기질 금속단백분해효소(MMP)는 기저막이나 간질성 조직 등에 있는 세포외기질 성분을 분해하여 상처 치유, 태아의 발생, 종양세포의 침윤과 전이 등을 포함하여 조직이 재구성되는 과정에서 생리학적 및 병리학적인 과정 양쪽 모두에 매우 중요한 역할을 한다. 본 연구에서는 MMP-9 유전자의 발현을 억제하는 물질을 천연물에서 탐색하였고, 탐색된 시료 중에서 높은 역가를 가진 낙우송과의 Metasequoia glyptostroboides가 선택되었다. 여러 가지 flavonoid 성분을 가지고 있는 Metasequoia glyptostroboides를 이용하여 4개의 biflavonoid와 2개의 monoflavonoid 구조의 물질을 분리하였고, 이 flavonoid들은 sciadopitysin, isoginkgetin, bilobetin, 2, 3-dihydrohino-kiflavone, luteolin, apigenin으로 정제하여 구조를 밝히고 zymography, WST-1을 이용한 세포독성시험, northern blot 등의 실험을 통하여 각 화합물의 구조적인 특성과 함께 MMP-9 유전자 발현을 억제하는 효과가 있는 것을 확인하였다.
Matrix metalloproteinases (MMPs) have been known to affect to cell migration, proliferation, morphogenesis and apoptosis by degrading the extracellular matrix. In the previous studies, undifferentiated mouse embryonic stem cells (ESCs) were successfully proliferated inside the extracellular matrix (ECM) analog-conjugated three-dimensional (3D) poly ethylene glycol (PEG)-based hydrogel. However, there is no report about MMP secretion in ESCs, which makes it difficult to understand and explain how ESCs enlarge space and proliferate inside 3D PEG-based hydrogel constructed by crosslinkers containing MMP-specific cleavage peptide sequence. Therefore, we investigated what types of MMPs are released from undifferentiated ESCs and how extracellular signals derived from various niche conditions affect MMP expression of ESCs at the transcriptional level. Results showed that undifferentiated ESCs expressed specifically MMP2 and MMP3 mRNAs. Transcriptional up-regulation of MMP2 was caused by the 3D scaffold, and activation of integrin inside the 3D scaffold upregulated MMP2 mRNAs synergistically. Moreover, mouse embryonic fibroblasts (MEFs) on 2D matrix and 3D scaffold induced upregulation of MMP3 mRNAs, and activation of integrins through conjugation of extracellular matrix (ECM) analogs with 3D scaffold upregulated MMP3 mRNAs synergistically. These results suggest that successful proliferation of ESCs inside the 3D PEG-based hydrogel may be caused by increase of MMP2 and MMP3 expression resulting from 3D scaffold itself as well as activation of integrins inside the 3D PEG-based scaffold.
자외선은 가시 광선보다 파장이 짧은 전자기파이며, 오존층을 통과하는 자외선이 피부 노화의 주요 원인이다. 피부 조직을 자외선에 만성적으로 노출 시키면 인간 각질 형성 세포에서 Mitogen-activated Protein Kinases (MAPKs) 신호 전달 경로가 활성화되어 매트릭스 metalloproteinases (MMPs) 생성이 증가한다. 본 연구에서는 제주도의 초본 추출물을 대상으로 자외선 자극을 통해 인간 각질 형성 세포(HaCaTs)의 항 노화 효과에 대해 조사 하였다. 우리는 자외선 유도 된 각질 형성 세포에서 MMP-1 유전자와 단백질 발현 수준을 측정 하였으며 그 결과, Thymus quinquecostatus 추출물(TQE)은 자외선 조사된 HaCaT 세포에서 의해 농도 의존적으로 MMP-1의 발현을 유의하게 감소시키는 것으로 확인되었다. 또한, TQE는 MAPK 신호 전달요소 인 ERK1/2, JNK1/2 및 p38 단백질의 자외선 유도에 따른 인산화를 유의 적으로 감소시켰다. 이러한 결과는 MAPKs 경로가 인간 각질 세포에서 UV 유도 MMP-1 생산에 대한 TQE의 억제 효과에 기여할 수 있음을 시사한다. 우리의 결과는 아마도 인간 각질형성 세포에서 TQE가 자외선 유도된 MMP-1 생산을 억제하는 피부 노화 소재로 활용될 수 있음을 시사한다.

