The Selective Visualization of Lignin Peroxidase, Manganese Peroxidase and Laccase, Produced by White Rot Fungi on Solid Media

  • A visual method for the selective screen Eng of lignin degrading enzymes, produced by white rot fungi (WRF), was investigated by the addition of coloring additives to solid media. Of the additives used in the enzyme production media, guaiacol and RBBR could be used for the detection of lignin peroxidase (LiP), manganese peroxidase (MnP) and lactase. Syringaldazine and Acid Red 264 were able for the detection of both the MnP and lactase, and the LiP and laccase, respectively, and a combination of these two additives was able to detect each of the ligninases produced by the WRF on solid media.

Utilization of Robinia pseudoacacia as Sawdust Medium for Cultivation of Edible and Medicinal Mushrooms

  • This study was undertaken to examine the feasibility of black locust (Robinia pseudoacacia) as substrates for several edible mushrooms. For the cultivation of several edible and/or medicinal fungi on black locust, optimum bulk densities, synthetic or semisynthetic additives, natural additives and pretreatment methods were investigated. Fruit body yields of the fungi on various sawdust media composed of different wood species were also analyzed for testing the capability of black locust as a substrate for mushroom production. Mycelial growths decreased proportional when the bulk density increased. The most suitable carbon and nitrogen sources as additives to promote the mycelial growth were sucrose (2%, w/w) and ammonium phosphate (0.2%, w/w) respectively. When corn-powder and beer-waste as natural additives were added to sawdust of black locust showed the significant growth of mycelia. And the optimum mixing ratio was 10:2:1 (sawdust: corn-powder: beer-waste, w/w). Black locust after cold water treatment showed the outstanding mycelial growths. Any significant changes of pH, moisture content (%) and dry-weight losses (%) could not be found among culture substrates (sawdust of black locust, oak and poplar wood) examined before and after harvesting of fruit bodies. Yield of fruit bodies on black locust culture media were comparable with those culture media composed with oak and poplar wood. The present work indicated strongly the potentiality of black locust as raw materials for edible and medicinal mushrooms.

Factors influencing somatic embryo maturation, high frequency germination and plantlet formation in Terminalia chebula Retz.

  • The factors influencing somatic embryo maturation, high frequency somatic embryo germination, and plantlet formation were studied in Terminalia chebula Retz. Maturation of somatic embryo were influenced by a number of factors such as in vitro culture passage, concentrations of sucrose, levels of abscisic acid (ABA), basal media and media additive combinations. Maximum frequency of somatic embryo maturation ($57.22{\pm}2.02$), was obtained on MS medium supplemented with 50 g/l sucrose. Different factors such as strengths of MS nutrients, plant growth regulators, media additives and their combinations controlling somatic embryo germination and plantlet formation were studied. High frequency of germination and plantlet formation ($58.80{\pm}1.47$) were achieved by subsequent subculture of mature somatic embryos on MS medium containing 30 g/l sucrose and 0.5 mg/l benzyl-adenine (BA). However, although duration of in vitro passage of the callus tissue was critical, contribution of the combinations of plant growth regulators and media additives showed nugatory effect on somatic embryo maturation and germination as evident from variable responses.

A Study on the Recognition and Education of Food Additives in Middle School Students (중학생들의 식품첨가물에 대한 인식과 교육에 관한 연구)

  • The purpose of this study is to offer youth with the basic materials for developing nutrition education programs and to help domestic science teachers in schools implement effective dietary education by examining their awareness of food additives. In the source of acquiring knowledges on food additives, the results were through mass media, technology and home economics teachers, and family members in order. The majority of students thought that they don't need the education about food additives. As effective teaching methods, they first selected the use of visual media, followed by experimental cooking classes, field trip, and inquiry lessons using processed foods. As a result of analyzing the education on food additives in accordance with demographic factors, more number of female students, compared to male students depicted the need for education on food additives. Further, the lower the cost students spend on purchasing processed foods per day, the higher the necessity of the education they indicate. The necessity of education content on food additives showed significant difference depending on the cost of buying processed foods, and the degree of interest and help real-life according to gender indicated significant differences. The satisfaction with education on food additives, which was implemented in home economics education revealed significant differences according to gender. This study aims to provide the basic data for the development and research of educational programs regarding good eating habits among the general youth. However, there are limitations to the presentation of the practical training program. For this reason, based on the results of this study, further studies should follow this study in order to develop and study educational programs related to food additives for teaching and learning purposes.

