TRAIL Based Therapy: Overview of Mesenchymal Stem Cell Based Delivery and miRNA Controlled Expression of TRAIL

  • Rapidly increasing number of outstanding developments in the field of TRAIL mediated signaling have revolutionized our current information about inducing and maximizing TRAIL mediated apoptosis in resistant cancer cells. Data obtained with high-throughput technologies have provided finer resolution of tumor biology and now it is known that a complex structure containing malignant cells strictly coupled with a large variety of surrounding cells constitutes the tumor stroma. Utility of mesenchymal stem cells (MSCs) as cellular vehicles has added new layers of information. There is sufficient experimental evidence substantiating efficient gene deliveries into MSCs by retroviral, lentiviral and adenoviral vectors. Moreover, there is a paradigm shift in molecular oncology and recent high impact research has shown controlled expression of TRAIL in cancer cells on insertion of complementary sequences for frequently downregulated miRNAs. In this review we have attempted to provide an overview of utility of TRAIL engineered MSCs for effective killing of tumor and potential of using miRNA response elements as rheostat like switch to control expression of TRAIL in cancer cells.

  • Background and Purpose : To investigate the wild plants to be used for survival on tactical activity of Special Forces soldiers at deep enemy's territory Material and Methods : To investigate the wild plants, I selected various native plants in the forests and fields that can be used for medical cure and eatables. And I selected some poisonous plants and some plants containing water or salts, after due consideration of tactical activity and use in the battle-fields. Results and Conclusion : There are many wild plants to be used for multiple purpose in our country. We can get from these plants emergency medicines, foods, even water and salts. I chose 6 species of digestives, 11 species of analgesics, 9 species of antiphlogistics, 7 species of hemostatics, 5 species of antidiarheal, 5 species of diuretics, 2 species of laxatives, 7 species of antipyretics, 1 species of vermicide and 45 species of eatable plants and 12 species of poisonous plants. and I summarized general name, scientific name, component, picking season, used portion, taking method of these plants. Building up accurate knowledge on the wild plants could contributed to raise survival rate of Special Forces soldiers in the battle-fields.

익모초 추출물의 항산화 작용에 관한 연구 (Study of Antioxidation Action of Lenonuri herba Extract)

  • 익모초(Leonurus sibiricus L)는 한국, 중국, 일본, 아시아 각지에 야생으로 자생하는 2년생 초본으로 꿀풀과(Labiatae) 식물로 높이 1.5${\~}$2m 정도 자란다. 약효로는 부인병의 각종 자궁질환, 활혈, 조경, 이뇨, 소증 효과는 물론 혈변, 혈뇨, 치질, 대${\cdot}$소변 불통 등의 치료제로 사용한다. 그 외에 고혈압, 강심작용, 항암연구도 활발하다. 익모초의 $CHC1_3$ 엑스와 MeOH 엑스를 $H_2O$, $30\%$ MeOH, $60\%$ MeOH, $100\%$ MeOH로 분획하였고, 이 5가지 분획물들에 대하여 activity guided fractionation 방법에 따라 DPPH free radical에 대한 scavenging activity와 LDL의 lipid peroxidation을 이용한 TBARS assay로 항산화 활성실험을 측정하였다. 그 결과 익모초의 MeOH 엑스는 $30\%$ MeOH, $60\%$ MeOH 분획물이 항산화 활성이 증가하였으며, Vitamin C와 비교해 볼 때 강한 활성을 나타내었다. 또한 pyrogaliol을 이용한 SOD 활성에서도 250 ppm에서는 Vitamin C 보다 우수한 유사 활성을 보였다. 이상과 같이 익모초로부터 비교적 강한 항산화 활성을 확인할 수 있어 천연항산화 기능성 화장품의 개발이 가능하리라 보여 진다.

P53 Polymorphism at Codon 72 is Associated with Keratocystic Odontogenic Tumors in the Thai Population

  • Objective: To clarify the association between the p53 polymorphism at codon 72 and susceptibility to the sporadic keratocystic odontogenic tumor (KCOT). Design: One hundred KCOTs and 160 match-group healthy controls were genotyped to ascertain the frequency of the p53 codon 72 polymorphism using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), confirmed by direct sequencing. Results: The frequencies of the Pro/Pro, Arg/Pro, and Arg/Arg genotypes were 23.8%, 49.4%, and 26.9%, respectively, in the controls, while the KCOT cohort demonstrated 43.0%, 39.0%, and 18.0%, respectively. Further analysis suggested that p53 Pro could be a KCOT-susceptible allele (OR (95%CI)=1.77 (1.22 to 2.59), p=0.0024), with a sex-adjusted OR (95%CI) of 1.71 (1.17-2.50), p=0.0046. Moreover, the results indicated that p53 codon 72 Pro homozygous was associated with a two-fold risk of developing KCOT (adjusted OR (95%CI) =2.17(1.23-3.84), p=0.0062). Conclusions: The C/C genotype of P53 gene codon 72 increases the risk of developing sporadic KCOT and may be a useful tool for screening and diagnostic purposes.

