On the Chemical Properties of Nursery Soil in Cultivation of Panax ginseng (인삼포상토(人蔘圃床土)의 화학적(化學的) 성상(性狀)에 관(關)한 연구(硏究))

  • The cultivation of ginseng plant (Panax ginseng C.A. Meyer) in Korea as an eminent medicinal herb may be traced far back in history. However, the practices in cultivation have not much improved in terms of efficiency and scientific farming. In the present study some experiments were undertaken for the search of the soil and nutrition conditions, because of the nutritional requirement of ginseng plant shaws quite unique compared with other crops. In both the seed bed and the field 'Yakto' has been traditionally employed or the prime source of nutrition of the crop. Yakto is a complex matter prepared from raw foliage of the broad-leaved trees as the main portion with the admixture of a variety of organic nitrogen source through fermentative processes. The composition of Yakto may be classified coarsely into the decomposed and undecomposed substances, the former being further fractionated according their solubilities, comprising also various colloidal matters whose composition and structure are yet to be known. The Yakto-fractions were subjected to analyze for search of its nature and coarse composition in terms of the distribution of nitrogen, contents of organic functional groups such as -COOH, phenolic-OH, alcholic-OH and methoxyl and hydrolysable sugars. Furthermore, absorption-spectra of each fraction were determined in visible and infrared region and compared the results each other.

Physiological Activities using Root and Stem Extracts of Cymbidium (심비디움 뿌리 및 줄기 추출물의 생리 활성)

  • Cymbidium is one of perennial herbs belonging to the Orchidaceae and is known as a medicinal plant. However, its scientific data are insufficient. The purpose of this study is to extract from root and stem of Cymbidium, to investigate the biological effects of them. Cymbidium antibacterial effects of the extracts were performed by antibacterial test against Staphylococcus aureus (S. aureus), Staphylococcus saphrophyticus (S. saprophyticus), Proteus vulgaris (P. vulgaris) and Klebsiella pneumonia (K. pneumonia). Antioxidant effects of the extracts were carried out by DPPH radical scavenging. Total phenolic contents were also determined. Moreover, Cell viability of extracts against MTT assay was cell viability against HepG2 cell and also measures Cholesterol adsorptivity of extracts. In this study, the extracts inhibited the growth of bacteria. Particularly Cymbidium root extracts by only ethanol extraction showed highest antimicrobial effect against S. aureus. The Cymbidium stem extracts by both ethanol extraction and sonication for 1 hour had higher antioxidant activites as well as total phenolic contents. Cell cytotoxicity showed higher than $50{\mu}g/mL$. Cholesterol adsorptivity showed lower than 20%. These results suggest that the Cymbidium might be a source of anti-bacterials and anti-oxidants.

Characteristics of Tofu Added with Soybeans Cultured by Mycelia of Pleurotus eryngii (큰느타리버섯 균사체를 증식시킨 콩을 첨가제조한 두부의 품질특성)

  • The effects of the addition of soybeans cultured by mycelia of Pleurotus eryngii (SMP) on the characteristics of tofu were studied. Protein and ash contents of SMP were higher than those of untreated soybeans: 43.23 and 6.34% for treated soybeans, respectively, and 40.42 and 5.90% for untreated soybeans. But lipid and carbohydrate contents of SMP were lower. For minor elements, Mg and Ca contents of SMP were higher and P was lower than control. Tofu could be manufactured when SMP was added below 25% to the untreated soybean. In scanning electron microscopic observation, tofu tended to break down as the ratio of SMP was over 25% to the untreated soybean. The addition of SMP $(5{\sim}15%)$ increased the yield of tofu to $1.5{\sim}3.5%$. In textural characteristics, hardness of tofu increased as the ratio of SMP increased up to 20%. Cohesiveness, chewiness, springiness and gumminess were high in tofu which was made with $5{\sim}15%$ SMP. Savory taste and overall acceptability of the tofu prepared with $5{\sim}15%$ SMP were higher than those of control when evaluated by sensory test.

