Isolation and Characterization of Exogenously Expressed Calmodulin from Endogenous Tobacco Calmodulin by Anion-exchange Fast Protein Liquid Chromatography

  • A Mono Q HR 5/5 anion-exchange column with a FPLC system was used to separate exogenously expressed calmodulin from endogenous tobacco calmodulins. Transgenic tobacco calmodulins were purified by fractionation with ammonium sulfate, precipitation with sulfuric acid and hydrophobic chromatography on phenyl-Sepharose CL-4B. The purified calmodulins were chromatographed in the FPLC using the column. This method was selected because of the slight differences in the net charge of foreign and endogenous plant calmodulins due to amino acid sequence differences. By this approach, the exogenously expressed calmodulin was isolated from endogenous tobacco calmodulins. The isolated calmodulin was characterized by amino acid composition analysis as well as methylation analysis.

Synthesis of Model Microspheres and Adsorption Study of Bovine Albumin (모델 Microspheres의 합성 및 Bovine Albumin의 흡착)

  • Microspheres are expected to be applied to biomedical areas such as solid-phase immunoassays, drug delivery systems, Immunomagnetic cell separation. To synthesize micro-spheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio or synthesized microspheres was always smatter than 1.05. And the surface charge density (or the number of ionizable functional groups) of the microspheres synthesized by "two stage shot growth method" was 6-13 times higher than thats of the ml crospheres synthesized by conventional seeded batch copolymerization. As a previous step for biomedical application, adsorption experiments of bovine albumin on microspheres were carried out under various conditions. The maximum adsorbed amount was obtained in the neighborhood of pH 4.5. Isoelectric point of bovine albumin Is pH 5.0, so experimental result shows that it shifted to acid area. The adsorption Isotherm was obtained, the plateau region was always reached at 2.Og/L (bulk concentration of bovine albumin ) . The effect of the kind and the amount of surface functional group was also examined.p was also examined.

  • Endoscope system is the device that observe interior the body. It has some demerits because it use of optical fibers. The resolution of images depends on the number of optical fibers, it is impossible that several people observe at the same time and it needs special camera in order to record images. In order to overcome these demerits, electro endoscope system using CCD(Charge Coupled Device) has been developed recently. If the diameter of insertion tube is thinner, it is easier to be examined by thin endoscope system. At the present time, we develope these type of electro endoscope system in the department of SNU biomedical engineering. Moreover, we are researching an algorithm of the 3D-reconstruction of interior of the gaster. In this paper, a method for 3D information detection using electroendoscopic image sequence has been presented.

  • The purpose of this paper is to review recent developments in lower limb exoskeletons. The exoskeleton system is a human-robot cooperation system that enhances the performance of the wearer in various environments while the human operator is in charge of the position control, contextual perception, and motion signal generation through the robot's artificial intelligence. This system is in the form of a mechanical structure that is combined to the exterior of a human body to improve the muscular power of the wearer. This paper is followed by an overview of the development history of exoskeleton systems and their three main applications in military/industrial field, medical/rehabilitation field and social welfare field. Besides the key technologies in exoskeleton systems, the research is presented from several viewpoints of the exoskeleton mechanism, human-robot interface and human-robot cooperation control.

  • Digital FHR monitoring system based on the personal computer combined wi th the digital signal processing (DSP) board was implemented. The DSP board acquires and digitally processes ultrasound fetal Doppler signal for digital rectification, envelope detection, autocorrealtion function calculation and its peak position detection. The personal computer interfaced with the DSP board is in charge of graphic display, hardcopy, data taransmission and on-line analysis of fetal heart rate change including on-line warning system, base-line estmation, acceleration, deceleration and variability. The advantages of PC-DSP based system can be summarized as follows: 1) Minimum hardware realization. 2) Increased flexibility. 3) Total care system.

  • In the electronic, medical, aerospace and automobile industries, many products and parts are manufactured by joining. Recently, as these get smaller, micro joining is becoming more and more important. In this study, micro wire-to-micro wire parallel joining was performed using single mode fiber laser. Maximum power of the fiber laser is 100 W. The CCD(Charge- Coupled Device, CCD) camera to observe the specimen was made up. The objective was applied to micro joining system to make a small spot size of laser beam. In order to control the target position, micro-multi-axis-stage was set up. This paper presents results for the single mode fiber laser joining of micro wires.

  • Finasteride, ($5{\alpha}$, $17{\beta}$)-N-(1,1-dimethylethyl)-3-oxo-4-azaandrost-1-ene-17-carboxamide), is a 5a-reductase enzyme inhibitor. This enzyme converts testosterone to the more potent androgen, a-dihydrotestosterone. This molecules a logical medical treatment for benign prostatic hyperplasia (BPH), as it induced a reduction in serum dihydrotestosterone and prostatic specific antigen levels with a concomittant increase in blood testosterone concentration . Despite its widespread use, little has been published concerning its molecular properties. Therefore, in this study, in order to explain characteristics of finasteride, total energy, net charge, vibrational mode of melatonin are calculated by PM3 methods of HyperCam 8.0.

  • Functional dysphonia (FD) is a voice disorder in the absence of structural or neurologic laryngeal pathology. FD is not a single disease but a disease entity. Therefore several voice disorders, which have completely different pathogenesis, are included in this category. The first step of treatment of FD is differentiating patient's voice symptoms from other organic voice disorders and other functional voice problems. Several different treatment modalities are included in the managements of FD. Voice therapy is in charge of the main role in treatment of FD. Medical treatment is also necessary when patient has general problems which would affect voice production. Vocal folds mucosal lesions can cause FD even the lesion is minor. In this case proper surgical intervention helps to improve the symptom of FD. Psychiatric consultation should be considered when the patient has psychological problems.

  • Delayed luminescence of plant leaves was imaged by a 2-D cooled charge-coupled device. We report the delayed luminescence imaging of normal/injured leaves picked ami the leaves intact. The luminescent intensity was lower in leaf veins, scars and edge cut. The intensity of delayed luminescence from intact leaves was higher than that of picked leaves. These results indirectly support the argument that the delayed luminescence of a photosynthetic system is closely related to the electron transfer process of PSII in the thylakoid membrane.