Antibacterial potential of the extracts derived from leaves and in vitro raised calli of medicinal plants Pterocarpus marsupium Roxb., Clitoria ternatea L., and Sanseveiria cylindrica Bojer ex Hook

  • Pterocarpus marsupium, Clitoria ternatea, and Sanseveiria cylindrica are some of the important and endangered medicinal plant species of India. Despite of medicinal properties, antibacterial potential of the plants have not yet been explored. The present study was designed to optimize the in vitro technique for micropropagation and to screen the extracts from leaves and in vitro raised calli for antibacterial properties. Excised leaf-explants from the parent plants were surface sterilized and cultivated on Murashige & Skoog's (MS) medium containing $N^6$-benzyladenine (BA) in concentrations of 1, 2, 5, and $10{\mu}M$. Optimal growth of calli was noticed at a concentration of $5{\mu}M$, therefore the extracts from calli grown at this concentration were further studied for antibacterial activity. Both alcoholic and aqueous extracts from leaves of respective plants, and their in vitro raised calli were tested for antibacterial activity by agar well diffusion method against a range of Gram-positive and Gram-negative bacteria. Aqueous extracts showed antibacterial activity against limited number of bacterial species; notably the extracts of C. ternatea which showed antibacterial activity against Streptococcus pyogenes, Bacillus subtilis and Bacillus cereus. Alcoholic extracts of all three plants showed antibacterial activity against a wider range of bacteria. Among the Gram-positive bacteria, extracts from C. ternatea showed strong antibacterial activity against Bacillus spp., whereas the extracts of S. cylindrica showed good antibacterial potential for Staphylococcus aureus, S. epidermidis and S. pyogenes. The extracts from all three plants showed antibacterial activity against Gram-negative bacteria, including, Salmonella spp. and Shigella dysenteriae; organisms causing enteric fever and dysentery. In most of the cases, the extracts from respective calli showed comparable, and in some cases better, result in comparison to the extracts from parent leaves. To the best of our knowledge this is the first preliminary report on antibacterial potential, especially through calli extracts, of these plants; and in vitro cultivation of the explants may be used to obtain phytotherapeutic compounds.

  • The HyangyakBoncho(鄕藥本草) in Vol. 76-85 of the Hyangyakjipseongbang(鄕藥集成方) deals with the Hyangyak, medicinal stuffs produced in the territory of Chosun from the late times of Corea to the early times of Chosun. It also includes whole knowledges and the way to process(?製) of Oriental medical botany(本草) in that period of Chosun and China. It contains all the knowledges of the Hyangyak and the Boncho(鄕藥本草學) so that it can be helpful for the Chosun to apply to medicines. This study shows the characters of HyangyakBoncho(鄕藥本草) through analyzing. In future it is supposed that this report is the first approach to the core of the Hyangyakjipseongbang(鄕藥集成方), which describes medicines with prescriptions made up of the Hyangyak mainly.

  • The Purpose of this study was to find the characteristics of the medicine mainly using the Hyangyak(鄕藥) in the early Chosun by analyzing the text in ${\ulcorner}$Hyangyakjipseongbang(鄕藥集成方)${\lrcorner}$ and comparing it with other medical books. To start out this grand investigation, the research was focused on ${\ulcorner}$Sanghanmun(傷寒門)${\lrcorner}$, Vol.5-8 of ${\ulcorner}Hyangyakjipseongbangs{\lrcorner}$. The conclusion made in this study is as follows: 1. All of the prescriptions in ${\ulcorner}Sanghanmun{\lrcorner}$, was made up of the Hangyak listed in ${\ulcorner}$HyangyakBoncho(鄕藥本草)${\lrcorner}$, Vol.76-85 of ${\ulcorner}Hyangyakjipseongbangs{\lrcorner}$ 2. The main purpose of ${\ulcorner}Sanghanmun{\lrcorner}$ was to make sure it was easy to use for people with no medical backgrounds. To have a more specific understanding on the medicine mainly llsing the Hyangyak, further research should be done on how the catergory of the Hangyak changed. This should be done by comparing the books written after the ${\ulcorner}Hyangyakjipseongbangs{\lrcorner}$, and also on the characteristics of the usage of prescriptions in ${\ulcorner}Hyangyakjipseongbangs{\lrcorner}$.