Trends of Perception and Information Needs on Food Additives of Children and Parents by Analyzing the Safety Assessment Reports of Food Additives in 2008~2013 (어린이와 학부모의 식품첨가물에 대한 인식 및 정보요구도 추이 분석: 2008~2013 식품첨가물 섭취 안전성 평가 연구 결과를 중심으로)

  • This study was performed in order to grasp the trends of elementary school children and their parents on their purchasing behavior of processed foods, awareness of food additives and its education experience by analyzing the safety assessment reports of food additives in 2008~2013. The most important factor in purchasing processed foods was safety in both groups followed by nutrition in parents and taste in children, respectively. While purchasing foods, the first item that is checked has been shifted from food additives to the origin of the products. Parents still perceived food additives as the most hazardous factor for food safety; however, recently, children began to regard microbial contaminants as being most hazardous, which is regarded as a desirable educational effect. The most concerned food additives were preservatives, synthetic seasoning and colorants in both groups. However, the awareness level on food additives still remained low as 3.0~3.1/5.0 for parents and 2.4~2.9/5.0 for children. Educational experience on food additives increased in children from 12% in 2008 to 25% in 2013; however, it decreased in parents from 23% in 2008 to 15% in 2013. Information needs for food additives by education and promotion were very high both in parents (4.2~4.5) and children (3.8~4.1). Both groups had an interest in the safety, legal standards of food additives, and foods with food additives, in order. The most reliable resource institutions on food additives were university/research institute and hospital for parents, but, hospital and government for children. The preferred media on food additives were TV and the internet for parents, and school newsletter and TV for children. Overall, the above results demonstrated that the perceptions on food additives did not change much with parents during the last 6 years; however, children's perceptions began to show improvement with the increase of educational experience. Hence, the government needs to make efforts to increase the trust level of consumers by developing educational tools and providing educational experiences including mass media for the promotion of risk communication on food additives.

Comparative Analysis on the Perceptions for Food Additives Between Elementary School Teachers and Nutrition Teachers (식품첨가물에 대한 초등교사와 영양교사의 인식 비교)

  • Literacy on food additives of elementary school teachers (ET) and nutrition teachers (NT) could be influential factor on safe dietary education for school children. Therefore, the perceptions and information needs on food additives were surveyed from 351 elementary school teachers and nutrition teachers in metropolitan area of Korea, and the basic data for the promotion of risk communication on food additives among them were obtained. Compared to ET who consider 'taste' (39.1%) as the most important factor while purchasing food, NT considered 'safety' (68.1%) first (p < 0.001). Among the food labelling items, the level of understanding on food additives was the lowest both in ET (3.53) and NT (4.17), and NT showed better levels of understanding overall on food labels. Both ET and NT regarded hazardous factors of food as environmental pollutants, foodborne pathogens, and food additives in order, and tended to select 'no additives' or 'no artificial color' products while purchasing processed food. Although NT answered that they know all food additives had been passed the evaluation of safety and effectiveness tests (100%) and have standards of use (81.9%), majority of them (87.5%) believed the consumption of food additives are harmful on human health. ET (75.2%) also regarded food additives as dangerous materials. Above results suggested the necessity of proper and enough risk communication for both ET and NT. Both ET and NT wanted to have information on the safety or hazard of food additives. Most preferred media to get the information on food additives was TV (3.80) among ET and lecture (3.65) among NT. ET and NT trusted hospital, research institution/universities or the personnels working in these institutions as the provider of information on food additives. The result that the trust levels of ET and NT on government were relative low suggested the weakness of risk communication in Korean government. Although ET and NT answered that they do not trust mass media, their behaviors were affected by them such as reading food labels in ET (39.4%) and reducing the consumption of food additives in NT (50%). They also indicated mass media's problem of sensitive approach on food additives and asked the urgent reaction of government by providing sound information through experts on food additives. Above results revealed that ET and NT have different perceptions and information needs on food additives, therefore, proper risk communication should be provided for them to serve as dietary educators for elementary school children.

