Abalone Haliotis discus hannai Intestine Digests with Different Molecule Weights Inhibit MMP-2 and MMP-9 Expression in Human Fibrosarcoma Cells

  • The abalone Haliotis discus hannai, is one of the economically important species in the fisheries industry. Abalone intestines are one of the by-products of its processing. To investigate its bioactive potential, abalone intestine was digested using an in vitro gastrointestinal (GI) digestion system containing pepsin, trypsin, and ${\alpha}$-chymotrypsin. The abalone intestine G1 digests (AIGIDs) produced by the GI digestion system were fractionated into AIGID I (> 100 kDa), AIGID II (10-100 kDa), and AIGID III (1-10 kDa) using an ultrafiltration membrane system. Of the three digests, AIGID II and AIGID III exhibited inhibitory effects against matrix metalloproteinase-2 and -9 (MMP-2, MMP-9) in HT1080 human fibrosarcoma cells. Both fractions potently inhibited gelatine digestion by MMP-2 and MMP-9 treated with phorbol 12-myristate 13-acetate (PMA) and migration of HT1080 cells in dose dependently. Furthermore, AIGID II and III attenuated expression of p65, a component of nuclear transcription factor kappa B. These results indicate that of the abalone intestine digests inhibit MMP-2 and MMP-9. Thus, the AIGIDs or their active components may have preventive and therapeutic potential for diseases associated with MMP-2 and MMP-9 activation in fibrosarcoma cells.

Characterization of Matrix Metalloproteinase Expression in Triglyceride Treated THP-1 Macrophages

  • Elevated blood triglyceride (TG) levels correlate with development of atherosclerosis suggesting that TG may promote the development of this disease. During atherosclerosis, TG is taken up by tissue macrophages which result in dramatic changes in various secreted factors. One such factor is the family of matrix metalloproteases (MMP) which are involved in tissue remodeling during both physiological and pathological processes. In this study, we examined the MMP expression profile in PMA-differentiated THP-1 macrophages treated with TG. We found that TG-treated THP-1 macrophages showed decreased expression of MMP-3, MMP-7, MMP-8 and MMP-9 in a time- and dose-dependent manner. In contrast, expression of MMP-1, MMP-2, and MMP-10 remained relatively unchanged after TG treatment. In addition, we found that expression of select MMPs was affected by various inhibitors of signaling pathways. In particular, expression of MMP-3 was slightly recovered by cRAF and PLC signaling pathway inhibitors. These data suggests a possible role of MMPs in macrophages during TG-induced atherosclerosis.

The effect of lead on matrix metalloproteinase-9 expression in rat primary glial cells

  • Lead has long been considered as a toxic environmental pollutant, which severely damages central nervous system. Lead can cause hypo- and de-myelination, and glial cells are closely related with myelination or demyelination. Matrix metalloproteinases (MMPs) are proteolytic enzymes that are involved in the remodelling of the extracellular matrix in a variety of physiological and pathological processes. MMPs also seem to be important in the pathogenesis of inflammatory demyelinating diseases of the central and peripheral nervous system. In this study, we investigated whether lead affects MMP-9 expression in rat primary glial cells. Treatment of 0.1-5 ${\mu}$M lead dose- and time-dependently increased MMP-9 expression in rat primary glial cells. The activity of MMPs was determined using zymography. Lead activated Erk(1/2) but neither of the other endogenous MAP kinases, p38 or JNK. Inhibition of Erk(1/2) activation by PD98059, a MEK inihibitor, prevented lead-induced expression of MMP-9. The results of the present study suggest that lead intoxication may adversely affect brain function at least in part by inducing MMP-9 expression through Erk(1/2) activation in primary glial cells.

Transglutaminase-2 Is Involved in All-Trans Retinoic Acid-Induced Invasion and Matrix Metalloproteinases Expression of SH-SY5Y Neuroblastoma Cells via NF-κB Pathway

  • All-trans retinoic acid (ATRA) is currently used in adjuvant differentiation-based treatment of residual or relapsed neuroblastoma (NB). It has been reported that short-term ATRA treatment induces migration and invasion of SH-SY5Y via transglutaminase-2 (Tgase-2). However, the detailed mechanism of Tgase-2's involvement in NB cell invasion remains unclear. Therefore we investigated the role of Tgase-2 in invasion of NB cells using SH-SY5Y cells. ATRA dose-dependently induced the invasion of SH-SY5Y cells. Cystamine (CTM), a well known tgase inhibitor suppressed the ATRA-induced invasion of SH-SY5Y cells in a dose-dependent manner. Matrix metalloproteinase -9 (MMP-9) and MMP-2, well known genes involved in invasion of cancer cells were induced in the ATRA-induced invasion of the SH-SH5Y cells. Treatment of CTM suppressed the MMP-9 and MMP-2 enzyme activities in the ATRA-induced invasion of the SH-SY5Y cells. To confirm the involvement of Tgase-2, gene silencing of Tgase-2 was performed in the ATRA-induced invasion of the SH-SH5Y cells. The siRNA of Tgase-2 suppressed the MMP-9 and MMP-2 activity of the SH-SY5Y cells. MMP-2 and MMP-9 are well known target genes of NF-${\kappa}B$. Therefore the relationship of Tgase-2 and NF-${\kappa}B$ in the ATRA-induced invasion of the SH-SY5Y cells was examined using siRNA and CTM. ATRA induced the activation of NF-${\kappa}B$ in the SH-SY5Y cells and CTM suppressed the activation of NF-${\kappa}B$. Gene silencing of Tgase-2 suppressed the MMP expression by ATRA. These results suggested that Tgase-2 might be a new target for controlling the ATRA-induced invasion of NBs.

