In this study, a mechanistic model of cutting force components in up and down end milling process is presented. Using this cutting force model of 4-tooth endmills with various helix angles, cutting force variation of inconel 718 has been predicted. Predicted values of cutting force components are coincide well with the measured ones. As helix angle increases, overlapping effects of the active cutting edges increase. In up endmilling the magnitudes of radial and feed cutting force componts FX and FY are lowest when the helix angle is $40\{\circ}$, but in down endmilling the magnitudes of these values increase slightly as helix angle becomes large.
The baculovirus/insect cell expression system is of great value in the study of structure-function relationships in mammalian glucose-transport proteins by site-directed mutagenesis and for the large-scale production of these proteins for mechanistic and biochemical studies. In order to exploit this, the effects of substitution at the highly conserved residue glutamine 282 of the human erythrocyte-type glucose transporter have been examined by in vitro site-directed mutagenesis. The modified human transport protein has been expressed in Spodoptera frugiperda 21 cells by using the recombinant baculovirus AcNPV-GTL. To assess the functional integrity of the expressed transporter, measurements of the transport inhibitor cytochalasin B binding were performed, involving the membranes prepared from 4 days post infection with no virus, with wild-type virus or AcNPV-GTL virus. Data obtained showed that there was little or no D-glucose-inhibitable binding in cells infected with the wild type or no virus. Only the recombinant virus infected cells exhibited specific binding, which is inhibitable by D- but not by L-glucose. However, there was a notable reduction in the affinity for the potent inhibitor cytochalasin B when binding measurements of AcNPV-GTL were compared with those of AcNPV-GT, which has no substitution. It is thus suggested that although the modified and unmodified human transporters differed slightly in their affinity for cytochalasin B, the glutamine substitution did not interfere the heterologous expression of the human transporter in the insect cells.
The market of rice transplanter has already been changed for riding and to be turned large size of chassis. Because an automation is rapidly expanded, It is strongly need to analyze a mechanism and develop a hydraulic circuit. In this study, we analyzed the mechanism of working equipment of rice transplanter for riding and developed hydraulic circuits. We proposed the operating mechanism of rice transplanter for riding through the mechanistic analysis of working equipment. And the simulation and experiment were performed. In order to up and down the working equipment of rice transplanter for riding, we designed the mechanism which was installed hydraulic circuit and hydraulic cylinder, and it was manufactured. The pressure of developed hydraulic circuit was set by 800 $N/cm^2$. In the field testing, the hydraulic response presented at 50 msec in the maximum driving velocity, 0.8 m/sec of rice transplanter for riding, and it was well performed. The results of experiments showed the system characteristics sufficient to use as the hydraulic mechanism for a rice transplanter for riding.

We isolated a lignan, kobusin from Geranium thunbergii and studied its effect on the expression of inducible nitric oxide synthase (iNOS) gene in a monocyte/macrophage cell line, RAW264.7 cells. Kobusin inhibited lipopolysaccharide (LPS)-stimulated NO production and the expression of iNOS in a concentration-dependent manner. To identify the mechanistic basis for its inhibition of iNOS induction, we examined the effect of kobusin on both the luciferase reporter activity using $NF-{\kappa}B$ minimal promoter and the nuclear translocation of p65. Kobusin suppressed the reporter gene activity and the LPS-induced movement of p65 in to nucleus. $NF-{\kappa}B$ activation is controlled by the phosphorylation and subsequent degradation of $I-{\kappa}B{\alpha}$, and in the present study, we found that $I-{\kappa}B{\alpha}$ phosphorylation was also inhibited by kobusin. Our findings indicate that kobusin may provide a developmental basis for an agent against inflammatory diseases.
Control of plant diseases is largely dependent on use of agrochemicals. However, there are widening gaps between our knowledge on plant diseases gained from genetic/mechanistic studies and rapid translation of the knowledge into target-oriented development of effective agrochemicals. Here we propose that the time is ripe for computer-aided drug discovery/design (CADD) in molecular plant pathology. CADD has played a pivotal role in development of medically important molecules over the last three decades. Now, explosive increase in information on genome sequences and three dimensional structures of biological molecules, in combination with advances in computational and informational technologies, opens up exciting possibilities for application of CADD in discovery and development of agrochemicals. In this review, we outline two categories of the drug discovery strategies: structure- and ligand-based CADD, and relevant computational approaches that are being employed in modern drug discovery. In order to help readers to dive into CADD, we explain concepts of homology modelling, molecular docking, virtual screening, and de novo ligand design in structure-based CADD, and pharmacophore modelling, ligand-based virtual screening, quantitative structure activity relationship modelling and de novo ligand design for ligand-based CADD. We also provide the important resources available to carry out CADD. Finally, we present a case study showing how CADD approach can be implemented in reality for identification of potent chemical compounds against the important plant pathogens, Pseudomonas syringae and Colletotrichum gloeosporioides.
