Genomic Heterogeneity of Chicken Populations in India

  • A comprehensive genome profiling study was undertaken based on automated genotyping and analysis of 20 microsatellite markers that involved 155 birds representing eight different populations. The distribution of microsatellite markers in each of these breeds helped us to decipher genetic heterogeneity, population genetic structure and evolutionary relationships of the present day chicken populations in India. All the microsatellite loci utilized for the analysis were polymorphic and reasonably informative. A total of 285 alleles were documented at 20 loci with a mean of 14.25 alleles/locus. A total of 103 alleles were found to be population/strain specific of which, only 30 per cent had a frequency of more than 10. The mean PIC values ranged from 0.39 for the locus ADL158 to 0.71 for loci MCW005 or ADL267 across the genomes and 0.55 in Dahlem Red to 0.71 in Desi (non-descript), among the populations. The overall mean expected and observed heterozygosity estimates for our populations were 0.68 and 0.64, respectively. The overall mean inbreeding coefficients (FIS) varied between -0.05 (Babcock) and 0.16 (Rhode Island Red). The pairwise FST estimates ranged from 0.06 between Aseel and Desi (non-descript) to 0.14 between Dahlem Red and Babcock. The Nei's genetic distance varied from 0.30 (WLH-IWD and WLH-IWF) to 0.80 (Dahlem Red and Babcock. Phylogenetic analysis grouped all the populations into two main clusters, representing i) the pure breeds, Dahlem Red and Rhode Island Red, and ii) the remaining six populations/strains. All the chicken populations studied were in the state of mild to moderate inbreeding except for commercial birds. A planned breeding is advised for purebreds to revive their genetic potential. High genetic diversity exists in Desi (non-descript), local birds, which can be exploited to genetically improve the birds suitable for backyard poultry.

Left Atrial Velocity Vector Imaging Can Assess Early Diastolic Dysfunction in Left Ventricular Hypertrophy and Hypertrophic Cardiomyopathy

  • BACKGROUND: The function of left atrium (LA) is difficult to assess because of its ventricle-dependent, dynamic movement. The aim of this study was to assess LA function using velocity vector imaging (VVI) and compare LA function in patients with hypertrophic cardiomyopathy (HCMP) and left ventricular hypertrophy (LVH) with normal controls. METHODS: Fourteen patients with HCMP (72% male, mean age of 52.6 ± 9.8), 15 hypertensive patients with LVH (88% male, mean age of 54.0 ± 15.3), and 10 age-matched controls (83% male, mean age of 50.0 ± 4.6) were prospectively studied. Echocardiographic images of the LA were analyzed with VVI, and strain rate (SR) was compared among the 3 groups. RESULTS: The e' velocity (7.7 ± 1.1; 5.1 ± 0.8; 4.5 ± 1.3 cm/sec, p = 0.013), E/e' (6.8 ± 1.6; 12.4 ± 3.3; 14.7 ± 4.2, p = 0.035), and late diastolic SR at mid LA (-1.65 ± 0.51; -0.97 ± 0.55; -0.82 ± 0.32, p = 0.002) were significantly different among the groups (normal; LVH; HCMP, respectively). The e' velocity, E/e', and late diastolic SR at mid LA were significantly different between normal and LVH (p = 0.001; 0.022; 0.018), whereas LA size was similar between normal and LVH (p = 0.592). The mean late diastolic peak SR of mid LA was significantly correlated with indices of diastolic function (E/e', e', and LA size). CONCLUSIONS: The SR is a useful tool for detailed evaluation of LA function, especially early dysfunction of LA in groups with normal LA size.