  • 노화에 미치는 자외선의 영향에 대해 많은 연구가 진행되어 왔지만, 천연물에 대한 연구는 별로 알려진 것이 없다. MMPs는 광노화 과정에 매우 중요한 역할을 하는 것으로 알려져 있다. 본 연구에서는 MMP-1의 발현과 활성 및 항산화 효과에 미치는 $1,2,4,6-tetra-O-galloyl-{\beta}-{_D}-glucopyranose$와 3,4,5-trihydroxybenzoic acid의 효과를 측정하였다. 이들 화합물은 박과의 백렴으로부터 분리하였으며, 자유 라디칼과 활성산소 소거활성이 매우 높은 것으로 나타났다. 이들 화합물의 DPPH 라디칼과 활성산소를 50% 소거하는 활성$(SC_{50})$은 각각 $3.9{\mu}M,\;13.3{\mu}M,\;4.3{\mu}M,\;4.0{\mu}M$로 나타났다. 또한, 이들 화합물은 단백질 수준에서의 MMP-1 발현 및 활성을 처리농도 의존적으로 저해하였지만, 유전자 수준에서는 저해활성이 없는 것으로 나타났다 따라서, 이들 화합물은 단백질 수준에서의 우수한 MMP-1 발현 저해능과 높은 항산화 활성을 가지는 것을 알 수 있다. 결론적으로 이들 화합물은 새로운 항노화 소재로 적용될 수 있을 것으로 기대된다

Chondroprotective Effects of Wogonin in Experimental Models of Osteoarthritis in vitro and in vivo

  • We evaluated the chondroprotective effects of wogonin by investigating its effects on the gene expression and production of matrix metalloproteinase-3 (MMP-3) in primary cultured rabbit articular chondrocytes, as well as on production of MMP-3 in the rat knee. Rabbit articular chondrocytes were cultured in a monolayer, and RT-PCR was used to measure interleukin-$1{\beta}$ (IL-$1{\beta}$)-induced expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), and type II collagen. In rabbit articular chondrocytes, the effects of wogonin on IL-$1{\beta}$-induced production and proteolytic activity of MMP-3 were investigated using western blot analysis and casein zymography, respectively. The effect of wogonin on MMP-3 protein production was also examined in vivo. In rabbit articular chondrocytes, wogonin inhibited the expression of MMP-3, MMP-1, MMP-13, and ADAMTS-4, but increased expression of type II collagen. Furthermore, wogonin inhibited the production and proteolytic activity of MMP-3 in vitro, and inhibited production of MMP-3 protein in vivo. These results suggest that wogonin can regulate the gene expression and production of MMP-3, by directly acting on articular chondrocytes.

Type I Collagen-induced Pro-MMP-2 Activation is Differentially Regulated by H-Ras and N-Ras in Human Breast Epithelial Cells

  • Tumor cell invasion and metastasis are often associated with matrix metalloproteinases (MMPs), among which MMP-2 and MMP-9 are of central importance. We previously showed that H-Ras, but not N-Ras, induced invasion of MCF10A human breast epithelial cells in which the enhanced expression of MMP-2 was involved. MMP-2 is produced as a latent pro-MMP-2 (72 kDa) to be activated resulting the 62 kDa active MMP-2. The present study investigated if H-Ras and/or N-Ras induces pro-MMP-2 activation of MCF10A cells when cultured in two-dimensional gel of type I collagen. Type I collagen induced activation of pro-MMP-2 only in H-Ras MCF10A cells but not in N-Ras MCF10A cells. Induction of active MMP-2 by type I collagen was suppressed by blocking integrin ${\alpha}2$, indicating the involvement of integrin signaling in pro-MMP-2 activation. Membrane-type (MT)1-MMP and tissue inhibitor of metalloproteinase (TIMP)-2 were up-regulated by H-Ras but not by N-Ras in the type I collagen-coated gel, suggesting that H-Ras-specific up-regulation of MT1-MMP and TIMP-2 may lead to the activation of pro-MMP-2. Since acquisition of pro-MMP-2 activation can be associated with increased malignant progression, these results may help understanding the mechanisms for the cell surface matrix-degrading potential which will be crucial to the prognosis and therapy of breast cancer metastasis.

