Methyl violet, used extensively in the commercial textile industry and as a biological stain, is a hazardous recalcitrant. Aspergillus sp. strain CB-TKL-1 isolated from a water sample from Tsumoriri Lake, Karzok, Ladakh, India, was found to completely decolorize methyl violet within 24 h when cultured under aerobic conditions at $25^{\circ}C$. The rate of decolorization was determined by monitoring the decrease in the absorbance maxima of the dye by UV-visible spectroscopy. The decolorization of methyl violet was optimal at pH 5.5 and $30^{\circ}C$ when agitated at 200 rpm. Addition of glucose or arabinose (2%) as a carbon source and sodium nitrate or soyapeptone (0.2%) as a nitrogen source enhanced the decolorization ability of the culture. Furthermore, the culture exhibited a maximum decolorization rate of methyl violet after 24 h when the C:N ratio was 10. Nine N-demethylated decolorized products of methyl violet were identified based on UV-visible spectroscopy, Fourier transform infrared (FTIR), and LC-MS analyses. The decolorization of methyl violet at the end of 24 h generated mono-, di-, tri-, tetra-, penta-, and hexa-N-demethylated intermediates of pararosaniline. The variation of the relative absorption peaks in the decolorized sample indicated a linear decrease of hexa-N-demethylated compounds to non-N-demethylated pararosaniline, indicating a stepwise N-demethylation in the decolorization process.
Barren ground is an abnormal phenomenon in coastal ecosystems where seaweeds are destroyed and largely replaced by coralline algae containing calcium carbonate components. To restore the seaweed forest, we attempted reconstruction in an area of Samcheok, Korea, where barrenness is extensive. We developed two methods of seaforest construction underwater longline and seed transplantation for the brown seaweed Laminaria japonica, a fast-growing edible seaweed. The blade length of L. japonica attached to an underwater long line was $93.9{\pm}38.2cm$ and blade weight $7.6{\pm}2.1g$ in February 2004. Seaweed size declined after August 2004. A Similar pattern was observed using the transplantation method with maxima of $179.3{\pm}40.3cm\;and\;14.9{\pm}3.2g$ for blade length and weight, respectively in July 2004. The transplanted seaweeds matured in October as did a wild population. These results indicate that transplanted seaweed acclimate to new environments.
Ultraviolet spectrophotometric investigations have been carried out on the systems of benzene, naphthalene and phenanthrene with iodine in carbon tetrachloride. The results reveal the formation of the one to one molecular complexes. The equilibrium constants were obtained in consideration of that absorption maxima due to the formation of the charge transfer complexes shift to blue with increasing temperature. Thermodynamic parameters for the formation of the charge transfer complexes were calculated from these values. These results indicate that the stabilities of the complexes formed between polynuclear aromatics and iodine increase with the number of aromatic rings. This may be a measure of the relative basicities of polynuclear aromatics toward iodine, which is explained in terms of the resonance of the interaction between aromatics and iodine. The results in this study were, additionally, compared with those of the polymethylbenzene series to be discussed the reason for them.
Effect of NiO and $MnO_2$ addtivies on the dielectric piezoelectrics and polarization-reversal properties of $(Pb_{0.936} La_{0.064})$$(Zr_{0.60}Ti_{0.40})O_3$ ceramics have been investigated. The specimens were prepared by the mixed oxide techni-que and atmosphere sintering method. The room temperature X-ray diffraction studies show that perfect perovskite solution with tetragonal structure was obtained from PLZT and its additives. The dielectric constant and dissipation factor decreased with the addition of both NiO and $MnO_2$ The Curie of Curie temperature was not observed but they displayed broadened maxima. The planar coupling factor was improved by addition of NiO and also increased with increasing sintering time carried out at 105$0^{\circ}C$ Addition of $MnO_2$ yielded a markedly high mechanical quality factor. The space-charge field decreased with the addition of NiO but increased with the addition of $MnO_2$ The planar coupling factor and space-charge field showed same dependence on the additivies. The tetragonality Curie temperature and planar coupling factor of $(Pb_{0.936} La_{0.064})$$(Zr_{0.60}Ti_{0.40})O_3$ were higher than those of $(Pb_{0.936} La_{0.064})$$(Zr_{0.568}NU_{0.032}Ti_{0.40})_{0.984}O_3$ but the grain size lattic parameter dielectric constant dissipation factor mechanical quality factor and space-charge field of the former were lower than those of the latter.

Amorphous $MnO_{2}{\cdot}nH_{2}O$ in 1M KOH aqueous electrolyte proves to be an excellent electrode for a faradic electrochemical capacitor cycled between -0.5 and +1.0 versus Ag/AgCl. The effect of thermal treatment on the crystalinity, particle structure, and corresponding electrochemical properties of the resulting xerogel remained amorphous as Mn(OH)2 up to 160$^{\circ}C$. With an increase in the temperature above 200$^{\circ}C$, both the surface area and pore volume decreased sharply, because the amorphous Mn(OH)2 decomposed to form MnO that was subsequently oxidized to form crystalline Mn3O4. In addition, the changes in the crystallinity, and particle structure all had significant but coupled effects on the electrochemical properties of the xerogels. A maximum capacitance of 160.6F/g was obtained for an electrode prepared with the MnOx Xerogel calcined at 150$^{\circ}C$, which was consistent with the maxima exhibited in both the surface area and pore volume. This capacitance was attributed solely to a surface redox mechanism.