Purpose: Matrix metalloproteinases (MMPs) have been implicated in atherosclerosis, and therefore, are considered risk factors for metabolic dysfunction in adults. However, there is little data on circulating levels of MMPs and tissue inhibitors of MMPs (TIMPs) with regard to obesity-related biomarkers in the general adolescent population. In the present study, we determined the associations of MMP-8, MMP-9, and TIMP-1 levels and MMP-8/TIMP-1 and MMP-9/TIMP-1 ratios with obesity-related biomarkers in apparently healthy adolescent boys. Methods: We measured MMP and TIMP concentrations in plasma samples using the enzyme-linked immunosorbent assay and analyzed their associations with obesity-related biomarkers, such as liver enzymes and lipid profiles, in a sample of 91 Korean boys aged 13-14 years who participated in a general health check-up. Results: The mean age of the boys was $13.8{\pm}0.3years$; 72 boys were normal weight and 19 were overweight/obese. The Pearson correlation coefficients revealed a significant correlation between MMP-8 and aspartate aminotransferase (r=0.217, P=0.039) and alanine aminotransferase (r=0.250, P=0.017) and between TIMP-1 and aspartate aminotransferase (r=0.267, P=0.011). In a multivariate linear regression analysis, serum alanine aminotransferase was positively associated with the MMP-8 level. There were no significant differences in the MMP-8, MMP-9, and TIMP-1 levels or MMP-8/TIMP-1 and MMP-9/TIMP-1 ratios between control and overweight/obese subjects. Conclusion: We found a significant association between the MMP-8 level and alanine aminotransferase in the apparently healthy adolescent boys. These findings indicate that there may be a pathophysiological mechanism underlying the relationship between MMP-8 and liver enzymes in young adolescents.
It has been shown that the accumulation of prion in the cytoplasm can result in neurodegenerative disorders. Synthetic prion peptide 106-126 (PrP) is a glycoprotein that is expressed predominantly by neurons and other cells, including glial cells. Prion-induced chronic neurodegeneration has a substantial inflammatory component, and an increase in the levels of matrix metalloproteinases (MMPs) may play an important role in neurodegenerative development and progression. However, the expression of MMPs in PrP induced rat astrocytes and microglia has not yet been compared. Thus, in this study, we examined the fluorescence intensity of CD11b positive microglia and Glial Fibrillary Acidic Protein (GFAP) positive astrocytes and found that the fluorescent intensity was increased following incubation with PrP at 24 hours in a dose-dependent manner. We also observed an increase in interleukin-1 beta (IL-$1{\beta}$) and tumor necrosis factor alpha (TNF-${\alpha}$) protein expression, which are initial inflammatory cytokines, in both PrP induced astrocytes and microglia. Furthermore, an increase MMP-1, 3 and 11 expressions in PrP induced astrocytes and microglia was observed by real time PCR. Our results demonstrated PrP induced activation of astrocytes and microglia respectively, which resulted in an increase in inflammatory cytokines and MMPs expression. These results provide the insight into the different sensitivities of glial cells to PrP.
최근에 해양자원에 있는 동물, 해조류 곰팡이 세균에서 신규 잠재적인 후보약효제가 조사되어 왔다. 본 연구에서는 치료제를 탐색하기 위하여 암전이, 관절염, 만성염증 및 주름형성에 주요한 역할을 하는 기질금속단백질분해효소(s) (MMPs)를 목적효소로 이용하였다. 5종의 녹조류, 18종의 홍조류, 4종의 갈조류를 포함한 다양한 해조류가 사람피부섬유아세포 및 섬유아육종세포로부터 유래된 기질금속단백질효소에 미치는 영향을 gelatin zymography를 이용하여 조사하였다. 사람피부섬유아세포에서는 홍조류중에서 Laurencia okamurae, Polysiphonia japonica, Grateloupia lanceolate 및 Sinkoraena lancifolia에서 MMP-2 억제효과가 관찰되었다. 반면에 녹조류의 Enteromorpha compressa와 Enteromorpha linza, 갈조류의 Peltaronia bighamiae and Sargassum thunbergii에서는 MMP-2 활성화가 관찰되었다. 사람섬유아육종세포에서는 MMP-9 활성화가 갈조류인 Sargassum thunbergii, 홍조류의 Polysiphonia japonica, 녹조류의 Enteromorpha compressa와 Enteromorpha linza의 존재 하에서는 감소되었다. 본 연구에서 흥미로운 발견은 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 정상세포에서는 MMP-2에 대하여 활성화 효과를 나타내었으나, 암세포에서는 MMP-9응 억제하는 효과를 나타낸 것이다. 이러한 결과는 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 항암 효능을 발휘할 수 있는 성분을 함유하고 있다는 것을 암시하고 있다.
