagonistic action at the presynaptic ${\alpha}_2$-adrenoceptors. The $pA_2$values of yohimbine and YSL-3S were 7.66$\pm$0.13 and 6.64$\pm$0.18, respectively. The results indicate that YSL-3S acts as a competitive antagonist at presynaptic ${\alpha}_2$ -adrenoceptors with a potency approximately ten times lower than yohimbine, but is devoid of binding affinity for neuronal nicotinic cholinergic receptors.