생강나무 추출물의 암전이 억제효과 (Effect of Lindera obtusiloba extract on cancer metastasis)

  • 목 적 : 본 연구에서는 생강나무 메탄올 추출물이 암전이 억제에 미치는 영향을 조사하고자 하였다. 방 법 : 생강나무 추출물의 암전이 억제능을 확인하기 위해서 B16F10 흑색종 세포를 이용하여 금속단백분해효소의 활성 및 발현을 측정하였으며, 암세포의 이동능이나 침윤능도 조사하였다. 폐전이 동물모델에서 생강나무 추출물이 미치는 영향을 조사하여 활성을 최종적으로 확인하였다. 결 과 : 1. 생강나무 추출물은 B16F10 흑색종 세포에서 뚜렷한 금속단백분해효소의 효소활성 및 발현 억제효과를 보였으며 이는 NF-${\kappa}B$의 활성 억제에서 기인한 것임을 확인하였다. 2. 흑색종 세포의 이동이나 침윤 역시 생강나무 추출물 투여에 의해 현저히 감소하였다. 3. 폐전이 동물 모델에서도 생강나무 추출물에 의해 폐로 전이되 집락의 수가 감소하였다. 결 론 : 이상의 결과로 생강나무 추출물은 뛰어난 암전이 억제효과가 있는 것을 확인할 수 있었으며, 전이성 암치료에 있어서 유용하게 사용될 수 있을 것으로 사료된다.

Indole-3-carbinol에 의한 OVCAR-3 인체 난소암세포의 침윤 억제 (Inhibition of Cell Invasion by Indole-3-Carbinol in OVCAR-3 Human Ovarian Cancer Cells)

  • 본 연구에서는 식물체에 널리 분포하는 indole-3-carbinol (I3C)에 의한 OVCAR-3 인체 난소암세포의 이동성 및 침윤성 억제 가능성과 이와 연관된 기전을 조사하였다. 본 연구의 결과에 의하면 I3C에 의한 OVCAR-3 세포의 증식억제는 세포의 이동성 억제와 연관이 있었으며, 이를 wound healing 및 matrigel invasion assay로 확인 하였다. 아울러 I3C 처리에 의하여 transepithelial electrical resistance가 증가되었으며, cellular paracellular permeability는 감소되었는데, 이는 I3C 처리에 의해 세포 내 치밀결합(tight junctions, TJs)의 tightness가 증가되었음을 의미한다. RT-PCR 및 immunoblotting 결과에 의하면, I3C는 TJs의 구성 성분이면서 paracellular transport의 선택적 투과성을 조절하는 주요 인자인 claudin-3 및 -4의 발현을 유의적으로 억제하였다. 또한 matrix metalloproteinase (MMP)-2 및 -9의 활성이 I3C 처리에 의하여 매우 억제되었는데, 이는 그들의 mRNA 및 단백질 수준에서의 발현 감소와 연관성이 있었다. 따라서 I3C에 의한 OVCAR-3 난소암세포의 침윤성 억제는 TJs 기능의 강화와 MMP 활성의 저하가 주요 인자로 작용함을 알 수 있었다.

Clinical Implication of Aortic Wall Biopsy in Aortic Valve Disease with Bicuspid Valve Pathology

  • Background: Although unique aortic pathology related to bicuspid aortic valve (BAV) has been previously reported, clinical implications of BAV to aortopathy risk have yet to be investigated. We looked for potential differences in matrix protein expressions in the aortic wall in BAV patients. Methods: Aorta specimens were obtained from 31 patients: BAV group (n=27), tricuspid aortic valve (TAV) group (n=4). The BAV group was categorized into three subgroups: left coronary sinus-right coronary sinus (R+L group; n=13, 42%), right coronary sinus-non-coronary sinus (R+N group; n=8, 26%), and anteroposterior (AP group; n=6, 19%). We analyzed the expression of endothelial nitric oxide synthase (eNOS), matrix metalloproteinase (MMP)-9, and tissue inhibitor of matrix metalloproteinase (TIMP)-2. Results: Based on the mean value of the control group, BAV group showed decreased expression of eNOS in 72.7% of patients, increased MMP-9 in 82.3%, and decreased TIMP in 79.2%. There was a higher tendency for aortopathy in the BAV group: eNOS $(BAV:TAV)=53%{\pm}7%:57%{\pm}11%$, MMP-9 $(BAV:TAV)=48%{\pm}10%:38%{\pm}1%$. The AP group showed lower expression of eNOS than the fusion (R+L, R+N) group did; $48%{\pm}5%$ vs. $55%{\pm}7%$ (p=0.081). Conclusion: Not all patients with BAV had expression of aortopathy; however, for patients who had a suspicious form of bicuspid valve, aortic wall biopsy could be valuable to signify the presence of aortopathy.