탄성계수 감소곡선에 근거한 철도노반의 회복탄성계수 모델 개발 및 평가 (Development and Assessment for Resilient Modulus Prediction Model of Railroad Trackbeds Based on Modulus Reduction Curve)

  • 본 연구에서는 국내 철도 토공노반 재료로 가장 흔히 사용되는 입도조정쇄석, 화강풍화토, 암버럭-토사 혼합 재료에 대해 평균유효주응력과 축변형률의 함수로 표현되는 회복탄성계수 예측모델을 결정하였다. 회복탄성계수 예측모델은 대표적인 동적물성치인 변형률에 따른 전단탄성계수 감소곡선의 표현과 같이 최대영탄성계수와 정규화 영탄성계수 감소곡선으로 구성된다. 평균유효주응력의 함수로 표현되는 최대영탄성계수의 모델인자는 $A_E$$n_E$이고, 비선형 영역의 정규화 영탄성계수 감소곡선은 기준변형률(${\varepsilon}_r$)과 곡률계수(a)를 모델인자로 하는 수정 쌍곡선 모델로 표현된다. 제안된 회복탄성계수 예측모델을 검증하기 위해 3차원 다층탄성해석 프로그램(GEOTRACK)을 이용하여 평택 시험 철도노반의 탄성거동을 평가하였고, 화물열차 및 여객열차가 시험구간을 통과할 때 계측한 노반의 수직 탄성변위와 비교하였다. 현장계측은 자갈도상 아래의 재료가 각각 입도조정쇄석과 양질의 화강풍화토인 두 개소에서 수행되었다. 자갈도상 아래에서 계산된 수직 탄성변위는 대략 0.6mm 이내였고 계측 결과와 잘 일치하였다. 본 연구를 통해 제안된 회복탄성계수 예측모델이 열차하중에 의한 노반의 탄성거동을 적절히 표현하고 있음을 확인하였다.

Effect of Lactobacillus plantarum FH185 on the Reduction of Adipocyte Size and Gut Microbial Changes in Mice with Diet-induced Obesity

  • This study aimed to investigate the effects of Lactobacillus plantarum FH185 on the reduction of adipocyte size and gut microbial changes in mice with diet-induced obesity. The strain was found to have a lipase inhibitory activity of 70.09±2.04% and inhibited adipocyte differentiation of 3T3-L1 cells (18.63±0.98%) at a concentration of 100 µg/mL. To examine the effect of the strain supplementation on gut microbial changes in mice with diet-induced obesity, male C57BL/6J mice were fed on four different diets (i.e., A, normal diet (ND); B, high-fat diet (HFD); C, HFD with ABT-3 (109 CFU/day); and D, HFD with L. plantarum FH185 (109 CFU/day)) for 6 wk. According to the results of fecal pyrosequencing, the ratio of Firmicutes to Bacteroidetes in groups C and D was lower than in the control groups at the phylum level. At the family level, Lactobacillaceae in groups C and D was observed to dominate, while Lachnospiraceae in groups A and B was observed to dominate. At the genus level, Lactobacillus in groups C and D was comparatively higher than in groups A and B. To examine the effects of strain supplementation on the reduction of adipocyte size, the left and right epididymal fat pads were quickly isolated after the animals were sacrificed, and the adipocyte sizes were measured. In groups A, C and D, the percentage of 2,000 m2 of adipocyte was higher than in the other size of adipocyte, while the percentage of over 5,000 m2 of adipocyte was highest in group B. The mean adipocyte size of group D was significantly larger than that of group A, but smaller than that of group B.

Insecticidal Activity of Ethyl Acetate Extracts from Culture Filtrates of Mangrove Fungal Endophytes