라이프니츠와 인삼 (Leibniz and ginseng)

  • 위대한 철학자이자 미적분을 발견한 수학자로 잘 알려진 라이프니츠(Gottfried Wilhelm Leibniz, 1646~1716)가 인삼에 대해 물었다는 사실은 거의 알려져 있지 않다. 유럽의 계몽주의를 열었던 라이프니츠는 왜 인삼에 관심을 가지게 되었을까? 이 글은 라이프니츠가 남긴 방대한 기록 속에서 인삼을 찾아내고, 개인적 생애와 당대의 지적 흐름을 엮어 그가 인삼에 관심을 갖게 된 경로를 추적한 작업이다. 16세기부터 유럽에서는 약물지와 같은 고대 그리스 텍스트의 재발견과 해외에서 들여온 새로운 식물들로 인해 본초학이 크게 발달하게 되었다. 같은 맥락에서 '구대륙' 중국에서 만병통치약으로 이름을 떨치던 인삼 또한 여행기 등을 통해 유럽에 소개되기 시작했다. 중상주의 기치 하에 유용한 약용식물에 대한 관심이 커 가던 유럽에서는 과학단체들을 중심으로 본격적으로 인삼에 대한 연구가 시작되었다. 라이프니츠는 그런 연구의 중심지였던 왕립학회와 프랑스 왕립과학원을 방문하고 학자들과 교류를 나누었으며 독일에 그와 같은 연구기관을 설립하는 것을 평생의 목표로 삼았다. 라이프니츠는 로마를 방문했을 때 예수회 선교사들과 긴밀하게 교류하는 기회를 갖게 되었다. 그 과정에서 중국에 대한 지적 호기심이 한층 더 깊어갔고, 그 맥락에서 인삼에 대한 관심도 커갔다. 라이프니츠는 그리말디, 부베 등 중국선교에서 핵심적인 활동을 했던 학자들과 인삼의 효능에 대한 정보를 주고받게 된다. 이 논문은 유럽의 계몽주의에서 전혀 주목받지 못했던 인삼이 17세기 말 유럽의 학문적 지형과 지식인들 사이의 교류, 중상주의적 해외팽창을 얽어내는 단초가 될 수 있음을 제시한 것이다.

Cytotoxic Activity of Biosynthesized Gold Nanoparticles with an Extract of the Red Seaweed Corallina officinalis on the MCF-7 Human Breast Cancer Cell Line

  • Background: Nano-biotechnology is recognized as offering revolutionary changes in the field of cancer therapy and biologically synthesized gold nanoparticles are known to have a wide range of medical applications. Materials and Methods: Gold nanoparticles (GNPs) were biosynthesized with an aqueous extract of the red alga Corallina officinalis, used as a reducing and stabilizing agent. GNPs were characterized using UV-Vis spectroscopy, transmission electron microscopy (TEM), energy dispersive analysis (EDX) and Fourier transform infra-red (FT-IR) spectroscopy and tested for cytotoxic activity against human breast cancer (MCF-7) cells cultured in Dulbecco's modified Eagle medium supplemented with 10% fetal bovine serum, considering their cytotoxicty and effects on cellular DNA. Results: The biosynthesized GNPs were $14.6{\pm}1nm$ in diameter. FT-IR analysis showed that the hydroxyl functional group from polyphenols and carbonyl group from proteins could assist in formation and stabilization. The GNPs showed potent cytotoxic activity against MCF-7 cells, causing necrosis at high concentrations while lower concentrations were without effect as indicated by DNA fragmentation assay. Conclusions: The antitumor activity of the biosynthesized GNPs from the red alga Corallina officinalis against human breast cancer cells may be due to the cytotoxic effects of the gold nanoparticles and the polyphenolcontent of the algal extract.

First Report of the Ash Dieback Pathogen Hymenoscyphus fraxineus in Korea

  • In the past two decades, European ash trees (Fraxinus spp.) have been severely damaged due to ash dieback disease, which is caused by the fungal species Hymenoscyphus fraxineus (Chalara fraxinea in the anamorphic stage). Recent molecular phylogenetic and population genetic studies have suggested that this fungus has been introduced from Asia to Europe. During a fungal survey in Korea, H. fraxineus-like apothecia were collected from fallen leaves, rachises, and petioles of Korean ash and Manchurian ash trees. The morphological and ecological traits of these materials are described with the internal transcribed spacer rDNA sequence comparison of H. fraxineus strains collected from Korea, China and Japan.