그래핀(graphene)은 원자 하나의 두께를 가지는 흑연(graphite)의 단층으로서 탄소구조체들 중 하나이다. 그래핀은 최근 의학분야에서 광열요법을 이용한 암 발생의 예방효과와 DNA의 산화에 대한 보효효과를 가진다고 밝혀진 바 있다. 본 연구에서는 사람 섬유아육종세포(HT1080)에서 산화 스트레스와 MMPs에 대한 그래핀의 효과가 조사되었다. 항산화 효과에 대한 연구에서 그래핀은 DNA 산화에 대한 억제효과를 특이하게 나타내었으나 다른 산화 스트레스는 억제하지 않았다. 뿐만 아니라 그래핀은 세포 내 과산화수소를 생성시키는 phenazinemethosulfate(PMS)에 의하여 자극된 MMP-2 및 MMP-9의 발현과 활성을 감소시켰다. 특히 superoxide dismutase(SOD-2)와 같은 항산화 효소의 발현이 HT1080세포에서 감소하였는데, 이것이 시사하는 바는 SOD 발현수준의 감소가 그래핀의 항산화 효과로부터 기인 되었다는 것을 나타낸다. 이상의 결과로 그래핀의 존재에서 산화스트레스의 억제효과가 HT1080 세포에서 MMP-9의 활성과 발현을 감소시킬 수 있다는 것을 암시하고 있다. 이러한 연구 결과를 바탕으로 그래핀은 암 전이와 관련 있는 MMP-2 및 MMP-9의 활성과 발현의 억제를 통하여 암 억제에 도움을 줄 수 있어, 산업화를 위한 하나의 우수한 생의학 응용소재로 이용될 수 있으리라 기대된다.
피부는 자외선, 감염 등과 같은 외부 요인에 의해 피부 노화가 진행된다. 이러한 요인들에 의해 피부의 섬유아세포는 단백질 분해효소인 matrix metalloproteinases (MMPs)를 분비한다. MMPs는 세포외기질에 위치하는 콜라겐의 분해를 유도하여 노화에 직접적인 영향을 미친다. 으름덩굴(Akebia quinata Decaisne) 줄기는 항산화, 항염증 효과가 있는 것으로 보고되었다. 하지만 으름덩굴 줄기 에탄올 추출물(AQSEE)의 항노화 효과에 대해서는 알려지지 않았다. 따라서 인간 섬유아세포에서 TNF-α로 유도된 MMP-1 억제 효과를 연구하였다. MTT asaay를 통해 AQSEE의 세포 생존율을 확인한 결과 400 ㎍/mL까지 독성을 나타내지 않았다. RT-qPCR과 ELISA를 통해 MMP-1 mRNA와 단백질 분비를 억제하는지 확인한 결과 100, 200, 400 ㎍/mL 농도에서 유의하게 감소하였다. 또한, western blotting을 통해 MAPKs 신호전달경로와 전사인자의 인산화가 감소하는지 확인하였다. 그 결과 p38, c-Jun, p65의 인산화가 모든 농도에서 유의하게 감소하였다. AQSEE의 radical 소거능을 확인하기 위해 DPPH, ABTS assay를 진행한 결과 모든 농도에서 유의하게 감소하였다. 본 연구 결과를 통해 MMP-1 억제 효과와 radical 소거능을 확인하였으며, 이는 항노화 물질로 사용될 수 있음을 시사한다.
서해안에 서식하는 해양생물인 전복은 항산화 및 항염증 효과가 있으며, 항생제 개발 및 화장품 원료 등 다양한 산업에 활용될 수 있는 잠재력을 가진 자원이다. 본 연구에서 우리는 서해안의 다양한 해양 생물 중에서 전복을 선택하였으며, 전복에서 분리한 항균펩타이드(AMP)를 이용하여 항균펩타이드 유도체인 Ab4-7를 규명하고 생리활성을 연구하였다. Ab4-7의 항염증 효능을 확인하기 위해 염증이 유도된 RAW 264.7에 Ab4-7을 처리한 결과, 염증성 사이토카인, TLR4, TNF-α, IL-1β, COX-2, iNOS 등을 억제하고 항산화와 관련된 유전인자인 HO-1의 mRNA 발현을 증가시켰다. Ab4-7의 강력한 항산화 및 항염증 효과를 바탕으로, 염증이 유도된 RAW 264.7 세포에서 Ab4-7이 extrocellular matrix metalloproteinase (ECM) 분해에 관여하여 다양한 염증성 피부질환을 유도하는 유전자인 matrix metalloproteinase (MMPs)를 조절하는 효과를 RT-PCR을 통해 확인하였다. Ab4-7은 강력한 항염증 및 항산화 효과를 가지고 있으며, 항염증 및 항산화 효과를 통한 MMPs 관련 유전인자 조절을 통해 다양한 염증성 피부질환 치료제의 기능성 재료로 활용될 수 있다고 사료된다.
야관문의 항산화 활성과 항주름 활성을 조사하기 위하여 30% 에탄올 추출물과 각종 용매 분획물을 이용하여 DPPH 및 ABTS radical 소거활성과 matrix metalloproteinase MMP1과 MMP3 생성억제 효과를 조사하였다. 에탄올 추출물과 각종 용매 분획물 중 에틸아세테이트 분획물에서 페놀 및 플라보노이드 함량이 가장 높았다. 야관문 추출물의 항산화 활성과 피부 주름을 유발하는 MMP1, MMP3의 생성을 억제 효과는 페놀 및 플라보노이드 함량과 높은 상관관계를 보였으며, 페놀과 플라보노이드 함량이 가장 높은 에틸아세테이트 분획물의 항산화 활성과 MMPs 생성 억제 효과가 가장 높았다.
