Study on Growth Characteristics of Insects using a Food waste-Derived Dry Feed (음식폐기물 건조사료를 이용한 곤충의 생육특성 연구)

  • In this study, growth characteristics of insects for mass production was investigated. For Hermetia illucens, the biomass was increased by the supply of the mixture of FWDF with hop cake and typical feed at the early growth stage, however, no difference was shown between 100% of FWDF and the mixture at the late stage. The growth of Housefly larvae was retarded with the 100% of FWDF, resulting in abnormal paputation, hence the amount of FWDF shoud be lowered to 20% to 30% for the efficient growth. Super mealworm larvae was found to grow with little influence by the kind of feed, and none of them was died or inhibited by the supply of FWDF as a feeding source.

  • This study was conducted to establish a nce insect pest control system, ~ es.ys tematic application of insecticides using carbohran and buproiezin, in Korea. The effects oi various dates of application and rates of buprofezin (25% WP) after carbofuran (3G) soil incorpombon in late May on the pop~~lation densities of the brown planthoppa (BPH) immigrating in July were investigated Appropriate application tune of buprofezin for the BPH that had evaded insecticidal effect of 5011 incorporated carbofuran was late July-early August. Application rate of buprofezln at 7.0g a.i/lOa was enough to suppress the BPH density. Buprofezin treatment after carbofumn soil incorporation could also suppress the whlte backed planthopper population but did not affect the densities oi the paddy rice spiders. Considering the charactenstics of occurring patterns of the nce insect pests in Korea. buprofezin treatment m late July or early August after carbofuran soil incorporation in late May can be a useful application system of ir~sectic~deins controlling early season Insect pests and migmtoly planthoppers on rice.

Dietary inclusion of mealworm (Tenebrio molitor) meal as an alternative protein source in practical diets for rainbow trout (Oncorhynchus mykiss) fry

  • Background: An 8-week feeding trial was designed to evaluate the potential of yellow mealworm (MW; Tenebrio molitor) as a locally available nutrient-rich feedstuff for rainbow trout fry (Oncorhynchus mykiss). Methods: Triplicate groups of fish (mean ± SE; 1.11 ± 0.01 g) were assigned to each of the five isonitrogenous and isocaloric practical diets containing graded level of a full fat MW (0, 7, 14, 21, and 28%) at the expense of fish meal (designated as MW0, MW7, MW14, MW21, and MW28, respectively). Results: Fish growth performance in terms of weight gain and specific growth rate significantly increased with increasing dietary MW level up to 14% and then declined when dietary MW levels further increased to 28%. Significantly higher protein efficiency ratio and lower feed conversion ratio were found in fish fed with diets containing MW compared to fish fed the control MW0. Myeloperoxidase activity was significantly higher in fish fed MW7 diet compared to fish fed the MW0 diet. Fish fed the MW14 and MW28 diets had significantly higher lysozyme activities than those fed the MW0 diet. Conclusions: Overall, the efficacy of MW as promising alternative to fish meal in practical diets for rainbow trout fry has been proved not only in relation to growth rates and feed utilization, but also from the viewpoint of immunopotentiation effects.

Characteristics of digestive enzyme activity, antibiotic resistance, and pathogenicity of bacteria inhabited in animal feed resources (사료자원에 서식하는 세균의 소화효소활성, 항생제내성 및 병원성에 관한 특성)

  • Among different types of spoilage, microbial contamination can cause feed decomposition, which results in decreases in feed intake and productivity, infection, and breeding disorder. During the storage time, various microbes have a chance to inoculate with depreciation of feed and to infect the animals. We investigated bacteria that inhabit diverse feed ingredients and complete feed which have been stored for a few months. We isolated and identified 30 genera and 62 species of bacteria. Among these 62 species, 21 species were of non-pathogenic bacteria, 18 species were of pathogenic bacteria, 9 species were of opportunistic pathogens, and 14 species were of unknown bacteria. Pantoea allii and 24 species showed proteolytic enzyme activity. We also confirmed that 6 species including Pseudomonas psychrotolerans showed ${\alpha}$-amylase activity, and 29 species including Burkholderia vietnamiensis showed cellulase activity. Microbacterium testaceum and 3 species showed resistance to Ampicillin, Kanamycin, Streptomycin, Gentamicin, Carbenicillin, and Erythromycin ($50{\mu}g/mL$). Using mealworm larvae (Tenebrio molitor L.) as a model for pathogenicity, we confirmed that 8 species including Staphylococcus xylosus had pathogenicity for mealworm larvae. Especially, Enterobacter hormaechei, Staphylococcus xylosus, and Staphylococcus hominis were reported as being pathogenic for humans. This research suggests that hygienic management of animal feed is essential because beneficial and harmful bacteria can inhabit animal feed differently during storage and distribution.