We investigated the effects of ${\beta}$-glucan purified from Paenibacillus polymyxa JB115 on the viability and proliferation of splenocytes. Splenocytes play a critical role in host immunity. MTT assays and trypan blue exclusion tests revealed that ${\beta}$-glucan significantly promoted the viability and proliferation of splenocytes over a range of concentrations. However, there was no specific subset change. ${\beta}$-glucan protected splenocytes from cytokine withdrawal-induced spontaneous cell death. For further mechanistic studies, ELISA assay revealed that ${\beta}$-glucan enhanced the expression of anti-apoptotic molecules and interleukin 7 (IL-7), a cytokine critical for lymphocyte survival. We also investigated the IL-2 dependency of ${\beta}$-glucan-treated splenocytes to determine if treated cells could still undergo clonal expansion. In flow cytometric analysis, ${\beta}$-glucan induced increased levels of the activation marker CD25 on the surface of splenocytes and ${\beta}$-glucan-treated splenocytes showed higher proliferation rates in response to IL-2 treatment. This study demonstrates that ${\beta}$-glucan can enhance the survival of splenocytes and provides valuable information to broaden the use of ${\beta}$-glucan in research fields.
Plasma ionized calcium ($Ca^{2+}$) concentrations are tightly regulated in the body and maintained within a narrow range; thus it is challenging to quantify calcium absorption under normal physiologic conditions. This study aimed to develop a mechanistic model for the parathyroid hormone (PTH) response after calcium intake and indirectly compare the difference in oral calcium absorption from PTH responses. PTH and $Ca^{2+}$ concentrations were collected from 24 subjects from a clinical trial performed to evaluate the safety and calcium absorption of Geumjin Thermal Water in comparison with calcium carbonate tablets in healthy subjects. Indirect response models (NONMEM Ver. 7.2.0) were fitted to observed $Ca^{2+}$ and PTH data, respectively, in a manner that absorbed but unobserved $Ca^{2+}$ inhibits the secretion of PTH. Without notable changes in $Ca^{2+}$ levels, PTH responses were modeled and used as a marker for the extent of calcium absorption.

Various analytical procedures for the structural behavior of concrete pavement system are studied in order to identify a suitable method which will be incorperated in the pavement management system using nondestructive test and mechanistic evaluation. A typical four layered system is adopted and analyzed using 3 dimensional finite elements, plane strain elements, plates with Winkler foundation, and plates with Burmister foundation. Numerical analysis results of various structural analysis procedures are compared and analized based on displacements and stresses. It is concluded, after analysis of merits and demetits of the procedures, that Burmister foundation analysis procedure is the most sui-table procedure for implementation for the analysis of stresses and displacements because of its accuracy and simplicity.
Natural pterocarpan Medicarpin (Med) has been shown to have various beneficial biological roles, including inhibition of osteoclastogenesis, stimulation of bone regeneration and induction of apoptosis. However, the effect of the Med on lipolysis in adipocytes has not been reported. Here, we show the effect of Med on lipolysis in different mouse adipocytes and elucidate the underlying mechanism. We observed that Med treatment promoted release of glycerol in the media. Differentiated mouse brown adipose tissue cells were treated with Med. RNA-Seq analysis was performed to elucidate the effect of med and subsequently was confirmed by qRT-PCR and western blotting analyses. Med treatment increased both protein and gene expression levels of hormone-sensitive lipase (Hsl) and adipose triglyceride lipase (Atgl), which are two critical enzymes necessary for lipolysis. Mechanistic study showed that Med activates Protein Kinase A (PKA) and phosphorylates Hsl at PKA target position at $Serine^{660}$. Silencing of PKA gene by short interfering RNA attenuated the Med-induced increase in glycerol release and Hsl phosphorylation. The results unveil that Med boosts lipolysis via a PKA-dependent pathway in adipocytes and may provide a possible avenue of further research of Med mediated reduction of body fat.
Hydrogen safety is one of important issues for future public usage of hydrogen. When hydrogen is released in a compartment, the occurrence of detonation must be prohibited. In order to evaluate the possibility of DDT (Deflagration to Detonation Transition) in the compartment with the hydrogen release, sigma-lambda criteria which were developed from experimental data are commonly used. But they give a little conservative results because they do not consider the detailed geometrical effect of the compartment. This is the main reason of the need to mechanistic combustion model for evaluation of hydrogen flame propagation and acceleration. In this study, sigma-lambda criteria and combustion model were systematically applied to evaluate a possibility of DDT in a IRWST compartment of APR1400 nuclear power plant during a hypothetical accident. A combustion model in an open source CFD code OpenFOAM has been applied for analyses of hydrogen flame propagation. The model was validated by evaluating the flame acceleration tests conducted in FLAME facility. And it was applied to evaluate the characteristics of a hydrogen flame propagation in the IRWST compartment of APR1400.