  • In the search for novel potent fungi-derived bioactive compounds for bioinsecticide applications, crude ethyl acetate culture filtrate extracts from 110 mangrove fungal endophytes were screened for their toxicity. Toxicity tests of all extracts against brine shrimp (Artemia salina) larvae were performed. The extracts with the highest toxicity were further examined for insecticidal activity against Spodoptera litura larvae and acetylcholinesterase (AChE) inhibition activity. The results showed that the extracts of five isolates exhibited the highest toxicity to brine shrimp at 50% lethal concentration ($LC_{50}$) values of 7.45 to 10.24 ppm. These five fungal isolates that obtained from Rhizophora mucronata were identified based on sequence data analysis of the internal transcribed spacer region of rDNA as Aspergillus oryzae (strain BPPTCC 6036), Emericella nidulans (strains BPPTCC 6035 and BPPTCC 6038), A. tamarii (strain BPPTCC 6037), and A. versicolor (strain BPPTCC 6039). The mean percentage of S. litura larval mortality following topical application of the five extracts ranged from 16.7% to 43.3%. In the AChE inhibition assay, the inhibition rates of the five extracts ranged from 40.7% to 48.9%, while eserine (positive control) had an inhibition rate of 96.8%, at a concentration of 100 ppm. The extracts used were crude extracts, so their potential as sources of AChE inhibition compounds makes them likely candidates as neurotoxins. The high-performance liquid chromatography profiles of the five extracts differed, indicating variations in their chemical constituents. This study highlights the potential of culture filtrate ethyl acetate extracts of mangrove fungal endophytes as a source of new potential bioactive compounds for bioinsecticide applications.

Intronic Sequences for the Discrimination of Silkworm Strains in Silkworm Powder

  • After a serial study on the therapeutic efficacy of the powdery silkworm for diabetics was positively resulted in, many powdery silkworm products were on the market in Korea. Up to now, however, no causal method is available to discriminate the strain of silkworms that is a major ingredient for manufacturing powdery silkworm, even though the quality of the powdery silkworm differs greatly by source and origin of strains. We previously were successful in identifying 25 silkworm strains kept in Korea using nine intronic sequences. In this study, we tested the utility of the nine intronic sequences to distinguish the most widely reared silkworm strains originated from Korea and China. Two intron regions, PTTH Intron3 and PTTH Intron3, showed a substantial sequence divergence (mean sequence divergence of 3.13% in PTTH Intron3 and 4.99% in PTTH Intron3). These two intronic sequences provided no identical sequences among the seven strains tested. Thus, these sequences each along can be used to discriminate the seven strains tested in this study. Furthermore, other intron regions, except for VDP Intron4 allowed us to discriminate $2{\sim}4$ strains by strain-specific unique insertion/deletion or substitution.

모래의 미소변형 전단탄성계수에 대한 고결영향 분석 (Analysis of cementation effects on Small-strain Shear Modulus of Sand)

  • The small-strain shear modulus ($G_{max}$) of uncemented sand is affected by the the mean principal stress and void ratio, and it has been known that the cementation and aging also affect to $G_{max}$ of sand. For extensive understanding about the effect of cementation on the $G_{max}$ of sand, a series of bender element tests was conducted on the cemented specimens prepared in a large calibration chamber by pluviation of the sand-gypsum mixture. It was observed from the experimental results that the $G_{max}$ of cemented sand is higher above 10 times than value of uncemented one, and it increases exponentially with the gypsum content increases. Whereas, the increase of the vertical stress from 50kPa to 200kPa and the relative density from 40% to 80% result in 20~30% and 2 times increase of $G_{max}$, respectively. It means that the gypsum content, that is cementation level, is the most influential factor on the $G_{max}$ of cemented sand. In addition, the effect of relative density on $G_{max}$ was more apparent on cemented sand than uncemented one.

파래첨가 사료가 이스라엘계 잉어(Cyprinus carpio)의 성장 및 혈액성상에 미치는 영향 (Effects of Dietary Enteromorpha compressa on Growth and Blood Properties in Israeli Strain of Common Carp (Cyprinus carpio))

  • 이스라엘 잉어에 대하여 파래첨가 사육시 성장 및 혈액성상에 미치는 영향을 조사하기 위하여 파래분말을 각각 0, 1, 5 및 $10\%$ 첨가한 사료를 이용하여 4개월간 사육하면서 어체의 증체율, 비만도, 사료효율 및 혈액학적인 성상을 비교 조사하였다. 증체율은 파래 첨가농도에 비례하여 감소하였으며, 투여기간에 따라서 약간의 차이가 있었으나 전기간동안 증체율의 감소를 보였다. 비만도는 대조구에 비해서 $1\%$ 첨가군에서는 전사육기간에 약간 증가되는 경향을 보였으나, $5\%$ 첨가군에서는 전 사육간을 통하여 현저한 증가를 보였다. 한편, $10\%$첨가군에서는 사육기간에 비례감소 되었다. 사료효율은 대조구에 비해서 파래 첨가농도 및 사육일수에 무관하게 저하되었으며, 특히 $10\%$ 군에서는 사육일수에 비례하여 현저하게 저하되었다. 혈액성분은 첨가군에서 총단백, Hb, GOT 및 GPT등은 차이가 인정되지 않았으며, 알부민 및 글루코스치등은 증가되었으나, Ht값은 첨가농도에 비례하여 감소되었다.