Effects of brewer's spent grain on the growth and nutrition of the giant mealworm beetle, Zophobas atratus

  • The giant mealworm beetle, Zophobas atratus (Coleoptera: Tenebrionidae), is a tropical beetle. As this beetle can be used as an ingredient in animal feed, the effects of brewer's spent grain (BSG) on the development and nutritional value of Z. atratus were investigated. As results, there were no significant differences on the larval survival rate, body weight, and duration at different content of BSG, mixed with conventional feed, wheat bran (WB). Based on these results, BSG can be mixed with wheat bran as a Z. atratus food ingredient without any problems. The nutritional value was compared between 100% wheat bran control and 50% BSG experimental groups. The moisture, crude protein, carbohydrates, and amino acids were 1.1-1.4 times higher in the 50% BSG group. In the BSG group, the linoleic acid content was 1.6 times higher than that in the control group. In the wheat bran group, the oleic acid content was 38.4%, which was 1.3 times higher than that in the BSG group. As minerals, the control and BSG groups showed high potassium and phosphorus contents. In terms of hazardous materials, four heavy metals (lead, mercury, arsenic, and cadmium) and two microorganisms (Escherichia coli and Salmonella spp.) were not detected. There were no significant differences in developmental characteristics between the wheat bran and BSG mixed groups, and the nutritional values were better in the BSG mixed group. Therefore, BSG can be used as alternative food source for rearing Z. atratus.

Enzymatic Preparation and Antioxidant Activities of Protein Hydrolysates from Tenebrio molitor Larvae (Mealworm) (갈색거저리 유충 단백가수분해물의 제조 및 항산화 활성)

  • The present study was carried out to evaluate the applicability of Tenebrio molitor larvae (mealworm) as a health functional food material in order to contribute to the development of the domestic insect industry and health functional food industry. Protein hydrolysates were prepared from mealworm powder by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain), and the hydrolysates were then tested for their antioxidant activities. Based on available amino group contents and sodium dodecyl sulphate-polyacrylamide gel electrophoresis analyses, mealworms treated with alcalase ($4,781.39{\mu}g/mL$), flavourzyme ($5,429.35{\mu}g/mL$), or neutrase ($3,155.55{\mu}g/mL$) for 24 h showed high degree of hydrolysis (HD) value, whereas HD values of bromelain ($1,800{\mu}g/mL$) and papain-treated ($1,782.61{\mu}g/mL$) mealworms were much lower. Protein hydrolysates showing high HD values were further separated into > 3 kDa and ${\leq}3kDa$ fractions by a centrifugal filter system and then lyophilized, and the production yields of the low molecular weight protein hydrolysates (${\leq}3kDa$) by alcalase, flavourzyme, and neutrase were 42.05%, 26.27%, and 30.01%, respectively. According to the RC_{50} values of the protein hydrolysates (${\leq}3kDa$) obtained from three different antioxidant analyses, all three hydrolysates showed similar antioxidant activities. Thus, alcalase hydrolysates showing the highest production yield of low molecular weight protein hydrolysates were further tested for their inhibitory effects on peroxidation of linoleic acid by measuring thiobarbituric acid values, and the results show that peroxidation of untreated linoleic acid increased dramatically during 6 days of incubation. However, pretreatment with the hydrolysates ($100{\sim}800{\mu}g/mL$) significantly inhibited linoleic acid peroxidation in a dose-dependent manner over 6 days.

Tenebrio molitor (Mealworm) Extract Improves Insulin Sensitivity and Alleviates Hyperglycemia in C57BL/Ksj-db/db Mice (C57BL/Ksj-db/db 제 2형 당뇨모델을 이용한 갈색거저리 유충(밀웜) 추출물의 인슐린 감수성 및 혈당개선효과)

  • Diabetes is one of the serious chronic metabolic diseases caused by Westernized eating habits, and the goal of diabetes treatment is to keep blood glucose at a normal level and prevent diabetic complications. This study was designed to investigate the anti-diabetic effects of a mealworm (Tenebrio molitor larva) extract (MWE) on hyperglycemia in an animal model with type 2 diabetes. Diabetic C57BL/Ksj-db/db mice were divided into three groups: diabetic control, rosiglitazone, and MWE. The mice supplemented with MWE showed significantly lower blood levels of glucose and glycosylated hemoglobin when compared with the diabetic control mice. The homeostatic index of insulin resistance was significantly lower in mice supplemented with MWE than in diabetic control mice. MWE supplementation significantly stimulated the phosphorylation of insulin receptor substrate-1 and Akt, and activation of phosphatidylinositol 3-kinase in insulin signaling pathway of skeletal muscles. Eventually, MWE increased the expression of the plasma membrane glucose transporter 4 (GLUT4) via PI3K/Akt activation. These findings demonstrate that the increase in plasma membrane GLUT4 expression by MWE promoted the uptake of blood glucose into cells and relieved hyperglycemia in skeletal muscles of diabetic C57BL/Ksj-db/db mice. Therefore, mealworms are expected to prove useful for the prevention and treatment of diabetes, and further studies are needed to improve type 2 diabetes in the future.