Quality and Sensory Characteristics of White Bread added with Various Ginseng Products (인삼제품의 첨가에 따른 제빵적성 및 관능평가)

  • This study investigated the effect of ginseng products on the baking properties of white breads. Flour was substituted by ginseng products (ginseng at levels 2, 4, 6, and 8%, all w/w, of flour levels). Both ginseng powder (GP) and red ginseng powder (RGP) were used. Similarly, red ginseng extract (RGE) was substituted at levels of 1, 2, 3, and 4% (all w/w) of flour. The physical properties of each dough were assessed using farinograms, extensograms, and amylograms. Water absorption increased as the proportions of ginseng products rose. The dough surface areas and R/E (resistance/extensibility) values decreased, as did peak viscosities, at the proportions of ginseng products increased. The pH values during fermentation decreased as the proportions of ginseng products increased. The fermentation power of dough with GP was lower than that of control, and higher than that of dough with RGP or RGE, but the addition of ginseng products beyond certain levels weakened the gas retention power: The specific loaf volumes of breads with 2% GP were the highest at 5.41 mL/g. In breads with RGE, the specificloaf volume increased from 5.52 mL/g to 5.82 mL/g as RGE levels rose from 0% to 4%. Hardness increased with rising GP and RGP levels in breads with GP and RGP, but decreased with RGE levels in breads with RGE. The moisture contents of breads during storage tended to be higher than control in breads with ginseng products. Lightness increased with addition of GP and decreased upon addition of RGP or RGE, while redness and yellowness increased after addition of any ginseng products. In sensory evaluation tests, the sensory scores for texture, color, mouth feel, and overall acceptability, were high, when any ginseng product (GP, RGP or RGE) was present at 2%. Of these breads, the bread with 2% RGE attained the highest sensory score.

Biosynthesis of Organic Germanium Using Cordyceps militaris (동충하초를 이용한 유기게르마늄의 생산)

  • In the present study, an attempt has been made to produce a high quality medicinal mushroom Cordycops militaris supplemented with organic Ge. Cordycops militaris was cultivated in SDAY liquid medium containing yeast extract 10 g, peptone 10 g, glucose 40 g per liter and chrysalis media supplemented with inorganic Ge at 100, 500, 1000, and 5000 ppm concentrations. The greatest organic Ge production of 442.4 ppm/g and 284 ppm/g were observed in SDAY liquid and Chrysalis media cultures supplemented with 100 ppm inorganic Ge respectively. Similarly, 4,509.7 ppm/g and 1,058 ppm/g of organic Ge were obtained from liquid and chrysalis media cultures supplemented with 5,000 ppm and 1,000 ppm inorganic Ge, respectively. In addition, higher concentration of organic Ge was obtained in mycelia than fruiting bodies. These results indicate that the concentration of organic Ge increase with decreasing inorganic Ge concentration in the medium. This is the first report on production of high valuable Cordyceps militaris contained with organic Ge.

  • This study was conducted to evaluate the effect of systematic herbicide application and polyethylene(PE) film mulching in direct seeded Angelica gigas Nakai. Freshly-collected-seed showed 86% of emergence rate and required 18 days of emergence duration. Cold treatment of seeds increased emergence rate by more than 90% and shortened 2 days of emergence duration. Paraquat) 1,1'-dimethyl-4,4'-bipyridylium dichloride was the most effective when applied just before emergence of A. gigas and provided 96% of emergence rate and 91% of weeding effect when applied 23 days after seeding. Soil applied herbicides used did not cause any phytotoxicity on emergence with application of 2-fold recommended rate and gave more than 90% of weeding effect. Black PE film mulching gave excellent emergence rate and weeding effect, and shortened emergence duration by 8 days.

Effects of Size of Rhizome on Top Part Growth in Saururus chinensis Baill (종근 크기가 삼백초의 경엽생육에 미치는 영향)

  • This study was conducted to provide basic data for productivity of Saururus chinensis Baill. depending on rhizome size. The kinds of the treatment were 4 including below 5mm of rhizome size. Major results were summarized as follows. Sprouting was earlier in above 9mm of rhizome size and the number of last sprouting was increased $84{\sim}109%$ compared with 8.0 $individual/m^2$ of 6mm of rhizome size. Growth of stem showed a good tendency in 6mm of rhizome size with the longer stem length and the thicker stem but the number of tiller was more rhizome was more thicker. Though the number of commercial leaf was more above 9mm of rhizome size leaf length, leaf width and the number of noncommercial leaf did not showed difference by rhizome size. Dry weight of commercial foliar in 9mm of rhizome size over was increased $35{\sim}37%$ compared with 6mm of rhizome size. Therefore, rhizomes size above 9mm showed most effective for increasing sprout and wight of stem and leaves of Saururus chinensis.