불포화토에 대한 반복삼축압축시험의 요소시뮬레이션을 위한 응력-변형율 관계의 수립 (Inducing stress-strain relationship for element simulation of cyclic triaxial test on unsaturated soil)

  • 부(-)의 간극수압으로서 불포화토 내에 작용하는 석션은 입자간 응력을 증가시키며, 이에 따라 토립자 골격의 항복응력 및 소성전단강성을 증대시키는 등, 불포화토의 역학적 특성에 지대한 영향을 미친다. 따라서, 본 연구에서는 이러한 석션의 효과를 지진 등의 동적 하중조건에서 고려하기 위하여 불포화토에 대해 확장된 반복탄소성구성식에 근거한 응력-변형율 관계를 제 1 항복함수 및 제 2 항복함수를 고려하여 유도하였다. 본 관계를 2차원 및 3차원 수치해석에서 액상화 이후의 압밀거동 예측 등에 적용하는 경우에는 평균골격응력(Mean skeleton stress)의 변화를 반영하는 제 2 항복함수의 도입이 필요하나, 요소시뮬레이션에서는 제 1 항복함수만으로도 수치해석을 위한 각 물성치 및 석션 파라미터 결정이 가능할 것으로 사료된다. 본 관계와 응력반전을 반영한 반복재하 루틴을 함께 코딩(Coding)할 경우 불포화조건 하에서의 반복삼축압축시험에 대한 수치해석적 모사가 가능할 것으로 보이며, 본 연구결과는 동적 하중이 작용하는 불포화토 거동 예측의 정확도 제고에 기여할 것으로 전망된다.

서열 환경에서 농작업 모자 착용에 따른 체온 조절 및 주관적 반응 (Thermal and subjective responses by sun hats for farmer in a hot climatic chamber)

  • This study examined the effects of two kinds of functional sun hats through a head-manikin test and a climatic chamber trial for farm workers in summer. Experiment was composed of four conditions. The first condition was the condition without any hat (Control). The second was the condition with a sun hat on the market (Hat A). The third was the condition with a functional sun hat made of reflective fabric (Hat B). The last was the condition with a functional sun hat having a ventilating structure as well as reflective fabric (Hat C). For the subjects in the climatic chamber trials, 12 healthy males volunteered. Air temperature, relative humidity and globe temperature in the chamber was maintained at $33{\pm}0.5^{\circ}C,\;65{\pm}5%RH\;and\;39{\pm}1^{\circ}C\;(WBGT\;33^{\circ}C)$. Subjects did a simulated red pepper-work (50-min work and 10-min rest, twice repetition) for 120 min. As the result of head-manikin test, the surface temperature on middle of back-neck was the lowest in Hat B of four conditions and the surface temperature on top of head was the lowest in Hat C. As the result of climatic chamber trials, there were apparent differences between with (Hat A, Hat B, Hat C) and without a sun hat (Control). In rectal temperature ($T_{re}$), mean skin temperature ($\={T}_{sk}$), heart rate (HR), total sweat rate (TSR), The physiological heat strain was less in the condition with hats than in the condition without a sun hat. As the increasing rate in Tre, Hat B is the most effective hat for alleviation heat strain. As the subjective responses, Hat B was the most effective hat for thermal comfort even though the difference was not significant. Hat C was less effective than Hat B and the reason might be the increase of weight due to inserting the ventilating structure.