Developmental Characteristics of Zophobas atratus (Coleoptera: Tenebrionidae) at Varing Temperatures (온도에 따른 아메리카왕거저리(Zophobas atratus)의 발육 특성)

  • We investigated the developmental characteristics of super mealworm (Coleoptera: Tenebrionidae) at four different temperatures (25℃, 27℃, 30℃, and 33℃). The rearing conditions were 9L/15D, 65% RH, and 1,330 to 1,800 lux with wheat bran. The length of developmental period of 1 to 18 instars at each temperature showed that 30℃ was the shortest at 120.0±5.8 days, compared to that at 27℃ (132.6±10.7 days), 33℃ (136.5±9.2 days), and 25℃ (156.7±7.5 days). The larval developmental period was statistically significantly longer at 25℃, 27℃, and 30℃ compared to the length at 33℃. However, the death rate of larvae at 33℃ was 2.7-3.3 times higher than the rate at other temperatures. Body weight was heaviest at 30℃ followed by 27℃, 33℃, and 25℃. The patterns of head capsule, body capsule, and body length were similar to that of body weight. Regression analyses of developmental period, larval body weight, and length according to temperature revealed 29-30℃ as the most suitable temperature. The prepupa rate was 43.1% in 17 instars, 30.3% in 18 instars, 15.4% in 16 instars, 7.1% in 19 instars, 2.2% in 15 instars, and 1.9% in 20 instars, accounting for 88.8% in 16-18 instars. Prepupal period was longer at lower temperatures. For the average prepupal period of 15-18 instars, prepupa time was 18.8±1.9 days at 27℃, 18.8±2.3 days at 30℃, 23.0±2.4 days at 33℃, and 23.1±2.9 days at 25℃. The average pupal period of females and males was 11.1±2.2 and 11.6±2.4 days, respectively. The data indicate that the most suitable rearing temperature of super mealworm was 30℃.

Temperature-dependent Development Model of Larvae of Mealworm beetle, Tenebrio molitor L. (Coleoptera: Tenebrionidae) (갈색거저리(Tenebrio molitor L.) 유충의 온도발육 모형)

  • The developmental times of mealworm beetle larvae, Tenebrio molitor were studied at six temperatures ranging from 15 to $30^{\circ}C$ with 60~70% RH, and a photoperiod of 14L:10D. Mortality of larval period was very low at 17 and $20^{\circ}C$ but did not die over $22^{\circ}C$. Developmental time of larva was decreased with increasing temperature. The total developmental time of T. molitor larvae was longest at $17^{\circ}C$ (244.3 days) and shortest at $30^{\circ}C$ (110.8 days). Egg and larvae were not developed at $15^{\circ}C$. The lower developmental threshold and effective accumulative temperatures for the total larval stages were $6.0^{\circ}C$ and 2564.1 degree-days, respectively. The relationship between developmental rate and temperature was fitted by a linear model and nonlinear model of Logan-6($r^2$=0.95). The distribution of completion of each development stage was well described by the 2-parameter Weibull function ($r^2$=0.8502~0.9390).

Effect of Tenebrio molitor Larvae Fermented with Saccharomyces cerevisiae M1 (KACC 93023) on Non-alcoholic Fatty Liver-induced Rats (Saccharomyces cerevisiae M1 (KACC 93023)으로 발효한 갈색 거저리 유충이 비 알코올성 지방간 유발 흰 쥐에 미치는 영향)

  • In this study, Tenebrio molitor (brown mealworm) larvae powder was fermented with Saccharomyces cerevisiae M1 (KACC 93023). The fermented and non-fermented powders were subsequently administered to rats with induced non-alcoholic fatty liver disease to compare the degree of improvement. The rats exhibited abnormal lipid metabolism due to orotic acid, and the group that were fed the fermented larvae powder showed similar weight to normal rats. AST, ALT, ALP, and LDH activities, all known indicators of liver health, and various other lipid-related indicators appeared positive, suggesting improvement. The lipid peroxide and glutathione content in the liver tissue and serum were similar to or better than those of the normal group. Finally, morphological observations of liver tissue using H&E and oil red O stainings revealed that the fermented powder group showed markedly reduced fat along with normal hepatocyte shape and arrangement. To summarize, it was confirmed that dietary brown mealworm larvae improve non-alcoholic fatty liver function, and, when fermented with yeast, most indicators showed positive improvements to almost normal levels. As such, the fermented powder can be considered to improve non-alcoholic fatty liver, suggesting that it may be used as a material for various products in the future.