Selecting marker substances of main producing area of Codonopsis lanceolata in Korea using UPLC-QTOF-MS analysis (UPLC-QTOF-MS분석를 이용한 국내산 더덕 주산지의 표지물질 선정)

  • Codonopsis lanceolata (Deoduk) was grown in East Asia, including Korea, China, Japan, and Russia, and the roots of C. lanceolata have been used as functional foods and traditional medicine to treat symptoms of cough, bronchitis, asthma, tuberculosis, and dyspepsia. The phytochemicals of C. lanceolata have been reported such as phenylpropanoids, polyacetylenes, saponins, and flavonoids that are involved in pharmacological effects such as anti-obesity, anti-inflammation, anti-tumor, anti-oxidant, and anti-microbial activities. Selecting marker substances of the main producing area by MS-based metabolomics analysis is important to ensure the beneficial effect of C. lanceolata without side-effects because differences in cultivated areas of plants were related not only to the safety of medicinal plants but also to changes in chemical composition and biological efficacy. In our present study, ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry combined with multivariate statistical analysis was applied to recognize the main producing area of C. lanceolata in South Korea. As a result of Principal Component Analysis and loading plot analysis of three groups, Inje (Kangwon-do), Hoengseong (Kangwon-do), and Muju (Jeonlabuk-do), several secondary metabolites of C. lanceolata including tangshenoside I, lancemaside A, and lancemaside G, were suggested as potential marker substances to distinguish the place of main producing area of C. lanceolata.

Inhibitory Effects of Ethanol Extract of Rhodiola Sacra on Endoplasmic Reticulum Stress in Neuro-2A Cells (설치류 Neuro-2A 신경세포에서 홍경천 에탄올 추출물의 소포체 스트레스 억제효과)

  • Growing evidence suggests that mediating apoptotic cell death of ER stress plays an important role in pathological development of neurodegenerative diseases including Alzheimer's disease. The ethanol extract of Rodiola sacra (ERS) investigates whether ER stress protects neuroinvasive neuro-2A cells from homocysteine (Hcy) cell death and ER stress. In neuronal cells, Hcy markedly decreased the viability of the cells and induced the death of Annexin V-positive cells as confirmed by MTT assay. The Hcy cell viability and apoptotic loss pretreated with ERS were attenuated, and Hcy showed stress in the expression of C / EBP homologous protein, 78-kDa glucose regulatory protein and the junction of X-box binding protein-1 (xbp1) mRNA. ESR decreased Hcy-induced mRNA binding, GRP78 and CHOP cells induced Hcy-induced ER stress and apoptosis, and Western blotting revealed expression of heme oxygenase-1 and HO-1 enzyme activity Inhibition is indicative of therapeutic value for neurodegenerative diseases such as decreased cell death by hemin.

Spatial Distribution and Host Plants of the Ramulus koreanus (Phasmida; Phasmatidae) in Korean Thistle Cultivation (고려엉겅퀴 재배지에서 발생한 우리대벌레 공간분포 및 기주식물)

  • The Korean thistle (Cirsium setidens) is grown in Gangwon province as a food ingredient called gondre. Based on regular monitoring of Korean thistle cultivation, we detected local outbreaks of a stick insect causing severe damage. Here we report the identification of the stick insect and its ecological characteristics. The survey involved 3 farms located in Jeongeon-gun, Gangwon province and lasted from May 28 to October 1 in 2019. According to morphological and DNA analyses the stick insect was identified as Ramulus koreanus Kwon Ha and Lee. a member of the family Phasmatidae. Its outbreak in the Korean thistle farms was observed from June 11 to August 22 with peak density on July 23. Spatial distribution analyses, using Taylors power law and Greens index showed that R. koreanus was uniformally distributed. Food consumption rate of Korean thistle leaves was estimated as 60.98±4.35 ㎠/day/adult. The main food plants were identified as Juglans mandshurica and Aronia melanocarpa. Further study needs to pursue the mechanism of the outbreak and its possible impact on agricultural and forest ecosystems. Additionally, means to relieve crop damage need to be identified.