The structural and functional aspects of ervatamin B were studied in solution. Ervatamin B belongs to the $\alpha+\beta$ class of proteins. The intrinsic fluorescence emission maximum of the enzyme was at 350 nm under neutral conditions, and at 355 nm under denaturing conditions. Between pH 1.0-2.5 the enzyme exists in a partially unfolded state with minimum or no tertiary structure, and no proteolytic activity. At still lower pH, the enzyme regains substantial secondary structure, which is predominantly $\beta$-sheet conformation and shows a strong binding to 8-anilino-1-napthalene-sulfonic acid (ANS). In the presence of salt, the enzyme attains a similar state directly from the native state. Under neutral conditions, the enzyme was stable in urea, while the guanidine hydrochloride (GuHCl) induced equilibrium unfolding was cooperative. The GuHCl induced unfolding transition curves at pH 3.0 and 4.0 were non-coincidental, indicating the presence of intermediates in the unfolding pathway. This was substantiated by strong ANS binding that was observed at low concentrations of GuHCl at both pH 3.0 and 4.0. The urea induced transition curves at pH 3.0 were, however, coincidental, but non-cooperative. This indicates that the different structural units of the enzyme unfold in steps through intermediates. This observation is further supported by two emission maxima in ANS binding assay during urea denaturation. Hence, denaturant induced equilibrium unfolding pathway of ervatamin B, which differs from the acid induced unfolding pathway, is not a simple two-state transition but involves intermediates which probably accumulate at different stages of protein folding and hence adds a new dimension to the unfolding pathway of plant proteases of the papain superfamily.
In the design and condition assessment of bridges, it is usually necessary to take into consideration the extreme conditions which are not expected to occur within a short time period and thus require an extrapolation from observations of limited duration. Long-term structural health monitoring (SHM) provides a rich database to evaluate the extreme conditions. This paper focuses on the extrapolation of extreme traffic load effects on bridges using long-term monitoring data of structural strain. The suspension Tsing Ma Bridge (TMB), which carries both highway and railway traffic and is instrumented with a long-term SHM system, is taken as a testbed for the present study. Two popular extreme value extrapolation methods: the block maxima approach and the peaks-over-threshold approach, are employed to extrapolate the extreme stresses induced by highway traffic and railway traffic, respectively. Characteristic values of the extreme stresses with a return period of 120 years (the design life of the bridge) obtained by the two methods are compared. It is found that the extrapolated extreme stresses are robust to the extrapolation technique. It may owe to the richness and good quality of the long-term strain data acquired. These characteristic extremes are also compared with the design values and found to be much smaller than the design values, indicating conservative design values of traffic loading and a safe traffic-loading condition of the bridge. The results of this study can be used as a reference for the design and condition assessment of similar bridges carrying heavy traffic, analogous to the TMB.

Surveillance cameras installed in tunnels capture the various video frames effected by dynamic and variable factors. In addition, localizing and managing the cameras in tunnel is not affordable, and quality of capturing frame is effected by time. In this paper, we introduce a new method to detect and track the vehicles in tunnel by using surveillance cameras installed in a tunnel. It is difficult to detect the video frames directly from surveillance cameras due to the motion blur effect and blurring effect on lens by dirt. In order to overcome this difficulties, two new methods such as Differential Frame/Non-Maxima Suppression (DFNMS) and Haar Cascade Detector to track cars are proposed and investigated for their feasibilities. In the study, it was shown that high precision and recall values could be achieved by the two methods, which then be capable of providing practical data and key information to an automatic accident detection system in tunnels.
In a $Co_{67}Fe_{4.5}Nb_{2}Si_{10}B_{15}$ amorphous alloy ribbon the effect to two-step annealing on the soft magnetic properties has been studied. By two-step annealing method which the second annealing at low temperature of $310^{\circ}C$ for 2 hours is undertaken after the primary annealing at high temperature above $480^{\circ}C$ for 20 minutes at the vacuum state, the coercive force and the squareness are not changed nearly but the initial permeability at d.c. and the effective permeability at a.c. are remark-ably increased compared with the one-step annealing. The maxima of the initial permeability and the effective permeability at 1 kHz after the two-step annealing are 290,000 and 41,000, respectively, which are 30% higher than those of the one-step annealing. The change of magnetic properties with annealing temperature is discussed in terms of the residual stress, the domain size, the cluster and the crystalline phase.

Magnetic and magnetostrictive properties of Tb-Fe and Tb-Fe-B thin films are systematically investigated over a wide composition range from 40.2 to 68.1 at. % Tb. The films were fabricated by rf magnetron sputtering using a composite target which consists of an Fe plate and Tb chips. The microstructure, examined by X-ray diffraction, mainly consists of an amorphous phase and, at high Tb contents, a pure Tb phase also exists. A progressive change in the direction of anisotropy from the perpendicular to in-plane occurs as the Tb content increases and the boundary at which the anisotropy change occurs shifts significantly towards to higher Tb contents with the addition of B. The saturation magnetization exhibits maxima at the Tb contents of 42 and 48 at. % for Tb-Fe and Tb-Fe-B thin films, respectively, and it is decreased by the addition of B. The coercive force, measured in the easy direction, decreases monotonically with the Tb content. Excellent magnetostrictive characteristics, particularly at low magnetic fields, are achieved in both Tb-Fe and Tb-Fe-B thin films; for example, a magnetostriction of 138 ppm is obtained in a Tb-Fe-B thin film at a magnetic field as low as 30 Oe. The excellent magnetostrictive properties of the present thin films are supported by the equally excellent magnetic softness, the coercivity below 10 Oe and a typical squared-loop shape with the saturation field as low as 1 kOe. Due to the excellent low field magnetostrictive characteristics, the present Tb-Fe based thin films are thought to be suitable for Si based microdevices.