Production of Environment-friendly Artificial Media for Agriculture Using Urban Sludge (도시발생 슬러지를 이용한 환경친화적 인공배지 생산)

  • Large amount of sludge have been generating in the process of water and wastewater treatment in urban area, and it has been making many environmental problems. Currently almost of sludge is landfilled, and since sludge is difficult to handle and dehydrate, the permeated water from the filled-in ground contaminate the surrounding soil and groundwater which may cause serious environmental and sociological problems. The organic component in sludge can be almost removed through the heat treatment process, and the final product is called artificial soil or artificial media according to the temperature control. To produce artificial media using sludge, chabazite and lime were used as an additive, and the mixture of sludge & additives was thermally treated in the firing kiln at about 800~1, 100。C for about fifteen minutes. The physical and chemical characteristics of the produced artificial media were analyzed, and it showed that it can be used as an artificial media for plant production or soil conditioner for farmland. The concentrations of the toxic heavy metals in the artificial media were lower than the soil quality standard for farmland. The characteristics of produced artificial media, using the mixture of sludge and additives through the heat treatment, is similar to the natural chabazite and soil. The analyzed result of the mineral composition of artificial media showed that it has a characteristics similar to natural stable soil, so the produced artificial media may be applied to farmland or water culture without causing adverse effect. Therefore this study showed that the above process can be a feasible alternative for sludge treatment.

  • The fundamental dispersion property of Si2N4 and a combination of AlN and Nd2O3 as sintering additives in a variety of organic solvents such as alcohols, hydrocarbons, ketones, and ethers was investigated. The stabilization mechanism and interaction between organic functional groups of the various organic additives were studied to clarify the dispersibility of the ceramic particles in the nonaqueous suspending medium. characterization of the suspensions was based mainly on electrokinetic sonic amplitude(ESA) measurements and the flow curves obtained from the rheological studies as well as estimated Hamaker constants. It was found that the contribution of electrostatic repulsive forces to the Si3N4, AlN and Nd2O3 stabilization in organic media is appreciably greater than anticipated and is dependent on the physicochemical properties of organic solvents.

Adsorption Kinetics for Polymeric Additives in Papermaking Aqueous Fibrous Media by UV Spectroscopic Analysis

  • The general objective of the present study was to investigate the potential application of the UV spectroscopic method for determination of the polymeric additives present in papermaking fibrous stock solutions. The study also intended to establish the surface-chemical retention model associated with the adsorption kinetics of additives on fiber surfaces. Polyamide epichlorohydrin (PAE) wet strength resin and imidazolinium quaternary (IZQ) softening agents were selected to evaluate the analytical method. Concentrations of PAE and IZQ in solution were proportional to the UV absorption at 314 and 400 nm, respectively. The time-dependent behavior of polymeric additives obeyed a mono-molecular layer adsorption as characterized in Langmuir-type expression. The kinetic modeling for polymeric adsorption on fiber surfaces was based on a concept that polymeric adsorption on fiber surfaces has two distinguishable stages including initial dynamic adsorption phase and the final near-equilibrium state. The simulation model predicted not only the real-time additive adsorption behavior for polymeric additives at high accuracy once the kinetic parameters were determined, but showed a good agreement with the experimental data. The spectroscopic method examined on the PAE and IZQ adsorption study could potentially be considered as an effective tool for the wet-end retention control as applied to the paper industry.
